해바라기침선충(Paratylenchus projectus)에 의한 상추 피해 발생 보고 (First Report of Corky Roots of Lettuce (Lactuca sativa) Associated with Paratylenchus projectus)

  • 상추 시설재배지에서 해바라기침선충(Paratylenchus projectus)에 의한 피해가 발생하였다. 피해발생 시설에서 약 3개월 정도 상추를 재배하고 있었는데 상품성이 있는 상추는 전혀 생산되지 않았다. 피해 상추는 잔뿌리가 거의 없었고 굵은 뿌리는 푸석하게 부풀어 코르크화 현상이 발생했었다. 본 시설하우스에서 발견된 침선충 밀도는 토양 100 ㎤ 당 평균 3,296마리(320-11,600)로 매우 높았고 본 시설하우스의 흙을 사용하여 상추를 정식하여 실험한 결과, P. projectus 접종구에서 30% 정도 상추 생육의 저하가 발생하였다. 따라서 이 증상을 해바라기 침선충(P. projectus)에 의한 "상추 뿌리 코르크병"으로 명명하고자한다.

  • A rotting bacterium was isolated from decayed root of ginseng (Panax ginseng Meyer), cultured purely, and its pathogenicity was confirmed by reinoculation test. The strain causing ginseng root rot was identified as Pseudomonas fluoresens biotype II. The strain was somewhat different from P.marginalis and P.talaasii, considering the number of flagella, pathotype and ability of indole production. The strain did not exhibit pathogenicity to other plants tested, such as red kidney bean(Phasolus vulgaris L.), soy bean (Glycine max Merr.), cucumber (Cucumis sativus L.) lettuce (Lactuca sativa L.) and cowpea bean (Vigna sinensis Savi.).

형질전환된 상추내에서 GUS 유전자의 발현 및 후대검정 (Expression of $\beta$-Glucuronidase (GUS) Gene in Transgenic Lettuce (Lactuca sativa L.) and Its Progeny Analysis)

  • 상추의 종자 무균발아후 4일된 자엽조직을 GUS 유전자가 도입된 A. tumefaciens LBA 4404와 2일간 공동배양한 다음 0.1mg/L NAA, 1.0mg/L 2ip, 50mg/L kanamycin, 500mg/L carbenicillin이 첨가된 MS 배지에 배양하여 식물체를 재분화시켰다. PCR 분석결과 GUS 유전자가 형질전환된 식물체의 게놈상에 삽입되어 있음을 확인하였다. 해부학적 GUS 활성을 분석하여 형질전환된 식물체의 줄기, 잎 그리고 뿌리에서 GUS 유전자의 발현을 확인하였다. 형질전환체로 확인된 식물체를 자가수정시켜 얻어진 종자의 GUS 활성을 분석하여 GUS 유전자가 발현되는 것을 확인하였다.

Allelopathy of Tagetes minuta L. Aqueous Extracts on Seed Germination and Root Hair Growth

  • Present paper showed allelopahtic effects of Tagetes minuta aqueous extracts on seed germination and root hair development. Allelopathy of aqueous extracts derived from T. minuta examined using two test plant species (Lotus comiculatus var. japonicus and Lactuca sativa). The seeds of test species were inoculated in petri dishes containing 0, 10,50 and 100% aqueous extracts from T. minuta. At day 5, the relative seed germination ratio to control was evaluated, and the development of seedling root hairs was observed through light microscopy. Seed germination of L. comiculatus var. japonicus was significantly inhibited proportional to the concentrations of aqueous extract, but that of L. sativa wasn't inhibited. The inhibitory allelopathic effect of T. minuta was found in the development and growth of seedling root hairs. It was concluded that the inhibitory allelophatic effects have been to be investigated using various bioassay, for the allelopathy of plant species shows species-specific and organ-specific.

Growth-Inhibiting Effects of Vegetable Extracts on Beneficial and Harmful Human Intestinal Bacteria

  • Ethanol extracts of 38 vegetables were subjected to an in vitro screening for their growth-inhibitory activities towards Bifidobacterium bifidum, B. longum, Clostridium perfringens, Lactobacillus acidophilus, L. casei, and Escherichia coli using paper disc agar diffusion methods under anaerobic conditions. The responses varied with both bacterial strain and vegetable species. In a test with 20 mg/disc, Zingiber officinale extracts showed significant growth-inhibitory responses against B. bifidum, and strong inhibitions against L. casei were detected in the extracts of Chrysanthemum coronarium var. spatiosum and Lactuca sativa. The extracts of Allium sativum, Capsicum annuum, L. esculentum, L. esculentum var. cerasiforme, and Z. officinale showed strong inhibitory activities against C. perfringens, while moderate growth-inhibitory responses were observed in the extracts of C. ffutescens, Cucurbita moschata, Daucus carota var. sativa, and Rubus coreanus. However, all vegetable extracts showed no inhibitions against B. longum, L. acidophilus, and E. coli. In tests with 5 mg/disc, moderate inhibitions were observed in the extracts of C. coronarium var. spatiosum and L. sativa against L. casei and Z. officinale against B. bifidum. Vegetables extracts, except for C. coronarium var. spatiosum, L. sativa, and Z. officinale, did not affect the growth of beneficial bacteria. Strong inhibitory responses against C. perfringens were detected in the extracts of C. annuum and L. esculentum var. cerasiforme. Daily intake of vegetables may be important in the prevention of human diseases caused by the intestinal bacteria.

Transgenic Lettuce Expressing Chalcone Isomerase Gene of Chinese Cabbage Increased Levels of Flavonoids and Polyphenols

  • Flavonoid are large group of the polyphenolic compounds which are distinguished by an aromatic or phenolic ring structure and the phenolic compounds are induced by microbial infection, ultraviolet radiation, temperature and chemical stress. They are known for their antioxidant activity, anti-allergic, anti-inflammatory, anti-microbial and anti-cancer activities. In this study, changes in flavonoid content were investigated using heterologous chalcone isomerase (CHI) expression system. Also, phenolic compounds level was measured to examine the relation between flavonoids and phenols contents. Explants of lettuce (Lactuca sativa L.) were transformed with Agrobacterium tumefaciens LBA 4404 strain containing pFLH-CHI (derived from pPZP2Ha3) vector constructed with CHI gene from Brassica rapa. The putative transgenic plants were confirmed by genomic DNA PCR analysis. Also the transcription levels of the gene were analyzed by semi-quantitative RT-PCR with gene specific primers. The total flavonoid contents were increased at $T_0$ and $T_1$ generations over 1.4 and 4.0 fold, respectively. Total phenol contents also increased at $T_1$ generation. These results indicate that CHI gene plays an important role to regulate the accumulation of flavonoids and its component changes.

볏짚에서 추출한 짚초액의 식물성장에 대한 영향과 항균 효과 (Effect on Plant Growth and Antibiosis of Rice Straw Liquor Extracted from Rice Straw)

  • Rice straw liquid was prepared from rice straw using simple pyrolysis furnace, and its effects on plant growth and antibiosis were investigated. Effects of straw liquid on plant growth of Oryza sativa L., Glycine max Merr. and Lactuca sativa L. and antibiosis of bacteria and mold of rice straw liquid were studied. Th rice straw liquid showed good results on plant growth and multiplication. Rice straw liquid showed a little antibiosis on bacteria but non antibiosis on mold. Rice straw liquid had minimal inhibitory concentration of 2.5-5% for bacteria.

Effects of Inoculation of Rhizomicrobial Strains on Plant Growth at the Early Germination Stage

  • Plant-growth-promoting rhizobacteria can affect plant growth by various direct and indirect mechanisms. This study was conducted to determine the ability of some rhizobacterial strains to enhance the seed germination of Lactuca sativa (lettuce) and Raphanus sativus (radish). Seeds were inoculated using a spore suspension ($1{\times}10^7cfumL^{-1}$) and incubated in a growth chamber at $28^{\circ}C$ under dark conditions and 65% RH. Azotobacter chroococcum and LAP mix inoculation increased the plumule length of L. sativa by 1.3, 0.8, and 0.7 cm, respectively, in comparison to the uninoculated control. Pseudomonas putida showed an increase of only 0.6 cm in plumule length when compared to the control. Inoculation of A. chroococcum, P. putida, and LAP mix enhanced the seed germination rate of R. sativus, by 10, 5, and 30%, respectively, in comparison with the uninoculated seeds. The results demonstrated that the inoculation of seeds by select rhizobacterial strains showed remarkable enhancement to the radicle length of lettuce and radish seedlings.

유전자총을 이용한 상추 내로의 배추 Glutathione Reductase (GR)유전자의 도입 (Transformation of Chinese Cabbage Glutathione Reductase (GR) gene into Lettuce (Lactuca sativa L.) with Particle Bombardment)

  • 상추에 particle bombardment를 이용한 배추 Glutathione Reductase (GR)유전자의 형질전환효율 향상 및 형질전환체의 유전분석에 관한 실험을 수행하여 얻은 결과를 요약하면 다음과 같다. Particle bombardment 이용시 사출 전 4시간, 사출 후 16시간 동안 삼투조절제인 0.6M serbitolfmannitol 을 처리하여 형질전환시킨 상추 자엽조직에서 GUS 반응 spot이 가장 많았다. 0.3, 0.6, 1.5 kbp로 나타나는 3가지 종류의 primer를 사용하여 PCR한 결과 각각의 primer에 homology를 타나내는 band를 확인하여 355 promoter와 BcGRl유전자가 삽입이 된 것을 확인하였다. 형질전환된 상추의 자식 종자를 채종하여 kanamycin 200mg/L이 첨가된 MS배지에 파종하여 형질전환시키지 않은 상추의 자식 종자와 비교해 본 결과, 형질전환된 상추의 종자에서 발아된 유묘의 약 70%정도가 kanamycin에 대해 내성을 나타내었다.

생물반응기 산삼 배양액의 진세노사이드 분석 및 엽채류 응용에 따른 생물학적 변화 (Ginsenoside Analysis of Panax ginseng C. A. Meyer Culture Broth in a Bioreactor and Its Application in Inducing Biological Changes in Leafy Vegetables)

  • Background: The aim of this study was done to identify whether mass produced wild ginseng culture broth prepared from cultivated wild ginseng roots could have an application in enhancing the agricultural utility value of leafy vegetables. Methods and Results: Leafy vegetables Lactuca sativa and Brassica juncea were treated with wild ginseng culture broth. Plants were examined and treatment (100 ㎖) applied twice a week over an eight week period. Total phenolic and flavonoid content of treated plants was then measured. Wild ginseng culture broth treatment resulted in phenolic and flavonoid content of 0.40 mg·GAE/㎖ and 0.36 mg·QE/㎖, respectively in L. sativa. When treated with wild ginseng culture broth, free radical scavenging ability was found to be higher in both L. sativa and B. juncea whereas antimicrobial activity was found to be higher in B. juncea (625 ㎍/㎖) than in L. sativa. Inorganic element analysis of L. sativa and B. juncea showed that Ca and Mg were higher in the wild ginseng broth treatment group, whereas harmful elements such as As were reduced. Conclusions: Rather than discarding the wild ginseng culture broth, it can be used as a fresh biomaterial by reprocessing it as agricultural products that can promote growth and improve functionality in plants.
