Relaxation Effects of Epimedium Koreanum Nakai in Isolated Rabbit Corpus Cavernosum Smooth Muscle (음양곽(淫羊藿)의 토끼 음경해면체 평활근 이완효과)

  • This study aimed to investigate the relaxation effects and its underlying mechanisms of Epimedium koreanum Nakai(EK) in phenylephrine(PE) treated isolated rabbit corpus cavernosum smooth muscle. The dose-dependent relaxation responses of phenylephrine(PE, $1{\times}10^{-6}M$)-precontracted strips to EK at $0.01-3.0mg/m{\ell}$ were measured and also observed after endothelial denudation using organ bath. To analyze the underlying mechanisms of EK-induced relaxation, $N{\omega}$-nitro-L-arginine(L-NNA), methylene blue(MB), tetraethylammonium chloride(TEA), indomethacin(IM) were pretreated before EK extract infused into precontracted strips induced by PE. To investigate cytotoxic activity and nitric oxide(NO) concentration of EK extract on EA.hy926 cells, mitochondrial dehydrogenase activity(MTT) assay and nitric oxide detection kit were used. The cavernous strips were significantly relaxed by EK extract at $0.3mg/m{\ell}$, $1.0mg/m{\ell}$, $3.0mg/m{\ell}$ and the relaxation responses of PE-precontracted strips denuded endothelium also inhibited in comparison with intact endothelium. The pretreatment of L-NNA, MB, TEA reduced EK extract-induced endothelium-dependent relaxation, but the pretreatment of IM didn't affect EK extract-induced endothelium-dependent relaxation. When EK extract was applicated on EA.hy926 cells, the NO concentration was increased. Our findings have shown that EK extract exerts a relaxing effect on corpus cavernosum in part by suppressing influx of extracellular $Ca^{2+}$ through activating the NO-cGMP system.

Mechanism of Corni Fructus Induced Vasorelaxation in Rabbit Carotid Artery (산수유의 혈관이완효과 기전에 대한 연구)

  • This study is conducted to investigate vasorelaxant effect of Corni Fructus(CF) on rabbit carotid artery. To determine vasorelaxant effect of CF on rabbit carotid artery, arterial sections with intact or removed endothelium were used in this organ bath study. After being contracted by phenylephrine(PE), arterial sections were treated with CF extract in a dose-dependent manner. To identity its mechanism, the contracted arterial sections by PE were pretreated with indomethacin(IM), tetraethylammonium chloride(TEA), Nω-nitro-L-arginine(L-NNA) or methylene blue(MB) and 1.0 ㎎/㎖ CF extract. We also studied to confirm the effect on influx of extracellular calcium chloride(Ca2+) of the CF extract in rabbit carotid artery. To measure the cytotoxicity of the CF extract, cell viability of human umbilical vein endothelial cell(HUVEC) was measured by MTT assay. Generation of nitric oxide(NO) was also measured by Griess reagent. The arterial sections with intact endothelium were relaxed significantly by CF extract, but this effect was inhibited in the arterial sections with damaged endothelium. The vasorelaxant effect was inhibited significantly when arterial sections were pretreated with IM, TEA, L-NNA, MB. In Ca2+-free krebs solution, increasing of arterial contraction by Ca2+ was also inhibited by CF significantly. The treatment of CF extract increased NO concentration in HUVEC. This study suggested that the vasorelaxant effect of CF extract would be related with endothelium derived relaxing factor(EDRF) such as NO, prostacyclin(PGI2), endothelium derived hyperpolarization factor(EDHF).

Effect of Aqueous Extract of Polygoni Multiflori Radix on Hypertension and Arterial Contraction in Animal Models (하수오(何首烏)가 고혈압과 수축혈관에 미치는 영향)

  • This research was aimed to examine the effect of Polygoni Multiflori Radix extract on the blood pressure in spontaneous hypertensive rat (SHR) and norepinephrine - induced arterial contraction in rabbit. In order to investigate the effect of Polygoni Multiflori Radix on rabbit's contracted vascular ring detached from common carotid artery, vascular ring with intact or damaged endothelium was used for the experiment using organ bath. To analyze the mechanism of Polygoni Multiflori Radix-induced relaxation, Polygoni Multiflori Radix extract was infused into contracted vascular ring which had been pretreated by $N{\omega}$-nitro-L-arginine(L-NNA), Methylene blue(MB), and $Ca^{2+}$ was infused into contracted vascular ring induced by NE after treatment of Polygoni Multiflori Radix extract in $Ca^{2+}$-free solution. The results were as follows: Systolic blood pressure was significantly attenuated by administration of Polygoni Multiflori Radix. Blood flow and aldosterone were significantly decreased, but velocity and renin were not affected by Polygoni Multiflori Radix. Polygoni Multiflori Radix had an effective relaxation to the contracted vascular ring by NE in 0.03 mg/ml, 0.1 mg/ml and 0.3 mg/ml level. Polygoni Multiflori Radix had an effective relaxation to the intact endothelium vascular ring, but when endothelium was removed, vascular ring did not relax. Polygoni Multiflori Radix-induced relaxation was inhibited by the pretreatment of L-NNA and MB. Pretreatment of Polygoni Multiflori Radix extract inhibit the contraction by influx of extra-$Ca^{2+}$ in contracted vascular ring induced by NE in $Ca^{2+}$-free solution. As mentioned above, we suggest that Polygoni Multiflori Radix relaxes vascular ring through suppress influx of extra-cellular $Ca^{2+}$ by the action of nitric oxide from endothelium.

Effect of lithium on endothelial-dependent relaxation to melatonin in rat aorta (흰쥐 대동맥에서 melatonin의 내피 의존적 혈관 이완 작용에 대한 lithium의 영향)

  • Melatonin, the principal hormone of the vertebral pineal gland, participates in the regulation of cardiovascular system in vitro and in vivo. Lithium inhibits both inositol polyphosphate phosphatase (IPPase) and inositol monophosphatase (IMPase), which are involved in a wide range of signal transduction pathways. The aim of the present study was to assess the effect of lithium on endothelial-dependent relaxation to melatonin and on the melatonin-induced inhibition of contraction by phenylephrine (PE) in isolated rat aorta. Melatonin induced a concentration-dependent relaxation in PE-precontracted in endothelium-intact (+E) aortic rings. Melatonin inhibited a PE-induced sustained contraction in +E aortic rings. These effects of melatonin on relaxation and contractile responses were inhibited by pretreatment with lithium. In PE-precontracted +E aortic rings, the melatonin-induced vasorelaxations and the inhibitory effects of melatonin on maximal contractions were inhibited by endothelium removal or by pretreatment with L-$N^G$-nitro-arginine (L-NNA), 1H-[1,2,4] oxadiazolo-[4,3-a] quinoxalin-1-one (ODQ) and nifedipine and verapamil, but not by tetrabutylammonium, clotrimazole and glibenclamide, However, in endothelium-denuded (-E) aortic rings and in the presence of L-NNA and ODQ in +E aortic rings, the melatonin-induced residual relaxations and the melatonin-induced residual contractile responses to PE were not affected by lithium. It is concluded that the inositol phosphate pathway may be involved in endothelial-dependent relaxation induced by melatonin.

The Endothelium-Dependent Vasorelaxation Effect of Cynomorii Herba (쇄양(鎖陽)의 내피세포 의존성 혈관이완효과)

  • Objectives : The purpose of this study was to investigate the endothelium-dependent vasorelaxation effect of Cynomorii Herba(CH) extract on contracted rabbit carotid artery.Methods : To clarify the vasorelaxation effect of CH extract, arterial strips with intact was used, to endothelium -dependent vasorelaxation effect of CH extract, arterial strips damaged endothelium was used for experiment using organ bath. Arterial strips was contracted with phenylephrine(PE) before treated with CH extract(0.01, 0.03 and 0.1 ㎎/㎖). To study mechanisms of CH-induced vasorelaxation effect, CH extract infused into arterial rings after treatment by indomethacin(IM), tetraethylammonium chloride(TEA), Nω-nitro-L-arginine (L-NNA), methylene blue(MB) for comparing with non-treated. And calcium chloride(Ca2+) 1 mM was treated into precontracted arterial ring induced by PE after treatment of CH extract in Ca2+-free krebs solution. Cytotoxic activity of CH extract on human umbilical vein endothelial cell(HUVEC) was measured by MTT assay, and nitric oxide(NO) concentration was measured by Griess reagent.Results : PE-induced arterial strips was significantly relaxed, but the damaged endothelium arterial ring wasn't relaxed by CH extract. Pretreatment of IM, TEA didn't inhibit the vasorelaxation of CH extract, but pretreatment of L-NNA, MB inhibited the vasorelaxation of CH extract. Pretreatment of CH extract reduced the increase of contraction by influx of extracellular Ca2+ in contracted arterial ring induced by PE, CH extract increased nitric oxide concentration on HUVEC significantly.Conclusions : This study shows that CH extract have the vasorelaxation effect by blocking the influx of extracellular Ca2+ through the activating NO-cGMP system.

Effects of Nelumbo nucifera on the Regional Cerebral Blood Flow and Blood Pressure in Rats (연근의 열수 추출물이 흰쥐의 국소 뇌혈류량과 혈압에 미치는 영향)

  • The purpose of this study was to measure the changes of regional cerebral blood flow(rCBF) and blood pressure(BP) in rats, following the intravenous injection of Nelumbo nucifera water extract. The measurement was continually monitored by laser-doppler flowmeter and pressure transducer in anesthetized adult Sprague-Dawley rats for about 2 to 2 and half hours through the data acquisition system composed of MacLab and Macintosh computer. The results of this experiment were as follows. Nelumbo nucifera significantly increased the changes of rCBF in rats. The rCBF of Nelumbo nucifera was not changed by pretreated propranolol, atropine, L-NNA and indomethacin. But the rCBF of Nelumbo nucifera was decreased by pretreated methylene blue. Nelumbo nucifera decreased the BP significantly. The BP of Nelumbo nucifera was not changed by pretreated propranolol, atropine, L-NNA and indomethacin. But the BP of Nelumbo nucifera was decreased by pretreated methylene blue. These results indicated that Nelumbo nucifera might increase the rCBF and decrease the BP which related to guanylyl cyclase activity.

Effects of Sasim-tang Water Extract on Blood Pressure and Regional Cerebral Blood Flow Volume in Rats (사심탕(瀉心湯)이 백서(白鼠)의 혈압(血壓) 및 국소(局所) 뇌혈류량(腦血流量)에 미치는 영향(影響))

  • Objective : The purpose of this study was to investigate the effects of Sasim-tang water extract on the cardiovascular systems including changes of blood pressure and regional cerebral blood flow volume (rCBF) in male Sprague-Dawely rats. Methods : The changes of blood pressure were recorded by data acquisition system composed of MacLab and Macintosh computer. The changes of rCBF were observed by Laser-Doppler flowmetry through a opened cranial window. Results : 1. The changes of blood pressure was not affected by Sasim-tang extract intravenous injection in rats. 2. After pretreatment with propranolol(3mg/kg), L-NNA(10mg/kg) and 000(10mg/kg), the changes of blood pressure was not affected by Sasim tang extract intravenous injection in rats. 3. The changes of rCBF was increased in dose-dependent manner by Sasim-tang extract intravenous injection in rats. 4, After pretreatment with propranolol(3mg/kg), ODQ(10mg/kg) and L-NNA(10mg/kg), rCBF was significantly decreased in dose- dependent manner by Sasim-tang extract intravenous injection in rats. Conclusions : These results suggest that Sasim-tang was related to the regulation of the sympathetic nerve system, nitric oxide synthesis and synthesis of cyclic GMP.

Effects of Artemisia iwayomogi on the Airway Smooth Muscle (더위지기가 기관지평활근에 미치는 영향)

  • Artemisia iwayomogi has been used in Korea for many centuries as a treatment for anemia. The effect of Artemisia iwayomogi on tracheal smooth muscle is not known. The purpose of the present study is to determine the effect of Artemisia iwayomogi on histamine induced tracheal smooth muscle contraction in guinea pigs. Guinea pigs(500g, male) were killed by CO₂ exposure and a segment (8-10mm) of the thoracic trachea from each rat and guinea pig was cut into equal segments and mounted ‘in pairs’ in a tissue bath. Contractile force was measured with force displacement transducers under 0.5g loading tension. The dose of histamine (His) which evoked 50% of maximal response (ED/sub 50/) was obtained from cumulative dose response curves for histamine (10/sup -7/~10/sup -4/M). Contractions evoked by His (ED/sub 50/) were inhibited significantly by Artemisia iwayomogi. In guinea pig tracheal smooth muscle, the mean percent inhibition of histamine induced contraction was 31.9% (p<0.05) after 100㎕/㎖ Artemisia iwayomogi. L-NNA slightly but significantly attenuated the inhibitory effects of Artemisia iwayomogi. Following treatment with L-NNA, the mean percent inhibition caused by 100㎕/㎖ Artemisia iwayomogi fell to 44.6% in guinea pig induced by histamine contraction. Propranolol, indomethacin and methylene blue did not significantly alter the inhibitory effect of Artemisia iwayomogi. These results indicate that Artemisia iwayomogi can relax histamine induced contraction of guinea pig, and that this inhibition related to nitric oxide formation.
