Metagonimus yokogawai 세르카리아의 감각유두에 관한 연구 (Study on Sensory Papillae of Metagonimus yokogawai Cercaria)

  • A number of studies on the papillae of cercariae of trematodes reported that the papillar patterns (or chaetotaxy) of cercariae might be an excellent method to attain better understanding of the digenetic trematodes (Richard, 1971 ; Short and Cartrett, 1973; Bayssade-Dufour, 1979) . The present study was aimed to determine the number, distribution pattern and structure of the sensory papillae of Metagonimus yokogawai cercariae, and to elucidate the chaetotaxy of this digenetic trematode. M. yokogawai cercariae were pipetted from a vial in which infected snails (Semisulcospira libertina) had been kept for 3 hours. The snails were collected from an endemic area of M. yokogawai, Boseong river in west-southern part of Korea. Observations of papillae were based on light microscopy of those stained with silver nitrate, and on scanning electron microscopy The results are summarized as follows: 1, All papillae observed were uniciliated. 2. Cilia in anterior tip were shorter than the others in other portions. 3. The body papillae were arranged in essentially symmetrical patterns, Total number of the papillae was 126(63 pairs) in average; anterior tip 40(20 pairs), ventral 20(10 pairs), lateral 42(21 pairs), and caudal 8(4 pairs). 4. The chaetotany of M. yokogawai cercaria was: Ci cycle ($3+3C_{I}V,{\;}2+2C_{I}L,{\;}2+3C_{I}D),{\;}C_{II}{\;}cycle(2C_{II}V,{\;}1C_{II}L,{\;}2C_{II}D),{\;}C_{lll}{\;}cycle{\;}(1+lC_{III}V,{\;}1C_{IlI}L),{\;}C_{IV}{\;}cycle{\;}(1C_{IV}V,{\;}IC_{lV}L){\;}in{\;}cephalic{\;}region:{\;}A_I(1A_{IV}V,{\;}1+2A_{I}L,{\;}1A_{I}D),{\;}A_{II}(1A_{II}V,{\;}1+3A_{II}L,{\;}1A_{II}D),{\;}A_{III}(1A_{III}V,{\;}1+1A_{III}L,{\;}1A_{III}D){\;}and{\;}A_{IV}(1A_{IV}V,{\;}2A_{IV}L)$ in antacetabular region: $1M_{I}V{\;}and{\;}2M_{I}L$ in median: $1+1P_{I}L,{\;}1P_{II}L,{\;}1P_{II}D,{\;}1P_{III}L,{\;}1P_{IV}L{\;}and{\;}1P_{IV}D$ in postacetabular region: 2-2-2-2 in caudal region.

배양환경에 따른 Thraustochytrium aureum(ATCC34304)의 DHA(docosahexaenoic acid)생산과 성장특성

  • 배양 조건으로 온도는 4, 11, 18, 24, $32^{\circ}C$로 배양하여, $24^{\circ}C$에서 Lipid 내에 약 42.3%의 DHA 최대 수율을 보였으며, 교반속도는 50, 100, 150, 200rpm 으로 실험하여 l00rpm에서 최대수율로 42.1% 올 얻었다. 초기 당농도는 5, 8, 11, 14, 17, 23, 29, 35g/L로 주어 8g/L에서 Lipid 내 DHA 농도가 43.1%로 나타났으며, N/C 비율은 glucose 5 g/L로 고정하여0.1, 0.2, 0.4, 0.6, 0.8 에서 38-40 % 로 거의 일정하게 나타났다.

  • Let L/F be a Galois quadratic extension such that F contains a primitive n-th root of 1. Let N = L(${\alpha}^{{\frac{1}{n}}$) where ${\alpha}{\in}L{\ast}$. We show that if $N_{L/F}({\alpha})\;{\in}L^n{\cap}F$, and [N : L] = m, then $G(N/ F) {\simeq}D_m$ or generalized quaternion group whether $N_{L/F}({\alpha})\;{\in}\;F^n\;or\;{\notin}F^n$, respectively.

Screening of Immune-Active Lactic Acid Bacteria

  • The purpose of this study was to investigate the effect of lactic acid bacteria (LAB) cell wall extract on the proliferation and cytokine production of immune cells to select suitable probiotics for space food. Ten strains of LAB (Lactobacillus bulgaricus, L. paracasei, L. casei, L. acidophilus, L. plantarum, L. delbruekii, Lactococcus lactis, Streptococcus thermophilus, Bifidobacterium breve, and Pedicoccus pentosaceus) were sub-cultured and further cultured for 3 d to reach 7-10 Log colony-forming units (CFU)/mL prior to cell wall extractions. All LAB cell wall extracts failed to inhibit the proliferation of BALB/c mouse splenocytes or mesenteric lymphocytes. Most LAB cell wall extracts except those of L. plantarum and L. delbrueckii induced the proliferation of both immune cells at tested concentrations. In addition, the production of TH1 cytokine (IFN-γ) rather than that of TH2 cytokine (IL-4) was enhanced by LAB cell wall extracts. Of ten LAB extracts, four (from L. acidophilus, L. bulgaricus, L. casei, and S. thermophiles) promoted both cell proliferating and TH1 cytokine production. These results suggested that these LAB could be used as probiotics to maintain immunity and homeostasis for astronauts in extreme space environment and for general people in normal life.

배지응고제가 시호캘러스의 Saikosaponin함량에 미치는 영향 (Effects of Gelling Agents on the Saikosaponin Content of Bupleurum falcatum Callus)

  • In order to product the saikosaponin which is one of the secondary product from Bupleurum falcatum efficiently through the tissue culture, several levels of agar and gellan gum as the gelling agent, 2,4-D as the growth regulator, and L-phenylalanine as the precursor were used with single or combination treatment on MS basal medium. Gellan gum was more effective than agar as the gelling agent in fresh and dry weight increase of callus induced from Bupleurum falcatum leaf segment. Gellan gum medium supplemented with L-phenylalanine produced 1.6 times of fresh weight more than that of agar. The fresh weight was remarkably high in gellan gum when the calli was treated with the combination of 2,4-D and L-phenylalanine similar to the single treatment of 2,4-D or L-phenylalanine. However, the saikosaponin content in callus was high in gellan gum with the single treatment of L-phenylalanine. Especially, the saikosaponin content in gellan gum supplemented with 1.0mg/L L-phenylalanine was 2 times(2.4 mg/g) higher than that in agar medium supplemented with 0.1 mg/L 2,4-D(0.9 mg/g).

양파의 성숙배 배양을 통한 체세포배발생 캘러스 유기 및 식물체 재분화 (Induction of Embryogenic Callus and Plant Regeneration by Mature Embryo Culture of Onion (Allium cepa L.))

  • 양파의 재분화 체계 확립을 위해 성숙된 양파의 종자로부터 성숙배를 적출한 후 2,4-D와 BA의 다양한 농도를 이용하여 체세포배발생 캘러스의 형성율을 조사하였다. 체세포 배발생 캘러스는 쉽게 부서지는 특성을 가지고 있었으며 비체세포배발생 캘러스는 물기를 많이 포함하고 있어 쉽게 구분이 가능하였다. 체세포배발생 캘러스는 picloram $2.5\;\cal{mg/L}$ 또는 $5\;\cal{mg/L}$가 단독으로 포함된 MS배지를 이용하여 $25^{\circ}C$ 암배양 4주 후 가장 높은 유도율을 나타내었다. 그러나 유도된 캘러스를 shoot로 재분화 시킨 결과 2,4-D와 BA $1\;\cal{mg/L}$가 혼합된 처리구에서 가장 높은 shoot 유기율을 나타내었다. 따라서 다른 처리에서 유도된 캘러스를 2,4-D와 BA $1\;\cal{mg/L}$가 포함된 액체배지에 현탁배양을 시도하였다. 현탁배양은 캘러스의 증가뿐만 아니라 캘러스의 cell cycle을 synchronized 함으로서 이후 형질전환이 용이할 것으로 판단되었다. 현탁 배양된 캘러스는 Kinetin $1\;\cal{mg/L}$가 포함된 1/2 MS 배지에서 60일간 광배양 하여 shoot 유기하였으며 유도된 shoot는 NAA $0.5\;\cal{mg/L}$가 포함된 발근배지에서 발근 후 순화하여 완전한 식물체로 재생할 수 있었다.

활엽수림, 침엽수림 및 혼효림 지역의 강우시 비점오염물질 유출특성 (Runoff Characteristics of Non-point Source Pollutants from Different Forest Types During Rainfall Events)

  • Long-term monitoring was conducted to identify the runoff characteristics of non-point source according to the three forest types (deciduous forest, coniferous forest and mixed forest) in this study. Rainfall events of each deciduous forest, coniferous forest, and mixed forest were 10, 8, 12, respectively. Average runoff depth and coefficients of each forest type were founded to be coniferous forest and were followed by others in turns : deciduous forest, and mixed forest because various conditions (i.e., rainfall property, Antecedent Precipitation Index (API), soil property, slope, and forest management) could change runoff characteristics. In the analysis of the first flush phenomenon, it showed that SS and T-P were sensitive for the first flush phenomenon. The first flush phenomenon of them were showed differently by rainfall intensity, rainfall duration, and amount of rainfall. The research results indicated that range of the Event Mean Concentration (EMC) values in deciduous forest were 0.8~2.4 mg/L for $BOD_5$, 2.0~13.4 mg/L for $COD_{Mn}$, 1.3~2.9 mg/L for DOC, 1.150~3.913 mg/L for T-N, 0.010~0.350 mg/L for T-P and 3.1~291.8 mg/L for SS and in coniferous forest were 0.8~2.2 mg/L for $BOD_5$, 1.9~3.6 mg/L for $COD_{Mn}$, 1.0~2.0 mg/L for DOC, 1.025~2.957 mg/L for T-N, 0.002~0.084 mg/L for T-P and 0.8~5.4 mg/L for SS. Also, range of the EMC values in mixed forest were 1.3~2.3 mg/L for $BOD_5$, 2.4~4.8 mg/L for $COD_{Mn}$, 1.1~2.1 mg/L for DOC, 0.385~2.703 mg/L for T-N, 0.016~0.080 mg/L for T-P and 2.3~30.0 mg/L for SS.

Site-directed mutagenesis를 이용한 BCTV ORF L4의 기능 분석 (Functional Analysis of BCTV ORF L4 by Site-directed Mutagenesis)

  • Beet curly top virus (BCTV) mutant has been constructed in vitro that contain G-to-T transversions at nucleotide 2727 within overlapping open reading frames (ORFs) L1 and L4. The mutations introduce termination codon in ORF L4 without affecting the amino acid encoded by ORF L1. When agroinoculated into Arabidopsis thaliana the mutant caused mild stunting and stem curling, but not the callus induction and hyperlasia on infected tissues of Sei-O ecotype. However, this mutant was not infectious on Col-O. Levels of single stranded DNA forms were similar in mutant and wild type BCTV infections. The DNA quantitation data showed that the DNA of BCTV-L4 mutant virus was accumulated in shoot tips, infection origin and roots with similar levels to those of wild type virus infected. Three tissues of asymptomatic ecotype Col-O also had as much as virus DNA from wild type virus infections. In both ecotypes infected with BCTV-Logan and BCTV-L4 mutant, root tissues contained more virus DNA than any other tissues by the Southern hybridization data. The results suggest that ORF L4 encodes a functional protein that is a major determinant of pathogenesis that might affect the hyperplastic response of the host to BCTV infection.

Synthesis of a Di-N-cyanoethylated Tetraaza Macrocycle Containing Eight C-Methyl Groups and Its Nickel(II) Complex: Effects of the Methyl Groups on Their Properties

  • A new di-N-cyanoethylated 14-membered tetraaza macrocycle 1,8-bis(2-cyanoethyl)-3,5,7,7,10,12,14,14-octamethyl-1,4,8,11-tetraazacyclotetradecane $(L^2)$ and its nickel(II) complex $[NiL^2(OAc)]^+$ have been prepared. The square-planar complex $[NiL^2](C IO_4)_2$ can be prepared by addition of $HClO_4$ to a hot aqueous solution of $[NiL^2(OAc)]^+$ The Ni-N (tertiary amino group) bond distances $(2.008{\AA})$ of $[NiL^2](C IO_4)_2$ are relatively long, and the complex exhibits a d-d transition band at unusually long wavelength (ca. 515 nm). The complex $[NiL^2](C IO_4)_2$ rapidly reacts with acetate ion or ethylenediamine (en) to produce $[NiL^2(OAc)]^+$ or [Ni(en)_3]^{2+}$, respectively, and is readily decomposed in NaOH (0.01 M) solution. The chemical properties of $[NiL^2]^{2+}$ as well as its synthetic procedure are quite different from those for other related 14-membered tetraaza macrocyclic complexes. LH-모멘트에 의한 극치홍수량의 빈도분석을 위한 적정분포형 유도 (Derivation of Optimal Distribution for the Frequency Analysis of Extreme Flood using LH-Moments)

  • This study was conducted to estimate the design flood by the determination of best fitting order of LH-moments of the annual maximum series at six and nine watersheds in Korea and Australia, respectively. Adequacy for flood flow data was confirmed by the tests of independence, homogeneity, and outliers. Gumbel (GUM), Generalized Extreme Value (GEV), Generalized Pareto (GPA), and Generalized Logistic (GLO) distributions were applied to get the best fitting frequency distribution for flood flow data. Theoretical bases of L, L1, L2, L3 and L4-moments were derived to estimate the parameters of 4 distributions. L, L1, L2, L3 and L4-moment ratio diagrams (LH-moments ratio diagram) were developed in this study.
