The Conformation of the Jindo (Canis familiaris), Korean Native Dog - the trunk and limb (진도개의 체형 - 몸통과 다리)

  • The trunk and limb of the Jindo, Korean native dog was measured in the present study. One hundred and ninety five Jindoes were randomly selected from the 1,985 dogs in the Jindo County in August 1998, which were screened by the judging committee of the County and registered by inserting electronic chips in the withers. They were about a year and half old, and were white and fawn dogs. A total of 18 parts of the trunk and limb were measured using calipers and tapes. The average height at the withers was 50.67 cm in dogs and 47.48 cm in bitches. The average body length was 52.99 cm in dogs and 50.33 cm in bitches. And the average height : body length ratio was 104.70 in dogs and 106.08 in bitches. No differences were observed between the Jindoes of the two coat colors in these measurements. The results of this study would be useful for compiling the official standard for the adult Jindo.

An Intersex with Os Clitoris and Ovotestes in Korean Native Jindo Dog (진도개에서 발생한 Os clitoris와 Ovotestes를 특징으로 하는 양성자 사례 연구)

  • Hermaphroditism occurs rarely in human and animal populations. Previously, in Korea, The hermaphrodite dog in American Cocker Spaniel dogs have been reported in six cases. However, in this case, we found a hermaphrodite dog in Korean native Jindo dog. The characteristics were its enlarged os clitoris that was shown a reddish fingerlike structure protruding from the vulva. The dog was diagnosed as hermaphroditism with ovotestes. To our knowledge, this report is the first case of hermaphroditism in Korean native dog.

Flow cytometry analysis of DNA ploidy of transmissible venereal tumors in the Jindo dogs (유식세포 분석법에 의한 진도개 전파성 성기육종의 DNA Ploidy 유형분석)

  • Transmissible venereal tumor(TVT) is a naturally occurring contagious neoplasm which can be transmitted by mechanical contact during mating in dogs and transplanted as intact viable cells to dogs and other members of canine family such as coyotes, jackals, wolves, and foxes. The incidence of this tumors tends to increase in Korean native Jindo dogs. This is probably due to the high density and unrestrained management system. With time, TVT reaches the maximum size and then tends to regress spontaneously unless individuals are immunologically compromised. It consists of different types of cells depending on the stage. In this study, 10 tumors were selected from Jindo dogs. These were histologically calssified into three stages; progressive, steady-state, and regressive. Mitotic figures were counted, and their histological appearance at each stage is compared with their DNA ploidy. Histologically, 5 tumor cases were calssed as the progressors, 3 cases as the steady-state tumors, and 2 cases as regressors. Progressors were composed of round cells with large nuclei containing conspicuous nucleoli and frequent mitotic figures. A few spindle-shaped cells and inflammatory cells including mainly lymphocytes, a few neutrophils and macrophages were also seen. In the steady-state tumors, there was an increased number of spindle shaped cells and mitotic figures were rare. Six tumors were diploid and four were aneuploid with the variation coefficient of 7.02. Two of five progressive tumors were aneuploid. Two of three steady-state tumors were aneuploid while both tumors at the regressive stage were diploid. Progressive and steady-state tumors had a much larger S/G2M fraction and a higher mitotic index than regressive tumors. Two tumors which persisted for more than one year were aneuploid. These results suggest that the progressive and steady-state tumors had more active cell division than the regressive neoplasms.

Development of Artificial Vessels with Autologous Bone Marrow Cells and Polymers (자기 골수세포와 고분자 폴리머를 이용한 인공 혈관의 개발)

  • Bakcground: To treat anastomosis site stenosis and occlusion of the artificial vessels used in vascular surgery, tissue-engineered artificial vessels using autologous cells have been constructed. We developed artificial vessels using a polymer scaffold and autologous bone marrow cells and performed an in vivo evaluation. Material and Method: We manufactured a vascular scaffold using biodegradable PLCL (poly lactide-co-${\varepsilon}$-caprolactone) and PGA (poly glycolic acid) fibers. Then we seeded autologous bone marrow cells onto the scaffold. After implantation of the artificial vessel into the abdominal aorta, we performed an angiography 3 weeks after surgery. After the dogs were euthanized we retrieved the artificial vessels and performed histological analysis. Result: Among the six dogs, 2 dogs died of massive bleeding due to a crack in the vascular scaffold 10 days after the operation. The remaining four dogs lived for 3 weeks after the operation. In these dogs. the angiography revealed no stenosis or occlusion at 3 weeks after the operation. Gross examination revealed small thrombi on the inner surface of the vessels and the histological analysis showed three layers of vessel structure similar to the native vessel. Immunohistochemical analysis demonstrated regeneration of the endothelial and smooth muscle cell layers. Conclusion: A tissue engineered vascular graft was manufactured using a polymer scaffold and autologous bone marrow cells that had a structure similar to that of the native artery. Further research is needed to determine how to accommodate the aortic pressure.

Physical Characteristics and Microsatellite Polymorphisms in Miryang Native Dogs (밀양지방 토종개의 형태학적 특징 및 유전적 다양성 연구)

  • This study was carried out to investigate the physical characteristics (height, body lenght, chest depth, head type, ear type, body color, eye type and tail type) and genetic diversity using 15 microsatellite DNA markers (PEZ 1, 5, 8, 10, 11, 12, 13, 15, 16, 17, 20, 21, FHC 2010, FHC 2054 and FHC 2079) in 44 random Miryang native dogs(6 months${\sim}$12 years old). The height, body lenght, and chest depth of Miryang native dogs were 43-55 cm(mean 49.5 cm), 45-60 cm(mean 54.3 cm), and 50-64 cm(mean 57.9 cm), respectively. Miryang native dog was medium sized. The head and eye type were reverse-triangle(100%), triangle (90.9%) and newborn moon(9.1%), respectively. Most of body color had white coat color(93.2%), light pink tongue color(100%), light black anal color(90,9%) and pink claw color(100%). The ear type showed erect ear(100%), and half-curled(56.8%), upward(34.1%), curled(9.1%) in tail type, respectively. Number of alleles observed at a single locus ranged from 2 (PEZ 21 and FHC 2010) to 14 (PEZ 13), with average number of alleles per locus of 6.13. The expected heterozygosities of 15 microsatellite loci were estimated based on gene frequencies. The highest expected heterozygosity, 0.863 was estimated in PEZ 13 locus and the lowest, 0.455 in PEZ 21 and FHC 2010 locus. And the mean expected heterozygosity of 15 microsatellite markers was calculated as 0.635. Polymorphic information content (PIC) values were ranged from 0.348 (PEZ 21 and FHC 2010) to 0.837 (PEZ 13), and the mean PIC value was calculated as 0.570. Of the 15 markers, PEZ 10, PEZ 13, PEZ 17 and FHC 2054 loci have relatively high PIC value (> 0.7) in Miryang native dog. In order to determine the efficieney of parentage control, exclusion probabilities (PE) were calculated for each allele. The highest PE 1 and PE 2 in PEZ 13 locus was caculated to 0.548 and 0.710, respectively. And the total exclusion power in PE 1 and PE 2 was calculated to 0.9895 and 0.9996, respectively. These results can give basic information for perservation and research in Miryang native dog, and phylogenetic relationships of the Korean native dog and Asian dog breeds.

Genetic structure analysis of domestic companion dogs using high-density SNP chip

  • Background: As the number of households raising companion dogs increases, the pet genetic analysis market also continues to grow. However, most studies have focused on specific purposes or native breeds. This study aimed to collect genomic data through single nucleotide polymorphism (SNP) chip analysis of companion dogs in South Korea and perform genetic diversity analysis and SNP annotation. Methods: We collected samples from 95 dogs belonging to 26 breeds, including mixed breeds, in South Korea. The SNP genotypes were obtained for each sample using an AxiomTM Canine HD Array. Quality control (QC) was performed to enhance the accuracy of the analysis. A genetic diversity analysis was performed for each SNP. Results: QC initially selected SNPs, and after excluding non-diverse ones, 621,672 SNPs were identified. Genetic diversity analysis revealed minor allele frequencies, polymorphism information content, expected heterozygosity, and observed heterozygosity values of 0.220, 0.244, 0.301, and 0.261, respectively. The SNP annotation indicated that most variations had an uncertain or minimal impact on gene function. However, approximately 16,000 non-synonymous SNPs (nsSNPs) have been found to significantly alter gene function or affect exons by changing translated amino acids. Conclusions: This study obtained data on SNP genetic diversity and functional SNPs in companion dogs raised in South Korea. The results suggest that establishing an SNP set for individual identification could enable a gene-based registration system. Furthermore, identifying and researching nsSNPs related to behavior and diseases could improve dog care and prevent abandonment.

Description of Clinicopathologic Changes during the Development and Clinical Resolution of Experimentally induced Canine Nephrotic Syndrome (실험적으로 유발한 개 신증후군의 진행 및 치유과정에서의 임상병리학적 변화)

  • The purpose of this study is to evaluate urine protein-to-creatinine ratio as a parameter for early detection of nephrotic syndrome and as a parameter for monitoring effectiveness in early course of treatment. Nine healthy dogs were sensitized by intravenous injection with 1 $\mu$g of endotoxin and 5 mg of native bovine serum albumin. After 1 week, 120 mg of cationized bovine serum albumin was injected intravenously 5 times a week. Among nine dogs, five dogs were confirmed as having developed glomerulonephritis and nephrotic syndrome by increase of urine protein-to-creatinine ratio(>1.0), hypoalbuminemia (<1.5 g/dl), hypercholesterolemia (> 240 mg/dl) and azotemia (BUN>40 mg/dl). During the induction of glomerulonephritis and the progression to nephrotic syndrome, the increase of urine protein-to-creatinine ratio was firstly detected. 1 to 4 weeks later, hypoalbuminemia, hypercholesterolemia, and azotemia were detected. Prednisolone (2.2 mg/kg, bid) was administered orally to the dogs with induced nephrotic syndrome. In early stage of treatment, the increase of serum albumin and decrease of serum cholesterol were detected. 1 to 4 weeks later, decrease of urine protein-to-creatinine ratio was detected. It was concluded that urine protein-to-creatinine ratio is a useful parameter for early detection of nephrotic syndrome, and serum albumin and cholesterol are useful parameters for the monitoring in early course of treatment in nephrotic syndrome.

In Vivo Experiment of Tissue-Engineered Artificial Vessel (조직공학적 인조혈관의 생체 내 이식 실험)

  • The number of patients with coronary artery disease and peripheral vascular disease are increasing, and the need of small diameter vessel is also increasing. We developed small diameter artificial vessel and experimented in vivo. We got allogenic valve from mongrel dogs, and removed all cells from the allogenic valve. Then, we seeded autologous bone marrow cells onto the decellularized scaffold. After implantation of artificial vessel into the canine carotid artery, we performed angiography regularly. In case of vessel occlusion or at 8 weeks after operation, we euthanized dogs, and retrieved the implanted artificial vessels. Control vessels were all occluded except one (which developed aneurysmal dilatation). But autologous cell seeded vascular graft were patent by 4 weeks in one, by 6 in one and by 8 weeks in two. Histologic examination of patent vessel revealed similar structure to native artery. Tissue-engineered vascular graft manufactured with decellularized allogenic matrix and autologous bone marrow cells showed that tissue engineered graft had similar structure to native artery.

Experimental Studies on the Acupuncture Anesthesia in Dogs (개의 전침마취(電針痲醉)의 통전량(通電量)에 관한 실험적연구(實驗的硏究))

  • In the present study, the optimal electric current and induction time of the acupuncture anesthesia were determined and the clinical signs during and after the acupuncture anesthesia were observed in 90 Korean native dogs. The results were summarized as fellows. 1. Optimal electric current was 11-13 Hz at 1.0 volt initially with gradual increase up to 22-26 Hz at 5.0-6.0 volts after 5 minutes. 2. The induction time was 7-20 minutes in all cased; 55.8% out of them showed the induction time of 7-10 minutes. 3. No differences were found in the analgesic effect and clinical signs between ages and between body weights. There were individual differences in the induction time and the optimal electric current. The respiratory and pulse rates were lower at 5 minutes after the removal of the needles than 5 minutes before attachment of needles to the electrodes.

Toxicity of ivermectin in Jindo-dogs 1. Clinical and hematological observation (진돗개에서 ivermectin의 독성 I. 임상증상과 혈액학적 변화 관찰)

  • Ivermectin is a synthetic derivative of the naturally occurring avermectin $B_{1a}$ (22, 23-dihydroavermectin $B_{1a}$) and $B_{1b}$ (22, 23-dihydroavermectin $B_{1b}$), It is widely used as antiparasitic and pesticidal agents because of its remarkably potent and broad spectrum of antiparasitic activity. Although the drug has shown excellent anthelmintic effects, development of toxicosis in some animals such as the Collie species of dog is well documented. However, no studies have been reported on the toxic effects of the drug in Korean native animals such as the Jindo dog. The toxic effect of ivermectin was evaluated in 25 Jindo dogs divided into five groups which were orally administered with ivermectin at dosage levels of $200{\mu}g/kg$, $300{\mu}g/kg$, $600{\mu}g/kg$ and $2,500{\mu}g/kg$ of body weight, respectively. Toxic signs were not observed in the groups receiving $200{\mu}g/kg$ and $300{\mu}g/kg$ B.W. ivermectin. One dog developed mild clinical signs of toxicosis in the group receiving $600{\mu}g/kg$ dosage of ivermectin. In the group with $2,500{\mu}g/kg$ dosage, all dogs developed mild (salivation, drooling, vomiting, mydriasis, and/or confusion) and/or moderate (ataxia and tremors) clinical signs of toxicosis. Hematologic changes were not observed in the groups receiving $200{\mu}g/kg$, $300{\mu}g/kg$ and $600{\mu}g/kg$ B.W. ivermectin. In the groups receiving $2,500{\mu}g/kg$ B.W., total erythrocyte counts, total and differential leukocyte counts and hemoglobin levels were not affected by drug. Aspartate aminotransferase levels were increased after administration of ivermectin, while serum cholesterol and blood glucose levels were decreased.
