Anti-Androgenic Activity of Phthalate Esters (Di(2-ethylhexyl) Phthalate, Di(n-butyl) Phthalate, and Butylbenzyl Phthalate) in the Rodent 10-day Hershberger Assay using Immature Castrated Male Rats

  • The rodent Hershberger assay is considered as a potential short term in vivo screening method for the detection of androgenic or anti-androgenic compounds. The objective of this study was to evaluate the anti-androgenic activities of di(2-ethylhexyl) phthalate (DEHP), di(n-butyl) phthalate (DBP), and butylbenzyl phthalate (BBP). A 10-day Hershberger assay was performed using immature Sprague-Dawley male rats castrated at 6 weeks of age. Tastosterone propionate (TP, 0.4 mg/kg/day) was administered s.c. to castrated male rats and followed by flutamide (1, 5, 10, or 20 mg/kg/day) treatment for 10 days by oral gavage. Similarly, DEHP, DBP, or BBP were also administered by oral gavage at 250, 500, or 1000 mg/kg/day after TP (0.4 mg/kg/day) administration. As expected, flutamide significantly inhibited the TP-induced re-growth of seminal vesicles, ventral prostate, and Levator ani plus bulbocavernosus muscles (LABC) at 1 mg/kg/day and above, and Cowper's glands and glans penis at 5 mg/kg/day and above. DEHP significantly (p<0.05) decreased the seminal vesicles, ventral prostate, LABC and Cowper's glands weights at 1000 mg/kg/day. BBP at 1000 mg/kg/day significantly inhibited TP-induced re-growth of the LABC in the immature castrated male rats, whereas ventral prostate, seminal vesicles, and Cowper's glands weights were unaffected. In contrast to DEHP, DBP did not affect accessory sex organ weights at any concentration. Body weights, combined adrenal glands, and kidney weights were not affected, but liver weights were significantly increased at high dosages in the DEHP, DBP, and BBP treatment groups. Our observations strongly suggest that DEHP acts as an androgen antagonist at the high dose (i.e., 1000 mg/kg/day).

Optimization of the Processing Conditions for Heated Garlic Juice by Response Surface Methodology

  • This study was designed to determine the optimum conditions of heating temperature, heating time and dilution rates for producing heated garlic juice by using central composite design of response surface methodology. Garlic 3Department of Biosystems Engineering, Chungbuk National University, Cheongju 361-763, Korea was heated using a high temperature and pressure treatment apparatus. Total soluble solid contents ranged from 4.4 at $130^{\circ}C$ 3 hr and 6 fold dilution to 5.89 at $115^{\circ}C$ for 2 hr and 8 fold dilution. The highest total acidity was 0.55% at $120^{\circ}C$ for 3 hr and 2 fold dilution. The pH ranged from 4.01 at $130^{\circ}C$, 3 hr and 6 fold dilution to 5.85 at $120^{\circ}C$, 1 hr and 6 fold dilution. From the results of statistical analysis on the sensory evaluation the predicted optimum processing conditions for best color, taste, flavor and overall acceptance were $119.41^{\circ}C$, 3.11 hr, 5.85 fold dilution, $118.23^{\circ}C$, 3.57 hr, 3.15 fold dilution, $120.54^{\circ}C$, 3.47 hr, 5.01 fold dilution and $119.54^{\circ}C$, 3.18 hr, 5.66 fold dilution, respectively. The application of response surface methodology for preparing heated garlic juice processing showed a good correlation with high significance.

복분자 열매(Rubus coreanus Miquel)의 항산화 활성 및 생리활성물질 (Antioxidant Activity and Effective Compounds of Black Raspberry (Rubus coreanus Miquel) Extracted by Different Solvents)

  • The black raspberry (Rubus coreanus Miquel) contains anthocyanin, tannins, gallotannin, gallic acid, ferulic acid and phenolics. It brightens the eyes and protects the liver and kidneys. It was effective for anti-aging. Thus, the purpose of this study was to inform the excellence of black raspberry and to screen antioxidant activity to ensure the possibility as a functional material. In this study, bioactive compounds in black raspberry were determined. Additionally, black raspberry was extracted by CM (chloroform:methanol, 2:1, v/v), 70% methanol and 70% ethanol, and were investigated and compared in vitro methods. Total phenol and flavonoid contents were measured to compare each different solvents. In addition, DPPH radical scavenging activity and ABTS radical scavenging activity, ferric reducing antioxidant power, reducing power were determined to measure the antioxidant activity. The results were the highest in 70% ethanol extracts, and the higher the concentration showed a significantly higher antioxidant capacity. When observed the relationship between the study, antioxidant activity of black raspberry was supposed to affect by the anthocyanin, phenol and flavonoid contents.

농축 콩단백질 분리 및 추출에 관한 연구 (Studies on the characteristics of concentrated soy protein)

  • 본 연구는 우리나라에서 재배되는 대두, 녹두, 적두 및 완두 4종의 콩 원물을 선발하여 단백을 추출한 후 얻은 농축콩단백의 영양학적 및 기능적 특성을 분석하여 4종 단백 소재의 품질 특성을 알아보고자 하였다. 4종 농축콩단백의 영양학적 특성을 살펴본 결과, 단백질 함량은 72.58-78.69%로 나타났고, 아미노산의 함량은 4종 농축콩단백 모두 aspartic acid, glutamic acid, lysine에서 높은 함량을 나타났으며, 농축녹두단백의 총 필수 아미노산 함량이 3종 농축콩단백에 비해 유의하게 높았다(p<0.05). 4종 농축콩단백의 기능적 특성을 알아본 결과, pH와 겉보기 밀도는 농축녹두단백과 농축적두단백이 다른 농축콩단백에 비해 유의하게 높았고(p<0.05), 수분흡수력은 농축완두단백이, 유지흡수력은 농축대두단백이, 유화 활성은 농축녹두단백이, 유화안전성은 농축녹두단백과 농축완두단백이 다른 농축콩단백에 비해 유의하게 높았다(p<0.05). 따라서 본 연구를 종합해 볼 때, 4종 농축콩단백의 각 기능적 특성을 고려하여 대체육 제품 산업에 적용한다면, 식품소재로서 활용가치가 있을 것으로 판단된다.

동결 전처리가 매실의 착즙과 건조 특성에 미치는 영향 (Effects of Freezing Pretreatment on Juice Expression and Drying Characteristics of Prunus mume Fruit)

  • 매실(Prunus mume)의 착즙과 건조 특성에 동결 전처리가 미치는 영향을 조사하기 위하여, 매실을 동결처리($-20^{\circ}C$)하거나 처리하지 않고 착즙수율과 주스의 품질특성 및 건조속도와 분말의 품질특성을 각각 측정하였다. 매실의 착즙수율과 건조속도는 동결처리구가 무처리구보다 유의적으로 높은 수준을 나타내었다. 압착주스와 건조분말의 L과 b값 및 a값은 동결처리구가 무처리구보다 유의적으로 낮고 높은 수준을 각각 보였다. 압착주스와 건조분말의 유리당과 유기산 함량은 동결 전처리에 따른 유의적인 차이를 나타나지 않았다. 주스의 가용성 고형분 함량, 적정산도 및 pH는 동결 전처리의 영향을 받지 않은 것으로 나타났다. 이로써 동결 전처리는 매실의 착즙과 건조 효율을 증대시킬 수 있는 유효한 방법이지만 착즙액과 건조분말의 갈변도가 높은 문제점이 있는 것으로 확인되었다.

원료 배합비율을 달리한 순무 발효액의 품질특성 (Physicochemical Properties of Fermented Turnip Juice with Different Mixture Ratio of Materials)

  • The objective of this study was the development of the preparation method of fermented turnip juice. The yields and soluble solids of fermented turnip juice were 41.2, 52.8, 56.4 and 57.4%, and 34.3, 42.4, 48.6 and 54.7 $^{\circ}Brix$ with 40, 60, 80 and 100% addition rate of turnip quantity, respectively. The major organic acids in turnip juice were succinic acid, isobutyric acid and fumaric acid, and these organic acids were decreased after fermentation. On the other hand, lactic acid, formic acid, acetic acid, propionic acid and butyric acid were produced during the fermentation. Soluble solid and pH value in fermented turnip decreased by time-dependently. Free radical scavenging activity was increased by fermentation of turnips and nitrate scavenging effects were the highest in pH 3 and increased with adding Omija or Japanese apricot. The best preparation method of fermented turnip juice was fermentation at $24^{\circ}C$ 12-15 days after adding 40% of sucrose to total quantity. As adding Omija, organic acid contents were increased and sensory evaluation was improved in fermented turnip juice.

  • Lyophyllum decastes의 발효사료 제조를 위한 cellulase 생산은 $30^{\circ}C$, pH 6.0, 수분 $60{\sim}70%$ 범위에서 I5일간 배양하는 것이 좋았다. 부원료로 미강 $30{\sim}40%$$(NH_4)_2HOP_4$ 0.72%를 첨가하고 볏짚을 4% alkaline peroxide로 전처리하면 cellulase 생산은 증가하였다. 볏짚에 Lyophyllum decastes를 40일간 발효시켰을 때 lignin은 19.9% 분해되있고 총질소는 대조구의 1. 1%에서 1.6%로 증가되었다. 발효가 진행됨에 따라 볏짚의 in vitro 소화율은 증가하여 35일 발효 후에는 대조구에 비하여 21. 1%증가하였다.

대두의 수화속도와 콩나물의 발아특성에 대한 동결처리 효과 (Effect of Freezing on Hydration Rate and Sprouting Characteristics of Soybeans)

  • 은하콩과 태광콩의 수화속도 및 콩나물 생육 특성에 대한 동결처리의 효과를 수화반응에 대한 속도론적 연구와 콩나물의 재배실험을 통하여 조사하였다. 수화시간에 대한 동결처리의 영향은 침지온도에 따라 달랐는데, $60^{\circ}C$ 이하의 침지온도에서는 수화시간이 15-50% 정도 감소하였으며, $60^{\circ}C$ 이상의 침지온도에서는 수화시간이 감소하지 않았다. 동결 처리한 은하콩과 태광콩의 수화반응에 대한 활성화 에너지는 각각 39.79 및 39.25 kJ/mol로서 동결처리를 하지 않은 콩에 비해 약 20%정도 감소하였다. 은하콩과 태광콩은 동결처리에 의해 콩나물의 발아율, 길이 및 무게는 증가하였으며, 콩나물의 수율은 약간 감소하였고, 굵기는 콩의 품종에 따라 달랐다.

향토음식의 스토리텔링 적용 사례연구 (A Case Study on Storytelling Application of Native Local Foods)

  • The purpose of this study is to present the scheme that can enhance the value of native local foods by analyzing the case into which storytelling is incorporated in order to revitalize native local foods. Towards this end, the cases of native local foods throughout the country were recommended through professional survey and one region among them was finally selected through the field study. The survey on the native local foods was conducted through qualitative study. The four stages of storytelling: the findings of storytelling, the production of storytelling, the experience of storytelling, and sharing of storytelling were analyzed by stage and the contents of the survey by stage were developed. In the findings of story, the origins and background of the main family's foods and food-related stories were derived. The production of stories deals with how to convey the stories of surveyed native local foods to consumers and it presented the stories about the place and food menu. In the experience of storytelling, the physical environment and non-physical environment, in which people visit native local eating houses and experience native local foods were analyzed. Finally, in the sharing of story, how the tourists who have experienced the native local foods make stories and disseminate and share them was analyzed. The significance of this study is that through the storytelling technique it presented the findings, production, experience and sharing of native local foods in the level that can apply to practical business. Although the research is in the stage of inquiry, this study is meaningful in that it presented the basic methodology that can induce the revitalization of the native local food industry through examining the tradition and significance of native local foods that can be found in all the regions of the country.

E-screen assay 및 자궁비대반응시험 (Uterotrophic assay)을 이용한 di-(2-ethylhexyl) adipate의 에스트로겐성 작용에 관한 연구 (Study on the Estrogenic Activity of Di-(2-Ethylhexyl) Adipate in E-Screen Assay and Uterotrophic Assay)

  • 랩 등 PVC 제품의 가소제인 DEHA의 에스트로겐 활성을 검색하기 위하여 내분비계 장애작용 중 에스트로겐성 작용을 검색하는 대표적인 방법인 in vitro E-screen assay와 OECD 및 미국 EPA EDSTAC에서 권고하고 있는 in vivo 난소절제 랫드를 이용한 자궁 비대반응시험(uterotrophic assay)을 실시하였다. 시험결과 $DEHA(5{\times}10^{-9}{\sim}5{\times}10^{-4}\;M)$는 MCF-7세포의 증식을 유발하지 않았고 난소절제 랫드에 200 mg/kg/day 용량까지 투여하여도 자궁 및 질 무게에 영향을 미치지 않았다. 따라서 본 실험결과 DEHA는 E-screen assay 및 자궁비대반응시험에서 에스트로겐성 작용이 없는 것으로 확인되었으나, 이들 물질의 에스트로겐성에 대한 결론을 내리기 위해서는 여러 종류의 in vitro 시험 data 및 in vivo 시험 data가 더 보충되어야 할 것으로 생각된다.
