Streptozotocin으로 유발한 당뇨 마우스에서 식물성 지방 급원에 따른 혈액 및 간조직의 지질 조성 (Effect of Different Kind of Plant Oil Sources on Serum and Hepatic Lipid levels of Streptozotocin-induced Diabetic Mice)

  • 본 연구는 식물성 지방급원이 당뇨모델의 지질대사에 미치는 영향을 검토하고자 수행하였다. 실험 모델은 STZ의 투여로 유발된 인슐린의존형 당뇨 마우스를 이용하였고, 4종류(라아드 10%에 각각 대두유, 쌀눈기름, 쌀겨 기름, 참기름 10% 첨가)의 실험식이를 7주간 급여하여 체내 지질 함량 및 대변으로의 배설량을 조사하였다. 그 결과 실험동물의 식이섭취량과 체중 및 장기 무게는 실험군 사이에 큰 차이가 없었으나, 혈청 중성지방은 쌀눈기름 및 쌀겨기름 첨가군에서 다른 실험군들보다 낮은 경향을 보였고, 간의 총콜레스테롤 함량은 쌀눈기름 첨가군에서 대조군보다 낮은 경향을 보였으며, 간의 총지질 함량도 쌀눈기름 첨가군에서 대조군보다 낮은 경향을 보였다. 그리고 대변으로의 총지질 및 총콜레스테롤 배설량은 쌀눈기름 첨가군과 쌀겨기름 첨가군에서 대조군인 대두유군보다 유의하게 많은 것으로 나타났다. 이상의 결과로부터 쌀눈기름과 쌀겨기름 급여군에서 체내 지질 함량이 낮았고 쌀눈기름군에서 쌀겨기름군보다 낮은 경향을 보였다. 그러므로 쌀눈기름은 인슐린의존형 당뇨마우스에서 체내 지질 함량 저하 효과가 있는 것으로 밝혀졌으며, 이는 쌀눈기름의 대변중 총지질 및 총콜레스테롤 배설량 증가와 관련된 것으로 평가되었다. 따라서 쌀눈기름은 대두유나 참기름보다 당뇨병 모델의 체내 지질대사에 개선 효과가 있는 것으로 보인다.

Effect of Temperature Variables on Growth and Inorganic Nutrient Contents of Codonopsis lanceolata

  • This study was conducted to investigate the effects of temperature and shade, which are basic environmental conditions, on growth, yield, inorganic components, and general components of Codonopsis lanceolata, in order to obtain basic data for improving yield capacity. In natural light, in the 15, 20, and $25^{\circ}C$ groups, the plant heights ranged between 218.9 cm and 223.9 cm, and there was no significant difference between groups. However, the leaf size was larger in shade, and the leaf area was significantly larger in the 15 and $30^{\circ}C$ groups. In natural light, root length and diameter were shorter and thinner when the temperature was higher, and growth was highly suppressed at $30^{\circ}C$. With regards to macroelements, the contents of Na, Mg, and P increased as temperature increased, regardless of the plant part; however, no constant tendency was observed in K and Ca according to temperature. The contents of Mg and Ca (from highest to lowest) were in the order leaf>stem>root, whereas the contents of Na, P, and K were in the order stem>leaf>root. Contents of general components varied according to temperature, and were highest at $30^{\circ}C$. While the plant height was increased under the constant $25^{\circ}C$+DIF (Difference between day and night temperature) condition, growth was suppressed in the -DIF group, in which the night temperature was higher than the day temperature, which suggests that a change in night temperature is one of the factors that affects the growth of C. lanceolata. As in the growth of the above-ground parts, fresh weight of the root was high in the constant $25^{\circ}C$ group and +DIF group. Notably, it was more than 2.5 times the fresh weights in the constant $15^{\circ}C$ group, constant $20^{\circ}C$ group, and -15 DIF group.

미나리즙이 고지방식이를 급여한 흰쥐의 혈청지질구성에 미치는 영향 (Effect of Oenanthe javanica Sap on Plasma Lipid Composition in Rats with High-Fat Diet)

  • 미나리즙 투여가 고지방식이를 급여한 흰쥐의 혈장과 지질과산화에 미치는 영향에 대하여 검토하였다. Sprague-Dawely계의 횐쥐를 I(정상군)과 II(대조군:고지방식이급여군), 그리고 미나리즙의 처리수준에 따라 III(고지방식이급여군+미나리즙(1.0 ml) 과 IV(고지방식이급여군+미나리즙(1.5 ml)의 4군으로 나누어 7주간 사육하였다. 실험종료일의 체중은 고지방식이 급여로 모두가 정상군보다 높은 수치를 보였다(p>0.05). 그러나 대조군과 미나리즙군 간에는 미나리즙군이 다소 감소하는 경향이었으나 유의적인 차이는 없었다(p>0.05). 식이섭취량 및 효율은 각 처리군간에 유의한 차이를 나타내지 않았다 (p>0.05). Total-cholesterol도 미나리즙 투여에 의해 감소하는 경향을 나타내었으나 유의적인 차이는 없었다(p>0.05), HDL-cholesterol은 미나리즙(1.0 ml) 급여군이 대조군에 비하여 감소하였다. GPT는 처리군간에 유의차가 없었으나 GOT활성은 처리군이 대조군에 비해 감소하는 경향을 나타내었다. 항산화효소인 glutathione peroxidase의 활성은 처리군 간에 유의한 차이는 없었다(p>0.05). Lipid peroxide는 고지방식이 급여군에서 증가하였으나(p>0.05), 미나리즙의 투여는 lipid peroxide를 감소시키지 못하였다.

Changes of Plasma and Hepatic Lipids, Hydroxy-Methyl-Glutaryl CoA Reductase Activity and Acyl-CoA : Cholesterol Acyltransferase Activity by Supplementation of Hot Water Extracts from Rosa rugosa, Crataegus pinnatifida and Polygonum cuspidatum in High-Cholesterol Fed Rats

  • Lipid lowering properties from three plant water extracts, Rosa rugosa, Crataegus pinnatifida and Polygonum cuspidatum, were tested by supplementing a 1% high-cholesterol diet with them in rats. Plasma triglyceride levels in Rosa fugosa, Crataegus pinnatifida and Polygonum cuspidatum groups were significantly lower compared to that of the control. by 29% , 24% and 47% respectively. hepatic trigylceride levels in Rosa rugosa and Crataegus pinnatifida groups were significantly lower compared to the control by 11% and 15% respectively. Hepatic HMG-CoA reductase activity in Rosa rugosa group was significantly greater compared to the control by 406%. Hepatic ACAT activity was significantly lower in Polygonum cuspidatum group compared to the control by 28%. by multiple regression results, only plasma cholesterol was associated significantly (p<0.05) with liver HMG-CoA reductase activity. Plasma cholesterol explained 12% of thevariance of the liver HMG-CoA redctase activity. In conclusion, we have showen that hot water extracts from Rosa rugosa, Crataegus pinnatifida and Polygonum cuspidatum lowered plasma triglycerides in rats fed on a high-cholesterol diet. Data suggests that these extracts could potentially prevent or treat hypertriglyceridemia induced by a high fat diet and fatty liver.

국내 담수어류 45종의 먹이원 분석(문헌자료 분석을 중심으로) (Analysis of Food Resources of 45 Fish Species in Freshwater Ecosystems of South Korea (Based on Literature Data Analysis))

  • 국내 담수 어류 12과, 28속, 45종의 먹이원을 문헌 조사한 결과, 동물류 먹이원은 16문, 24강, 54목, 126과, 212속으로 조사되었으며 식물류 먹이원은 10문, 18강, 42목, 63과, 82속으로 나타났다. 동물류 먹이원 중 가장 많은 어류가 섭식한 먹이원은 분류군에 따라 절지동물문, 곤충강, 파리목, 깔따구과로 조사되었으며, 식물류 먹이원은 돌말문, 윷돌말강, 반달돌말목, 반달돌말과로 조사되었다. SOM을 이용하여 45종 어류의 유형화 결과, 어류를 주로 포식하는 어류와 저서무척추동물을 포식하는 어류, 동물플랑크톤을 섭식하는 어류, 식물플랑크톤을 섭식하는 어류, 잡식성 어류 5가지 유형으로 나타났다. 네트워크 분석의 허브 점수가 높은 상위 5종의 어류는 블루길, 몰개, 수수미꾸리, 남방종개, 새코미꾸리 순으로 나타났으며 먹이원 중 허브 점수가 높은 상위 5 종류의 먹이원은 파리목, 하루살이목, 이지목, 김발돌말목, 쪽배돌말목 순으로 조사되었다.

Framingham Risk Score에 의한 한국 성인 남성의 영양소 및 식품 섭취의 다양성 평가 (Evaluation of Nutrient Intake and Food Variety in Korean Male Adults according to Framingham Risk Score)

  • The purpose of this study was to evaluate dietary intake according to the risk of coronary heart disease (less than 10% = low-risk group; 10~20% = middle-risk group) based on Framingham risk score (FRS), on 122 male adult subjects. The body weight and body mass index were not significantly different between the groups, while height of the low-risk group was shown to be significantly high compared to that of the middle-risk group. The daily energy intake was shown to be significantly high in the low-risk group with 1,910.88 kcal, compared to 1,606.63 kcal of the middle-risk group. As a result of analyzing nutrient intake per 1,000 kcal of energy, while the low-risk group had significantly high intake of animal protein, fat, and animal fat compared to the middle-risk group, the intake of plant protein, carbohydrate, and plant iron was found to be significantly low. The daily food intake was shown to be significantly high in the low-risk group (1,445.16 g), compared to the middle-risk group (1,075.12 g). The low-risk group was found to have significantly high intake of sugars, eggs, and beverages compared to the middle-risk group, while mushrooms intake was significantly high in the middle-risk group. Dietary variety score (DVS) was significantly high in the low-risk group with 26.42, compared to 22.66 of the middle-risk group. Dietary diversity score (DDS) was indicated to be significantly high in the low-risk group with 3.70, compared to 3.27 of the middle-risk group. The low-risk group was indicated to have significantly high score in DDS of dairy products and fruit group, compared to the middle-risk group. In the correlation between diversity index of food intake (DVS and DDS) and FRS, DDS was shown to have significantly negatively correlation with FRS after adjusting for confounding factors. To sum up these results, the adult males with low-risk of coronary heart disease had more various consumptions of fruits and milk, compared to the subjects with the middle-risk. The proportion of consuming major food groups such as cereals, meat group, milk, fruits, and vegetables more than a fixed quantity was indicated to be high. Accordingly, dietary habit for intake of various food seems to be necessary, to prevent coronary heart disease.

화학적 변성 및 생감자 전분이 고지방식이로부터 유도된 비만 흰쥐의 지질대사 및 항산화계에 미치는 영향 (The Effect of Dietary Modified Potato Starch By Chemically Denatured Treatment and Potato Starch on the Weight Loss, Lipid Metabolism and Redox Antioxidant System in High Fat Diet-Induced Obese Rats)

  • AIN-93G 식이조성을 기본으로 corn starch를 potato starch로 100% 대치한 고지방식이를 42일간 rat에 급여하여 비만을 유도 후, GPS군, SPS군, EZ군, H40군으로 나누어 70일 급여한 후 지질대사에 미치는 영향을 살펴보았다. 그 결과, 체중증가량은 GPS, EZ, H40군에서 차이가 없었고 식이효율 또한 유의적으로 차이가 없었다. 혈중 혈당농도, TC, LDL-cholesterol 농도는 변성 전분 섭취 군(EZ, H40군)에서 높았고, atherogenic index는 H40이 다른 군에 비교해 높게 측정되어 H40군이 비만, 동맥경화에 미치는 영향이 크게 나타났다. 간과 신장 조직의 MDA 생성량은 H40군이 혈청 MDA 생성량이 다른 군에 비해 매우 높았고, 간과 신장 조직에서는 EZ, H40군이 낮았다. Glutathione 함량은 각 조직별로 경향이 달랐으나, GP-x활성은 SPS군에서 낮았다. 본 연구에서 전분을 달리하여 첨가한 고지방 식이가 비만한 식이에 미치는 영향을 살펴본 결과, 알감자 전분을 100% 대치하여 제조한 SPS군 및 효소 처리한 변성전분(EZ)을 100% 대치하여 급여한 군에서 비만억제 효과가 나타났다. 따라서 변성전분의 심혈관계 질환 및 혈중 지질 패턴 변화에 관련된 기전 연구가 지속적으로 필요하며, 생리활성이 규명된 후 다양한 가공식품 개발이 이루어져야 할 것으로 사료된다.

Genetic Diversity in the Coat Protein Genes of Prune dwarf virus Isolates from Sweet Cherry Growing in Turkey

  • Sweet cherry is an important fruit crop with increasing economical value in Turkey and the world. A number of viruses cause diseases and economical losses in sweet cherry. Prune dwarf virus (PDV), is one of the most common viruses of stone fruits including sweet cherry in the world. In this study, PDV was detected from 316 of 521 sweet cherry samples collected from 142 orchards in 10 districts of Isparta province of Turkey by double antibody sandwich-enzyme linked immunosorbent assay (DAS-ELISA). The presence of PDV in ELISA positive samples was confirmed in 37 isolates by reverse transcription- polymerase chain reaction (RT-PCR) method. A genomic region of 862 bp containing the coat protein (CP) gene of PDV was re-amplified from 21 selected isolates by RT-PCR. Amplified DNA fragments of these isolates were purified and sequenced for molecular characterization and determining genetic diversity of PDV. Sequence comparisons showed 84-99% to 81-100% sequence identity at nucleotide and amino acid level, respectively, of the CP genes of PDV isolates from Isparta and other parts of the world. Phylogenetic analyses of the CP genes of PDV isolates from different geographical origins and diverse hosts revealed that PDV isolates formed different phylogenetic groups. While isolates were not grouped solely based on their geographical origins or hosts, some association between phylogenetic groups and geographical origins or hosts were observed.

서울 지역 여대생의 식사 섭취 상태 및 식사의 질 평가 -과체중군과 정상군의 비교를 중심으로 - (Evaluation of Nutrient and Food Intake Status, and Dietary Quality - Focused on Comparison with Overweight and Normal Female University Students -)

  • The purpose of this study was to evaluate nutrient and food intake status and dietary, quality according to obesity based on nutrient and food group intake, in female university students. Surveys were conducted using questionnaires and 3 days, dietary record in 360 female university students residing in Seoul. The subjects were divided into two groups by percent of body fat: an overweight group(percent of body fat more than 23%, n=249) and normal group(percent of body fat below 23%, n=111). The overweight group had a larger proportion of subjects who binged compared to the normal group. There was no significant difference in energy intake between the two groups. The densities of plant calcium and dietary fiber in the overweight group were significantly lower than those of the normal group. However, animal fat density was significantly higher in overweight subjects. The Korean's dietary diversity score(KDDS) of the overweight group was 4.65, and that of the normal group was 4.67, indicating no significant difference. There was no significant difference of DQI-I between the overweight group(50.8) and normal group(51.1). However, the macronutrient ratio score was significantly lower in the overweight group. These results indicate that overweight female college students may have improper dietary habits, and have lower overall balance aspect macronutrient ratios.

Antioxidant Effects of Elsholtzia splendens Extract on DMBA-induced Oxidative Stress in Mice

  • The present study was conducted to investigate the effects of flowers ethanol extract of Elsholtzia splendens (ESE) on the antioxidant defence system in mice with 7,12-dimethylbenz(a)anthracene (DMBA)-induced oxidative stress. The ESE was pre-administered orally to 2 groups of mice at 10 and 50mg/kg body weight (BW) for 5 weeks. Subsequently, mice with pretreatment of ESE received DMBA intragastrically at a dose of 34 mg/kg BW twice a week for 2 weeks. In DMBA alone group, biomarkers of oxidative stress (TBARS value, carbonyl content, and serum 8-OH-dG) were significantly increased. Also, the antioxidant enzymes were down-regulated. ESE significantly restored the TBARS value and carbonyl content at both doses, while a decrease in the elevated serum 8-OH-dG content was observed only at the higher dose. The DMBA-induced decreases in catalase and superoxide dismutase (SOD) activities were restored to nearly control levels by ESE. Glutathione peroxidase (GSH-px) and glutathione reductase (GR) activities, as well as the reduced glutathione (GSH)/oxidized GSH (GSSG) ratio, were significantly affected by ESE at the higher dose. These results suggest that ESE possesses antioxidant activity, which plays a protective role against DMBA-induced oxidative stress.
