Qi-deficiency in the Patients with Allergic Disorders

  • ■ Introduction Allergy refers a hypersensitive immune response to the various environmental factors and Qi is considered as vit al energy to defend oneself from environments in Traditional Chinese Medicine. Therefore, this study was performed to assess the relationship between allergy and Qi-deficiency ■ Methods We enrolled the subjects with allergic disorders and the healthy controls without allergic disorders. We got the information on the general characteristics, and the score of Qi-deficiency from all of the subjects, and measured their total Immunoglobulin E(IgE) level. ■ Results There were one hundred forty three subjects with allergic disorders and 211 of the control. The educational level, total IgE, and the scores of the Qi-deficiency were higher in the allergy group than in the control. ■ Conclusion Allergic subjects tend to have more severe Qi-deficiency. This could suggest the clinical effectiveness of some herbal medicines for Qi-deficiency on allergic disorders.

데이터베이스를 이용한 식료찬요(食療纂要)의 통계적 연구 (A Statistical Study on Sikryo-chanryo by Applying Database)

  • 본 연구의 목적은 한국 고유의 전통지식에 기반하여 음식을 이용한 건강증진 방법에 대한 지식을 체계화하고 통계적으로 활용할 수 있는 방법에 대하여 연구하는 데 있다. 이를 위하여 한국 고유의 음식을 이용한 질병치료에 대하여 언급한 고문헌인 식료찬요(食療纂要)의 지식을 DB로 구축하였다. 데이터의 처리는 Relational Data Model을 이용하였고, 9개의 데이터 테이블을 사용하였다. 소프트웨어는 MS사의 Access 2014를 사용하였다. 그 결과, 데이터베이스 쿼리문장을 이용하여 식재료, 약재별, 증상, 병명, 효능, 변증 정보를 유기적으로 조건을 만들어 통계적 결과를 즉시 확인할 수 있었다. 따라서 이러한 Relational Data Model을 이용하여 기존 방식의 연구논문을 대체할 수 있었다.

The culture conditions for mycelial growth and sclerotial formation of Polyporus umbellatus

  • Polyporus umbellatus (Syn. Grifola umbellata) is a sclerotium forming mushroom belongs to family Polyporaceae of Polyphorales, Basidiomycota. The sclerotia of P. umbellatus have long been used for traditional medicines in China, Korea and Japan. This study was initiated to obtain the basic data for artificial sclerotial production of P. umbellatus. Here, we investigated the favorable conditions for mycelial growth of P. umbellatus and its symbiotic fungus Armillaria mellea. We also evaluate the favorable carbon and nitrogen sources for sclerotial formation in dual culture between P. umbellatus and A. mellea. The favorable conditions for mycelial growth of P. umbellatus were $20^{\circ}C$ and pH 4, while optimal conditions for mycelial growth of A. mellea were $25^{\circ}C$ and pH 6. The carbon sources for optimal mycelial growth of P. umbellatus were fructose and glucose, while carbon sources for favorable mycelial growth of A. mellea were also fructose and glucose. The nitrogen sources for favorable mycelial growth P. umbellatus were peptone and yeast extract, while optimal mycelial growth of A. mellea were obtained in peptone and yeast extract. When P. umbellatus and A. mellea were dual cultured on carbon sources, sclerotia were induced on basal media supplemented with glucose, fructose and maltose at pH 4~6, while nitrogen sources inducing sclerotia were basal media supplemented with peptone and yeast extract for 60 days at $20^{\circ}C$ under dark condition.

Protective Effects of a Herb, Menthae Herba, against Radiation-induced Oxidative DNA Damage

  • As utilization of radiation in medicine, industry and biochemical research increases, the protection against radiation damage has become an important issue. Natural products such as herbal medicines are beginning to receive attention as modifiers on the radiation response. In the present study, the protective effect of a herb, Menthae Herba, against radiation-induced DNA damage was evaluated using alkaline single-cell gel electrophoresis (SCGE; comet assay) in the mouse peripheral blood Iymphocytes and the micronucleus formation test in the Chinese hamster ovary (CHO) cells. The tail moment, which was a marker of DNA damage in the SCGE, and the frequency of micronuclei was decreased in groups treated with Mentae Herba extract before exposure to 200 cGy of gamma-ray. We also confirmed its activities to scavenge DPPH and hydroxyl radicals. These experiments demonstrated that Menthae Herba was effective at reducing the radiation-induced damage of DNA and scavenging free radicals. It is plausible that scavenging of free radicals by Menthae Herba may have played an important role in providing the protection against the radiation-induced damage to the DNA. These results indicated that Menthae Herba might be a useful radioprotector and that radical scavenging appears to be one of the mechanisms of radiation protection.

Effects of Oleo Gum Resin of Ferula assa-foetida L. on Senescence in Human Dermal Fibroblasts - Asafoetida reverses senescence in fibroblasts -

  • Objectives: Based on data from Chinese and Indian traditional herbal medicines, gum resin of Ferula assa-foetida (sometimes referred to asafetida or asafoetida) has several therapeutic applications. The authors of various studies have claimed that asafetida has cytotoxic, antiulcer, anti-neoplasm, anti-cancer, and anti-oxidative effects. In present study, the anti-aging effect of asafetida on senescent human dermal fibroblasts was evaluated. Methods: Senescence was induced in in vitro cultured human dermal fibroblasts (HDFs) through exposure to $H_2O_2$, and the incidence of senescence was recognized by using cytochemical staining for the activity of ${\beta}$-galactosidase. Then, treatment with oleo gum resin of asafetida was started to evaluate its rejuvenating effect. The survival rate of fibroblasts was evaluated by using methyl tetrazolium bromide (MTT) assays. Real-time quantitative reverse transcription-polymerase chain reaction (qRT-PCR) and western blot assays were performed to evaluate the expressions of apoptotic and anti-apoptotic markers. Results: Our experiments show that asafetida in concentrations ranging from $5{\times}10^{-8}$ to $10^{-7}g/mL$ has revitalizing effects on senescent fibroblasts and significantly reduces the ${\beta}$-galactosidase activity in these cells (P < 0.05). Likewise, treatment at these concentrations increases the proliferation rate of normal fibroblasts (P < 0.05). However, at concentrations higher than $5{\times}10^{-7}g/mL$, asafetida is toxic for cells and induces cell death. Conclusion: The results of this study indicate that asafetida at low concentrations has a rejuvenating effect on senescent fibroblasts whereas at higher concentrations, it has the opposite effect of facilitating cellular apoptosis and death.

GC-MS 기반 대사체학 기법을 이용한 산수유의 산지판별모델 (Discrimination model of cultivation area of Corni Fructus using a GC-MS-Based metabolomics approach)

  • 생약의 원산지를 판별하는 논리적인 일련의 기준을 개발한다면, 현재 유통되는 한약을 좀 더 과학적으로 관리 할 수 있을 것이다. 이러한 노력은 전통적인 한약 산업 발전에 기여할 것이라고 사료된다. 산수유의 원산지 판별법을 개발하기 위해, 본 연구에서는 우선 국산 산수유와 중국산 산수유를 각각 수증기 증류하고 이 때 얻은 휘발성분을 GC/MS를 이용하여 분석하였다. NIST mass spectral library의 데이터베이스로부터 정성분석한 결과를 바탕으로 데이터를 범주화(binning)하여 변수를 얻고, 이에 대하여 PCA, OPLS-DA 등 다변량 통계 분석을 수행함으로써 신속, 정확하게 국산 산수유와 중국산 산수유의 산지를 판별할 수 있는 산지 판별모델을 확립하였다. 산지 판별모델 개발을 위해서 학습집합(n=53)을 분석하여 산지 판별모델을 수립한 후, 검증집합(n=12)을 산지 판별모델에 적용함으로써 그 타당성을 확인하였다. 더불어 1-ethylbutyl-hydroperoxide, nonadecane, butylated hydroxytoluene, 5β,7βH,10α-Eudesm-11-en-1α-ol, 7,9-bis (2-methyl-2-propanyl)-1-oxaspiro[4.5]deca-6,9-diene-2,8-dione, 그리고 2-decyldodecyl-benzene 등 6개의 마커성분을 선정할 수 있었다. 최근에 NMR을 활용한 산수유 원산지 판별에 대한 보고는 있었으나, GC/MS를 기반으로 한 대사체학 연구기법을 이용하여 산지판별 모델을 제시하는 것은 최초의 보고로서 그 의미가 크다. 본 연구결과를 활용하여 한약의 원산지 판별모델 확립과 산수유 원산지의 과학적인 관리에 적용할 수 있으리라 사료된다.

산머루 관련 정보수집 및 데이터베이스의 구축 (Data Mining and Construction of Database Concerning Effects of Vitis Genus)

  • The database for the oriental medicine had been existed in documentation in past times and it has been developed to the database type for random accesses in the information society. However, the aspects of the database are not so diversified and the database for the bio herbal material exists in widened type dictionary style. It is a situation that the database which handles the in-depth raw herbal medicines is not sufficient in its quantity and quality. Korean wild grape is a deciduous plant categorized into the Vitaceae and it was found experimentally that it has various medical effects. It is one of the medical materials with higher potentiality of academic study and commercialization recently because it has a bigger possibility to be applied into diverse industrial fields including the medical product for health, food and beauty. We constituted the cooperative system among the Muju cluster business group for Korean mountain wild grapes, Physiology Laboratory in Kyung Hee University Oriental Medicine and Medical Classics Laboratory in Kyung Hee University Oriental Medicine with a view to focusing on such potentiality and a database for Korean wild grapes was made a touchstone for establishing the in-depth database for the single bio medical materials. First of all, the literatures based on the North East Asia in ancient times had been categorized into the classical literature (Korean literature published by government organization, Korean classical literature, Chinese classical literature and classical literature fro Korean and Chinese oriental medicine) and modern literature (Modern literature for oriental medicine, modern literature for domestic and foreign herbal medicine) to cover the eastern and western research records and writings related to Korean wild grapes and the text-mining work has been performed through the cooperation system with the Medical Classics Laboratory in Kyung Hee University Oriental Medicine. First of all, the data for the experiment and theory for Korean wild grape were collected for the Medline database controlled by the Parliament Library of USA to arrange the domestic and foreign theses with topic for Korean wild grapes and the network hyperlink function and down load function were mounted for self-thesis searching function and active view based on the collected data. The thesis searching function provides various auxiliary functions and the searching is available according to the diverse searching/queries such as the name of sub species of Korean wild grape, the logical intersection index for the active ingredients, efficacy and elements. It was constituted for the researchers who design the Korean wild grape study to design of easier experiment. In addition, the data related to the patents for Korean wild grape which were collected from European Patent Office in response to the commercialization possibility and the system available for searching and view was established in the same viewpoint. Perl was used for the query programming and MS-SQL for database establishment and management in the designing of this database. Discrimination and Authentication of Eclipta prostrata and E. alba Based on the Complete Chloroplast Genomes

  • Eclipta prostrata and E. alba are annual herbal medicinal plants and have been used as Chinese medicinal tonics. Both species are widely distributed in tropical and subtropical regions as well as in Korea. Both species have similar morphological features but E. alba has smoother leaf blade margins compared with E. prostrata. Although both species are utilized as oriental medicines, E. prostrata is more widely used than E. alba. Morphological semblances have confounded identification of either species. Here, we report the complete chloroplast genomes of both species to provide an authentication system between the two species and understand their diversity. Both chloroplast genomes were 151,733-151,757 bp long and composed of a large single copy (83,285-83,300 bp), a small single copy (18,283-18,346 bp), and a pair of inverted repeats (25,075-25,063 bp). Gene annotation revealed 80 protein coding genes, 30 tRNA genes and four rRNA genes. A phylogenetic analysis revealed that the genus Eclipta is grouped with Heliantheae tribe species in the Asteraceae family. A comparative analysis verified 29 InDels and 58 SNPs between chloroplast genomes of E. prostrata and E. alba. The low chloroplast genome sequence diversity indicates that both species are really close to each other and are not completely diverged yet. We developed six DNA markers that distinguish E. prostrata and E. alba based on the polymorphisms of chloroplast genomes between E. prostrata and E. alba. The chloroplast genome sequences and the molecular markers generated in this study will be useful for further research of Eclipta species and accurate classification of medicinal herbs.

방사선조사 마우스에서 소장움세포 및 조혈세포 생존에 미치는 사물탕 및 사군자탕의 영향 (Effect of Si-Wu-Tang and Si-Jun-Zi-Tang on the Survival of Jejunal Crypt Cells and Hematopoietic Cells in Irradiated Mice)

  • 한방 보혈제의 기본처방인 사물탕과 보기제의 기본 처방인 사군자탕의 방사선 조사 마우스에서의 방사선 방호 효과를 소장움 생존(고선량 방사선, 12 Gy), 조혈세포 생존(중간선량 방사선, 6.5 Gy), apoptosis 형성(저선량 방사선, 2 Gy) 실험법을 적용하여 관찰하였다. 방사선조사 전 사물탕투여군(경구투여: 음수 mL당 2 mg씩 7일간, 복강내주사: 마리당 1 mg씩 방사선 조사 24시간 전)에서 소장움의 생존이 증가되었으며 (방사선대조군: $38.48{\pm}4.34$, 복강내 주사군: $77.98{\pm}16.11$, P<0.005), endogenous spleen colony의 형성도 높게 나타났고(방사선대조군: $3.5{\pm}4$, 경구투여군: $10.7{\pm}7.730$, 복강내주사군: $8.667{\pm}5.244$, P<0.05), 소장움에서의 apoptotic cell의 발생율은 억제되었다(방사선대조군: $5.111{\pm}0.529$, 복강내주사군 $3.919{\pm}0.214$, P<0.01). 사군자탕 병행투여군에서는 이상의 실험에서 통계적으로 유의성 있는 효과는 없었다. 이상의 결과는 사물탕이 방사선방호 식품으로 활용될 수 있는 가능성을 제시하였으며, 각 구성약재 및 성분의 효과에 대한 연구가 계속되어야 할 것이다.

련화향(蓮花香) 정유액이 glioma cell에 미치는 효과 (Effects of the Essential Oil of Nelumbo nucifera Flower on Glioma Cells)

  • Objective : Herb medicines are potential sources of useful edible and medicinal plants. They are used as a drug because of their various biological activities such as immunomodulatory, antiviral, and antitumor functions. Nelumbo nucifera have been applied in Chinese herbal prescriptions to improve tissue inflammation. However, it has not been elucidated on the effect of the flower of Nelumbo nucifera in cells. Method : In the present study, to examine the effect of that on glioma cells, U87, the essential oil was extracted from the flower of Nelumbo nucifera (NN essential oil). U87 cells were exposed to different concentrations of 2-40 ug/ml of NN essential oil in ethanol. Cell viability was measured by MTT assay at 24 h. To find out the intracellular target signal molecule(s) for this antiproliferative activity of NN essential oil, phosphorylation of Akt, ERM, MAPK or p38 proteins were examined by Western blot analysis. To study long term effect of NN essential oil in U87 cells, the image of cells treated with NN essential oil for 4 days were obtained. Results and Conclusion : NN essential oil was shown to exhibit antitumor activity in glioma cells, at a broad range of concentrations of 10-40 ug/ml. The phosphorylation of Akt and Endoplasmic Reticulum Matrix (ERM) proteins which known to be involved in the cell death, were gradually decreased to 2 hours after addition 20 ug/ml of NN essential oil. However, the phosphorylation of mitogen-activated protein (MAPK) and p38 was found to increase in NN essential oil treated cells. NN essential oil treated cells showed decreased glioma cell number. These results provide a possible NN essential oil-induced inhibitory signal for tumor cell proliferation that is initiated by the decrease in Akt activity. Moreover, it is likely that the activation of p38 is required for the NN essential oil-induced inhibition of tumor proliferation.
