Elution Profiles of Volatile Compounds and Free Amino Acids during Alcohol Soaking of Garlic(Allum sativum L.) (마늘의 alcohol 침지 중 휘발성 향기성분과 침출유리아미노산 함량)

  • Free amino acids and volatile compounds of fresh garlic and its liqueur were investigated to search elution profile of those components as basic data for development of garlic liqueur. The garlic was soaked in 20% alcohol solution and then sampled every week for 5 weeks. The major free amino acids were L-aspartic acid, L-glutamic acid, L-arginine, L-alanine, L-proline, L-asparagine and L-serine. Neutral amino acids such as L-threonine, L-proline, L-valine and L-leucine, and aromatic amino acids such as tyrosine and phenylalanine were eluted over 80% of those content in fresh garlic after 3 weeks of soaking, but acidic, basic and sulfur containing amino acids were below 80% even after 5 weeks. Sulfide compounds such as diallyl trisulfide, diallyl disulfide, methyl allyl disulfide, 2-vinyl-4H-1,3-dithi in, 3-vinyl-3,4-dihydro-1,2-dithiin, 3,5-diethyl-1,24-trithiolane, isobutyl isothiocyanate and diallyl sulfide were identified as major volatile compounds of fresh garlic by using GC/MS. Among volatile compounds of fresh garlic, allyl alcohol, diallyl disulfide, 3,5-diethyl-1,2,4-trithiolane, diallyl trisulfide and 3,4-dimethoxy furan were eluted to liqueur, but those compounds except 3,5-diethyl-1,2,4-trithiolane were lowered in liqueur during soaking. Furfural, 5-methylfurfural, 5-hydroxymethylfurfural, dimethyl pyrazine, furfuryl alcohol, 3-hydroxy-2-bytanone and 2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyr-an-4-one were generated newly and their content increased in liqueur during soaking.

D(-)- and L(+)-Lactic Acids Contents of Commercial Yogurts (시판 요구르트 중의 D(-)- 및 L(+)-락트산 함량)

  • The ranges of D(-)- and L(+)-lactic acids contents in commercial liquid yogurt were $6.1{\sim}535.8mg/100ml\;and\;70.0{\sim}664.6mg/100ml$, respectively. The ratio of L(+)-lactic acid to D(-)-lactic acid was $0.2{\sim}109.0$. The ranges of D(-)- and L(+)-lactic acids contents in commercial semi-solid yogurt were $10.1{\sim}418.3mg/100g\;and\;515.8{\sim}792.1mg/100g$ respectively. The ratio of L(+)-lactic acid to D(-)-lactic acid was $1.2{\sim}78.4$.

L1 Cell Adhesion Molecule Promotes Migration and Invasion via JNK Activation in Extrahepatic Cholangiocarcinoma Cells with Activating KRAS Mutation

  • Extrahepatic cholangiocarcinoma (ECC), a malignant tumor of biliary origin, has a poor prognosis with limited treatment options. The KRAS oncogene is the most commonly mutated gene in ECC and one of the factors that predicts a poor prognosis and low survival rate. L1 cell adhesion molecule (L1CAM) is expressed in ECC cells and acts as an independent poor prognostic factor in predicting patient survival. In this study we investigate the functional significance of L1CAM in ECC cells with activating KRAS mutation. We selected an ECC cell line, EGI-1, with activating KRAS mutation, and then confirmed its expression of L1CAM by RT-PCR, western blot analysis, and flow cytometry. The suppression of L1CAM expression (using a specific lentivirus-delivered shRNA) significantly decreased the migratory and invasive properties of EGI-1 cells, without altering their proliferation or survival. Analyses of signaling effectors in L1CAM-depleted and control EGI-1 cells indicated that L1CAM suppression decreased the levels of both phosphorylated MKK4 and total MKK4, together with c-Jun N-terminal kinase (JNK) phosphorylation. Further, exposure to a JNK inhibitor (SP600125) decreased migration and invasion of EGI-1 cells. These results suggest that L1CAM promotes cellular migration and invasion via the induction of MKK4 expression, leading to JNK activation. Our study is the first to demonstrate a functional role for L1CAM in ECC carrying the activating KRAS mutation. Given that KRAS is the most commonly mutated oncogene in ECC, L1CAM may serve as an attractive therapeutic target for ECC cells with activating KRAS mutation.

The Anti-diabetic Effects of Kocat-D1 on Streptozotocin-Induced Diabetic Rats (Kocat-D1의 streptozotocin으로 유도한 당뇨모델에 대한 항당뇨 활성)

  • This study was conducted to investigate the anti-diabetic activity of Kocat-D1, which is widely used in traditional medicine to treat diabetes in Shandong, China. Sprague Dawley rats (8 weeks of age) were separated into 4 groups: a normal control, streptozotocin (STZ)-induced diabetic rat group (DM control), Kocat-D1-1 (diabetic rat treated with 0.25 g/kg/day hot water extract), and Kocat-D1-2 (diabetic rat treated with 1 g/kg/day hot water extract). After eight weeks of treatment, the fasting blood glucose levels of the Kocat-D1-1 ($334.3{\pm}32.9\;mg/dL$) and Kocat-D1-2 group ($259.5{\pm}35.0\;mg/dL$) were significantly lower when compared to the DM control group ($451{\pm}42.6\;mg/dL$). Furthermore, the levels of glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), albumin and high-density lipoprotein (HDL) cholesterol in the serum of the Kocat-D1-2 group were significantly normalized when compared to the DM control group. However, significant differences were not observed between the Kocat-D1-1 group and the DM control group. Histochemical staining of the liver of the Kocat-D1-2 group revealed no fat accumulation. The insulin level was significantly upregulated in the Kocat-D1-2 group ($0.13{\pm}0.02\;ng/mL$) when compared to the DM control group ($0.05{\pm}0.04\;ng/mL$). The relative volume of $\beta$-cells in the pancreas of the Kocat-D1-2 group ($49.4{\pm}4.2%$) also increased significantly when compared to the DM control group ($12.9{\pm}7.9%$). These results suggest that Kocat-D1 exerts an anti-hyperglycemic effect through the enhancement of insulin secretion.

Runoff Characteristics of Non-point Source Pollutants from Road Area During Rainfall Events (강우시 도로지역 비점오염물질 유출특성)

  • 본 연구에서는 강우시 도로지역에서 발생하는 비점오염물질 유출 및 수질특성을 살펴보고자 도로지역(국도 46호선)을 대상으로 총 16회의 강우사상에 대해 분석하였다. 모니터링 기간(2014년 5월~9월)동안 연구대상지역에는 3.2~80.3 mm의 강우가 발생하였으며, 강우강도는 0.39~11.29 mm/hr의 범위로 나타났다. 선행무강우일수는 0.3~20.1일이며, 총 유출량은 0.1~6.8 ㎥, 유출율은 0.24~0.85(평균 0.6)의 범위로 나타났다. 강우 모니터링 결과, 유량가중평균농도(Event Mean Concentration, EMC)는 TOC 4.9~56.2 mg/L(평균 18.0 mg/L), BOD 3.1~21.5 mg/L(평균 7.5 mg/L), COD 6.7~58.6 mg/L(평균 23.6 mg/L), SS 2.1~281.9 mg/L(평균 59.4 mg/L), T-N 1.07~13.46 mg/L(평균 4.89 mg/L) 그리고 T-P 0.065~0.680 mg/L(평균 0.23 mg/L)의 범위로 나타났으며, 강우계급별로 살펴보면 0~10 mm일 때 BOD 9.3 mg/L, COD 30.5 mg/L, SS 84.1 mg/L, T-N 5.42 mg/L, T-P 0.27 mg/L로, 10~30 mm일 때 BOD 6.6 mg/L, COD 22.0 mg/L, SS 29.0 mg/L, T-N 4.9 mg/L, T-P 0.20 mg/L로, 50 mm 이상의 강우에서는 BOD 3.6 mg/L, COD 7.1 mg/L, SS 46.4 mg/L, T-N 3.42 mg/L, T-P 0.10 mg/L로 강우계급별 EMC는 대부분 수질항목에 있어 10 mm 이하 계급의 평균 EMC가 높은 수준으로 나타났다. 그리고 각 강우사상에 대한 단위면적당 오염부하는 TOC 0.06~3.5 kg/ha, $BOD_5$ 0.03~1.6 kg/ha, CODMn 0.09~4.74 kg/ha, SS 0.09~35.99 kg/ha, T-N 0.012~2.600 kg/ha, T-P 0.001~0.062 kg/ha의 범위로 산정되었다. 도로지역은 불투수층 면적비율이 높아 누적강우량 10 mm 이하에서도 유출이 발생하는 것으로 분석되었으며, 작은 강우에도 초기유출이 발생하고 유출되는 오염물질 농도가 높은 경향을 나타내었다.

  • This study was conducted to derive optimal design floods by Gumbel, GEV, GLO and GPA distributions for the annual maximum series at sixteen watersheds. Adequacy for the analysis of flood data used in This study was established by the tests of Independence, Homogeneity, detection of Outliers. Parameters were estimated by the Methods of L, L1 and L2-moments. Design floods obtained by Methods of L, L1 and L2-moments using Gringorten methods for plotting positions in GEV distribution were compared by the Relative Mean Errors and Relative Absolute Errors.

  • The Lie algebra analogue of Schur's result which is proved by Moneyhun in 1994, states that if L is a Lie algebra such that dimL/Z(L) = n, then $dimL_{(2)}={\frac{1}{2}}n(n-1)-s$ for some non-negative integer s. In the present paper, we determine the structure of central factor (for s = 0) and the factor Lie algebra $L/Z_2(L)$ (for all $s{\geq}0$) of a finite dimensional nilpotent Lie algebra L, with n-dimensional central factor. Furthermore, by using the concept of n-isoclinism, we discuss an upper bound for the dimension of $L/Z_n(L)$ in terms of $dimL_{(n+1)}$, when the factor Lie algebra $L/Z_n(L)$ is finitely generated and $n{\geq}1$.

  • In order to determine the content levels of trace metals in cow's milk, 79 market milk samples were collected from markets in major cities of Korea from July to August, 1985 and analyzed for content levels of Cu, Fe, Zn, Mn, Pb, Cd. 물상추를 이용한 중금속 제거에 관한 연구

  • 물상추에 의한 Pb 제거효율은 96시간 동안 0.5 mg/L 일 때 71.4%, 1mg/L일 때 65.6%, 1.5 mg/L일 때 81.2%로 나타났으며, 0.5 mg/L 일 때 24시간만에 24.6%로 가장 높은 제거효율을 나타냈다. 또한 농도가 높아질수록 높은 제거효율을 나타냈다. $Cr^{6+}$ 의 제거효율은 높은 농도에서 시간이 지날수록 제거효율이 높아졌다각 수조별 pH 변화는 전반적으로 높아지는 경향을 보였고 Pb의 경우는 1.5 mg/L에서 96시간 동안 6.7에서 7.73으로 가장 큰 변화를 보였다. 수온을 23 ~ 26$^{\circ}C$로 유지하면서 물상추의 비성장율을 살펴본 결과 Pb의 경우 1.5mg/L에서 24시간만에 0.046g $day^{-1}$에서 96시간 후에는 1.5 mg/L에서 0.114 g $day^{-1}$로 가장 높은 성장을 보였으며 $Cr^{6+}$ 의 경우도 1 mg/L에서 24시간 후에 0.0l2g $day^{-1}$에서 96시간 후에는 0.5 mg/L에서 0.07g $day^{-1}$로 가장 높은 성장률을 보였다. 중금속 수조의 $NH_3-N$, $NO_3$-N, T-P 제거효율을 보면 $NH_3-N$는 전체적으로 50%이상의 높은 제거효율을 보여 영양원으로서의 흡수가능성을 보였고 $NO_3$-N와 T-P의 경우는 전체적으로 낮은 제거효율을 보였다.

  • Objectives : This study wad designed to elucidate the short term effect of Rossa rugosae Radix on proliferation, differentiation and maturation of 3T3-L1 Preadipocyte. Methods : 3T3-L1 preadipocytes obtained from Korean Cell line Bank were cultured in a Dulbecco's modified eagle medium(DMED) culture colution containing 10% fetal bovine serum(FBS) and various concentration of aqueous extract of Rossa rugosae Radix on proliferation, differentiation and maturation of 3T3-L1 preadipocytes were investigate after treatment for 24 hours b measuring MTT, Oil Red O and latate dehydrogenase activity.. Results : The Rossa rugosae Radix extract inhibited significantly the proliferation of 3T3-L1 preadipocytes and tended to increase latate dehydrogenase activity in the media of differentiated 3T3-L1 preadipocytes & matured 3T3-L1 preadipocytes. the extract also inhibit the lipid accumulation of differentiated and maturaion of 3T3-L1 preadipocytes, suggesting that Rossa rugosae Radix has anti-obesity effect: however further in vivo study is needed to demonstrate its pharmacological effects.
