Toxicity Evaluation of Endocrine Disrupting Chemicals Using Human HepG2 Cell Line, Lumbricus rubellus and Saccharomyces cerevisiae (HepG2 인간 세포주, Lumbricus rubellus 및 Saccharomyces cerevisiae를 이용한 내분비교란물질의 독성평가)

  • Toxicity evaluation systems for various chemicals and their metabolites have been developed during last decades. In this study, the acute toxicity of endocrine disrupting chemicals, such as endosulfan, bisphenol A, vinclozolin, and 3,5-dichloroaniline, was evaluated using HepG2 cell line, Lumbricus rubellus and Saccharomyces cerevisiae, respectively. The extents of toxicity of the chemicals in different bioassay systems varied substantially, such as endosulfan>3,5-dichloroaniline> bisphenol A in HepG2 cell line system, endosulfan>bisphenol A>3,5-dichloro aniline in L. rubellus system, and 3,5-dichloroaniline>endosulfan>bisphenol A in S. cerevisiae system. Meanwhile, no cytotoxicity was observed by treatment of vinclozolin in the evaluation systems. Our results suggest that earthworm and yeast are useful to evaluate acute toxicity of endocrine disrupting chemicals, and direct comparison of toxicity data from different bioassay systems is unattainable. Based on our results, we propose that the bioassay system with earthworm or yeast, a rapid, simple and economic system, could be applied as pre-test for the toxicity evaluation using human cell line or animals.

The Comparison of Trunk Muscle Activities During Sling and Mat Exercise (요부 안정화 운동에 따른 몸통 근육들의 근활성도 비교)

  • The purpose of this study was to verify the most effective spinal stabilization exercises program by comparing the activities of muscles contributing to spinal stabilization during four types of exercises using a sling and a mat. Twenty healthy males were recruited and each subjects performed four types of exercises. Exercise 1 was performed in a quadruped position with the subjects lifting the left arm and the opposite leg on the mat. Exercise 2 was performed in a prone position while holding a sling with the right hand and the left knee was fully extended while lifting the left arm and right leg. Exercise 3 was performed in quadruped position while holding a sling with one the right hand and lifting the opposite arm and leg. In exercise 4, subjects were instructed to maintain a balance push-up position while holding slings with both hands in 10 cm forward reaching with extended elbows. Electromyographic(EMG) activities were recorded from the multifidus, external oblique, internal oblique, abdominal rectus, and erector spinalis muscles during the exercises. The EMG amplitude of each muscle was normalized to the amplitude in the maximal voluntary isometric contraction (MVIC) of each muscle. Repeated ANOVA and Bonferroni's tests were used to compare the differences in the muscle activity according to the types of exercise. The EMG amplitudes of all the muscles were significantly different according to the types of exercises (p<.05). The highest EMG activities of each muscle was as follow; multifidus was 73.38%MVIC in exercise 3, the erector spinalis was 40.03%MVIC in exercise 3, the external oblique was 135.88%MVIC in exercise 4, the internal oblique was 128.60%MVIC in exercise 4, and the rectus abdominalis was 95.24%MVIC in Exercise 4. The types of exercises showed a significant difference in composition rate of EMG amplitudes of each muscle (p<.05). EMG composition rate of the multifidus was high in exercise 1 and 3. However, EMG composition rates of the external oblique, internal oblique, and the rectus abdominals were high in exercise 2 and 4. These results showed differences in EMG activities of muscles contributing to trunk stabilization during different therapeutic exercises. Therefore, the type of exercise should be carefully selected to effectively strengthen a specific trunk stabilizer.

Analytical Methods on the Determination of Active Ingredients for Hair Nourisher Products (HPLC를 이용한 양모제 유효성분의 동시분석법)

  • The hair nourisher products are used for prevention of hair loss and classified as quasi-drug in Korea. As concerns about hair loss has been increased, the demand for hair nourisher products has been growing. It is difficult to analyze their main ingredients because they contain various ingredients including natural plant extracts, vitamins, preservatives and exfoliators. The purpose of this study was to develop and validate simultaneous analytical methods of active ingredients in hair nourisher products such as nicotinamide, tocopherol, salicylic acid, dexpanthenol and benzyl nicotinate by HPLC. The active ingredients were separated on a $C_{18}$ column by using acetonitrile/phosphate buffer as a mobile phase, and detected at UV 220, 270 and 300 nm. The calibration curve showed linearity in the range of $12.5{\sim}800\;{\mu}g/mL$ and the recoveries were 97.3 ~ 103.5 % (RSD 0.9 ~ 2.8 %) in liquid matrix and 101.9 ~ 115.9 % (RSD 0.7 ~ 7.7 %) in shampoo matrix. Validated method was applied to hair nourisher products obtained from distribution market. Fortunately, all samples met their criteria. This study might be expected to provide the method for determining active ingredients in hair nourisher products and lead to promote a rapid market entry.

The Anti-Inflammatory Effect of Trichilia martiana C. DC. in the Lipopolysaccharide-Stimulated Inflammatory Response in Macrophages and Airway Epithelial Cells and in LPS-Challenged Mice

  • A number of species of the genus Trichilia (Meliaceae) exhibit anti-inflammatory effects. However, the effect of Trichilia martiana C. DC. (TM) on lipopolysaccharide (LPS)-induced inflammation has not, to the best of our knowledge, yet been determined. Therefore, in the present study, the antiinflammatory effect of TM on LPS-stimulated RAW264.7 macrophages was evaluated. The ethanol extract of TM (TMEE) significantly inhibited LPS-induced nitric oxide (NO), prostaglandin 2 (PGE2), inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). TMEE also reduced the levels of inflammatory cytokines, including tumor necrosis factor-alpha (TNF-α), interleukin (IL)-1β and IL-6. The upregulation of mitogen-activated protein kinases (MAPKs) and NF-κB activation was revealed to be downregulated following TMEE pretreatment. Furthermore, TMEE was indicated to lead to the nucleus translocation of nuclear factor erythroid-derived 2-related factor 2 (Nrf2) and the expression of heme oxygenase-1 (HO-1). In H292 airway epithelial cells, the pretreatment of TMEE significantly downregulated the production of LPS-stimulated IL-1β, and TMEE was indicated to increase the expression of HO-1. In animal models exhibiting LPS-induced acute lung injury (ALI), treatment with TMEE reduced the levels of macrophages influx and TNF-α production in the bronchoalveolar lavage fluid (BALF) of ALI mice. Additionally, TMEE significantly downregulated the activation of ERK, JNK and IκB, and upregulated the expression of HO-1 in the lungs of ALI mice. In conclusion, the results of the current study demonstrated that TMEE could exert a regulatory role in the prevention or treatment of the endotoxin-mediated inflammatory response.

Changes in Quality Attributes of Sigumjang with Fermentation (시금장 발효기간에 따른 품질 변화)

  • The various components of sigumjang were analyzed according to fermentation time. Aerobic bacteria were increased with fermentation time. Yeasts and molds were increased until 3 days after fermentation and then not changed. Six free sugars, 3 volatile organic acids and 6 non-volatile organic acids were detected. The content of free amino acids was $1,407.9{\sim}3,053.9\;mg%$. Glutamic acid was most abundant component among the amino acids, followed by phenylalanine, arginine and valine. The ratio of essential amino acid was $34.4{\sim}37.0\;mg%$. The content of mineral was $12,966.7{\sim}13,864.5\;mg%$. Potassium was the most abundant in quantity among the minerals in sigumjang except sodium which was added artificially. The principal fatty acids were linoleic acid, palmitic acid and oleic acid. The ratio of unsaturated fatty acids was $76.3{\pm}1.1%$. As a result of sensory evaluation, the highest score was obtained 3 to 5 days after fermentation.

  • Probiotics have many effects such as antihypertensive, prevention of cancer, antioxidation, reduction of dermatitis symptoms, improvement of mineral absorption, reduction of allergic symptoms, and decrease of cholesterol, However, the main role of probiotics is that they balance intestinal microbials proportion. L. acidophilus is one of probiotics and microflora in intestine. It has an acidification activity, aroma production, texture formation and probiotics properties. We studied on the roles of L. acidophilus in mice. In this study, body weights of mice were decreased when administration of L. acidophilus ($1{\times}10^{10}$ CFU) and swimming ability has been raised than a normal group after feeding on L. acidophilus ($1{\times}10^{10}$ CFU). After taking L. acidophilus ($1{\times}10^{10}$ CFU), total white cells were increased than a normal group; hemoglobin and thrombocytes were increased. The level of cholesterol and triglyceride were decreased in blood analysis. We knew L. acidophilus is related to innate immune system. We found out the secretion of cationic peptide was increased in the Lysoplate assays as a result of L. acidophilus ($1{\times}10^{10}$ CFU) administration. Appearance rate of lysozyme was also increased than the normal group on an immunohistochemistry stain. We confirmed L. acidophilus contributes to host health through innate immune system stimulation. L. acidophilus more than $1{\times}10^{10}$ CFU are thought to be beneficial for the host health and prevention of intestinal diseases in field condition.

Maxillary sinus septum;panoramic radiographic and dental computed tomographic analyses in the planning of implant surgery (상악동 중격;임플란트 수술 계획시 파노라마와 치과용 전산화 단충촬영 분석)

  • Surgical intervention in the posterior maxillary region requires detailed knowledge of maxillary sinus anatomy and the possible anatomical variations. This study evaluated the incidence, location of maxillary sinus septa by using radiographic (panoramic radiography and computed tomography) findings and comparison of panoramic radography with CT in antral anatomical variation. This study was based on data from 70 sinuses in partial dentate maxilla. The sample consisted of 61 patients(25 women and 36 men, with ages ranging between 19 and 77 years and a mean age of $49.4{\pm}11.3$ years) who were being treatment-planned to receive implant-supported restorations. First, the panoramic images were examined for the presence of antral septa by radiologist and examiner who don't know about CT findings. And incidence of antral septa was evaluated using an axial plane of CT image. The incidence of septa was compared between panoramic radiography and CT. The accuracy of the incidence was compared between radiologists and dentists. A total of 20 septa were found in 70 sinuses on CT image and the prevalence of one or more septa per sinus was found to be 28.6%. The assumed incidence of septa on panoramic radiography was $27.6%{\pm}2.2%$ in radiologist and $31.9%{\pm}5.8%$ in dentists. Erroneous diagnosis rate was 11.42% in radiologist and 15.96% in dentists. 40% of antral septa were located in the anterior(premolar) region, 30% of septa were located in the middle(first molar) and posterior(second molar) region separately. Prior to implant placement, it seems appropriate to consider panoramic radiography as a standard radiographic examination and periapical radiographs may be used to complete the findings in regions not sharply depicted in the panoramic radiograph. And cross-sectional imaging should be used in sites with severe bone loss and close proximity of the maxillary sinus.

Comparison of Growth Characteristics Fruit Vegetable Seedlings Grown on Cylindrical Paper Pot Trays of Plug Trays (과채류 공정 육묘시 플러그 및 원통형 종이포트 육묘시스템 간의 생육특성 비교)

  • This study was conducted to examine the growth characteristics of seedlings of several major fruit vegetables and compare them between plants grown with the conventional plug seedling system (Plug) and those grown with the cylindrical paper pot system (CPP). There were no significant differences in shoot growth characteristics, such as plant height, leaf area, dry weight, and fresh weight, between tomato, cucumber, and watermelon plants grown with the CPP and Plug systems. The total root lengths of tomato and cucumber plants grown with Plug were longer than those grown with CPP at the beginning of seedling growth, and the total root lengths of watermelon grown with Plug were longer than those grown with CPP throughout the whole seedling period. The length of root that separated five steps according to the diameter of the root of tomato did not differ between CPP and Plug, but those of cucumber and watermelon were 20-251% longer with Plug than with CPP for all kinds of roots. The fresh weight of Plug-grown tomato roots was about 30% heavier than that of CPP-grown tomato during the seedling period, but the difference decreased as growth progressed. Finally, there was no difference between plants before and after transplanting. The fresh weights of the Plug-grown plants were about 20-30% heavier than those of CPP-grown ones in terms of the whole seedling. After transplanting to the Wagner pot, the shoot growth of Plug-grown watermelon plants in terms of dry weight was low until 7 days had passed. However, this tendency was reversed from 8 days after transplantation onward. In conclusion, there were no significance differences in the growth of shoots between plants grown with the CPP and Plug systems. However, root development was better with Plug than with CPP.

  • Epilepsy is characterized by the presence of spontaneous episodes of abnormal neuronal discharges and its pathogenic mechanisms remain poorly understood. Recently, we found that the expression of creatine kinase (CK) was markedly decreased in an epilepsy animal model using proteomic analysis. A human CK gene was fused with a HIV-1 Tat peptide to generate an in-frame Tat-CK fusion protein. The purified Tat-CK fusion protein was efficiently transduced into PC12 cells in a time- and dose-dependent manner when added exogenously to culture media. Once inside the cells, the transduced Tat-CK fusion protein was stable for 48 h. Moreover, the Tat-CK fusion protein markedly increased endogenous CK activity levels within the cells. These results suggest that Tat-CK provides a strategy for the therapeutic delivery of proteins in various human diseases including the delivery of CK for potential epilepsy treatment.

Application Method of GIS for Planning of Forest Road Network (임도망의 계획에 있어서 GIS 활용방안)

  • The application method of GIS for planning of forest road network can be save the efforts and cost by frequently change of forest road and evaluation the forest road route before construction. The purpose of this study is to suggest the proper method for planning an optimal forest road network in mountains forest using geographic information system(GIS) in the national forest of Mt. Kumsan at Namhae-gun, Gyungsangnam-do. In the forest road network planning by the minimum longitudinal gradient, The total length was 20.41km, and road density was 6.92m/ha. In the forest road network planning by mixed with the minimum longitudinal gradient and the maximization of investment effect, The total length was 21.15km, and road density was higher than that of the minimum longitudinal gradient as 7.17m/ha. The road length overlapped by cost path was more short than 3.52m/ha of the minimum longitudinal gradient as 1.73km. So, it appeared that forest road has an high effectiveness in yarding function. Therefore, it considered that the geographic information system could provide an effective and resonable solutions for planning of optimal forest road network.
