The difference of ginsenosides content according to placement of ginseng planting (line) under shading net in 5-year-old ginseng roots were examined. The total saponin ($Rb_1$, $Rb_2$, Rc, Rd, Re, and $Rg_1$) contents were 15.01 mg/g and 21.79 mg/g in the main roots, 35.93 mg/g and 43.32 mg/g in the lateral roots, 87.85 mg/g and 105.51 mg/g in the fine roots for the front $1st{\sim}2nd$ lines in Yunpoong and Landrace variety (purple-stem variant), respectively. In the middle $3rd{\sim}5th$ lines the total saponin contents were 18.73 mg/g and 23.19 mg/g in the main roots, 44.92 mg/g and 43.50 mg/g in the lateral roots, 92.97 mg/g and 110.70 mg/g in the fine roots in Yunpoong and Landrace variety, respectively. In the rear $6th{\sim}7th$ lines the total saponin contents were 21.88 mg/g and 26.68 mg/g in the main roots, 38.41 mg/g and 44.89 mg/g in the lateral roots, 101.03 mg/g and 107.06 mg/g in the fine roots in Yunpoong and Landrace variety, respectively. The differences in total and individual ginsenosides content in the main, lateral and fine roots among the lines were not significant but total ginsenosides contents in the main roots were different in case of Yunpoong variety. The ratios of protopanaxadiol (PD) type saponin to protopanaxatriol (PT) type saponin in roots were lower in the front lines compared to the middle and rear lines and the ratios were significantly different among the parts of roots.
This study was conducted to estimate heritabilities, repeatabilities and rank correlation coefficients among breeding values for litter size and sex ratio of Yorkshire and Landrace pigs using various single trait animal models. The analyses were carried out the data comprising 26,390 litters of Yorkshire and 26,173 litters of Landrace collected from the year 1998 to 2008 at a private swine breeding farm located in central part of Korea. Five different analytical models were used for genetic parameter estimation. Model 1 was most simple basic model fitted with year-month contemporary group fixed effect, random additive genetic effect and random residual effect. Model 2 was similar to the model 1 but permanent maternal environmental effect added as random effect, and model 3 was similar with the model 2 but linear and quadratic effects of sow age were added as fixed covariate effect. Model 4 was similar as model 2 except that the parity was added as fixed effect and model 5 was similar to model 3 or model 4 but covariate of sow age was nested within parity effect. The results obtained in this study are summarized as follows: The means and standard error of total number of pigs born per litter (TNB) and number of pigs born alive per litter (NBA) were $11.35{\pm}0.02$ and $10.04{\pm}0.02$ for Yorkshire, $10.97{\pm}0.02$ and $9.98{\pm}0.02$ for Landrace, respectively. The sex ratio (percentage of female per litter) was $45.75{\pm}0.11%$ and $45.75{\pm}0.11%$ for Yorkshire and Landrace, respectively. The heritability estimates of TNB (0.243) and NBA (0.192) from model 1 tended to be higher than those from any other models in both breeds. Differences in heritability and repeatability for TNB were not large among models 3, 4 and 5 and same tendency of negligible differences among estimates by models 3, 4 and 5 were observed for NBA, where heritability and repeatability ranged from 0.096 to 0.099 and from 0.188 to 0.193, respectively, in Yorkshire; and ranged from 0.092 to 0.098 and from 0.193 and 0.196, respectively, in Landrace. The heritability estimates for sex ratio were close to zero which was ranged from 0.002 to 0.003 for TNB and from 0.001 to 0.003 for NBA over the models applied. The rank correlation coefficients of breeding values by model 1 with those from other models (model 2, 3, 4 and 5), and breeding values by model 2 with those from other models (model 1, 3, 4 and 5) were highly positive but lower than the coefficients among breeding values by model 3, model 4 and model 5 which were high of 0.99, approximately, for TNB and NBA of both breeds.
This experiment was carried out to determine the effect of breed and sex on behavioral characteristics and economic traits of performance-tested pigs. Totally 32 tested pigs in 4 different breeds were assigned for behavioral observation. Behavior was recorded for 4 hours(each 2 hours, before and after none) of tested pigs in each tested pen. The average daily gain, age at 90 kg and feed efficiency on the basis of the performance data were collected from 8,477 performance-tested pigs in which pure breeds of Duroc, Yorkshire, Landrace and Berkshire at the Korea Swine Testing Station. The results obtained in this study are summarized as follows; The effect of breed was statistically significant for all traits studied. Average daily gain were the highest(P<0.05) in Duroc and ages at 90 kg were the highest(P<0.05) in Berkshire. In feed efficiency of male, Landrace and Yorkshire were the most efficient. The average daily gain and feed efficiency were superior to those of female, however, the age at 90 kg was not different. Overally, females were superior to males in various traits examined. Berkshire breed had more proportion of time spent in ventral tying and sitting whereas the proportion of time spent in walking, drinking and eating was less than that of other breeds. The male pigs showed more time spent in standing and social behavior.
This study was conducted to investigate the genetic relationships among four reproductive traits. Data for this study were 7616 records from 1910 Landrace (L) and 10,454 records from 2283 Yorkshire (Y) in a closed nucleus swine herd. Traits considered on this study were gestation length (GL), total number of piglets born (TNB), wean to first service (WFS), and number of stillborn per litter (NSB). Heritabilities and genetic correlations were estimated by using the Bayesian inferences via Gibbs sampling in a four trait linear-threshold repeatability animal mixed model by designating NSB as a categorical trait in the L and Y purebred populations. Effects on the statistical model were considered for parity, contemporary group as fixed and service sire, permanent environmental, animal additive genetic effects as random. Estimates of heritability were 0.21, 0.23, 0.16, and 0.09 for GL, WFS, TNB, and NSB in the L population and 0.35, 0.16, 0.14 and 0.10 for corresponding traits in the Y population, respectively. Genetic correlation for GL was -0.59 and -0.28 with TNB and -0.58 and -0.17 with NSB in the L and Y populations, respectively. The NSB was positively correlated with TNB in the L and Y populations in genetic and environmental aspects. Therefore, the NSB should be taken into account in selecting sows for improving prolificacy of dam line breeding swine stock.
The purpose of this study was to establish proper shipping weight and backfat thickness by applying the growth model to backfat thickness, measured by means of not only body weight, but also ultrasonography, and predicting the changes by age. Three breeds, i.e. Duroc, Landrace, and Yorkshie, were analyzed, and the Gompertz, logistic, and Von Bertalanffy model were used for inference with the parameter of the growth model being sex. As a result, both body weight and backfat thickness showed different growth curve parameters and characteristics at inflection points depending on model selection and sex. As for backfat thickness, in estimating the inflection point, unlike the case of body weight, the inflection ages of the boars of the Duroc breed was earlier than that of sows, whereas the inflection ages of the sows of the Landrace and Yorkshire breeds was earlier than that of boars. More than anything else, in the analysis of the changes in backfat thickness according to body weight, as the body weight reached 145kg, the backfat thickness showed much variation as great as 1.7-3.2 cm in each breed and sex. In addition, unlike the other breeds, the boars of the Landrace breed showed an exponential type of relationship between body weight and backfat thickness. As they grow to become 100 kg or heavier, abrupt change in back fat thickness was confirmed. If the growth of body weight and backfat thickness is understood and the genetic relationship is taken advantage of like this, it would be possible to set desired body weight and backfat thickness, and thus help effectively set the shipping time. If not only the phenotype, but also genetic parameters about growth characteristics are estimated and analyzed additionally, more effective data can be generated.
The amount of telomeric DNA was quantified across different breeds(Landrace, Duroc, Yorkshire and Berksire), at different ages(90 days old and 180 days old) and among sexes(male and female) in pigs raised at the Performance Testing Station of Korea Swine Association, Jinkyo, Korea. The telomeric DNA amount was quantified by Quantitative Fluorescence In Situ Hybridization(Q-FISH) using a porcine telomeric DNA probe on interphase nuclei of lymphocytes. Analysis revealed that the amount of telomeric DNA on the pig lymphocytes was found to decrease with age. The quantity of telomeres significantly differed among breeds at 90 days of age. The colored breeds such as Berkshire and Duroc had higher amount of telomeric DNA than the Yorkshire and Landrace breed. In addition, the amount of telomeric DNA in male lymphocytes was significantly higher than that of females. In the correlation coefficients between the telomere quantity and their productive traits; average daily gain, loin percent and index value were positively correlated, whereas body length, feed efficiency and back fat thickness correlated negatively. However, the correlation coefficients were very low and not significant. Therefore, this study suggests that the amount of telomeres on lymphocytes can be considered as a physiological marker but not as a productive marker in pig.
In this study, we conducted a next generation sequencing based microbial community analysis to investigate gut microbiota of the two commercially most available swine breeds, Yorkshire and Landrace. Bacterial 16S rRNA gene was amplified from fecal DNA using universal primer sets designed for V4 regions. Our comparison analysis of the gut microbiota of the two breeds suggested that their gut microbiota changed depending on the growth stages, while the difference between the two breeds was insignificant. However, there was a limited number of genera, the abundance of which was found to be different between the breeds. Those included the genus Xylanibacter in the Yorkshire samples, which was previously reported as a fiber digesting bacteria, likely increasing energy harvesting capacity of swine. In addition, others included opportunistic pathogens mostly found in the Yorkshire samples while the Landrace samples had significantly more prevalent Clostridium_IV species that were known to play a key role in systemic immunity of hosts. While microbial community shifts was found to be associated with growth stages, the difference between the two breeds seemed to be insignificant. However, there were several bacterial genera showing differential abundance, which may affect growth of hosts.
Currently, in pork auctions in Korea, only carcass weight and backfat thickness provide information on meat quantity, while the production volume of primal cuts and fat contents remains largely unknown. This study aims to predict the production of primal cuts in pigs and investigate how these carcass traits affect pricing. Using the VCS2000, the production of shoulder blade, loin, belly, shoulder picnic, and ham was measured for gilts (17,257 pigs) and barrows (16,365 pigs) of LYD (Landrace × Yorkshire × Duroc) pigs. Single and multiple regression analysis were conducted to analyze the relationship between the primal cuts and carcass weight. The study also examined the correlation between each primal cut, backfat thickness (1st thoracic vertebra backfat thickness, grading backfat thickness, and Multi-brached muscle middle backfat thickness), pork belly fat percentage, total fat yield, and auction price. A multiple regression analysis was conducted between the carcass traits that showed a high correlation and the auction price. After conducting a single regression analysis on the primal cuts of gilt and barrow, all coefficients of determination (R2) were 0.77 or higher. In the multiple regression analysis, the R2 value was 0.98 or higher. The correlation coefficient between the carcass weights and the auction price exceeded 0.70, while the correlation coefficients between the primal cuts and the auction prices were above 0.65. In terms of fat content, the backfat thickness of gilt exhibited a correlation coefficient of 0.70, and all other items had a correlation coefficient of 0.47 or higher. The correlation coefficients between the Forequarter, Middle, and Hindquarter and the auction price were 0.62 or higher. The R2 values of the multiple regression analysis between carcass traits and auction price were 0.5 or higher for gilts and 0.4 or higher for barrows. The regression equations between carcass weight and primal cuts derived in this study exhibited high determination coefficients, suggesting that they could serve as reliable means to predict primal cut production from pig carcasses. Elucidating the correlation between primal cuts, fat contents and auction prices can provide economic indicators for pork and assist in guiding the direction of pig farming.
Two hundred and sixty Korean soybean landrace accessions were analyzed for polymorphism at 92 simple sequence repeat(SSR) loci. The 995 identified alleles served as raw data for estimating genetic diversity and population structure. The number of alleles at a locus ranged from three to 27 with a mean of 10.4 alleles per locus. $F_{ST}$ values estimated by analysis of molecular variance(AMOVA) using SSR data set were 0.018, 0.027, and 0.016 for usage, collection site and maturity groups, respectively, indicating little genetic differentiation. The model-based clustering analysis placed the accessions into three clusters(K=3) with 0.0503 of $F_{ST}$, indicating moderate genetic differentiation. Duncan's Multiple Range Test at K = 3 on the basis of 18 quantitative traits revealed that one cluster was mainly differentiated from the other two clusters by seed related traits and the other two clusters were differentiated from each other by biochemical traits. Genetic structure of Korean soybean landraces was differentiated by model-based clustering and supported by their phenotypic traits in part. This preliminary study could be the first step towards more efficient germplasm management and utilization of soybean landraces and helpful in association studies between genotypic and phenotypic traits in Korean soybean landraces.
The purpose of this study was to investigate the Cytochrome P450 (CYP2A6) gene as a candidate gene for the traits related with meat fatty acid composition traits in pigs. Porcine CYP2A6 polymorphisms were detected and PCR-RFLP was performed for genotyping of Korean native pig (n=14), Landrace (n=3), Duroc (n=3), Berkshire (n=3), Yorkshire (n=8) and F2 population composed of 202 individuals from an intercross between Korean Native pig and Yorkshire. PCR primer set amplified a 612 bp fragment of CYP2A6 and digestion of the PCR products was performed with the restriction enzymes SchI. The CYP2A6 SchI polymorphism was only found in the KNP breed. The genotype frequencies of TT, TC and CC genotypes were 0.36, 0.56 and 0.08 in the KNP respectively and the other pig breeds were fixed with CC genotype (Duroc, Landrace, Berkshire and Yorkshire). Statistical association between genotypes and fatty acid composition traits were tested in the Korean native pig and Yorkshire crossed F2 pigs. The CYP2A6 SchI polymorphism was associated with only fatty acid composition C20:3n3 level (cis11,14,17-Eicosatrienoic acid, p=0.0252). The 'T' allele was associated with lower C20:3n3 level. Further study is required to validate the genotypic association and biological consequence of the CYP2A6 gene polymorphism in pigs.
