Leaf mesophyll protoplasts of Dianthus superbus were cultured in MSP1 liquid medium supplemented with 0.5 mg/L BAP, 2.0 mg/L NAA and 9% mannitol. Protoplast-derived colonies were formed after 3 to 4 weeks of culture in the dark at 27$^{\circ}C$. These colonies were kept under continuous illumination (21.5 $\mu$E. m-2 sec-1) for 2 weeks and finally most of the colonies became green microcalli, about 3 mm in diameter. When green microcalli were transferred to MS solidified medium with 2.0 mg/L 2,4-D, they formed embryogenic calli after 4 week of culture. These calli were then transferred onto $N_{6}$ medium containing 0.1mg/L 2,4-D, 0.1 mg/L NAA, 2.0 mg/L kinetin and 2.0 g/L casein hydrolysate and cultured under illumination. After 5 weeks of culture the calli gave rise to multiple shoots of 10 to 15 per callus. Upon transfer onto MS medium containing 2.0 mg/L NAA, they were noted. The regenerates were successfully transplanted into potting soil.
This study was conducted to investigate the antimicrobial effect of chlorine dioxide ($ClO_2$) on the vase life of cut rose 'Beast' (Rosa hybrida L.). Postharvest treatments to extend the vase life of cut roses were divided into two: holding solution treatment and pulsing solution treatment. In holding solution treatment, the cut roses were treated with preservative solutions containing tap water (TW, control), distilled water (DW), $ClO_2$ 2, 4, 6, and $8{\mu}L{\cdot}L^{-1}$, and compared with a commercialized antimicrobial compound of 8-HQS $200{\mu}L{\cdot}L^{-1}$. In pulsing solution treatment, cut roses were dipped into the $ClO_2$ solutions of 50, 100, 150, 200, and $250{\mu}L{\cdot}L^{-1}$ for 60 seconds and were placed in DW. The air temperature was $18.4^{\circ}C$, RH 51.5%, and light (photosynthetically active radiation, PAR) $3.6{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ with 12 hour day length. The longest vase life of cut roses was observed in the holding solution with $ClO_2$$4{\mu}L{\cdot}L^{-1}$ as 13.8 days and pulsing with $200-250{\mu}L{\cdot}L^{-1}$ as 13.5-13.7 days, where vase life were extended four days longer than TW. Whereas, the inclusion of 8-HQS $200{\mu}L{\cdot}L^{-1}$ in vase solution resulted in phytotoxicity. The relative fresh weight and water uptake have similar tendencies. Bacteria inhibition by $ClO_2$ and 8-HQS were very effective. But bacteria at TW and DW treatments on cut flower with stem were detected in $3.7{\times}10^5CFU{\cdot}L^{-1}$ and $6.3{\times}10^5CFU{\cdot}L^{-1}$, respectively (without stem in DW $1.4{\times}10^4CFU{\cdot}L^{-1}$). The $ClO_2$ contents in holding solution of all treatments were scavenged in two-four days after treatment. This study indicated that $ClO_2$$4{\mu}L{\cdot}L^{-1}$ holding solution treatment and $200-250{\mu}L{\cdot}L^{-1}$ pulsing solution treatment can be applied to extend the postharvest life of cut roses.
The resistance to Tobamovirus in Capsicum spp. has been known to be controlled by five different alleles ($L^0$, $L^1$, $L^2$, $L^3$, and $L^4$) of L locus on the telomere of long arm of pepper chromosome 11. To develop a set of molecular markers differentiating all the alleles of L locus, we used five pepper differential hosts including Capsicum annuum Early California Wonder (ECW, $L^0L^0$), C. annuum Tisana ($L^1L^1$), C. annuum Criollo de Morelos 334 (CM334, $L^2L^2$), Capsicum chinense PI 159236 ($L^3L^3$), and Capsicum chacoense PI 260429 ($L^4L^4$). Developing a series of CAPS or SCAR markers specifically linked to the alleles was allowed by the sequence comparison of PCR amplicons of the $L^3$-linked markers (189D23M, A339, and 253A1R) and BAC sequences (FJ597539 and FJ597541) in the pepper differentials. Genotypes deduced by these markers in 48 out of 53 $F_1$ hybrids of commercial pepper varieties were consistent with their phenotypes by bioassay using Tobamovirus pathotypes ($P_0$, $P_1$, and $P_{1,2$). Consequently, these markers can be useful to differentiate L alleles and for breeding Tobamovirus resistance in pepper with marker-assisted selection.
This study investigated the optimal conditions of enzymatic reaction for production of isomaltooligosaccharides (IMO) using rice flour. To manufacture IMO, commercial enzymes (Termamyl 2X, Maltogenase L, Promozyme D2, Fungamyl 800L and Transglucosidase L) were used. The sugar composition and amount of IMO were examined by HPLC with charged aerosol detector (HPLC-CAD) in each manufacturing process. Liquefaction reaction was performed according to different Termamyl 2X concentrations (0.025%, 0.05%, 0.075%, 0.1%) and reaction times (1 h, 2 h). As a result, the reducing sugar content was the highest at 138.26 g/L when 0.075% Termamyl 2X was added for 2 hours. In order to optimize simultaneous saccharification and transglucosylation, experiments on enzyme selection, enzyme concentration and enzyme reaction time were conducted. Reaction with 0.0015% Maltogenase L, 0.05-0.1% Promozyme D2 and 0.1% Tansglucosidase L was effective in decreasing glucose content and increasing content of IMO with a high degree of polymerization. A change in sugar content was observed every 6 hours to determine the optimal reaction time, and the highest IMO was produced after 36 hours of reaction (75.36 g/L). The IMO prepared under optimal conditions showed isomaltose, 35.11 g/L; panose, 11.97 g/L; isomaltotriose, 19.95 g/L; isomaltotetraose, 7.46 g/L; isomaltopentaose, 1.05 g/L at 18 brix and the ratio of IMO in the total sugar was 56.37%.
Immature hybrid embryos of H. hybridum, 'Picottee', 'White Christmas', 'Eldorado', 'Origin', 'Red Lion', 'elstar', 'Crypsy' were cultured on the MS medium supplemented with various concentrations of 2,4-D, NAA, BA and TDZ. Among the treatments, NAA were more effective for the shoot regeneration and bulblet formation than other treatment. Addition of 0.5 ㎎/L NAA was effective for bulblet induction from explant Shoot regeneration was most effective on the medium with 1.0㎎/L NAA and 2.0 ㎎/L TDZ. The addition of 1.0-2.0㎎/L TDZ induced numerous shoots per explant but strongly inhibited root development when compared to 1.0-2.0㎎/L BA. When bulb scale segments of 'Star Van Holland' was incubated, bulblet formation was the most effective on MS medium with 0.5㎎/L NAA.
Korean (L1)-English (L2) bilingual adults' and children's production of Korean and English stops was examined to determine the age effects and L2 experience on the development of L1 and L2 stop contrasts. Four groups of Seoul Korean speakers (experienced and inexperienced adult and child groups) and two groups of age-matched native English speakers participated. The overall results of voice onset time (VOT) and fundamental frequency (F0) of phrase-initial stops in Korean and word-intial stops in English showed a delay in the acquisition of L1 due to the dominant exposure to L2. Significantly longer VOT and lower F0 for aspirated stops as well as high temporal variability across repetitions of lenis stops were interpreted to indicate a strong effect of English on Korean stop contrasts for bilingual children. That is, the heavy use of VOT for Korean stop contrasts shows bilingual children's attention to the acoustic cue that are primarily employed in the dominant L2. Furthermore, inexperienced children, but not adults, were shown to create new L2 categories that are distinctive from the L1 within 6 months of L2 experience, suggesting greater independence between the two phonological systems. The implications of bilinguals' age at the time of testing to the degree and direction of L1-L2 interaction are further discussed.
A series of aluminum complexes supported by $\beta$-ketoamino, ligand-bearing, 3-position substituents $LAlEt_2$ ($L=CH_3C(O)C(Cl)=C(CH_3)NAr\;(L_1)$, $L=CH_3C(O)C(H)=C(CH_3)NAr\;(L_2)$, $L=CH_3C(O)C(Ph)=C(CH_3)NAr\;(L_3)$, and $L=CH_3C(O)C(Me)=C(CH_3)NAr\;(L_4)$, $Ar=2,6-^iPr_2C6H_3$) were synthesized in situ and employed in the ring-opening polymerization (ROP) of $\varepsilon$-caprolactone ($\varepsilon$-CL) and cyclohexene oxide (CHO). The 3-position substituents on the $\beta$-ketoamino ligand backbone of the aluminum complexes influenced the catalyst activity remarkably for both ROP of $\varepsilon$-CL and CHO. Aluminum $\beta$-ketoamino complexes displayed different catalytic behavior in ROP of $\varepsilon$-CL and CHO. The order of the catalytic activity of $LAlEt_2$ was $L_1AlEt_2$>$L_2AlEt_2$>$L_3AlEt_2$>$L_4AlEt_2$ for ROP of $\varepsilon$-CL, being opposite to the electron-donating ability of the 3-position substituents on the $\beta$-ketoamino ligand, while the order of the catalytic activity for ROP of CHO was $L_1AlEt_2$>$L_3AlEt_2$>$L_4AlEt_2$>$L_2AlEt_2$. The effects of reaction temperature and time on the ROP were also investigated for both $\varepsilon$-CL and CHO.
Cotyledon and epicotyl explants of P. aridum and P. zonale formed calli when cultured on MS medium supplemented with 2 mg/L NAA and 0.2 mg/L BA. Calli were subcultured on the same medium Upon transfer to MS medium with 0.1 to 0.5mg/L NAA and 0.25 to 2mg/L BA for P. aridum 0.1 to 0.5mg/L NAA and 1 to 2mg/L BA for P. zonale subcultured calli gave rise to the greatest number of shoots (0.78 shoot for P. aridum and 0.65 shoot per explant for P. zonale, respectively).Most shoots produced roots when cultured on 1/2MS basal medium. The regenerates were transferred to potting soil and grown to materity in a greenhouse.

The radio signal in GNSS was intentionally designed with two frequencies in order to combat the dispersion error caused by trans-ionospheric propagation. By measuring the path delay independently at the two, widely spaced GPS frequencies, L1 & L2, the TEC along the path from satellite to receiver can be measured directly. The issue with dual frequency measurement of the ionosphere is the calibration of L1/L2 interfrequency biases. L1/L2 interfrequency biases are generated because physical electric signal paths of L1 and L2 circuits are different from each other for both satellites and receiver. Conventionally L1/L2 interfrequency bias is estimated and broadcasted by 2D ionospheric model. In this paper, we estimated IFB (interfrequency bias) by 2D & 3D ionospheric models including real time filter methods and compared the result of those and concluded the merit of 3D tomography model to recover the problem of 2D thin shell model. We confirmed our conclusion by experimental data.
