Development of Measuring Technique for Milk Composition by Using Visible-Near Infrared Spectroscopy (가시광선-근적외선 분광법을 이용한 유성분 측정 기술 개발)

  • The objective of this study was to develop models for the predict of the milk properties (fat, protein, SNF, lactose, MUN) of unhomogenized milk using the visible and near-infrared (NIR) spectroscopic technique. A total of 180 milk samples were collected from dairy farms. To determine optimal measurement temperature, the temperatures of the milk samples were kept at three levels ($5^{\circ}C$, $20^{\circ}C$, and $40^{\circ}C$). A spectrophotometer was used to measure the reflectance spectra of the milk samples. Multilinear-regression (MLR) models with stepwise method were developed for the selection of the optimal wavelength. The preprocessing methods were used to minimize the spectroscopic noise, and the partial-least-square (PLS) models were developed to prediction of the milk properties of the unhomogenized milk. The PLS results showed that there was a good correlation between the predicted and measured milk properties of the samples at $40^{\circ}C$ and at 400~2,500 nm. The optimal-wavelength range of fat and protein were 1,600~1,800 nm, and normalization improved the prediction performance. The SNF and lactose were optimized at 1,600~1,900 nm, and the MUN at 600~800 nm. The best preprocessing method for SNF, lactose, and MUN turned out to be smoothing, MSC, and second derivative. The Correlation coefficients between the predicted and measured fat, protein, SNF, lactose, and MUN were 0.98, 0.90, 0.82, 0.75, and 0.61, respectively. The study results indicate that the models can be used to assess milk quality.

Antibacterial and Antioxidative Activities of Epimedium koreanum Nakai Extracts (삼지구엽초 추출물의 항균 및 항산화 활성에 대한 연구)

  • In this study, the antibacterial and antioxidative activities of Epimedium koreanum Nakai were investigated for applications as cosmetic ingredients. Minimum inhibitory concentrations (MICs) of fraction-bacterium, that showed high antibacterial activity from disc diffusion assay on human skin pathogens, were tested. The ethyl acetate fraction on Staphylococcus aureus, Bacillus subtilis, Propionibacterium acnes and 50% ethanol extract on S. aureus exhibited higher antibacterial activities than methyl paraben, well known as a preservative. The DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging activities of 3 fractions of E. koreanum Nakai were lower than (+)-${\alpha}$-tocopherol, known as a typical antioxidant. From the results of the scavenging activities of various ROS generated in $Fe^{3+}-EDTA/H_2O_2$ systems ($OSC_{50}$), 50% ethanol extract ($OSC_{50}=2.46{\pm}0.06{\mu}g/ml$) and aglycone fraction ($OSC_{50}=1.45{\pm}0.02{\mu}g/ml$) showed high activities similar to L-ascorbic acid ($OSC_{50}=1.50{\pm}0.85{\mu}g/ml$), used as reference. The cellular protective effects (${\tau}_{50}$) on photohemolysis by $^1O_2$ generated by photosensitization reaction were tested. The cellular protective effect of 50% ethanol extract (${\tau}_{50}=37.0{\pm}0.3$ min) was similar to (+)-${\alpha}$-tocopherol (${\tau}_{50}=38.0{\pm}1.8$ min), used as reference. In particular, the ${\tau}_{50}$ of aglycone fraction results were $165.9{\pm}7.2$ min. This is a high cellular protective effect, more than 4 times that of (+)-${\alpha}$-tocopherol. These results indicate that E. koreanum Nakai extract, and its fractions, could be utilized as a cosmetic ingredient possessing antibacterial and antioxidative activities.

Assessment of National Soil Loss and Potential Erosion Area using the Digital Detailed Soil Maps (수치 정밀토양도를 이용한 전국 토양 유실량의 평가 및 침식 위험지역의 분석)

  • This study was performed to estimate the soil loss on a national scale and grade regions with the potential risk of soil erosion. Universal soil loss equation (USLE) for rainfall and runoff erosivity factors (R), cover management factors (C) and support practice factors (P) and revised USLE for soil erodibility factors (K) and topographic factors (LS) were used. To estimate the soil loss, the whole nation was divided into 21,337 groups according to city county, soil phase and land use type. The R factors were high in the southern coast of Gyeongnam and Jeonnam and part of the western coast of Gyeonggi and low in the inland and eastern coast of Gyeongbuk. The K factors were higher in the regions located on the lower streams of rivers and the plain lands of the western coast of Chungnam and Jeonbuk. The average slope of upland areas in Pyeongchang-gun was the steepest of 30.1%. The foot-slope areas from the Taebaek Mountains to the Sobaek Mountains had steep uplands. Total soil loss of Korea was estimated as $50{\times}10^6Mg$ in 2004. The potential risk of soil erosion in upland was the severest in Gyeongnam and the amount of soil erosion was the greatest in Jeonnam. The regions in which annual soil loss was estimated over $50Mg\;ha^{-1}$ were graded as "the very severe" and their acreage was $168{\times}10^3ha$ in 2004. The soil erosion maps of city/county of Korea were made based on digital soil maps with 1:25,000 scale.

Soil Erosion Risk Assessment in the Upper Han River Basis Using Spatial Soil Erosion Map (분포형 토양침식지도를 이용한 한강상류지역 토양유실 위험성 평가)

  • This study was conducted to evaluate soil erosion risk with a standard unit watershed in the upper Han river basin using the spatial soil erosion map according to the change of landuse. The study area is 14,577 $km^2$, which consists of 10 subbasins, 107 standard unit watersheds. Total annual soil loss and soil loss per area estimated were $895{\times}10^4\;Mg\;yr^{-1}$ and 6.1 Mg $ha^{-1}\;yr^{-1}$, respectively. A result of analysis with a subbasin as a unit showed that annual soil losses and soil loss per area in Namhan river basins was more than in Bukhan river ones. Predicted annual soil loss according to the landuse ranked as Forest & Grassland > Upland ${\gg}$ Urban & Fallow area > Paddy field > Orchard. Upland area covered 6.2% of the study area, but the contribution of total annul soil loss was 40.6% and that of Forest & Grassland was 44.2%. As a evaluation of soil erosion risk using the spatial soil erosion map, we could precisely conformed the potential hazardous region of soil erosion in each unit watersheds. The ratio of regions, graded as higher "Moderate" for annual soil loss, were respectively 8.7%, 7.9% and 7.8% in 1001, 1002 and 1003 subbasins in Namhan river basin. Most landuse of these area was upland, and these area is necessary to establish soil conservation practices to reduce soil erosion based on the field observation.

An Analysis of Socio-Economic Factors Associated with Private Forest Owners' Knowledge of Forestry Practice (사유림소유자(私有林所有者)의 임업지식수준(林業知識水準)에 영향(影響)을 미치는 사회경제적(社會經濟的) 제요인(諸要因)의 분석(分析))

  • The study was carried out to investigate the relationship of certain socio-economic factors to the private owner's knowledge of forestry practices. The dependent variable was knowledge level in forestry, and the six independent variables were owner's age, educational level, forestland area, socio-economic status, communication and socio-psychological factors. For the study the data were collected by interviewing 258 private forest owners, and analized primarily by multiple correlation statistical technique. The sampled private owners were grouped into two categories: the common owners and model owners. 1) The average score of forestry knowledge of common owners was lower than that of model owners, and the difference was about 15 scores. The scores of other independent variables such as age, educational level, forestland area, socio-economic status, communication and socio-psychological factors of common owners were also significantly lower than those of model owners. 2) The common owner's knowledge of forestry practice were positively correlated with educational level, socio-psychological and communication variables. On the other hand, the model owner's were negatively correlated with the educational level, but positively with the socio-economic status and the socio-psychological variables. 3) The multiple correlation coefficient between the common owner's knowledge scores and six independent variables was 0.5667. About 31.46 percent of the variance of this knowledge could be explained by the communication variable, and other 1.42 percent by the socio-psychological factor. In the case of model owners, the multiple correlation was 0.5332, and about 16.62 percent of the variance of the knowledge could be explained by the communication variable, and ls.y percent by the socio-psychological factor. 4) The opportunity of communication contact through the mass media, personal communication and owner's social participation should be augmented for the effective forestry extension works, because it is one of the results of the study that the private forest owner's knowledge in forestry was greatly affected by communication variable.

A STUDY ON THE CHANCES OF THE SOFT TISSUE PROFILE FOLLOWING ORTHODONTIC TREATMENT BY DIGITAL SUBTRACTION METHOD (교정치료에 따른 측모 연조직의 변화에 관한 계수공제 영상측정법적 연구)

  • The propose of this study was to quantify the changes of soft tissue profile following orthodontic treatment and to evaluate the relationship of those to the skeletal elements. Pre-and post-treatment lateral cephalometric head films of 40 cases(20 extraction cases, 20 non-extraction cases) were traced, and the changes following treatment were measured and quantified by digital subtraction method, and statisticall analyzed. The obtained results were as follows; 1. in extraction group, the change of upper lip area(UL) was $558.60\pm355.17$ pixels, that of lower lip area(LL) was $941.15\pm364.07$ pixels. But, in non-extraction group the change of uper lip area(UL) was $125.65\pm404.16$ pixels, that of lower lip area(LL) was $104.05\pm440.93$ pixels, which was significantly lesser than those in extraction group. 2. In extraction group, there was significant correlationship between upper lip area change(UL) and difference of upper incisor point(${\Delta}UIP$). Lower lip area change(LL) was significantly correlated with difference of upper incisor(${\Delta}UIP$), difference of Franlrfort upper incisor angle(${\Delta}FUIA$) or difference of interincisal angle(${\Delta}IIA$). 3. In extraction group, the ratio of difference of upper incisor point(${\Delta}UIP$) to difference of labrale superius(${\Delta}LSP$) was 1.68; difference of lower incisor point(${\Delta}LIP$) to difference of labrale inferius(${\Delta}LI$) was 1.19; difference of upper incisor point(${\Delta}UIP$) to increment in upper lip thickness(${\Delta}TUL$) was 1.95. 4. In non-extraction group, there was a significant correlationship between upper lip area change(UL) and difference of upper incisor point(${\Delta}UIP$).

Antioxidative Effect and Tyrosinase Inhibitory Activity of Lindera obtusiloba Blume Extracts (생강나무 추출물의 항산화 효과 및 타이로시네이즈 저해 활성)

  • In this study, the antioxidative effects and inhibitory activities on tyrosinase of Lindera obtusiloba Blume (L. obtusiloba Blume) extracts were investigated. 50 % ethanol extract, ethyl acetate and aglycone fractions of L. obtusiloba Blume were used in experiments. The DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging activities of ethyl acetate fraction of L. obtusiloba Blume was higher than (+)-${\alpha}$-tocopherol, known as a typical antioxidant. Reactive oxygen species (ROS) scavenging activities of extract and fractions of L. obtusiloba Blume on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system were similar to L-ascorbic acid, well known as a strong antioxidant. The cellular protective effects of 50 % ethanol extract and ethyl acetate of L. obtusiloba Blume on the rose-bengal sensitized photohemolysis of human erythrocytes were increased in a concentration dependent manner ($1{\sim}25{\mu}g/mL$). Ethyl acetate fraction in $10{\mu}g/mL$ concentration showed the most protective effect among extracts (${\tau}_{50}$ = 361.0 min). The inhibitory effects on tyrosinase of ethyl acetate and aglycone fractions were higher than arbutin, known as a whitening agent. These results indicate that L. obtusiloba Blume extracts can be used as antioxidant, and could be applicable to functional cosmetic ingredient.

Improvement of L-Lysine Productivity by Using Cell Fusion and Immobilized System (세포융합과 고정화 시스템을 이용한 L-Lysine의 생산성 향상)

  • This studies were designed to improve the productivity of L-lysine by protoplast fusion and immobilized system of fusants using strains of Brevibacterium flavum ATCC 21528, Brevibacterium lactofermentum ATCC 21086 and Corynebacterium glutamicum 820. Mutants were isolated with concentration method of $300{\mu}g/ml$ penicillin-G after treatment of $250{\mu}g/ml$ N-methyl-N-nitro-N-nitrosoguanidine. B. flavum $37-2(Hos^-,\;Kan^r,\;AEC^r)$, B. lactofermentum $6-2(Ile^-,\;Val^-,\;Str^r,\;AEC^r)$ and C. glutamicum 57-5$(Met^-,\;Thr^-,\;Rif^r,\;AEC^r)$ were isolated from mutants. Protoplasts were induced by being incubated with $500{\mu}g/ml$ lysozyme of lysis solution for 6 hr and the ratio of protoplast formation and regeneration were ranging from 97-99% and 33-37%, respectively. Fusion frequencies of fusants of BBFL 21, BCFG 37 and BCLG 59 were shown in the range from $1.25{\times}10^{-6}\;to\;5.83{\times}10^{-7}$ under the optimum conditions. The fusant BBFL 21 showed the highest productivity of $411.1\;ng/ml{\cdot}hr$ L-lysine in the lysine productivity broth at $30^{\circ}C$ for 72hr. In the immobilization systems, fusant BBFL 21 was employed in various polymer matrices such as sodium alginate, polyacrylamide, agar and ${\alpha}-carrageena$. The immobilization of sodium alginate showed the highest productivity of $413\;ng/ml{\cdot}hr$ L-lysine in the batch system. Continuous fermentation of immobilization system by using tube fermentor was produced the highest productivity $416.7\;ng/ml{\cdot}hr $ L-lysine under optimum condition.

Effects of Allium Vegetable Intake on Levels of Plasma Glucose, Lipid and Minerals in Streptozotocin Induced Diabetic Rats (Allium속 식용식물의 섭취가 Streptozotocin 유발 당뇨 흰쥐의 혈당, 지질 및 무기질 수준에 미치는 영향)

  • The effects of Allium vegetables on blood glucose levels and lipid metabolism in streptozotocin (S12) induced diabetic rats were investigated. Diabetes mellitus was induced by S1'2 injection (45 mg/kg 5.w.) into the tail vein. Sprague-Dawley rats weighing $220\;{\pm}\;10\;g$ were randomly assigned to 7 groups: normal, S1'2-control and five Allium groups (Allium cepa, Allium fistulosum, Allium sativum, Allium tuberosum and Allium victorialisL Normal and S12control groups were fed an AIN-93 diet and five Allium groups were fed a modified diet containing. 10% Allium powder each for 4 weeks. Body weight, diet intake, food efficiency ratio (FER) and organ weights- were monitored. Activities of aspartate aminotransferase (AST) & alanine aminotransferase (ALT) were observed: Plasma lev~ls of glucose, free fatty acid, triglyceride and HDL-cholesterol were analyzed. Levels of glycogen, cholesterol and triglyceride in liver were determined. Levels of malondialdehyde (MDA) in liver, lung, kidney, and pancreas were assayed. The hepatic contents of chromium (Cr) , iron (Fe), zinc (Zn) and manganese (Mu) were measured. The Allium sativum group had weight gain and suppressed a hypertrophy of the kidney significantly. The activity of ALT was significantly lowered in the diabetic groups except Allium sativum group compared to STZ-control group. The Allium sativum and Allium tuberosum groups showed the hypoglycemic effects at 4 weeks. There were no significant differences between the control and all the other diabetic groups in the plasma levels of cholesterol, HDL-cholesterol, triglycerides and free fatty acids. Most of the Allium groups except Allium fistulosum were observed significantly lowered level of MDA in the lung compared to STZ-control group. The diabetic rats fed the Allium cepa and Allium sativum have shown significantly lowered hepatic Zn contents. The results suggested that the intake of the Allium vegetables may be effective in the antihyperglycemia by lowering blood glucose levels.

Application of Chlorophyll Fluorescence Parameters for the Detection of Water Stress Ranges in Grafted Watermelon Seedlings (수박접목묘의 건조스트레스 범위 탐지를 위한 엽록소형광 지수의 적용)

  • This study was carried out to quantify the drought stress in grafted watermelon seedlings non-destructively by using chlorophyll fluorescence (CF) imaging technique rather than the visual judgment. Six-day old watermelon seedlings were grown under uniform irrigation for 3 days, and then given drought stress. Afterward, the sensor for the measurement of water content in plug tray cell unit was used to classify the drought-stress level into nine groups from D1 (53.0%, sufficient moisture state) to D9 (15.7%, extremely dry stress), and the 16 CF parameters were measured. In addition, re-irrigation was performed on the drought stressed seedlings(D5 - D9) to determine the growth and photosynthesis recovery level, which was not confirmed by visual judgment. The kinetic curve patterns of CF in three different drought stressed seedling groups were found to be different for the early detection of drought stress. All the 16 CF parameters decreased continuously with exposure to drought stress and drastically decreased from D5 (32.1%) where the visual judgment was possible. The fluorescence decline ratio (Rfd_Lss) started to decrease from the initial drought stress level (D5 - D6), and the Maximum PSII quantum yield (Fv/Fm) was significantly decreased in the later extreme drought stress range (D7 - D9) by re-irrigation recovery test. Thus, Rfd_Lss and Fv/Fm parameters were finally selected as potent indicators of growth and photosynthesis recovery in the initial and later stages of drought stress. Also, to the differences in the numerical values of the individual chlorophyll fluorescence parameters, the drought stress level was intuitively confirmed through the image. These results indicate that Rfd and Fv/Fm can be considered as potential CF parameters for the detection of low and extremely high drought stress, respectively. Furthermore, Fv/Fm can be considered as the best CF parameters for recovery at re-irrigation.
