Effects of Korean traditional herbal formula for common cold on the activities of human CYP450 isozymes

  • Objectives: Most drug interactions are attributed to the inhibition or induction of the activity of cytochrome P450s (CYP450). Although the regulation of CYP450s by drugs has been widely reported, there have been few studies on influence of traditional herbal formulas on the drug-metabolizing enzymes. Because herbal formulas have been used traditionally to treat various diseases and because herb-drug interactions are crucial factors determining therapeutic efficacies, a systematic evaluation of the effects of herbal formulas is important. Methods: The effects of Galgeun-tang (GGT, gegen tang), Gumiganghwal-tang (GMGHT, jiuweiqianghuo tang), Insampaedok-san (ISPDS, renshenbaidu powder), Samsoeum (SSE, shensu drink), Socheongryong-tang (SCRT, xiaoqinglong-tang) and Sosiho-tang (SSHT, xiaochaihu tang) that are traditional herbal formulas used to treat common cold, on drug-metabolizing enzymes were evaluated through an in vitro CYP3A4, CYP2D6, CYP2C19 and CYP2E1 inhibition assay to assess its interaction potential with synthetic drugs. The inhibitory effects of herbal formulas were characterized with $IC_{50}$ values. Results: These six herbal formulas inhibited the activities of CYP3A4, 2C19, 2D6 and 2E1, in a concentration-dependent manner. Among the six herbal formulas, GGT critically inhibited CYP2C19, CYP2D6 and CYP2E1. GMGHT also inhibited CYP2D6 and CYP2E1 to a greater extent than the other CYP450 isozymes. Additionally, SSE and SSHT may change the effects of medicines that depend primarily on the CYP2C19 and CYP2E1 pathways. On the other hand, ISPDS and SCRT were not inhibited CYP3A4, CYP2C19, CYP2D6 and CYP2E1-mediated metabolism. Conclusions: These findings provide useful information regarding the safety and effectiveness of herbal formulas.

LC-MS/MS를 이용한 향소산 중 15종 성분의 정량분석 (Quantitative Analysis of the Fifteen Constituents in Hyangso-San by LC-MS/MS)

  • Hyangso-san is a traditional herbal medicine that consists of the seven herbal medicines, Cyperi Rhizoma, Perillae Folium, Atractylodis Rhizoma, Citri Unshius Pericarpium, Glycyrrhizae Radix et Rhizoma, Zingiberis Rhizoma Crudus, and Allii Fistulosi Bulbus. Hyangso-san has long been clinically used to treat the influenza, including headache, ferver, chills, and pantalgia. In this study, we were performed the simultaneous analysis of the 15 marker compounds (liquiritin apioside, liquiritin, ferulic acid, naringin, hesperidin, rosmarinic acid, liquiritigenin, kaempferol, glycyrrhizin, nobiletin, 6-gingerol, elemicin, atractylenolide III, nootkatone, and atractylenolide I) in Hyangso-san using liquid chromatography-electrospray ionization-mass spectrometry (LC-ESI-MS/MS). Column for the separation of the 15 ingredients was used a Waters Acquity UPLC BEH $C_{18}$ analytical column ($2.1{\times}100mm$, $1.7{\mu}m$) at $45^{\circ}C$ by using a mobile phase of 0.1% (v/v) formic acid in water and acetonitrile with gradient condition. Identifications of all analytes were performed using a Waters ACQUITY TQD LC-MS/MS system. The flow rate and injection volume were 0.3 mL/min and $2.0{\mu}L$, respectively. Correlation coefficient of the calibration curve was ${\geq}0.9958$. The values of limits of detection and quantification of the 15 components were 0.002-4.29 and 0.01-12.88 ng/mL, respectively. The result of an analysis using the established LC-MS/MS method, kaempferol and atractylenolide I were not detected, while other 13 compounds were 0.08-56.87 mg/g in lyophilized Hyangso-san sample.

한약의 수치에 관한 연구(제 7보) -치자의 수치에 의한 성분변화 및 생리활성- (Studies on the Processing of Crude Drugs(VII) -On the Constituents and Biological Activities of Gardeniae Fructus by Processing-)

  • We have conducted to characterize the physico-chemical change and pharmacological transformation of traditional herbal medicines by means of processing. Processed Gardeniae Fructus was prepared by heating of fruit of Gardenia jasminoides(GF) for $30{\sim}50\;minute$ in the roster designed for herb processing. The contents of drying loss, water extract, diluted ethanol extract, ether extract and geniposide in non-processed GF and processed GF were examined. The contents of drying loss, water extract and geniposide in processed GF showed a decrease as compared with those of non-processed GF, however the contents of dilute ethanol and ether extract showed a increase as compared with those of non-processed GF. The rate of decrease/increase of those index were in proportion to heating time. And, biological activities of methanol extract of non-processed GF and processed GF were investigated. DPPH scavenging effects and inhibitory effect of xanthine oxidate and hemolysis of processed GF exhibited more effective than those of non-processed GF in vitro. Accelerating effect of large intestinal transport and purgative action of non- processed GF were discriminated by processing of GF. Methanol extracts of non-pro- cessed GF and processed GF showed the protective effects against the hepatotoxicity induced by ${\alpha}-naphthylisothiocyanate$ in rats. These results suggested that the transformation of biological activities of GF by means of processing may be due to the physico-chemical change of the constituents in GF by heating.

한국산 둥굴레속 식물의 형태적 특성 및 엽록체 DNA 염기서열을 이용한 유연관계 분석 (Morphological Characteristics and Phylogenetic Analysis of Polygonatum Species Based on Chloroplast DNA Sequences)

  • Polygonatum is a genus placed in the family Liliaceae, distributed throughout the Northern Hemisphere and 16 of the species are grown naturally in Korea. In oriental medicine, the rhizomes of Polygonatum have been used as two different medicines, Okjuk (Polygonati odorati Rhizoma) and Hwangjeong (Polygonati Rhizoma). However, it is difficult to identify the morphological and chemical differences between the medicinal groups and thus easy to confuse the one with the other. Therefore, a clear classification standard needs to be established so as to be able to discriminate between them. In the study, the morphological characteristics of the plants, Polygonatum spp., were examined. Then, the differences in SNPs among the DNA sequences of 7 of the Polygonatum spp. and 1 of the Disporum spp. were analyzed by DNA barcoding with rpoC1, rpoB2, matK, and psbA-trnH of the cpDNA region. In the results, three regions, rpoC1, rpoB2, and matK were useful for discriminating the species, P. stenophyllum and P. sibiricum. Furthermore, it was possible to discriminate the individual germplasm within the species by using the combination of the results obtained from rpoB2, rpoC1, and matK.

Protective Effects of a Herb, Menthae Herba, against Radiation-induced Oxidative DNA Damage

  • As utilization of radiation in medicine, industry and biochemical research increases, the protection against radiation damage has become an important issue. Natural products such as herbal medicines are beginning to receive attention as modifiers on the radiation response. In the present study, the protective effect of a herb, Menthae Herba, against radiation-induced DNA damage was evaluated using alkaline single-cell gel electrophoresis (SCGE; comet assay) in the mouse peripheral blood Iymphocytes and the micronucleus formation test in the Chinese hamster ovary (CHO) cells. The tail moment, which was a marker of DNA damage in the SCGE, and the frequency of micronuclei was decreased in groups treated with Mentae Herba extract before exposure to 200 cGy of gamma-ray. We also confirmed its activities to scavenge DPPH and hydroxyl radicals. These experiments demonstrated that Menthae Herba was effective at reducing the radiation-induced damage of DNA and scavenging free radicals. It is plausible that scavenging of free radicals by Menthae Herba may have played an important role in providing the protection against the radiation-induced damage to the DNA. These results indicated that Menthae Herba might be a useful radioprotector and that radical scavenging appears to be one of the mechanisms of radiation protection.

"상한론(傷寒論)"에 수록된 탕제(湯劑)의 전탕법(煎湯法)에 관한 연구 (A Study on the Methods of Decocting and Taking Prescriptions in SANGHANRON)

  • This is a study on the methods of decocting prescriptions in SANGHANRON (傷寒論), and after this, numbers of results have been obtained. SANGHANRON was written by Chang-Ki (張機) in the 2nd century, so it reflexes the usage of prescriptions of previous age indirectly. And gave affects on the methods of decocting prescriptions to the oriental medicine doctors of next generation.Berore Han-DYnasty(漢代), there were not so many publications connected with Oriental Medicine. Besides, some books couldn't hand down to next generation due to the gap of time and space. As time goes by, letters in medical books changed little by little, so contents connected with decocting methods changed too. The effects of decoction and herb tea can be changed by the flexibility of methods of decocting medicines, so we have to decide what kinds of decocting methods should be taken and adapted to patients by the most effective way.In SANGHANRON, there are many kiny kinds of boiling methods, so DHAMG-Ki selected the most appropriate method considering deree and position of disease and condition of patient. But nowadays, due to inconvenient procedure of boiling and taking medicines, some methos are not in common in clinical medicin. So this study was started to look back upon the changes of decocting prescriptions and gave deffort to find out the propriety of variation of boiling prescriptions.The common method of decocting prescriptions in SANGHANRON is boiling down all the herbs at the same time. Except the commom method, there are some kinds of other methods - boiling down twice the prescription, boiling down some special herbs earlier of later than other herbs, complicated or special boiling methods of extract, etc. These kinds of decoting methods simplified as time pass by, but this expected therapeutic value. So we have to distinguish the methods -though complicated and troublesome- in clinical medicine to make perfection more perfect in treating patients, and further studies have to be followed to prove the propriety of these complicated methods.

Effects of Baicalin on Oral Pharmacokinetics of Caffeine in Rats

  • Scutellaria baicalensis is one of the most widely used herbal medicines in East Asia. Because baicalein and baicalin are major components of this herb, it is important to understand the effects of these compounds on drug metabolizing enzymes, such as cytochrome P450 (CYP), for evaluating herb-drug interaction. The effects of baicalin and baicalein on activities of ethoxyresorufin O-deethylase (EROD), methoxyresorufin O-demethylase (MROD), benzyloxyresorufin O-debenzylase (BROD), p-nitrophenol hydroxylase and erythromycin N-demethylase were assessed in rat liver microsomes in the present study. In addition, the pharmacokinetics of caffeine and its three metabolites (i.e., paraxanthine, theobromine and theophylline) in baicalin-treated rats were compared with untreated control. As results, EROD, MROD and BROD activities were inhibited by both baicalin and baicalein. However, there were no significant differences in the pharmacokinetic parameters of oral caffeine and its three metabolites between control and baicalin-treated rats. When the plasma concentration of baicalin was determined, the maximum concentration of baicalin was below the estimated $IC_{50}$ values observed in vitro. In conclusion, baicalin had no effects on the pharmacokinetics of caffeine and its metabolites in vivo, following single oral administration in rats.

한국내 고추나물의 하이퍼리신 유전자(HyH)의 탐색과 유럽의 서양고추나물과 비교 (Detect of Hypericin (HyH) gene in Hypericum erectum in Korea and Comparison of H. perforatum in Europe)

  • 하이퍼리신(Hypericin, HyH)은 예로부터 성 요한의 풀(St. John's Wort)로 널리 알려져 있는 서양고추나물(Hypericum perforatum)에서 추출되는 약리성분이다. 서양고추나물은 국내에서는 자생하지 않으나 같은 속의 고추나물(H. erectum) 등이 이속에 속하며 우리나라에 자생한다 . 고추나물을 조직배양으로 RNA를 추출하고 cDNA를 합성한 후 Hyp 유전자를 추출하여 서열화한 결과, 전체 크기는 732 bp로 나타났으며 서양고추나물의 HyH-1과 거의 99.8% 서열 일치를 나타내었다. 이 서열의 152개 아미노산 역시 서양고추나물의 항산화효소와 98% 일치하였으며 자작나무와 능금속 식물에서 유발되는 알레르기 유발유전자와 약 60% 일치를 나타내었다. 본 연구 결과 우울증치료제로 사용되는 하이퍼리신 추출에 우리나라 자생종인 고추나물이 이용될 수 있을 것으로 사료된다.

Effects of Medicinal herb Extracts and their Components on Steatogenic Hepatotoxicity in Sk-hep1 Cells

  • Herbal medicines are widely used in many countries for the treatment of many diseases. Although the use of herb extracts as alternative medicine is growing, their toxicological properties have not been thoroughly investigated. In this study, we have investigated the effects of water and ethanol extracts of 18 herbs on the hepatic lipid metabolism and steatogenic hepatotoxicity. Ethanol extracts of Cirsium japonicum, Carthamus tinctorius, Rehmanniae glutinosa (preparata), Polygala tenuifolia, Foeniculum vulgare, Polygonum multiflorum, and Acorus gramineus and water extracts of Polygonum multiflorum and Rehmanniae glutinosa induced lipid accumulation in Sk-hep1 human hepatoma cells as determined by Nile red staining. These extracts increased the luciferase activity of sterol regulatory element (SRE) and decreased that of peroxisome proliferator response element (PPRE), indicating the possibilities of enhanced fatty acid synthesis and decreased fatty acid oxidation. To identify the components responsible for the fat accumulation, we tested 50 chemicals isolated from the nine herbs. Apigenin, luteolin, pectolinarin and lupeol from Cirsium japonicum, 8-methoxypsoralen and umbelliferone from Foeniculum vulgare and pomonic acid and jiocerebroside from Rehmanniae glutinosa significantly increased the accumulation of lipid droplets. These results suggest that ethanol extracts of Cirsium japonicum, Carthamus tinctorius, Rehmanniae glutinosa (preparata), Polygala tenuifolia, Foeniculum vulgare, Polygonum multiflorum, and Acorus gramineus and water extracts of Polygonum multiflorum and Rehmanniae glutinosa can cause fatty liver disease by decreasing ${\beta}$-oxidation of fatty acid and increasing lipogenesis.

영월군 가전(家傳) 『찰병요결(察病要訣)』에 대한 연구 (The Study on the Chalbyungyogyul (察病要訣), Discovered in Yeongwol-gun)

  • Chalbyungyogyul (察病要訣) is a book of Korean Medicine which contains simple knowledges combined with experienced prescriptions. This study intends to introduce a manuscript of Chalbyungyogyul handed down in a family in Yeongwol-gun and analyze the Eui'an (醫案) in it. This book includes common sense in medicinal herbs and 142 cases of Eui'ans. The Eui'ans in Chalbyungyogyul can be divided into three types. Some Eui'ans state miraculous effects. External medical treatments are used in another group of Eui'ans. In the other Eui'ans, prescriptions composed by a single herb are applied. These characteristics are differentiated from those of preexisting Eui'ans in the history of Korean medicine which mainly include complexly prescribed internal medicines and medical discussions. This book vividly describes medical practice in the early 20th century and shows the realization of theoretical writings. Above all, it is a valuable discovery of Eui'an compilation which is rare in the history of Korean medicine.
