Polymorphism and Expression of Isoflavone Synthase Genes from Soybean Cultivars

  • Isoflavones are synthesized by isoflavone synthases via the phenylpropanoid pathway in legumes. We have cloned two isoflavone synthase genes, IFS1 and IFS2, from a total of 18 soybean cultivars. The amino acid residues of the proteins that differed between cultivars were dispersed over the entire coding region. However, amino acid sequence variation did not occur in conserved domains such as the ERR triad region, except that one conserved amino acid was changed in the IFS2 protein of the GS12 cultivar ($R_{374}G$) and the IFS1 proteins of the 99M06 and Soja99s65 cultivars ($A_{109}T$, $F_{105}I$). In three cultivars (99M06, 99M116, and Simheukpi), most of amino acid changes were such that the difference between the amino acid sequences of IFS1 and IFS2 was reduced. The expression profiles of three enzymes that convert naringenin to the isoflavone, genistein, chalcone isomerase (CHI), isoflavone synthase (IFS) and flavanone 3-hydroxylase (F3H) were examined. In general, IFS mRNA was more abundant in etiolated seedlings than mature plants whereas the levels of CHI and F3H mRNAs were similar in the two stages. During seed development, IFS was expressed a little later than CHI and F3H but expression of these three genes was barely detectable, if at all, during later seed hardening. In addition, we found that the levels of CHI, F3H, and IFS mRNAs were under circadian control. We also showed that IFS was induced by wounding and by application of methyl jasmonate to etiolated soybean seedlings.

Selection of Forage Soybean Cultivars in Jeju Region (제주지역에서 사료용 콩의 우량품종 선발)

  • This study was carried out to select superior soybean cultivars for forage production for soybean forage production in Jeju. The results obtained are summarized as follows; Eighteen soybean cultivars were grown from May 11 to September 11 in 2002 in Jeju to select superior cultivars for forage production. Days to flowering was shortest(58 days) for Seokryangkong and Keunolkong, and longest(93 days) for Danpaheukdu. Days to flowering for other cultivars ranged from 65 to 69 days. Pureunkong and Namhaekong had greater heights(129.3 and 124.6 cm, respectively) while Keunolkong, Hwaeomkong and Seokryangkong had shorter heights. The number of branches per plant was gratest for Sobaekkong, Manrikong, Pungsankong, Kwangankong and Geumgangkong. The number of leaves per plant was geatest for Sobaekkong, Iksankong and Namhaekong. Rueunkong and Jangmikong had thicker stems. Fresh forage yield was greatest for Iksankong, Sobaekkong and Namhaekong(39.5, 39.3 md 38.0 MT/ha respectively). DM yield of forage was featest for Baekunkong, Hnrunkong and Danpaheukdu. Danwonkong, Dawonkong, Seokryanfong and Pungsankong had ueatest forage crude protein content. Duyukong and Kwangankong had featest forage crude fiber content. Jangmikong and Keunolkong had feater crude fat content, while Sobaekkong and Kwangankong had greater crude ash content. Namhaekong, Seokryangkong, Keunolkong and Jinpumkong had greater NFE contents ranging 40.0 to 43.5 and TDN was featest in Keunolkong, Danpaheukdu, Namhaekong and Seokryangkong had greater NFE contents ranging from 59.8 to 60.9%. The best cultivars in Jeju for forage soybean appear to be Namhaekong, Danpaheukdu md Baehukong on the basis of crude protein, TDN, and dry matter yields.

Influence of Antibiotics on Shoot Regeneration and Agrobacteium Suppression Using Cotyledonary Node in Korean Soybean Cultivars

  • Mature dry seeds of Korean cultivars, Daepungkong, Muhankong, Myeongjunamulkong, Somyeongkong, Sowonkong, Jinpumkong, and Pungsannamulkong were used. The influence of antibiotics on elimination of Agrobacterium growth and shoot regeneration was estimated with cotyledonary node. Cefotaxime and timentin at the concentration of 250 and 500 mg/l suppressed Agrobacterium, especially cefotaxime was an efficient antibiotic to suppress Agrobacterium in all cultivars. While carbenicillin and timentin at the concentration of 50 and 100 mg/l were not sufficient to control the development of Agrobacterium, respectively. Cefotaxime and timentin represented high shoot formation rates compared with carbenicillin. Carbenicillin at low concentrations did not effectively suppress Agrobacterium and also had no effect on shoot development. Cefotaxime at the concentration of 250 mg/l showed maximum frequency of shoot regeneration in cvs. Somyeongkong and Sowonkong. Furthermore, on medium containing cefotaxime, shoot was more quickly formed than the other antibiotics. The use of cefotaxime was very useful for elimination of Agrobacterium growth with cotyledonary node of Korean soybean cultivars.

Protein Composition of Domestic and Glyphosate-Tolerant Soybean (국내산 및 제초제 내성 콩의 단백질 조성 분석)

  • In order to elucidate the differences of protein profiles among soybean cultivars, the protein composition of three conventional domestic soybean cultivars and two imported ones including glyphosate-tolerant HS2906 was analyzed by total nitrogen measurement, amino acid analysis and PAGE/densitometry. There were no statistically significant differences in the levels of any amino acid, including aromatic amino acids, between glyphosale-tolerant soybean and the conventional soybean WS82. In the extraction of protein, the SDS/buffer system was more efficient than the defatting/water system. The SDS-PAGE/densitometry analysis showed that there was a similar profile of proteins among cultivars, although the amount of total protein ranged from 380.2 mg/g to 423.9 mg/g. In addition, there was no discernable difference of protein profile between glyphosate- tolerant soybean (total protein amount, 380.2 mg/g) and the conventional soybean WS82 (390.2 mg/g), although the amount of ${\beta}$-conglycinin (55 kDa) was lower in glyphosate-tolerant soybean. Meanwhile, the amount of 25 kDa protein was greater in domestic soybean cultivars than imported ones. Thus, normal PAGE/ densitometry method would be useful to analyze the difference in protein profiles of soybean proteins, and furthermore Evaluate the protein profile of proteins between GMO and conventional soybean.

Quality Properties of Soy-paste Soybean Cultivar for Fermented Soybean Products (장류용 콩 품종별 발효물의 품질 특성)

  • This study evaluates the quality properties of soy-paste soybean cultivar for fermented soybean products. The six soybean varieties that include Jinpung, Saedanbaek, Daepung 2, Pyeongwon, Cheonga and Saeolkong were used in the experiment. The range of water uptake ratio, hardness after soaking and hardness after steaming were 117.00~131.33%, 1.65~3.30 kg and 0.05~0.14 kg, respectively. The physicochemical analysis indicated the following: Moisture content, 63.27~68.72%; pH, 6.43~6.60; total acidity, 0.27~0.45%. Color values for L value (lightness), a value (redness), and b value (yellowness) ranged from 39.07~67.92, 7.64~11.79, and 7.48~20.67, respectively. The amylase and protease activities of the Saedanbaek samples were the highest among all cultivars. The amount of viscous substance in the fermented soybean products by cultivars ranged from 5.93 to 8.37%, and Saedanbaek was the highest. The total viable cells counts for soybean fermented products were 9.11~9.42 log CFU/g. The amino-type nitrogen contents of all samples were in the range of 401.07 to 524.47 mg% and Saedanbaek cultivars showed the highest content (524.47 mg%). Based on the results, Saedanbaek will be suitable as a soy-paste soybean cultivar and the quality standards for the fermentation process of the fermented soybean products.

Occurrence of Web Blight in Soybean Caused by Rhizoctonia sol ani AG-l(IA) in Korea

  • Web blight symptoms were frequently observed on soybean plants grown in a farmer's fields located in Jincheon in Korea during a disease survey in August, 2005. Incidence of the disease was $5-20\%$ infected plants in two of four soybean fields investigated. A total of 31 isolates of Rhizoctonia sp. were obtained from leaves, leaf petioles, and pods of diseased soybean plants. The isolates were identified as Rhizoctonia solani AG-l(IA) by anastomosis test and based on the morphological and cultural characteristics. Three isolates of R. solani AG-l(IA) were tested for pathogenicity to five cultivars of soybean by artificial inoculation. All the isolates induced blight symptoms on the leaves of soybean and formed sclerotia on the lesions, which were similar to those observed in the field. The pathogenicity tests revealed that all the soybean cultivars tested were susceptible to the pathogen. There was no difference in the pathogenicity among the isolates. The present study first reveals that R. solani AG-l(IA) causes web blight of soybean in Korea.

Differential Response of Soybean Cultivars to Alachlor, Linuron, and Metribuzin (Alachlor, Linuron과 Metribuzin의 약해반응에 대한 대두품종간 차이)

  • Forth soybean (Glycine max (L.) Merr.) cultivars were evaluated to determine the extent of crop injury caused by differential sensitivity of cultivars to two rates of alachlor, linuron, and metribuzin. Most cultivars were relatively tolerant of alachlor at 120 and 240g/10a and also linuron at 62.5 and 125g/10a. However, many cultivars were highly sensitive to metribuzin at 50 and 100g/10a. Some of the most sensitive cultivars were ‘Kyungnam 2’, ‘Toyosuzu’, ‘Noki 1’, ‘Iwade 2’, and ‘Hampton’. Metribuzin also showed a very narrow margin of safety to soybean cultivars.

Fatty Acid and Free Amino Acid Composition of Major Domestic Soybean Cultivars (국내산 백태 품종의 지방산 및 유리아미노산 조성)

  • The fatty acid composition and free amino acid content of domestic soybean cultivars were analyzed to confirm the quality characteristics of protein and fat contained in soybeans. The saturated fatty acid content of soybeans included palmitic acid at 9.47~11.15%, followed by stearic acid and myristic acid. The total saturated fatty acid content in soybeans was 12.56~14.34%, with Taekwang having the lowest content, followed by Daewon, Seonyu, Cheonga, and Jinpung. The linoleic acid content, an unsaturated fatty acid, was 45.69~58.17%, with Taekwang showing the lowest composition and Jinpung showing the highest composition. Next was oleic acid at 14.69~33.86%. Jinpung had the highest linoleic acid composition, had the lowest and Taekwang which had the least linoleic acid, had the highest. The unsaturated fatty acid content was in the order of linolenic acid, eicosatrienoic acid, eicosadienoic acid, and eicosapentaenoic acid. The total free amino acid content was 217.28~456.66 mg%, with Daewon showing the highest free amino acid content, followed by Seonyu, Taekwang, Cheonga, and Jinpung. The free amino acid content varied depending on the cultivars, but in general, the free amino acids in the soybeans used in the experiment showed higher aspartic acid, glutamic acid, and proline contents than other amino acids.

Multiple Shoot Formation from Cotyledonary Nodes of Soybean Cultivars (대두 품종에 따른 자엽절에서의 다신초 형성)

  • For the plant regeneration of soybean (Glycine me L. Merr.), the shoot formation rate, optimal medium and tissue conditions were examined using Korean soybean cultivars. Among the parts of seedling, a node that includes one cotyledon showed the highest shoot formation rate among other tissues. Half-strength B5 medium was more efficient than full strength medium. Formation rates of pair shoots (1 to 2 shooting) were higher in the when benzyl adenine was supplemented. The formation rates of multiple shoots, that is, 4 to 5 in shooting, were high when thidiazuron was supplemented. Multiple shoot was de novo formed in cutting side of cotyledonary node. The effective concentration of thidiazuron for shoot induction treatment was 2 mg/L. Among the 27 cultivars, multiple shoot formation rates were high in the 11 cultivars including 'Heugcheongkong, and pair shoot formation rates were high in the 16 cultivars including 'Malikong'.

Qualities Analysis of Domestic Soybean Cultivars (국내산 두류 품종에 대한 품질 분석)

  • To analyze the quality characteristics of soybean (Glycine max), the approximate analysis, physical properties, mineral and free sugar content of domestic soybean cultivars were analyzed. The moisture content was about 9.15~11.84%. The fat content of Cheonga, Taekwang, and Jinpung cultivars were 17.93~18.37%, which was significantly higher than that of others. Protein content of Cheonga and Seonyu were 36.15~36.70%, significantly higher than that of others. As for the weight and shape of soybeans, the Daewon was the largest allele, and the seed cover ratio was the highest in the Seonyu. In terms of water absorption rate, the Seonyu showed the highest absorption rate at 246.19%, and the Daewon showed the lowest absorption rate. Soaked soybeans had a hardness of 2.80~4.73 kg/㎠, which did not show low hardness in the sample with high moisture absorption. The grinded soybeans showed higher lightness and yellowness values than the raw soybeans, and the redness was decreased. Soybean minerals contained high K, P, Mg, and Ca content. Overall, The Taekwang contained more minerals than others. Stachyose, sucrose, glucose and fructose were detected as free sugars in soybeans. The total free sugar content of Taekwang was the lowest at 3.47%, and the Cheonga (6.80%) was the highest.
