Alteration of X-linked Inhibitors of Apoptosis (XIAP) Expression in Rat Model with DEN-induced Hepatocellular Carcinogenesis

  • The X-linked inhibitor of apoptosis (XIAP) is a member of a novel family of inhibitors of apoptosis and has several BIR domains (BIR1, BIR2, and BIR3) and a carboxy-terminal RING zinc-finger. Since suppressionof apoptosis is fundamentally important for carcinogenesis and tumor growth, we investigated the expression and function of XIAP in DEN-induced carcinogenesis using rat model. Wistar rats were injected intraperitoneally with DEN at a dose of 50 mg/kg in twice a week for 12 weeks (Group II) and 16 weeks (Group III) followed by the recovery periods, respectively. The evaluation of DEN-induced carcinogenesis carried out the blood, RT-PCR, histopathological and western blot analysis. The level of blood chemistry including GOT/GPT, albumin, and total bilirubin were significantly exchanged comparing to control and Group I/Group II. The expression of albumin and collagen mRNA were significantly exchanged (P<0.05) in both groups. In addition, AFP mRNA expression decreased more after recovery periods than Group II. XIAP was expressed constitutively in normal rat liver as well as DEN-induced Groups I and Group II. In addition, XIAP expression increased more in Group I with 4 weeks recovery periods than Group I. However, XIAP expression shown to increase in Group lI, otherwise, it was decreased in Group II with 10 weeks repair periods. Taken together, these results suggest the alteration of XIAP expression could be involved in hepatocellular carcinogenesis.

Report on the 4th WHO Informal Consultation on Development of International Standard Acupuncture Points Locations (제 4차 WHO 경혈 위치 국제표준화 회의 보고)

  • Objective: To inform of the 4th WHO informal consultation meeting on the development of international standard acupuncture points locations, held in Daejeon, Korea, on April $23{\sim}26$, 2005. Results and Conclusions: 12 experts from Korea, China and Japan discussed the locations of 18 controversial points that were not agreed at the previous meeting, and 16 non-controversial points that all 3 nations agreed on the locations but still needed more discussion for their expressions, as well as other non-controversial points that were agreed on both the locations and the expressions but were suggested by Chinese party to be discussed again. Also, the guidelines for English translation of the standard acupuncture points locations and the development of the standard acupuncture points charts, diagrams, and dolls were discussed in this meeting. Through this meeting the locations of ST30, SP11, SP12, TE18, GB9, GB10, LR4, LR5, LR6, BL6, BL7, BL9, GB19 were agreed, but it was suggested that more research would be needed for the locations of LI20, GV26, PC8, PC9, LR7, GB30, LR12, and the expressions of ST36, ST38, ST39, BL1. Due to the time limitation, 16 acupuncture points out of those Chinese party suggested to have more discussion about were not discussed at this meeting and remained to be discussed at the next meeting.

Development of a new lactic acid bacterial inoculant for fresh rice straw silage

  • Objective: Effects of newly isolated Lactobacillus plantarum on the fermentation and chemical composition of fresh rice straw silage was evaluated in this study. Methods: Lactic acid bacteria (LAB) from good crop silage were screened by growing them in MRS broth and a minimal medium with low carbohydrate content. Selected LAB (LAB 1821) were Gram-positive, rods, catalase negative, and were identified to be Lactobacillus plantarum based on their biochemical characteristics and a 16S rRNA analysis. Fresh rice straw was ensiled with two isolated LAB (1821 and 1841), two commercial inoculants (HM/F and P1132) and no additive as a control. Results: After 2 months of storage at ambient temperature, rice straw silages treated with additives were well-preserved, the pH values and butyric and acetic acid contents were lower, and the lactic acid content and lactic/acetic acid ratio were higher than those in the control (p<0.05). Acidity (pH) was lowest, and lactic acid highest, in 1821-treated silage (p<0.05). The $NH_3-N$ content decreased significantly in inoculant-treated silage (p<0.05) and the $NH_3-N$ content in 1821-treated silage was lowest among the treatments. The dry matter (DM) content of the control silage was lower than that of fresh rice straw (p<0.05), while that of the 1841- and p1174-inoculant-treated silages was significantly higher than that of HM/F-treated silage. Microbial additives did not have any significant (p>0.05) effect on acid detergent fiber or neutral detergent fiber contents. Crude protein (CP) content and in vitro DM digestibility (IVDMD) increased after inoculation of LAB 1821 (p<0.05). Conclusion: LAB 1821 increased the CP, IVDMD, lactic acid content and ratio of lactic acid to acetic acid in rice straw silage and decreased the pH, acetic acid, $NH_3-N$, and butyric acid contents. Therefore, adding LAB 1821 improved the fermentation quality and feed value of rice straw silage.

Effects of dietary forage-to-concentrate ratio on nutrient digestibility and enteric methane production in growing goats (Capra hircus hircus) and Sika deer (Cervus nippon hortulorum)

  • Objective: Two experiments were conducted to determine the effects of forage-to-concentrate (F:C) ratio on the nutrient digestibility and enteric methane ($CH_4$) emission in growing goats and Sika deer. Methods: Three male growing goats (body weight $[BW]=19.0{\pm}0.7kg$) and three male growing deer ($BW=19.3{\pm}1.2kg$) were respectively allotted to a $3{\times}3$ Latin square design with an adaptation period of 7 d and a data collection period of 3 d. Respiration-metabolism chambers were used for measuring the enteric $CH_4$ emission. Treatments of low (25:75), moderate (50:50), and high (73:27) F:C ratios were given to both goats and Sika deer. Results: Dry matter (DM) and organic matter (OM) digestibility decreased linearly with increasing F:C ratio in both goats and Sika deer. In both goats and Sika deer, the $CH_4$ emissions expressed as g/d, g/kg $BW^{0.75}$, % of gross energy intake, g/kg DM intake (DMI), and g/kg OM intake (OMI) decreased linearly as the F:C ratio increased, however, the $CH_4$ emissions expressed as g/kg digested DMI and OMI were not affected by the F:C ratio. Eight equations were derived for predicting the enteric $CH_4$ emission from goats and Sika deer. For goat, equation 1 was found to be of the highest accuracy: $CH_4(g/d)=3.36+4.71{\times}DMI(kg/d)-0.0036{\times}neutral$ detergent fiber concentrate (NDFC,g/kg)+$0.01563{\times}dry$ matter digestibility (DMD,g/kg)-$0.0108{\times}neutral$ detergent fiber digestibility (NDFD, g/kg). For Sika deer, equation 5 was found to be of the highest accuracy: $CH_4(g/d)=66.3+27.7{\times}DMI(kg/d)-5.91{\times}NDFC(g/kg)-7.11{\times}DMD(g/kg)+0.0809{\times}NDFD(g/kg)$. Conclusion: Digested nutrient intake could be considered when determining the $CH_4$ generation factor in goats and Sika deer. Finally, the enteric $CH_4$ prediction model for goats and Sika deer were estimated.

The Efficacy of Conjugated Linoleic Acid and Carprofen in Progression of Early Stage of Experimentally Induced Osteoarthritis in Dogs (개에서 Conjugated Linoleic Acid와 Carprofen이 실험적 골관절염 초기 진행에 미치는 효능)

  • This study was performed to compare early stage changes following the administrations of Conjugated Linoleic Acid (CLA), Carprofen and their combinations in the experimental canine cranial cruciate ligament rupture. Twenty five mongrel dogs were divided into five groups; Groups 1, 2, and 3 received a sectioning the cranial cruciate ligament (CCL) of the right stifle joint, and were administered CLA (250 mg/kg/day/orally), carprofen (4.4 mg/kg/day/orally), and their combinations for 4 weeks beginning 4 weeks postsurgery, respectively. Group 4 received sectioning CCL and no treatment. Group 5 was composed of unoperated normal dogs. The macroscopic observation of cartilage erosions on both the condyles and plateaus were evaluated, and the severity of the cartilage lesions and synovial inflammation was examined histologically at eight weeks after surgery. Histological examinations including hematoxylin and eosin stain, standard toluidine blue method, PAS technique and Masson trichrome technique, hematologic and radiographic evaluation were perfonned after experimental surgery. Slight yellowish discoloration of the surface was found in some of the experimental dogs. However macroscopic findings showed no significant differences among the groups. In radiographic findings, cranial displacement of the proximal tibia relative to the femoral condyles in all groups was observed but no significant differences among the groups was seen. This study showed that oral administration of CLA, carprofen and their combinations revealed no early stage change in the canine stifle joint following experimental rupture of the cranial cruciate ligament.

Efficient Second Harmonic Generation of a High-power Yb-doped Fiber MOPA Incorporating MgO:PPSLT (MgO:PPSLT를 이용한 고출력 Yb 광섬유 레이저 빔의 고효율 이차조화파 변환)

  • In this paper, we report highly efficient second harmonic generation of continuous-wave Yb fiber lasers incorporating a periodically poled LiTaO3 device (MgO:PPSLT) as a frequency converter. The seed laser output from a Yb fiber master oscillator using a Fabry-Perot feedback cavity was amplified in a Yb fiber amplifier stage, yielding 28.5 W of linearly polarized output at 1064 nm in a beam with beam quality, M2, of ~1.07. Second harmonic generation was achieved by passing the laser beam through MgO:PPSLT. Under optimized conditions, we obtained 11.1 W of green laser output at 532 nm for an incident signal power of 25.0 W at 1064 nm, corresponding to a conversion efficiency of 44.4%. The detailed investigation to find the optimized operating conditions and prospects for further improvement are discussed.

Preparation and characteristics of Haliotis Discus Hannai Ino (abalone) viscera Jeotgal, a Korean fermented seafood (전복내장 젓갈의 제조 및 품질특성)

  • We prepared Jeotgal with Haliotis Discus Hannai Ino (abalone) viscera and the studied the physicochemical properties. Abalone viscus was fermented with varying amounts of salt for 60 days in order to prepare for the Jeotgal. During the fermentation, we measured the change of pH, volatile basic nitrogen (VBN), amino nitrogen (AN) and protease activity. After the fermentation, we examined the composition of free amino acids and sensory evaluation. The pH decreased with the fermentation, which was not significant (from 5.5 to 6.5). After the fermentation, the highest VBN was 96.7 mg/g, while the highest AN value was 406.3 mg/g. Unlike VBN and AN, the protease activity increased and reached the highest activity at the 30th day, and then decreased afterward. Based on the results, it was deduced that higher salinity restrained the fermentation and lowered the VBN, AN and protease activity. The total free amino acids of abalone Jeotgal, which were analyzed after the fermentation, (62.75 mg/g) was more than twice the amount in the abalone viscera before the fermentation (30.37 mg/g). We prepared abalone viscera Jeotgal and studied the characteristics for the first time. This will provide us with useful information for future related researches.

In Vitro and In Vivo Studies of Different Liposomes Containing Topotecan

  • Liposome as a carrier of topotecan (TPT), a promising anticancer drug, has been reported in attempt to improve the stability and antitumor activity of TPT. However, the biodistr ibution pattern of TPT liposome in vivo and PEG-modified liposome containing TPT have not been studied systemically. In this paper, the in vitro stability and in vivo biodistribution behavior of several liposomes containing TPT with different lipid compositions and PEG-modification were studied. Compared with the 'fluid' liposome (S-Lip) composed of soybean phosphatidylcholine (SPC), the 'solid' liposome (H-Lip) composed of hydrogenated soybean phosphatidylcholine HSPC decreased the leaking efficiency of TPT from liposome and enhanced the stability of liposome in fetal bovine serum (FBS) or human blood plasma (HBP). The results of biodistribution studies in S$_{180}$ tumor-bearing mice showed that liposomal encapsulation increased the concentrations of total TPT and the ratio of lactone form in plasma. Compared with free TPT, S-Lip and H-Lip resulted in 5- and 19- fold increase in the area under the curve (AUC$_{0\rightarrow\propto}$), respectively. PEG- modified H-Lip (H-PEG) showed 3.7-fold increase in AUC$_{0\rightarrow\propto}$ compared with H-Lip, but there was no significant increase in t$_{1/2}$ and AUC$_{0\rightarrow\propto}$ for PEG-modified S-Lip (S-PEG) compared with S-Lip. Moreover, the liposomal encapsulation changed the biodistribution behavior, and H-Lip and H-PEG dramatically increased the accumulation of TPT in tumor, and the relative tumor uptake ratios were 3.4 and 4.3 compared with free drug, respectively. There was also a marked increase in the distribution of TPT in lung when the drug was encapsulated into H-Lip and H-PEG. Moreover, H-PEG decreased the accumulation of TPT in bore marrow compared with unmodified H-Lip. All these results indicated that the membrane fluidity of liposome has an important effect on in vitro stability and in vivo biodistribution pattern of liposomes containing TPT, and PEG-modified 'solid' liposome may be an efficient carrier of TPT.

Comparison of Yield and Content of Salidroside with Application Rates of Nitrogenic Fertilizer under Forcing Culture of Rhodiola rosea L. (바위돌꽃(Rhodiola rosea L.)의 촉성재배시 질소 시비에 따른 수량과 Salidroside 함량)

  • This experiment was conducted to investigate the optimum nitrogen fertilization conditions for the production of high-quality Rhodiola rosea L. in forcing culture. Up until 48 kg N/10 a of both urea and ammonium sulfate, dry matter yield of root of Rhodiola rosea L. tended to increase with increase in application rates, however, it decreased thereafter in higher application rates. In the case of urea, the content of salidroside in the root of the Rhodiola rosea L. appeared to decrease rapidly from the application rates of 64 kg N/10 a and over. Meanwhile, the content of salidroside in the root tended to decrease gradually with the application rates exceeding 64 kg N/10 a of ammonium sulfate. The optimum fertilization rates of urea and ammonium sulfate was 45-8-20-10-10 (N-P-K-Ca-Mg) kg/10 a according to the curvilinear regression equation. However, considering the nitrogen accumulation in soil, nitrogen translocation into the plant, and dry matter yield and content of salidroside in the root of Rhodiola rosea L., the optimum fertilization rates of urea and ammonium sulfate would be 40-8-20-10-10 kg/10 a and 35-8-20-10-10 kg/10 a, respectively.

Effect of Light with Different Wavelengths on Nostoc flagelliforme Cells in Liquid Culture

  • The effects of lights with different wavelengths on the growth and the yield of extracellular polysaccharides of Nostoc flagelliforme cells were investigated in a liquid cultivation. N. flagelliforme cells were cultured for 16 days in 500 ml conical flasks containing BG11 culture medium under $27{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ of light intensity and $25^{\circ}C$ on a rotary shaker (140 rpm). The chlorophyll a, phycocyanin, allophycocyanin, and phycoerythrin contents in N. flagelliforme cells under the lights of different wavelengths were also measured. It was found that the cell biomass and the yield of polysaccharide changed with different wavelengths of light. The biomass and the yield of extracellular polysaccharides under the red or violet light were higher than those under other light colors. Chlorophyll a, phycocyanin, and allophycocyanin are the main pigments in N. flagelliforme cells. The results showed that N. flagelliforme, like other cyanobacteria, has the ability of adjusting the contents and relative ratio of its pigments with the light quality. As a conclusion, N. flagelliforme cells favor red and violet lights and perform the complementary chromatic adaptation ability to acclimate to the changes of the light quality in the environment.
