Induction of in vitro root tubers in Holostemma annulare (Roxb.) K. Schum. for the production of bioactive metabolites

  • Holostemma annulare (Family Asclepiadaceae) is an invaluable vulnerable medicinal plant; the root tubers are used in Ayurveda medicine and by folk healers to treat various ailments. In this study, Schenk and Hildebrandt medium fortified with the cytokinins 6-benzyl adenine, kinetin, and auxins, including indole 3-butyric acid, indole 3-acetic acid, α-naphthaleneacetic acid, and 2,4-dichlorophenoxyacetic acid, were checked for their efficiency on root tuber induction from different explants. Adventitious root tubers were more successfully induced from in vitro leaf segments and shoots when cultured in Schenk and Hildebrandt medium supplemented with 0.5 mg/l of α-naphthaleneacetic acid. In addition, preliminary phytochemical analysis of in vitro root tubers and identification of different secondary metabolites were conducted. Thin layer chromatography and high performance thin layer chromatography analysis of the crude methanolic extracts of the in vitro root tuber identified the presence of lupeol, a bioactive triterpene. Adventitious root tuber induction offers a novel method for the in vitro production of bioactive metabolites that can be scaled up by bioreactors, thus ensuring the conservation and sustainable utilization of H. annulare. The study warrants further scale-up production and pharmacological investigation that can be extended for pharmaceutical needs.

민속식물의 항균활성 및 산화적 스트레스 개선 효과 (Antimicrobial Activities and Free Radical Scavenging Effect of Korean Folk Plants)

  • We investigated the antioxidative and antimicrobial activities of the methanol extracts from Korean folk plants (MKs) in Chungcheong Province. Among 30 MKs, 16 plants at $100{\mu}g/ml$ showed over 90% scavenging activity of 1,1-diphenyl-2-picrylhydrazy (DPPH) and 30 plants exerted the hydroxyl radical scavenging effect over 55%. Fourteen plants at the concentration $50{\mu}g/30{\mu}l$ showed strong microbial inhibition activity against Escherichia coli and Staphylococcus aureus, with clear zone greater than 11 mm in disc assays. Furthermore, the protective effect against anti-inflammatory system using RAW 264.7 macrophage cell was also studied. The treatment of LPS & INF-${\gamma}$ to RAW 264.7 cell induced nitric oxide (NO), however inhibit the formation of NO less than 50% of 5 plants. The present result indicates that the 30 species of MKs exerts protective effect of oxidative stress, antimicrobial activities and anti-inflammatory. In particular, Rhus javanica and Cornus controversa showed stronger effect on not only radical scavenging activity and inhibits growth of S. aureus but also highest protective effects from inflammation.

울릉도 민속식물 추출물의 항염증 효과 (Anti-inflammatory Effect of Extracts from Folk Plants in Ulleung Island)

  • 울릉도 민속식물 31분류군 49점 추출물을 대상으로 최종농도 $50{\mu}g/mL$로 LPS에 의해 유도된 RAW 264.7 대식세포의 NO 생성량을 조사하였다. 그 결과, 다래(Actinidia arguta) 잎 가지, 헛개나무(Hovenia dulcis) 잎, 동백나무(Camellia japonica) 잎 가지, 말오줌나무(Sambucus sieboldiana var. pendula) 잎 가지, 왕호장근(Fallopia sachalinensis) 뿌리 순으로 항염 활성이 우수하게 나타났다. 이 중에서 다래 잎 가지, 헛개나무 잎, 동백나무 잎 가지 추출물을 LPS로 유도된 Raw 264.7 대식세포에서 최종농도가 10, 20, 40, $50{\mu}g/mL$로 처리하여 NO 생성량과 세포생존율, $PGE_2$ 생성량을 측정하였다. 그 결과 다래 잎 가지($IC_{50}=29.21{\mu}g/mL$), 헛개나무 잎($IC_{50}=27.34{\mu}g/mL$), 동백나무 잎 가지($IC_{50}=45.68{\mu}g/mL$) 모두 NO 생성을 농도 의존적으로 유의성 있게 억제하였다. 또한 $PGE_2$ 측정결과 다래 잎 가지($IC_{50}=21.06{\mu}g/mL$), 헛개나무 잎($IC_{50}=33.47{\mu}g/mL$), 동백나무 잎 가지($IC_{50}=43.90{\mu}g/mL$)에서도 유의성 있는 억제효과를 보였다. 본 연구결과 다래 잎 가지, 헛개나무 잎, 동백나무 잎 가지의 추출물은 염증 유발의 중요 인자인 NO 및 $PGE_2$ 생성을 저해하여 우수한 항염증 효과를 보였다. 이들 소재에 대한 염증억제 유효성분 규명 및 그 작용기전을 추가적으로 연구함으로써 만성 염증질환의 예방과 치료에 효과적으로 사용할 수 있을 것으로 기대된다.

  • Angelica flaccida Kom. is a herbal plant growing wild in the marsh of valley in Korea. It has been used for sedative and analgesic as a folk medicine, but its constituents have not been clarified yet. Five known coumarins such as isoimperatorin, decursidin, (-) anomalin, decursin, decursinol and two sterols such as ${\beta}-sitosterol$ and stigmasterol were isolated from the root of this plant, Their structures were identified by the UV, IR, NMR, mass data and the physico-chemical properties.

관능검사를 통한 씀바귀의 쓴맛 연구 (A Sensory Evaluation of the Bitter Compounds from Ixeris dentata Nakai)

  • Ixeris dentata Nakai (Compositae) is a perennial herb and the young shoots when prepared properly are edible as a bitter appetizing vegetable in the early spring. The whole plant has been used as a folk medicine in treating diabetes in Korea. As a part of our studies on bitter substances, we have investigated the bitterness of the plant through a taste-guided fractionation method. Bitter substances were extracted by the solvent fractionation and the bitterness was evaluated by 10 trained panelists consisted with graduate students of Dept. of Foods and Nutrition. It has been revealed that the bitter compounds were distributed throughout the extracting fractions and the higher intensity of the bitterness was seen in the n-BuOH fractions. The bitter compounds were subfractionated by the silica gel column chromatography and the bitterness was evaluated by the trained panelists. The subfractions which were indicated as the bitter compounds will be further studied to isolate the compounds. The H$_2$O fractions were grouped according to the molecular weight through the ultrafiltration and the group with 1,000 < m.w. < 30,000 was evaluated as the most intense bitter subfraction.

The Effect of Salvia plebeia on Murine Macrophage-mediated Cytotoxicity

  • Macrophages play an important role in host defense against tumors by killing tumor cells. Our work is directed toward studying the effect of the extracts of Salvia plebeia on induction of antitumor activity in macrophages, since it has been usezd as a folk-medicine for the treatment of hepatitis and tumors. The ability of macrophage treated with the plant extracts to inhibit the growth of tumor cells was assessed. The Extracts of the plant induced antitumor activity and could enhance the tumoricidal activity of macrophages when used in combination with $IFN-{\gamma}$. These results suggest that Salvia plebeia extract contain immunomodulatory factors responsible for the induction of the antitumor activity.

Protective Effects of Plathymenia reticulata and Connarus favosus Aqueous Extracts against Cadmium- and Mercury-Induced Toxicities

  • The extracts of Plathymenia reticulata and Connarus favosus are widely used in the folk medicine. The potential protective effects of these extracts have been evaluated against cadmium in the yeast Saccharomyces cerevisiae, and against mercurial contamination in zebrafish Danio rerio. In yeast, both extracts efficiently protected the ${\Delta}ycf1$ mutant strain exposed to cadmium chloride restoring the growth, the expression of stress-response genes and decreasing the level of oxidative stress. In zebrafish, the supplementation of methylmercury-contaminated diet with both plant extracts similarly protected fish through the suppression of the methylmercury-induced lipid peroxidation, decrease of acetylcholinesterase activity, and restoring the expression levels of stress-response genes. This study particularly demonstrates the protective potential of both aqueous extracts against methylmercury, and could represent an interesting alternative for the Amazonian fish-eating communities to cope with the impact of chronic exposure to contaminated diets.

Essential Oil of Marrubium vulgare: Chemical Composition and Biological Activities. A Review

  • Marrubium vulgare, plant species belonging to Marrubium genus, is widespread in the Mediterranean areas, introduced elsewhere and also cultivated in many countries. Its oil is recognized to possess a considerable biological activities with varied chemical composition. This paper aims to overview the chemical composition and biological activities of M. vulgare essential oil's considered as a medicinal plant, widely used in folk medicine overall the world. In essential oils of M. vulgare, germacrene D, ${\beta}$-caryophyllene, ${\beta}$-bisabolene, bicyclogermacrene and carvacrol are generally considered as either mains or minor constituents and each species presents its own composition. Sesquiterpenoids were the dominant fraction while monoterpenoids were present in appreciable or in trace amount. Oxygenated fractions dominated in monoterpenes however, hydrocarbon fraction overpowered in sesquiterpenes. These oils are biologically active, they exhibit an antioxidant and antimicrobial activities and other activities. Due to the variability of composition of essential oil, further studies are necessary, particularly regarding their chemical's which may cause an important change in the biological activities of oils and probably defined different chemotype.

국가지정 문화재 민속마을의 외래식물 식재 및 발생현황 - 하회.양동.한개마을을 대상으로 - (The Planting and Occurrence Status of Exotic Plants of the Folk Village as National Cultural Heritage - Focus in Hahoe.Yangdong.Hangae Villages -)

  • 역사공간의 진정성 제고와 보존관리에 활용될 기초자료 제공을 목적으로 국가지정 문화재인 경상북도 소재 안동 하회마을, 경주 양동마을, 성주 한개마을 등 3개 민속마을의 외래식물 분포현황을 조사 분석한 결과는 아래와 같다. 1. 민속마을의 전체식물상은 127과 430속 614종 5아종 100변종 33품종으로 총 752분류군이 확인되었으며, 그 중에 목본식물은 263분류군(35.0%), 초본식물은 489분류군(65.0%)으로 나타났다. 하회 양동 한개마을의 전체 식물상은 각각 총 534 479 408분류군이 확인되었고, 외래식물률은 30.1 38.2 37.0%로 나타났다. 유형별로는 관상외래식물이 135분류군, 풍치외래식물이 21분류군, 경작외래식물이 64분류군 그리고 귀화외래식물이 80분류군 으로 나타나, 관상외래식물의 비율이 탁월하게 높았다. 마을별로는 하회마을 161분류군(30.1%), 양동마을 183분류군(38.2%), 한개마을 151분류군(37.0%)으로 양동마을에서 가장 높은 비율의 외래식물이 확인되었다. 2. 하회마을내 단위문화재로 지정된 고택 정원에 식재된 관상외래식물은 총 30분류군이고, 옥연정사(8분류군) 양 진당(7) 하동고택(6) 충효당(5) 등의 순으로 외래식물수가 높았으며, 백목련 능소화의 식재율이 탁월하였다. 양동마을에서는 총 51분류군이 관찰되었는데, 두곡고택(16분류군) 수졸당(14) 무첨당(13) 상춘헌고택(12) 등의 순으로 나타났으며, 수국 불두화 상사화 등의 출현율이 높았다. 그리고 한개마을의 관상외래식물은 총 62분류군으 로 진사댁(35분류군), 교리댁(25) 한주종택(20) 하회댁(16) 등의 순이었으며, 골담초 뚝향나무 백목련 불두화 산당화 등의 출현률이 높았다. 3. 정원 외부를 살펴보면 하회마을에서는 만송정 숲 하부에는 환경부 지정 생태계교란식물인 돼지풀이 높은 우점도로 생육하고 있어, 겸암정사 인근의 가시박과 함께 제거대책의 마련이 시급하다. 또한 만송정숲 주변 낙동강 하상으로는 아까시나무림과 양버들군락 등의 풍치외래식물이 하안식생으로 띠숲을 이루고 있다. 양동마을에서는 마을 안길에 자연 발생한 가죽나무군락과 아까시나무와 족제비싸리가 비향토식생경관으로 인지된다. 또한 한개마을 외곽에 산발적으로 분포하는 가죽나무와 아까시나무의 세력 확산이 문제점으로 드러났다. 민속마을 간 발생 외래식물의 유사도는 47.0~48.6%로 이와 같은 이유는 연구대상지가 모두 경상북도에 위치하여 있는 관계로 식물의 생육조건이 유사하며, 관상용으로 판매되는 외래식물이 제한적이기 때문으로 판단됨에 따라 외래식물에 대한 마을별 관리대책과 공통 관리대책의 병행이 요구됨을 암시하였다.

갓 지상부에서 플라보노이드와 고급 알콜 화합물의 분리 (Isolations of Flavonoids and a Higher Alcohol from the Aerial Parts of Brassica juncea)

  • The aerial parts of Brassica juncea (Cruciferae) called by brown or oriental mustard have been widely used as a spice in food and also traditional folk medicine as stimulant, diuretic and expectorant agent. And Gatkimchi made of the aerial parts of this plant are very popular in Korea. The aerial parts of this plant was refluxed with MeOH and then fractionationed with $CH_2Cl_2$, EtOAc, n-BuOH and $H_2O$, successively. One higher alcohol compound and three flavonoids were isolated from the EtOAc fraction through silica gel and Sephadex LH-20 column chromatographies. Their structures were elucidated as n-hexacosanol(1), kaempferol(2), isorhamnetin $3-O-{\beta}-D-glucopyranoside(3)$ and isorhamnetin $3-O-(6'-O-acetyl)-{\beta}-D-glucopyranoside(4)$ by comparison of spectral data with those in references. And compounds 1 and 4 were firstly isolated from this plant.
