BACKGROUND/OBJECTIVES: The efficacy of Lab4 probiotic and vitamin C combination on the prevention of upper respiratory tract infections (URTIs) was investigated in two studies with children. Our objective was to pool dataset of 57 preschool children from the PROCHILD study (ISRCTN28722693) and the dataset of 50 preschool matched cohort from the PROCHILD-2 study (ISRCTN26587549) to evaluate the impact of probiotic/vitamin C combination on the prevention of upper respiratory tract symptoms and provide a more robust assessment of effect using detailed individual level data. SUBJECTS/METHODS: The children were supplemented daily for 6 months with either the multistrain probiotic (1.25×1010 cfu/tablet consisting of two strains of Lactobacillus acidophilus CUL21 and CUL60, Bifidobacterium bifidum CUL20 and Bifidobacterium animalis subsp. lactis CUL34) plus 50 mg vitamin C or a placebo. RESULTS: In the pooled analysis of the individual participant data (per protocol population), significant reductions were observed for the incidence (-25%; 95% confidence interval [CI], 0.66, 0.85; P < 0.0001) and duration (-14.9 days; 95% CI, -24.8, -5.1; P = 0.0030) of typical URTI symptoms in the active group compared with the placebo. The incidence rates of absenteeism from preschool (IR ratio, 0.75; 95% CI, 0.66, 0.86; P < 0.0001), paediatric visits (IR ratio, 0.56; 95% CI, 0.47; 0.68; P < 0.0001) and antibiotic usage (IR ratio, 0.53; 95% CI, 0.39, 0.71; P < 0.0001) were also significantly reduced. CONCLUSION: The pooled analysis findings of comparable preschool cohorts from two studies indicate that the supplementation with probiotic and vitamin C combination is beneficial in the prevention and management of URTI symptoms.
The aim of this study was to assess the effects of dietary complex probiotic (Lactobacillus acidophilus, $1.0{\times}10^7$ CFU/g; Saccharomyces cerevisae, $4.3{\times}10^6$ CFU/g; Bacillus subtilis $2.0{\times}10^6$ CFU/g) on growth performance, nutrients digestibility, blood characteristics and fecal noxious gas content in growing pigs. Ninety [(Duroc${\times}$Yorkshire)${\times}$Landrace] pigs with the average initial BW of 39.75${\pm}$1.97 kg were allocated into three treatments by a randomized complete block design. There were five pens per treatment with six pigs per pen. Dietary treatments include: 1) CON (basal diet); 2) CP1 (basal diet+complex probiotic 0.1%) and 3) CP2 (basal diet+complex probiotic 0.2%). During the entire experimental period of 6 weeks, results showed that addition of complex probiotic at the level of 0.2% to diet increased ADG significantly (p<0.05). Also, digestibilities of DM and N tended to increase, however, no significant differences were observed (p>0.05). Blood characteristics (IgG, Albumin, total protein, RBC, WBC and lymphocyte) of pigs were not affected (p>0.05) by complex probiotic supplementation. Fecal $NH_3$-N was decreased (11.8%) significantly by the addition of complex probiotic (p<0.05), but no effects were observed on fecal acetic acid, propionic acid and butyric acid concentrations (p>0.05). In conclusion, results in this experiment indicated that dietary complex probiotic supplementation had a positive effect on growing pigs performance and could decrease fecal $NH_3$-N concentration.
Background: Helicobacter pylori (H. pylori) remains an important cause of gastric cancer and peptic ulcer disease worldwide. Treatment of H. pylori infection is one of the effective ways to prevent gastric cancer. However, standard triple therapy for H. pylori eradication is no longer effective in many countries, including Thailand. This study was designed to evaluate the efficacy of adding bismuth and probiotic to standard triple therapy for H. pylori eradication. Materials and Methods: In this prospective single center study, H. pylori infected gastritis patients were randomized to receive 7- or 14-day standard triple therapy plus bismuth with probiotic or placebo. Treatment regimen consisted of 30 mg lansoprazole twice daily, 1 g amoxicillin twice daily, 1 g clarithromycin MR once daily and 1,048mg bismuth subsalicylate twice daily. Probiotic bacteria composed of Bifidobacterium lactis, Lactobacillus acidophilus and Lactobacillus paracasei. Placebo was conventional drinking yogurt without probiotic. CYP2C19 genotyping and antibiotic susceptibility tests were also done. H pylori eradication was defined as a negative $^{13}C$-urea breath test at least 2 weeks after completion of treatment. Results: One hundred subjects were enrolled (25 each to 7- and 14-day regimens with probiotic or placebo). Antibiotic susceptibility tests showed 36.7% metronidazole and 1.1% clarithromycin resistance. CYP2C19 genotyping revealed 40.8%, 49% and 10.2% were rapid, intermediate and poor metabolizers, respectively. The eradication rates of 7- or 14 regimens with probiotics were 100%. Regarding adverse events, the incidence of bitter taste was significantly lower in the 7- day regimen with the probiotic group compared with 7- day regimen with placebo (40% vs. 64%; p=0.04). Conclusions: The 7-day standard triple therapy plus bismuth and probiotic can provide an excellent cure rate of H. pylori (100%) in areas with low clarithromycin resistance such as Thailand, regardless of CYP2C19 genotype. Adding a probiotic also reduced treatment-related adverse events.
프로바이오틱스 제품에 대한 수요가 지속적으로 증가하고 있으며, Lactobacillus 균주가 가장 대중적인 프로바이오틱스로 널리 사용되고 있다. 프로바이오틱스는 기준에 적합한 균수의 확보가 중요하며 제조원가나 시간 등을 낮추기 위해 배양법의 개발이 필요하므로 Lactobacillus 생산을 위한 배양 조건이 최적화되었다. 반응표면방법론에 의한 통계적 최적화에서 반응 변수에 영향을 미치는 독립 변수의 최적 조건은 Lactobacillus acidophilus의 경우 22.55 시간(배양시간), 25℃(배양온도), 3.41%(프리바이오틱스 농도); Lactiplantibacillus plantarum의 경우 24시간, 30.86℃, 2.00%; Lacticaseibacillus rhamnosus의 경우 66.67시간, 35℃, 3.41%이었다. Lactobacillus의 최적 배양조건은 예측한 결과와 실제 결과가 밀접하게 일치하는 것을 확인하였다. 이러한 데이터는 수율 높은 Lactobacillus를 생산하는데 중요한 포인트를 제공할 것이다.
한우와 홀스타인의 분변으로부터 MRS 배지와 LAPT 배지를 이용하여 무작위 선발법으로 54균주의 유산균을 1차로 분리하였다. 1차로 분리된 54균주에 대해 내담즙성이 우수한 10균주를 분리한 다음 내산성을 조사한 결과 인공위액 pH 2.5에서 LS1, LS15 및 LL6 균주가 각각 66.5%, 82.6% 및 80.7%의 생존율을 나타내었다. Sal. typhimurium, Sta. aureus 및 Cl. perfringens의 병원균에 대해 가장 큰 항균력을 보인 균주는 LL6와 LL7이었다. API CHL kit로 동정한 결과 LS1, LS2 및 LMI 균주는 모두 L. fermentum, LL6와 LL7 균주는 L. acidophilus, LS3 균주는 L. plantarum으로 각각 동정되었고, 나머지 4균주는 Lactobacillus spp. 로 동정되어 분리된 10균주 모두 안전성 있는 유산간균임을 확인하였다. 10종류의 항생제에 대한 내성을 조사한 결과 ampicillin, amoxicillin과 erythromycin에 대해서는 감수성이 있으나 colistin과 ciprofloxacin에 대해 모두 내성을 나타내었다. LB1, LL6 및 LL7 균주는 gentamicin과 neomycin에 대해 내성을 보여 주었다. 분리 동정된 균주 중에 내산성, 내담즙성 및 병원성 균에 대한 특성을 기준으로 판단할 경우 probiotic 유산균으로 사용 가능성이 높은 것은 LL6인 L. acidophilus로 나타났다.
This study was conducted to investigate the pathogenicity and acute single toxicity of Lactobacillus spp. PSC101 (PSC101) isolated from pigs and L. acidophilus (LA) at 2.5$\times$$10^9$CFU or 2.5$\times$$10^{12}$colony forming units (CFU) in mice for 14 days. After oral administration of the bacteria into mice, we could not find their any specific pathogenicity from the standpoints of clinical signs, and changes in body weight and body temperature, as compared with the control group during 14 days. We further investigated the toxicity of concentrated culture broth ($\times$10) after fermentation of them for safe industrial process. As the results, we could not find any clinical signs, changes in body weight and body temperature, as compared with the control group (MRS broth) for 14 days. The results obtained in this study suggest that the potentially probiotic, PSC101, is non-toxic in mice and is therefore likely to be safe for pig use.
This study was undertaken to determine the suitability of tomato juice as a raw material for production of probiotic juice by four lactic acid bacteria (Latobacillus acidophilus LA39, Lactobacillus plantarum C3, Lactobacillus casei A4, and Lactobacillus delbrueckii D7). Tomato juice was inoculated with a 24h-old culture and incubated at $30^{\circ}C$. Changes in pH, acidity, sugar content, and viable cell counts during fermentation under controlled conditions were measured. The lactic acid cultures reduced the pH to 4.1 or below and increased the acidity to $0.65\%$ or higher, and the viable cell counts (CFU) reached nearly 1.0 to $9.0\times10^9/ml$ after 72 h fermentation. The viable cell counts of the four lactic acid bacteria in the fermented tomato juice ranged from $10^6\;to\;10^8\;CFU/ml$ after 4 weeks of cold storage at $4^{\circ}C$. Probiotic tomato juice could serve as a health beverage for vegetarians or consumers who are allergic to dairy products.

The immune reinforcement of the probiotic lactic acid bacteria Lactobacillus acidophilus, Streptococcus thermophilus and Bifidobacterium bifidum was studied in RAW 264.7 cell line treated with diluted solution (dilution to $2^{5}$) of the supernatnats of lactic acid bacteria. RAW 264.7 cell line was used as a macrophage model to assess the effects of lactic acid bacteria on the production of nitric oxide (NO) and cytokine tumor necrosis factor (TNF)-$\alpha$ and cell morphological changes. The production of NO and TNF-$\alpha$ were largely affected by lactic acid bacteria in dose-dependent manner in 24 or 48 hr cultures and cell morphological changes were also largely affected by lactic acid bacteria. Especially Bifidobacterium bifidum differentially stimulated the production of NO and TNF-$\alpha$. NO production was increased by Bifidobacterium bifidum 25 $\mu$l/ml more than LPS (20 ng/ml) control, and TW-$\alpha$ by Bifidobacterium bifidum 6.25 $\mu$l/ml more than LPS (10 ng/ml) control. The in vitro approaches employed here should be useful in further characterization of the effects of lactic acid bacteria on systemic immunity.
프로바이오틱스에 대한 연구는 주로 생균의 효과가 많이 알려져 있지만 가열 처리된 유산균인 사균체의 기능에 대한 연구도 활발히 이루어지고 있다. 본 연구에서는 락토바실러스 아시도필러스 CBT LA1의 사균체의 장관장벽 기능에 대하여 in vitro, in vivo에서 실험하였다. 이를 위하여, 세포표면 소수성 상호작용력(cell surface hydrophobicity), 자가응집력(autoaggregation), 세포에 부착하는 능력(cell adhesion)과 자가응집력(autoaggregation), LPS와의 결합력을 조사하였다. 또한 HT-29 장상피세포에서 LPS로 유도되는 IL-8의 발현을 억제하는 효과를 조사하였다. CBT LA1을 80도에서 121도까지 10분 동안 열을 처리하였을 때, 80도에서 열을 처리한 CBT LA1 사균체가 가장 높은 세포에 부착하는 능력을 보여 주었다. CBT LA1 생균과 비교했을 때, 80도에서 열을 처리한 CBT LA1 사균체는 높은 LPS와의 결합력, 소수성 상호작용력, 자가응집력, HT-29 세포에 부착하는 능력과 IL-8의 발현을 억제하는 능력을 보여주었다. In vivo 실험에서 FITC로 label된 LPS를 투여하였을 때 16시간 후, CBT LA1 사균체를 섭취한 동물의 장관 내 LPS가 가장 많이 제거되었다. 이러한 연구 결과들은 CBT LA1 생균처럼 CBT LA1 사균체도 장관장벽 기능을 가지며 이는, 파마바이오틱스로서 그 가능성을 시사한다.
