Transformation of PAT gene into Lettuce (Lactuca sativa L.) using Agrobacterium tumefaciens (Agrobacterium tumefaciens를 이용한 상추 (Lactuca sativa L.)의 PAT유전자 형질전환)

  • Agrobacterium tumefaciens MP90 harboring PAT (phosphinothricin acetyltransferase) and NPTII-GUS gene were used for the genetic transformation of lettuce (Lactuca Sativa L.). Shoot regeneration from cotyledon explants were obtained from the MS medium supplemented with 0.1 mg.L$^{-1}$ NAA, 1.0 mg.L$^{-1}$ 2ip, 50 mg.L$^{-1}$ kanamycin and 500 mg.L$^{-1}$ carbenicillin after cocultivation with A. tumefaciens for 2 days. Kanamycin resistance test of transgenic plants indicated that the NPTII gene was integrated into the lettuce genome and was stably expressed. PCR and northern blot analysis indicated that bialaphos resistance gene (PAT) was stably integrated into the lettuce genome. The transgenic plant sprayed with Basta (1500x) remained healthy with continuous growth, while the control group exhibited fatality.

Transgenic lettuce (Lactuca sativa L.) with increased vitamin C levels using GalUR gene (GalUR 유전자를 이용한 비타민 C 증대 상추 (Lactuca sativa L.) 형질전환체 개발)

  • L-Ascorbic acid (vitamin C) in vegetables is an essential component of human nutrition. The objective is to transform lettuce (Lactuca sativa L.) with GalUR gene that is involved in the vitamin C biosynthesis. The cotyledons of Hwoahong (Nongwoo Bio Co.) were used to induce the callus and shoot under the selection media with MS + 30 g/L Sucrose + 0.5 mg/L BAP + 0.1 mg/L NAA + 100 mg/L kanamycin + 200 mg/L lilacillin, pH 5.2. The shoot was developed from the cut side of the explants after 3 weeks on the selection media. We successfully transformed the lettuce with GaIUR gene and analyzed the levels of vitamin C. We found that some of the lettuce transgenic lines contained higher levels of vitamin C compared with the normal one (non-transformed). Especially, some of $T_1$ lettuces inserted by GalUR showed about $3{\sim}4$ times higher content of vitamin C compared to the non-transformed lettuce. This data support the previously work performed with GLOase transgenic $T_1$ lettuces from which several times higher content of vitamin C were identified. The $T_2$ lettuces with high content of vitamin C have been selected for further analysis.

Establishment of Efficient Regeneration System Through In Vitro Culture of Lettuce (Lactuca sativa)

  • An efficient regeneration system was established by using in vitro plantlets of germinated seedlings from different cultivars of lettuce (Lactuca sativa cv. Chongchima, Chongchuckmyun, Jeokchima, Jeokchuckmyun). Shoot formation were observed from all cultivars on MS medium supplemented with 0.1 mg/L NAA and 0.5 mg/L BA. In all cultivars, when cotyledon was cultured, the number of shoot per explant was more greater than that hypocotyl and leaf disc were cultured. Shoot formation rate (91.7%) was high in a cotyledon culture of cultivar, Chongchukmyun. The growth of multiple shoots derived from the cultivar, Chongchukmyun, was most effective on medium containing 0.5 mg/L BA and 1.0 mg/L GA$_3$. When shoots were transferred on MS medium without plant growth regulators, roots were effectively differentiated. Rooted plantlets were acclimated on pots for further propagation.

  • To induce adventitious buds, hypocotyl and cotyledonary explants from 7 to 10 day-old seedlings of lettuce (Lactuca sativa L.: two Japanese cultivars of crisphead lettuce and four Korean cultivars of leaf lettuce) were cultured or Murashige and Skoog (MS) and Schenk and Hildebrandt (SH) media supplemented with BA and NAA in the light for five weeks. Cotyledonary explants produced adventitious shoots at greater frequencies than hypocotyl explants. MS medium was more favorable to adventitious shoot formation than HS medium. Combination of 0.5 mg/L BA and 0.1 to 1 mg/L HPh in MS medium led to the greatest frequency (86%) in adventitious shoot formation. Creator than 95% of shoots excised from explants were rooted when cultured on MS basal medium.

Phytochemical constituents of Lactuca serriola leaves and their content analysis by HPLC-UV

  • This study aimed to identify the phytochemical constituents of Lactuca serriola leaves and perform quantitative analysis of the methanol (MeOH) extract of L. serriola, L. indica, L. raddeana, L. sativa, and L. triangulata. Six compounds were isolated from the MeOH extracts of L. serriola using open column chromatography and identified as protocatechuic acid (1), caffeic acid (2), cynaroside (3), apigenin 7-glucuronide (4), luteolin (5), and apigenin (6) using 1H-, 13C-nuclear magnetic resonance, and mass spectrometry. Quantitative analysis of the six compounds was performed on the MeOH extract of Lactuca species using high-performance liquid chromatography (HPLC) and an ultraviolet (UV). A reverse-phased column was used, and the UV absorbance was set to 280 nm. The contents of compounds 2 and 3 were the highest (1.58 and 2.64 mg/g ext., respectively) in L. serriola extracts. However, compounds 4 and 6 were higher (1.46 and 0.40 mg/g ext., respectively) in L. triangulata. These results provide quantitative data for the application of Lactuca species in the pharmaceutical and functional food industries.

Assessment of Allelopathic Potential and Antioxidant Activity of Leaf Extracts from Three Compositae Plants

  • Some Compositae plants are known to contain biologically active substances that are allelopathic to weeds species as well as antioxidant to foods. Aqueous extracts from leaves of 3 plant species, Cirsium japonica, Kalimeris yomena, and Lactuca saliva, were bioassayed against alfalfa (Medicago saliva) to determine their allelopathic effects. The extracts applied on filter paper in bioassay significantly inhibited root growth of alfalfa. Extracts of 20 g dry tissue $\textrm{L}^{-1}$ from Lactuca sativa showed the most inhibitory effect on alfalfa seedling growth and followed by Cirsium japonica and Kalimeris yomena. Oxidative stability by Rancimat method and antioxidant activity by TBA method for the ground samples were the greatest in Lactuca sativa although were less than that of a commonly used antioxidant, 1% ascorbic acid. Antioxidant activity of methanol extracts on storing meat was stably kept for 28days and was excellent compared to control. These results suggest that three Compositae plants have potent allelopathic and antioxidant effects, and that their activities differ, depending on plant species.

Genetic Relationships of Lactuca spp. Revealed by RAPD, Inter-SSR, AFLP, and PCR-RFLP Analyses

  • RAPD, Inter-SSR, and AFLP markers were used to assess the genetic diversity of lettuce cultivars and the phylogenetic relationships in Lactuca spp. A total of 216 polymorphic bands from seven RAPD primers, four Inter-SSR primers, and five AFLP primer combinations were used to elucidate the genetic similarity among lettuce cultivars. Forty-four lettuce accessions were subdivided into discrete branches according to plant type: crisphead, butterhead, and stem type, with some exceptions. The leafy- and cos-type accessions were intermingled in other groups with no discrete branch indicating that these are more diverse than others. Three accessions, including the Korean cultivar 'Cheongchima', the Korean local landrace 'Jinjam', and the German cultivar 'Lolla Rossa' were classified as the most diverse accessions. Twenty bands were unique in specific cultivars. Among these, three were specific in a plant type; one in Korean leafy type, one in crisphead type, and one in cos type lettuce. In the phylogenetic analysis among Lactuca species, L. saligna, L. serriola, and L. georgica clustered in a sister branch of the L. sativa complex. Two L. virosa accessions show the highest intra-specific relationships. L. perennis outlied from all the other Lactuca species at a genetic similarity of 0.53 and clustered with two Cichorium species, C. intybus and C. endivia, with genetic similarity of 0.67. The phylogenetic tree was supported by data from polymorphism of chloroplast genome which was revealed by PCR-RFLP.

  • Lettuce (Lactuca sativa L., Family Asteraceae) is highly ranked in production and economic value and is consumed either fresh or in salad mixes because of its important dietary source of bioactive phytochemicals. The world collection of Lactuca spp. leafy crops, maintained in NAC, includes 2,464 accessions from 71 countries around the world, of which 2,411 belong to L. sativa species, nineteen to L. saligna, and fifteen to L. serriolar. We aimed to investigate the lettuce germplasm with morphological and biochemical analyses and provide new material for breeding. The lettuce crop comprises seven main groups of cultivars (including oilseed lettuce) differing phenotypically. Agricultural characteristcs were investigated including time to bolting, time to flowering, seed color, flower color, leaf attitude, leaf color, leaf anthocyanin coloration, type of incision of margin, depth of incisions of margins, and leaf venation. Screening of the health beneficial metabolites like anthocyanin and bitter sesquiterpene lactones (lactucin and lactucopicrin) was also conducted. The range of anthocyanin and SLs were 0~563.78 mg/100g D.W. and 3.74~3311.66 ug/g D.W., respectively. The investigation of the degree of variation regarding phenotypic traits and biochemical revealed adaptive stable and highly variable use of trait collection.

Antioxidant Potential and Chlorogenic Acid Level of Lettuce (Lactuca sativa L.) Cultivars

  • Lettuce (Lactuca sativa L.) is known to contain water-soluble substances that improve antioxidant status due to the richness in antioxidants. Greenhouse experiment was carried out under different shading conditions during spring lettuce growing season. Shade significantly reduced shoot weight, number of leaves and chlorophyll content, while it increased shoot length of lettuce plants. The antioxidant potential of the individual fraction was in order of n-butanol > ethyl acetate > water > n-hexane fraction, although was less than that of commonly used antioxidants, BHT and ascorbic acid. Fractions from lettuce plants dose-dependently increased DPPH free radical scavenging activity, in vitro test. By means of HPLC analysis, BuOH fraction of cultivar 'Hwahyang' (57.93 mg $100g^{-1}$) had the highest amount of antioxidant chi orogenic acid. Shading treatment increased average amount of chlorogenic acid of all cultivars in BuoH, EtOAc, hexane and water fractions by 33, 120, 144, and $58\%$, respectively. These results suggest that lettuce plants had potent antioxidant activity, and their activities were differently exhibited depending on cultivar and fraction.
