Tylosin Production by Streptomyces fradiae Using Raw Cornmeal in Airlift Bioreactor

  • Using a 50-1 airlift bioreactor, for the effective production of tylosin from Streptomyces fradiae TM-224 using raw cornmeal as the energy source, various environmental factors were studied in flask cultures. The maximum tylosin concentration was obtained at $32^{\circ}C$ and pH between 7.0 and 7.5. When seed was inoculated after 24 h of culture, the maximum tylosin concentration, 5.7 g/l, was obtained after 4 days of culture. Various concentrations of raw cornmeal were tested to investigate the optimum initial concentration for the tylosin production. An initial raw cornmeal concentration of 80 g/l gave the highest tylosin concentration, 5.8 g/l, after 5 days of culture. Of the various nitrogen sources, soybean meal and fish meal were found to be the most effective for the production of tylosin. In particular, with the optimal mixing ratio, 12 g/l of soybean meal to 14 g/l of fish meal, 7.2 g/l of tylosin was obtained after 5 days of culture. To compare raw cornmeal and glucose for the production of tylosin in the 50-1 airlift bioreactor for 10 days, fed-batch cultures were carried out under the optimum culture conditions. When raw com meal was used as the energy source, the tylosin production increased with increasing culture time. The maximum tylosin concentration after 10 days of culture was 13.5 g/l, with a product yield from raw cornmeal of 0.123 g/g of consumed carbon source, which was about 7.2 times higher than that obtained when glucose was used as the carbon source.

Optimization of Extraction Conditions for Useful Components from Sweet Potato Leaves (고구마 잎의 유용성분 추출조건 최적화)

  • This study was performed to establish extraction conditions for useful components from sweet potato leaves. Extraction conditions were heating temperature ($40{\sim}60^{\circ}C$), ratio of solvent to sample (10~30 mL/g), and ethanol concentrations (50~90%). The maximum lutein content was $381.88{\mu}g/g$ under the following conditions: 81.23% ethanol concentration, $46.75^{\circ}C$ temperature, and 25.78 mL/g ratio of solvent. The maximum total polyphenol content was 58.44 mg GAE/g under 67.0%, $53.90^{\circ}C$, and 29.10 mL/g, whereas maximum total chlorophyll content was 769.14 mg/100 g under 93.10%, $69.90^{\circ}C$, and 25.98 mL/g, respectively. The highest antioxidant activity of electron-donating ability ($IC_{50}$) was 0.582 mg/mL under 64.56%, $54.63^{\circ}C$, and 19.57 mL/g, whereas total antioxidant activity was 32.54 mg AA eq/g under 64.42%, $44.51^{\circ}C$, and 26.01 mL/g, respectively. Under these conditions, experimental data matched well with the predicted data. Consequently, the present study was carried out to simulate and optimize sweet potato leaves for useful component extraction.

  • Carbon dioxide is one of the main causes of global warming. In order to develop a novel absorbent, the characteristics of amino acid salts solution as a solvent for $CO_2$ capture in continuous process were investigated. The cost of $CO_2$ capture is almost 70% of total cost of CCS (carbon dioxide capture and storage). In the carbon dioxide capture process, process maintenance costs consist of the absorbent including the absorption, regeneration, degradation, and etc. It is very important to study the characteristics of absorbent in continuous process. In this study, we have investigated the properties of potassium L-lysine (PL) for getting scale-up factors in continuous process. To obtain optimum condition for removal efficiency of $CO_2$ in continuous process by varying liquid-gas (L/G) ratio, concentration of $CO_2$ and absorbent (PL) were tested. The stable condition of absorber and regenerator (L/G) ratio is 3.5. In addition, PL system reveals the highest removal efficiency of $CO_2$ with 3.5 of L/G and 10.5 vol% $CO_2$ ($1.5Nm^3/h$).

Developmental Delay Effect of Harpacticoid Copepod, Tigriopus japonicus s.l. Exposure to 4-tert-octylphenol (4-tert -octylphenol에 노출된 저서성 요각류 Tigriopus japonicus s.l.의 발생지연 현상)

  • The ecotoxicological effects of 4-tert-octylphenol were observed on Harpacticoid copepoda Tigriopus japonicus s.l. gathered and cultured from tidal pool of Korean coast. There were no significant differences in survival rate (except 10 $\mu$g/L; 70.00%) and sex ratio (except 30 $\mu$g/L) on T. japonicus s.l. exposed to 4-tert-octylphenol. However, 4-tert-octylphenol induced developmental delay (copepodite emergence day and adult male mergence day) and retardation of first brooding day of adult female. Moreover the body size and biomass decreased at 4-tert-octylphenol exposure. As a result, detailed life-cycle research of T. japonicus s.l. may yield potential bioindicators for environmental monitoring and assessment.

Volatile Fatty Acid Production from Saccharina japonica Extracts by Anaerobic Fermentation: Evaluation of Various Environmental Parameters for VFAs Productivity (혐기성 발효에 의한 다시마 추출물로부터 휘발성 유기산 제조: 휘발성 유기산 생산성에 대한 환경적 영향인자 평가)

  • Volatile fatty acids (VFAs) production from marine brown algae, Saccharina japonica, was investigated in anaerobic dark fermentation. In order to evaluate the VFAs productivity, various experimental parameters (i.e., physicochemical pre-treatment, microorganism inoculation ratio, substrate concentration, and pH) were evaluated. According to the physicochemical pre-treatment methods, the maximum concentrations of VFAs were obtained in the order of sulfuric acid, subcritical water and subcritical water with lipid-extraction. Also, we investigated the operating parameters such as microorganism inoculation ratio (MV/M = 10 to 30), the substrate concentration (18.0 to 72.0 g/L) and pH (6.0 to 7.0) in sulfuric acid pre-treatment method. When the substrate concentrations were 18.0, 36.0, 54.0 and 72.0 g/L at $35^{\circ}C$, microorganism inoculation ratio 15, pH 7.0 for 372 hours, the maximum concentrations of VFAs were respectively 9.8, 13.9, 18.6 and 22.3 g/L. The change in VFAs concentrations was detected that acetic- and propionic acids increased according to increasing pH, while the butyric acid increased with decreasing pH. The VFAs obtained from concentration and separation process may be used as basic chemistry materials and bio-fuel, and they will expect to produce alternative energy of fossil fuel.

Enhanced Acidification Efficiency of Sewage Sludge by Seaweed Addition (해조류 첨가를 통한 하수슬러지 산발효 효율 증대)

  • In the present work, the synergistic effect of seaweed addition on organic acid production from sludge was investigated. The batch experiment was conducted at various mixing ratios of sewage sludge and seaweed (100:0, 75:25, 50:50, 25:75, 0:100 on a COD basis) under the substrate concentration of 20 g COD/L. The fermentation temperature was conducted under mesophilic condition ($35^{\circ}C$) and a heat-treated ($90^{\circ}C$ for 20 min) anaerobic digester sludge was used as a seeding source to suppress the methanogenic activity, The results showed that the amount of organic acid production increased as the content of seaweed increased: organic acids were 1.45, 3.22, 4.28, 5.24 and 4.82 g COD/L for the mixing ratio of 100:0, 75:25, 50:50, 25:75 and 0:100 respectively. The synergistic effect was calculated based on the organic acid production of individual sludge and seaweed, and was found to be 0.92, 1.14, 1.26 g COD/L at the mixing ratio of 75:25, 50:50 and 25:75, which indicates that 40% of synergy was obtained when 25% of seaweed was added. The synergistic effect could be ascribed to the high C/N ratio and biodegradability of seaweed.

Monitoring on Functional Properties of Solid Cheonggukjang Extracts by Using Response Surface Methodology (반응표면분석을 이용한 생청국장 추출물의 기능성에 대한 모니터링)

  • Extraction characteristics of solid Cheonggukjang and the functional properties of corresponding extracts were monitored by the response surface methodology (RSM). The maximum extraction yield of 12.08% was obtained under the specific extraction conditions, such as microwave power of 132.40 W, ratio of solvent to sample content of 7.57 g/mL and extraction time of 14.11 min. The maximum electron donating ability (EDA) was found as 33.53% at the conditions of 113.26 W microwave power, 16.39 g/mL of ratio of solvent to sample content and 12.67 min extraction time. The maximum superoxide dismutase (SOD) -like activity was 89.62% under the extraction conditions of 61.82 W, 17.06 g/mL and 9.03 min. Based on superimposition of three dimensional RSM with respect to extraction yield, electron donating ability and SOD-like activity obtained under the various extraction conditions, the optimum ranges of extraction conditions were found to be microwave power of $75{\sim}140W$, ratio of solvent to sample content of $3{\sim}12g/mL$ and extraction time of $1{\sim}11min$, respectively.

Optimization of C/N ratio for production of heteropolysaccharide-7 by Beijerinckia indica

  • Heteropolysaccharide-7 (PS-7) was produced by Beijerinckia indica HS-2001 under aerobic condition. Production of PS-7 was investigated under various ratios of glucose as carbon source to ammonium nitrate as nitrogen source. Maximal production of PS-7 was 7.13 g/l when concentrations of glucose and ammonium nitrate were 10 g/l and 0.3 g/l, respectively. But its conversion rate from glucose was as low as 7 %. The highest conversion rate of PS-7 was 46% when those of glucose and ammonium nitrate were 1.0 g/l and 0.3 g/l, respectively.

Quality Characteristics and Optimization of Rice Muffins prepared by Substituting Salt with Spergularia marina L. Griseb using Response Surface Methodology (반응표면분석법을 이용한 소금대용 세발나물 첨가 쌀머핀의 품질특성 및 최적화)

  • The purpose of this study was to determine the optimal composite recipe of rice muffins with 3 concentrations of Spergularia marina L. Griseb, sugar and grape seed oil, using central composite design. In addition, the mixing condition of rice muffins was optimized by subjecting it to sensory evaluation and mechanical and physicochemical analysis using response surface methodology (RSM). In regard to its antioxidant effects, Spergularia marina L. Griseb had a total phenol and flavonoid contents and DPPH free radical scavenging activity of 17.03 mg/g, 5.13 mg/g and 17.21%, respectively. The results of mechanical and physicochemical analysis showed significant values for lightness, redness, yellowness, hardness, springiness, chewiness, gumminess, cohesiveness, height, volume, weight, specific volume, loss rate, pH, moisture, sweetness and saltiness (p<0.05), and the results of sensory evaluation showed significant values for color, flavor, taste, softness, appearance and overall quality (p<0.05). As results, optimal sensory ratio was found to be 6.69 g of Spergularia marina L. Griseb, 41.89 g of sugar and 30.48 g of grape seed oil.

Performance of a submerged membrane bioreactor for wastewater mimicking fish meal processing effluent

  • The objective of this work was to analyze organic matter removal, nitrification, biomass growth and membrane fouling in a submerged flat-sheet membrane bioreactor, fed with synthetic wastewater, of similar composition to the effluents generated in a fish meal industry. After biomass acclimatization with saline conditions of 12 gNaCl/L and COD/N ratio of 15 in the bioreactor, results showed that the organic matter removal was higher than 90%, for all organic loading rates (0.8, 1, 1.33 and $2gCOD/L{\cdot}d$) and nitrogen loading rates (0.053, 0.067, 0.089 and $0.133gN/L{\cdot}d$) tested during the study. However, nitrification was only carried out with the lowest OLR ($0.8gCOD/L{\cdot}d$) and NLR ($0.053gN/L{\cdot}d$). An excessive concentration of organic matter in the wastewater appears as a limiting factor to this process' operating conditions, where nitrification values of 65% were reached, including nitrogen assimilation to produce biomass. The analysis of membrane fouling showed that the bio-cake formation at the membrane surface is the most impacting mechanism responsible of this phenomenon and it was demonstrated that organic and nitrogen loading rates variations affected membrane fouling rate.
