Synthesis and Cytotoxic Activity of 1-(1-Benzoylindoline-5-sulfonyl)-4-phenylimidazolidinones

  • The novel 1-(1-benzoylindoline-5-sulfonyl)-4-phenyl-4,5-dihydroimidazolones 2 shows highly potent and broad cytotoxicities. Their cytotoxicities against human lung carcinoma A549, human chronic myelogenous leukemia K562, and human ovarian adenocarcinoma SK-OV-3 are compatible with doxorubicin. Compound 2p (1-[(4-aminobenzoyl)indoline-5-sulfonyl])-4-phenyl-4,5-dihydroimidazolone) exhibits a cytotoxicity that is far more potent than doxorubicin and also exhibits highly effective antitumour activities against murine (3LL, Colon 26) and human xenograft (NCI-H23, SW620) tumor models.

  • Leptospiral lipopolysaccharide (L-LPS) has shown potency in activating toll-like receptor 2 (TLR2) in pig fibroblasts (PEFs_NCC1), and causes the expression of proinflammatory cytokines. However, the stimulation by L-LPS was weak eliciting the function of TLR2 sufficiently in pig innate immunity responses during Leptospira infection. In this study, the immune response of pig embryonic fibroblast cell line (PEFs_SV40) was investigated and was found to be the high immune response, thus TLR2 is the predominate receptor of L-LPS in pig cells. Further, we found a strategy using the antibody against L-LPS, to prevent L-LPS interaction with TLR2 in pig cells which could impact on immune activation.

Anti-Diabetic Effects of DA-11004, a Synthetic IDPc Inhibitor in High Fat High Sucrose Diet-Fed C57BL/6J Mice

  • DA-11004 is a synthetic, potent NADP-dependent isocitrate dehydrogenase (IDPc) inhibitor where $IC_{50}$ for IDPc is 1.49 $\mu$M. The purpose of this study was to evaluate the effects of DA-11004 on the high fat high sucrose (HF)-induced obesity in male C57BL/6J mice. After completing a 8-week period of experimentation, the mice were sacrificed 1hr after the last DA-11004 treatment and their blood, liver, and adipose tissues (epididymal and retroperitoneal fat)were collected. There was a significant difference in the pattern of increasing body weight between the HF control and the DA-11004 group. In the DA-11004 (100 mg/kg) treated group the increase in body weight significantly declined and a content of epididymal fat and retroperitoneal fat was also significantly decreased as opposed to the HF control. DA-11004 (100 mg/kg) inhibited the IDPc activity, and thus, NADPH levels in plasma and the levels of free fatty acid (FFA) or glucose in plasma were less than the levels of the HF control group. In conclusion, DA-11004 inhibited the fatty acid synthesis in adipose tissues via IDPc inhibition, and it decreased the plasma glucose levels and FFA in HF diet-induced obesity of C57BL/6J mice.

Exposure Level of Airborne Bacteria in the University Laboratories in Seoul, Korea

  • This study evaluated the bacterial concentrations and affecting factors at the laboratories of a university in Seoul, Korea. Thirty-three samples of total airborne bacteria (TAB) and eighteen samples of gram negative bacteria (GNB) were collected from both microbiology laboratories (7) and chemistry laboratories (6). GM (GSD) of TAB and GNB concentrations were 194 (2.52) $cfu/m^3$, 24 (4.1) $cfu/m^3$, respectively. TAB concentrations in the chemical laboratories (GM (GSD): 193 (2.0) $cfu/m^3$) were not significantly different from those in microbial laboratories (GM (GSD): 202 (2.7) $cfu/m^3$, (p>0.05)). GM (GSD) of TAB concentrationsat the top of sink, the center of laboratory, and the front of ventilation ventilation device within laboratories, 182 (3.2) $cfu/m^3$, 217 (2.2) $cfu/m^3$, 176 (2.4) $cfu/m^3$, respectively, were not significantly different (p=0.48). Related factors were measured such as temperature, relative humidity, floor of laboratory, number of persons and laboratory area. TAB concentrations were significantly related to temperature (r=0.36, p<0.05), and the floor of laboratory and temperature were also significantly related (r=0.49, p<0.001). However, other factors such as relative humidity, number of persons and laboratory area did not show any significant relationship with TAB concentrations (p>0.05). TAB concentrations were affected significantly by cleaning frequency (p<0.001) and floor of laboratory (p<0.05). There was also a significant difference (p<0.01) between TAB indoor concentrations and TAB outdoor concentrations. However, other factors such as general ventilation did not affect TAB concentrations (p>0.05) in this study.

Protective Effect of DA-9601, an Extract of Artemisiae Herba, against Naproxen-induced Gastric Damage in Arthritic Rats

  • Gastrointestinal irritation is the most frequent adverse effect in patients chronically taking nonsteroidal antiinflammatory drugs (NSAIDs) for the treatment of arthritic conditions. Gastroprotective effect of DA-9601, a new antiulcer agent from Artemisiae Herba extract, against NSAID was evaluated in a rat model of arthritis that is similar in many aspects to human rheumatoid arthritis. Daily oral dosing of naproxen (30 mg/kg), one of the most commonly used NSAID, induced apparent gastric lesions as well as a significant decrease in mucosal prostagiandin $E_2;(PGE_2)$ and prostagiandin F_${1{\alpha}}$$(PGF_{1{\alpha}})$ levels. Coadministration of DA-9601 prevents naproxen-induced mucosal injury and depletion of prostaglandins, in a dose-related manner. DA-9601 did not alter the antiinflammatory or analgesic effect of naproxen. The present results suggest that DA-9601 may be useful as a mucoprotectant against NSAIDs in clinical practice.

  • Analysis of interferon alpha (IFN) modified with high molecular weight branched PEG was performed by capillary electrophoresis (CE) and MALDI-TOF mass spectrometry (MALDI-TOF MS). IFN was modified by the reaction of amine residues with an active ester of monomethoxy polyethylene glycol at various molar ratios. As a CE method. capilary electrophoresis sodium dodecyl sulfate nongel sieving (CE-SDS NGS) was performed using an uncoated capilary filled with a hydrophilic replaceable polymer network matrix.   • Background and objectives: Chromosomal abnormalities play an important role in genesis of acute lymphoblastic leukemia (ALL) and have prognostic implications. Five major risk stratifying fusion genes in ALL are BCR-ABL, MLL-AF4, ETV6-RUNX11, E2A-PBX1 and SIL-TAL1. This work aimed to detect common chromosomal translocations and associated fusion oncogenes in adult ALL patients and study their relationship with clinical features and treatment outcome. Methods: We studied fusion oncogenes in 104 adult ALL patients using RT-PCR and interphase-FISH at diagnosis and their association with clinical characteristics and treatment outcome. Results: Five most common fusion genes i.e. BCR-ABL (t 9; 22), TCF3-PBX1 (t 1; 19), ETV6-RUNX1 (t 12; 21), MLL-AF4 (t 4; 11) and SIL-TAL1 (Del 1p32) were found in 82/104 (79%) patients. TCF3-PBX1 fusion gene was associated with lymphadenopathy, SIL-TAL1 positive patients had frequent organomegaly and usually presented with a platelets count of less than $50{\times}10^9/l$. Survival of patients with fusion gene ETV6-RUNX1 was better when compared to patients harboring other genes. MLL-AF4 and BCR-ABL positivity characterized a subset of adult ALL patients with aggressive clinical behaviour and a poor outcome. Conclusions: This is the first study from Pakistan which investigated the frequency of5 fusion oncogenes in adult ALL patients, and their association with clinical features, treatment response and outcome. Frequencies of some of the oncogenes were different from those reported elsewhere and they appear to be associated with distinct clinical characteristics and treatment outcome. This information will help in the prognostic stratification and risk adapted management of adult ALL patients.

  • Grinding in the cement industry is a very energy-exacting process, therefore it is essential that these systems should operate with the highest possible efficiency. But, Cement grinding process is a complicated nonlinear system with large dead time, very noisy signal and many stochastic disturbances. So, it is difficult to develope mathematic process model. This paper presents correlation analysis of process variables and construction of experimental model for a ball mill grinding process.
