홍경천의 생육특성검정 및 재배지역 탐색 (Growth Characteristics and Search for Eligible Cultivation Area of Rhodiola sachalinensis A. Boriss.)

  • 중국의 약용작물로 널리 알려진 홍경천의 충북도내 재배 가능성을 알아보기 위하여 수행 한 시험 결과를 요약하면 다음과 같다. 지상부 생육상황은 최고조에 달했던 7월의 경우 초장은 8.1­15.8cm, 줄기수는 3.1­3.6개였으며, 여름철엔 하고현상이 심하여 지상부의 고사율이 76­94%로 매우 높았다. 지하부 생육상황의 경우 근장은 단양지역이 6.8cm로 가장 컸으나 굵기는 청원이 10.0mm로 가장 굵었으며 , 생체중 또한 3.5g으로 제천, 단양 대비 3.5배 정도 높았다. 한편 뿌리 부패율이 62.3­86.5%로 매우 높아 당분간 농가재배는 어려울 것으로 생각된다. 개화기는 6월 중$.$하순으로 개화기간은 12­13일 정도 되었고, 개화율은 60­77%정도 되었으며, 복산형화서로 꽃색은 자색, 황녹색, 회자색, 자홍색 등 다양하였다. 종자는 미세종자로 성숙기는 8월 중순­9월 하순이었고, 색깔은 황갈색이었으며 종자의 길이는 1.5­2.0mm, 너 비 는 0 1­0.5mm, 천립중은 0.13­0.15g으로 조사되었다.

큰까치수영의 항산화 및 안지오텐신 전환 효소 저해 활성 (Antioxidant and Inhibitory Activities on Angiotensin Converting Enzyme in Lysimachia clethroides Duby)

  • 생리활성 보유 식물자원 발굴 및 이용을 위해 큰까치수영 지상부에 대한 항산화 및 항고혈압 활성을 검정하였으며 페놀화합물의 함량은 메탄올 추출물이 2.62%, 물 추출물이 0.23%였고 메탄올 추출물에서 얻어진 용매별 분획물 중에서는 에틸차세테이트 분획이 37.76%로 가장 함량이 높았다. Linoleic acid 자동산화에 대한 저해효과는 메탄올 추출물이 반응 6일째에 83%이상의 저해율을 보여 ${\alpha}-tocopherol$의 -2%보다 탁월하게 우수였으며 각 분획물 중에서는 핵산 및 에텔 분획이 매우 효과적으로 자동산화를 저해하였다. Superoxide anion 라디칼에 대헤서는 큰까치수영의 메탄올 추출물과 물 추출물이 $5{\sim}200\;{\mu}g/ml$의 농도에서 $86{\sim}109%$$96{\sim}122%$로 항산화제인 ascorbic acid의 $-4{\sim}69%$보다 월등하게 우수한 소거능을 보였으며 DPPH 라디칼에 대해서는 에틸아세테이트분획이 $26.8\;{\mu}g/ml$$RC_{50}$로 좋은 소거 효과를 나타내었다. ACE에 대한 저해활성은 $4,000\;{\mu}g/ml$의 농도에서 물 추출물(33%)보다는 메탄올 추출물(71%)이 효과적이었으며 분획물 중에서는 n-hexane fraction(133%), ether fraction(100%) 및 ethylacetate fraction(88%) 분획이 매우 효과적인 저해활성을 나타내었다.

밭 작물에서 분리한 균근균의 접종 효과 비교 (Comparative Study of Infection Effects with AMF (Arbuscular-Mycorrhizal Fungi) Isolated from Upland Plants)

  • AMF의 특징 및 접종 효과를 확인하기 위해 경기 지역에서 토양 시료를 채집하고 AMF 포자를 분리하여 포자 및 감염 구조의 특징을 관찰하였으며 고추에 접종 후 생육 비교 조사를 실시하였다. 각 토양 시료에서 분리된 AMF 포자들은 대체적으로 원형, 타원형의 형태였으며 다양한 포자 색깔 중에서는 연노란색의 포자가 많은 분포를 나타냈다. 포자의 크기는 평균 $48.4{\mu}m$였으며 낭상체의 크기는 평균 $99.7{\mu}m$였다. 옥수수에 접종 배양한 AMF의 뿌리 감염률은 13.3~83.3%의 범위를 나타냈으며 옥수수를 기주로 하는 토양 시료를 접종 후 배양했을 때 평균 75% 이상의 높은 감염률을 나타냈다. AMF를 고추에 처리했을 때 무처리구에 비해 생체중과 건물중, 초장은 다소 증가했지만 유의적인 차이를 나타내지 않았다. 하지만 3회에 걸친 수량 조사에서는 각각의 수확시기와 전체 평균 누적 수확량에서 모두 2배 이상 차이를 보여 AMF를 고추에 처리했을 때 수량성면에서는 뚜렷한 효과를 나타냈다. 이러한 결과를 바탕으로 밭작물을 중심으로 분리된 AMF 포자를 다른 작물에 적용했을 때 효과의 가능성을 기대할 수 있을 것으로 판단된다. 또한 AMF가 식물 생육을 촉진할 수 있는 친환경 균근균 비료 자원으로서 다양한 작물과 재배지에서 적용할 수 있는 AMF의 접종원 대량 생산 시스템 및 제품 개발을 위한 기초 자료로의 활용이 가능할 것으로 사료된다.

Lomens-P0 (mixed extracts of Hordeum vulgare and Chrysanthemum zawadskii) regulate the expression of factors affecting premenstrual syndrome symptoms

  • BACKGROUND/OBJECTIVES: Premenstrual syndrome (PMS) is a disorder characterized by repeated emotional, behavioral, and physical symptoms before menstruation, and the exact cause and mechanism are uncertain. Hyperprolactinemia interferes with the normal production of estrogen and progesterone, leading to PMS symptoms. Thus, we judged that the inhibition of prolactin hypersecretion could mitigate PMS symptoms. MATERIALS/METHODS: Hordeum vulgare L. extract (HVE), Chrysanthemum zawadskii var. latilobum extract (CZE), and Lomens-P0 the mixture of these extracts were tested in subsequent experiments. The effect of extracts on prolactin secretion at the in vitro level was measured in GH3 cells. Nitric oxide and pro-inflammatory mediator expression were measured in RAW 264.7 cells to confirm the anti-inflammatory effect. Also, the hyperprolactinemic Institute for Cancer Research (ICR) mice model was used to measure extract effects on prolactin and hormone secretion and uterine inflammation. RESULTS: Anti-inflammatory effects of and prolactin secretion suppress by HVE and CZE were confirmed through in vitro experiments (P < 0.05). Treatment with Lomens-P0 inhibited prolactin secretion (P < 0.05) and restored normal sex hormone secretion in the hyperprolactinemia mice model. In addition, extracts significantly inhibited the expression of pro-inflammatory biomarkers, including interleukin-1𝛽, and -6, tumor necrosis factor-𝛼, inducible nitric oxide synthase, and cyclooxygenase-2 (P < 0.01). We used high-performance liquid chromatography analyses to identify tricin and chlorogenic acid as the respective components of HVE and CZE that inhibit prolactin secretion. The Lomens-P0, which includes tricin and chlorogenic acid, is expected to be effective in improving PMS symptoms in the human body. CONCLUSIONS: The Lomens-P0 suppressed the prolactin secretion in hyperprolactinemia mice, normalized the sex hormone imbalance, and significantly suppressed the expression of inflammatory markers in uterine tissue. This study suggests that Lomens-P0 may have the potential to prevent or remedy materials to PMS symptoms.

Biosynthesis of rare 20(R)-protopanaxadiol/protopanaxatriol type ginsenosides through Escherichia coli engineered with uridine diphosphate glycosyltransferase genes

  • Background: Ginsenosides are known as the principal pharmacological active constituents in Panax medicinal plants such as Asian ginseng, American ginseng, and Notoginseng. Some ginsenosides, especially the 20(R) isomers, are found in trace amounts in natural sources and are difficult to chemically synthesize. The present study provides an approach to produce such trace ginsenosides applying biotransformation through Escherichia coli modified with relevant genes. Methods: Seven uridine diphosphate glycosyltransferase (UGT) genes originating from Panax notoginseng, Medicago sativa, and Bacillus subtilis were synthesized or cloned and constructed into pETM6, an ePathBrick vector, which were then introduced into E. coli BL21star (DE3) separately. 20(R)-Protopanaxadiol (PPD), 20(R)-protopanaxatriol (PPT), and 20(R)-type ginsenosides were used as substrates for biotransformation with recombinant E. coli modified with those UGT genes. Results: E. coli engineered with $GT95^{syn}$ selectively transfers a glucose moiety to the C20 hydroxyl of 20(R)-PPD and 20(R)-PPT to produce 20(R)-CK and 20(R)-F1, respectively. GTK1- and GTC1-modified E. coli glycosylated the C3-OH of 20(R)-PPD to form 20(R)-Rh2. Moreover, E. coli containing $p2GT95^{syn}K1$, a recreated two-step glycosylation pathway via the ePathBrich, implemented the successive glycosylation at C20-OH and C3-OH of 20(R)-PPD and yielded 20(R)-F2 in the biotransformation broth. Conclusion: This study demonstrates that rare 20(R)-ginsenosides can be produced through E. coli engineered with UTG genes.

Analysis of the Genome Sequence of Strain GiC-126 of Gloeostereum incarnatum with Genetic Linkage Map

  • Gloeostereum incarnatum has edible and medicinal value and was first cultivated and domesticated in China. We sequenced the G. incarnatum monokaryotic strain GiC-126 on an Illumina HiSeq X Ten system and obtained a 34.52-Mb genome assembly sequence that encoded 16,895 predicted genes. We combined the GiC-126 genome with the published genome of G. incarnatum strain CCMJ2665 to construct a genetic linkage map (GiC-126 genome) that had 10 linkage groups (LGs), and the 15 assembly sequences of CCMJ2665 were integrated into 8 LGs. We identified 1912 simple sequence repeat (SSR) loci and detected 700 genes containing 768 SSRs in the genome; 65 and 100 of them were annotated with gene ontology (GO) terms and KEGG pathways, respectively. Carbohydrate-active enzymes (CAZymes) were identified in 20 fungal genomes and annotated; among them, 144 CAZymes were annotated in the GiC-126 genome. The A mating-type locus (MAT-A) of G. incarnatum was located on scaffold885 at 38.9 cM of LG1 and was flanked by two homeodomain (HD1) genes, mip and beta-fg. Fourteen segregation distortion markers were detected in the genetic linkage map, all of which were skewed toward the parent GiC-126. They formed three segregation distortion regions (SDR1-SDR3), and 22 predictive genes were found in scaffold1920 where three segregation distortion markers were located in SDR1. In this study, we corrected and updated the genomic information of G. incarnatum. Our results will provide a theoretical basis for fine gene mapping, functional gene cloning, and genetic breeding the follow-up of G. incarnatum.

Comparative transcriptome and metabolome analyses of four Panax species explore the dynamics of metabolite biosynthesis

  • Background: The genus Panax in the Araliaceae family has been used as traditional medicinal plants worldwide and is known to biosynthesize ginsenosides and phytosterols. However, genetic variation between Panax species has influenced their biosynthetic pathways is not fully understood. Methods: Simultaneous analysis of transcriptomes and metabolomes obtained from adventitious roots of two tetraploid species (Panax ginseng and P. quinquefolius) and two diploid species (P. notoginseng and P. vietnamensis) revealed the diversity of their metabolites and related gene expression profiles. Results: The transcriptome analysis showed that 2,3-OXIDOSQUALENE CYCLASEs (OSCs) involved in phytosterol biosynthesis are upregulated in the diploid species, while the expression of OSCs contributing to ginsenoside biosynthesis is higher in the tetraploid species. In agreement with these results, the contents of dammarenediol-type ginsenosides were higher in the tetraploid species relative to the diploid species. Conclusion: These results suggest that a whole-genome duplication event has influenced the triterpene biosynthesis pathway in tetraploid Panax species during their evolution or ecological adaptation. This study provides a basis for further efforts to explore the genetic variation of the Panax genus.

Molecular Characterization of an Isolate of Bean Common Mosaic Virus First Identified in Gardenia Using Metatranscriptome and Small RNA Sequencing

  • Gardenia (Gardenia jasminoides) is a popular and economically vital plant known for its ornamental and medicinal properties. Despite its widespread cultivation, there has been no documentation of plant viruses on gardenia yet. In the present study, gardenia leaves exhibiting symptoms of plant viral diseases were sampled and sequenced by both metatranscriptome and small RNA sequencing. As a consequence, bean common mosaic virus (BCMV) was identified in gardenia for the first time and named BCMV-gardenia. The full genome sequence of BCMV-gardenia is 10,054 nucleotides (nt) in length (excluding the poly (A) at the 3' termini), encoding a large polyprotein of 3,222 amino acids. Sequence analysis showed that the N-termini of the polyprotein encoded by BCMV-gardenia is less conserved when compared to other BCMV isolates, whereas the C-termini is the most conserved. Maximum likelihood phylogenetic analysis showed that BCMVgardenia was clustered closely with other BCMV isolates identified outside the leguminous plants. Our results indicated that the majority of BCMV-gardenia virus-derived small interfering RNAs (vsiRNAs) were 21 nt and 22 nt, with 21 nt being more abundant. The first nucleotide at the 5' termini of vsiRNAs derived from BCMV-gardenia preferred U and A. The ratio of vsiRNAs derived from sense (51.1%) and antisense (48.9%) strands is approaching, and the distribution of vsiRNAs along the viral genome is generally even, with some hot spots forming in local regions. Our findings could provide new insights into the diversity, evolution, and host expansion of BCMV and contribute to the prevention and treatment of this virus.

면역억제 마우스 모델에서 왕느릅나무 유래 DEU-7의 면역기능 증강 (DEU-7 Derived from Ulmus macrocarpa Improved Immune Functions in Cyclophosphamide-treated Mice)

  • 고량강(Alpinia officinarum), 산초(Zanthoxylum schinifolium), 황금(Scutellaria baicalensis Georgi), 왕느릅나무(Ulmus macrocarpa Hance), 네 종류의 식물성 한방 약재의 면역증강효능을 cyclophosphamide를 처리한 동물모델을 이용하여 조사하였다. 실험은 cyclophosphamide로 면역억제를 유도한 후 네 종류의 한방 약재를 식이하여 cyclophosphamide에 의해 억제된 면역인자의 회복 여부를 조사하였다. 네 종류의 한방 약재를 동일한 방법으로 열수 추출한 후 동일한 농도로 마우스에 처리하였다. 이 중 왕느릅나무 열수 추출물(DEU-7)이 cyclophosphamide의 영향으로 면역 억제된 마우스에서 면역인자 혈중 농도를 정상치 준하는 또는 가깝게 유지하는 것으로 나타내었다. DEU-7에 의해 비장의 무게와 비장세포수는 증가하였으나 간과 흉선과 같은 다른 장기에는 통계적으로 유의한 변화가 없었다. Ex vivo 조건에서 DEU-7은 비장세포의 사멸을 지연시키는 것으로 나타났다. 중요한 면역인자인 IL-2와 IL-4 cytokine은 면역억제로 농도가 감소되었으나 DEU-7이 약 2배와 3배 향상시키는 것으로 나타났으며, 이는 정상치의 약 1/2 수준이었다. IgM과 IgG의 농도는 cyclophosphamide 처리로 정상치의 약 1/2로 떨어졌으나, DEU-7에 의해서 정상치와 동일한 농도로 증가하였다. IL-2와 IL-4 결과에서 DEU-7은 T 림프구에 영향을 줄 수 있다고 생각되며, 또한 IgM과 IgG의 결과로서 B 림프구에 영향을 줄 수 있다고 생각된다. 선천성 면역에 중요한 면역인자인 TNF-α, IL-12과 IFN-γ 역시 cyclophosphamide에 의해 농도가 감소되었으나 DEU-7에 의해 정상치에 가깝게 회복되었다. 따라서, DEU-7은 면역 억제 또는 감소된 상태를 정상 상태로 회복 또는 유지하는 기능이 있는 것으로 생각된다.

신품종(新品種)을 포함한 한국산(韓國産) Acanthopanax속(屬)의 분류(分類)(I) (A New Forma of Acanthopanax Species(I))

  • 1. The Korean Acanthopanax genus includes 12 kinds consisting of 9 species and 3 forma. 2. The Korean Oga-pi which is on market sale has been used as bark for the medicinal purpose, and Oga-pi shall use Radicis Cortex. That is why it is basic rule that herbalogy shall use Radicis Cortex. 3. The origin of Oga-pi on sale is Acanthopanax sessiliflorum forma chungbunensis C.S. Yook. 4. $C_{HUNG}\;and\;N_AKAI'S}$ report on A. koreanum told us that there are brown hair on the mid-leaf junction, but in addition to it, our investigation was resulted in the fact that there are thorn along mid-rib sometimes. 5. 2 kinds of new forma are similar to A. sessiliflorum, but are different in the view-point of chemotaxonomy, compared with A. sessiliflorum. In its morphology, we can find some difference between 2 kinds of new forma and A. sessiliflorum. Our effort of examination on documents tell us that the all plants growing in the central part of our country is A. sessiliflorum forma chungbunensis C.S. Yook. The one which has thorn on both side among the plants collected in Mt. Dukyu, is called A. sessiliflorum forma nambunensis C.S. Yook. 6. A. sessiliflorum is growing in the southern part in Korea, and most Chungbu Oga-pi A. sessiliflorum forma chungbunensis in the central part of our country. For the convenience of our study, the key of Korean Acanthopanax plant is classified into, I-IV, as shown on the following items: I. No hair on both side of leaf A.
