Parasporin-4, A Novel Cancer Cell-killing Protein Produced by Bacillus thuringiensis

  • Bacillus thuringiensis was isolated as a pathogen of the sotto disease of silkmoth larvae about a hundred years ago. Since then, this bacterium has attracted attentions of not only insect pathologists but also many other scientists who are interested in its strong and specific insecticidal activity. This has led to the recent worldwide development of B. thuringiensis-based microbial insecticides and insect-resistant transgenic plants, as well as a landmark discovery of par asp orin, a cancer cell-specific cytotoxin produced by B. thuringiensis. In this review, we describe examination of interaction between inclusion proteins of B. thuringiensis and brush border membrane of insects using a surface plasmon resonance-based biosensor, identification and characterization of parasporin-4, the latest parasporin produced by the B. thuringiensis A1470 strain, and an effective method for preparing the parasporin-4 from inclusion bodies expressed in the recombinant Escherichia coli cells.

  • RNA polymerase II C-terminal domain phosphatases are newly emerging family of phosphatases that contain FCPH domain with Mg+2-binding DXDX(T/V) signature motif. Its subfamily includes small CTD phosphatases (SCPs). Recently, we identified several interacting partners of human SCP1 with appearance of dephosphorylation and O-GlcNAcylation. In this study, using an established cell line with inducible CTDSPL2 protein (a member of the new phosphatase family), proteomic screening was conducted to identify binding partners of CTDSPL2 in nuclear extract through immunoprecipitation of CTDSPL2 with its associated. This approach led to the identification of several interacting partners of CTDSPL2. This will provide a better understanding on CTDSPL2.

Fabrication of Ozone Bubble Cleaning System and its Application to Clean Silicon Wafers of a Solar Cell

  • Ozone micro-bubble cleaning system was designed, and made to develop a unique technique to clean wafers by using ozone micro-bubbles. The ozone micro-bubble cleaning system consisted of loading, cleaning, rinsing, drying and un-loading zones, respectively. In case of the cleaning the silicon wafers of a solar cell, more than 99 % of cleaning efficiency was obtained by dipping the wafers at 10 ppm of ozone for 10 minutes. Both of long cleaning time and high ozone concentration in the wet-solution with ozone micro-bubbles reduced cleaning efficiency because of the re-sorption of debris. The cleaning technique by ozone micro-bubbles can be also applied to various wafers for an ingot and LED as an eco-friendly method.

  • Radiological criteria such as smooth, sharply defined interface, obtuse angles between lesion and lung and intimate effect on mediastinal contents were usually used to differentiate mediastinal lesion from parenchymal lung lesion. Recently, we experienced a 60-year-old female presenting with anterior mediastinal mass with cavitation. Grossly it was proven to be peripheral lung cancer adjacent to mediastinum and microscopically it was squamous cell carcinoma. The gross pathological findings of surgical specimen were very well correlated with radiological findings. The unique location such as lung periphery and attachment to mediastinum led us to misdiagnosis of anterior mediastinal mass such as germ-cell tumor and neurogenic tumor.

Roles of mitochondria in neuronal development

  • Mitochondria are ubiquitous and multi-functional organelles involved in diverse metabolic processes, namely energy production and biomolecule synthesis. The intracellular mitochondrial morphology and distribution change dynamically, which reflect the metabolic state of a given cell type. A dramatic change of the mitochondrial dynamics has been observed in early development that led to further investigations on the relationship between mitochondria and the process of development. A significant developmental process to focus on, in this review, is a differentiation of neural progenitor cells into neurons. Information on how mitochondria-regulated cellular energetics is linked to neuronal development will be discussed, followed by functions of mitochondria and associated diseases in neuronal development. Lastly, the potential use of mitochondrial features in analyzing various neurodevelopmental diseases will be addressed.

  • The isolation of the MeOH extract from the flower bud of Magnolia biondii Pamp. using various column chromatographies and HPLC led to eleven neoglignan derivatives (1 - 11). Their structures were mainly determined by 1D and 2D NMR spectral data analysis and physiological methods. The isolated compounds (1 - 11) were tested for anti-allergic effects using IL-2 inhibitory assay in Jurkat T cells.

Cytotoxic Phenylpropanoids from the Rhizomes of Alpinia galanga

  • A bioassay-guided fractionation of the n-hexane and chloroform extracts of the rhizomes of Alpinia galanga led to the isolation of two active compounds, 1'S-1'-acetoxychavicol acetate (1) and p-coumaryl alcohol $\gamma$-O-methyl ether (2). 1'S-1'-acetoxychavicol acetate (1) exhibited significant cytotoxicity against all human cancer cell lines tested (A549; $IC_{50}$ 8.14, SNU 638; 1.27, HCTl16; 1.77, HT1080; 1.2, HL60; $IC_{50}$ 2.39 ${\mu}g/ml$), whereas p-coumaryl alcohol $\gamma$-O-methyl ether (2) showed selective cytotoxicity against the SNU638 cell ($IC_{50}$ = 1.62${\mu}g/ml$).

Steroids and Triterpenoid from the Fruit Bodies of Ganoderma lucidum and Their Cytotoxic Activity

  • To determine the cytotoxic activity of natural compounds, chromatographic separation of the hexanesoluble fraction from the fruiting body of Ganoderma lucidum led to the isolation of four steroids and one triterpenoid. They were identified as ergosterol peroxide (1), stella sterol (2), ergosterol (3), 9(11)-dehydroergostrol peroxide (4), and ganodermanontriol (5) based on spectroscopic evidence and physicochemical properties. These compounds were examined for their cytotoxic activity against HL-60, MCF-7, and LLC cancer cell lines. Ganodermanontriol (5) showed cytotoxic activity with IC$_{50}$ values of 24.8 and 22.9 $\mu$g/mL against HL-60 and MCF-7 cancer cell lines, respectively, whereas compounds 1 - 4 were inactive.

Taxonomy and Fermentation of Kitasatosporia kimorexae Producing New Thiopeptide Antibiotics, Kimorexins

  • An isolate, 90-GT-302, was found to produce antibiotics inducing typical mycelial swelling in Magnaporthe grisea and Fusarium solani. This isolate formed yellow substrate and white rectiflexbiles aerial mycelia in the early stages of growth. The aerial mycelium gradually changed its color to white and finally formed a gray spore mass. Analysis of the cell wall acid hydrolysate revealed the presence of LL- and meso-diaminopimelic acids, glycine, and galactose, which indicated cell wall type X. This result placed our isolate in genus Kitasatosporia. A comparison of isolate 9O-GT-302 with reference strains of Kitasatosporia spp., which not only demonstrated several differences in their physiological properties but also novelty of the active compounds produced by this isolate, led us to designate the isolate as Kitasatosporia kimorexae.

Antitumor constituents from the sclerotium of Poria cocos

  • The bioactivity-guided fractionation of an active methylene chloride extract of the sclerotium of Poria cocos led to the isolation of compounds 1-5. These compounds were tested in the human colon carcinoma and human breast carcinoma cell lines, compounds 3, 4, and 5 exhibited IC50 values of 10.8, 15.4, and 5.1 $\mu\textrm{g}$/$m\ell$ against human colon carcinoma cell line. In addition, compounds 3, 4 and 5 showed moderate activities as inhibitors of Topoisomerase I and all compounds were inactive in the Topoisomerase II inhibition.
