Anti-Inflammatory Effect of Chondrus ocellatus Holmes Ethanol Extract on Lipopolysaccharide-induced Inflammatory Responses in RAW 264.7 Cells (Lipopolysaccharide로 유도된 RAW 264.7 세포와 마우스모델에 대한 진두발 에탄올 추출물의 항염증 효과)

  • This study aimed to investigate the anti-inflammatory effect of the ethanol extract from Chondrus ocellatus Holmes (COHEE) in RAW 264.7 cells and in a mouse ear edema model, by measuring the production of lipopolysaccharide-induced inflammatory response mediators. There were no cytotoxic effects on the proliferation of macrophages treated with COHEE compared with the control. COHEE inhibited the production of nitric oxide and pro-inflammatory cytokines [interleukin (IL)-6, tumor necrosis factor-α, and IL-1β]. The extract also reduced the expression of inducible nitric oxide synthase, cyclooxygenase-2, nuclear factor-κB p65, and phosphorylated mitogen-activated protein kinase in a dose-dependent manner. In the croton-oil-induced ear edema model, COHEE decreased the formation of mouse ear edema at the highest dose compared with the control, and histological analysis revealed that the epidermal/dermal tissue thickness and mast cell numbers were reduced. Therefore, these results suggest that COHEE may be a promising topical anti-inflammatory therapeutic material through its action of modulating NF-κB and the MAPK signaling pathway.

  • Agricultural sciences field in South Korea has many strong points such as numerous researchers, establishment of research infra-structure, excellence in research competitiveness and high technological level. However, there are also many weaknesses including insufficient leadership at related societies and institutes, deficiency of the next generation research group, and insufficiency in research productivity. There are many opportunities including increasing the importance of the biotechnological industry, activating international cooperation researches, and exploring the multitude of possible research areas to be studied. However, some threats still exist, such as pressure from the government of developed countries to open the agricultural market, the decrease of specialized farms, and intensification for researches to gratify economic and social demands. To encourage research activities in the agricultural sciences field in Korea, the following actions and systems are required: 1) formulation of a mid- and a long-term research master plan, 2) development of a database on the man power in related fields, 3) activation of top-down research topics, and associated increase of individual research grants, 4) development of special national programs for basic researches in agricultural sciences, 5) organization of a committee for policy and planning within the related societies, and 6) system development for the fair evaluation of the research results.

Anti-Inflammatory Activity of Dichloromethane Fraction from Katsuwonus pelamis Heart in LPS-Induced RAW 264.7 Cells and Mouse Ear Edema (Lipopolysaccharide로 자극된 RAW 264.7 세포와 마우스 귀부종 모델에 대한 참치 심장 Dichloromethane 분획물의 항염증 효과)

  • This study investigated the effect of the dichloromethane fraction form Katsuwonus pelamis heart on anti-inflammatory responses in lipopolysaccharide-stimulated RAW 264.7 cells and mouse models. Ethanol extract was partitioned with dichloromethane, ethyl acetate, butanol, and water. Among the fractions, the dichloromethane fraction showed a significant decrease in nitric oxide (NO) and pro-inflammatory cytokines [interleukin (IL)-6, $IL-1{\beta}$, and tumor necrosis $factor-{\alpha}$] production compared to ethanol extract. The dichloromethane fraction attenuated the expression of inducible nitric oxide synthase and nuclear $factor-{\kappa}B$ ($NF-{\kappa}B$) p65 proteins in a dose-dependent manner. In addition, the expression of phosphorylation of mitogen-activated protein kinases (MAPKs) was also inhibited by the dichloromethane fraction. Moreover, the administration of 10, 50, and 250 mg/kg body weight-dose dependently inhibited the formation of edema by croton-oil and the application of dichloromethane (2 mg/ear) significantly reduced epidermal and dermal thickness and the infiltrated mast cell numbers. Therefore, the dichloromethane fraction exhibited an anti-inflammation effect by inhibiting $NF-{\kappa}B$ and MAPK signaling activation in macrophages.

Changes in Biochemical Components of Several Tissues in Sinonovacula constricta, in Relation to Gonad Developmental Phases (가리맛조개, Sinonovacula constricta의 생식소 발달단계에 따른 일부 조직의 생화학적 성분 변화)

  • We investigated the reproductive cycle with gonad developmental phases of Sinonovacula constricta by histological observations, and seasonal changes in biochemical component of the adductor muscle, visceral mass, foot muscle and mantle were studied by biochemical analysis, from January to December, 2004. The reproductive cycle of this species can be classifed into five successive stages: early active stage (March to May), late active stage (May to July), ripe stage (July to September), partially spawned stage (August to October) and spentfinactive stage (October to March). Total protein content was the highest in the following order: adductor muscle, visceral mass, foot muscle, and mantle. Except for mantle, it was generally higher during the ripe and spawning stages, while lower during the spent/inactive stage. There were positive correlations in total protein contents among adductor muscle, foot muscle, and visceral mass. However, the correlations were not statistically significant. Total lipid content was the highest in the visceral mass; it was more than 5 or 6-fold higher than those in the adductor muscle, foot muscle, or mantle. The monthly change was also most dynamic in the visceral mass. It first Increased during the early active stage (March to May), decreased during late active stage (May to July), and then increased again rapidly during the spawning stage (September). There were a strong negative correlation in total lipid contents between foot muscle and adductor muscle (r = -0.634, p = 0.027), and a strong positive correlation between adductor muscle and mantle (r = 0.665, p = 0.018). Glycogen contents showed more or less similar pattern to total lipid contents in the adductor muscle, foot muscle, and visceral mass. It was higher during the early active and spawning stages, while lower during the late active and spent/inactive stages. There was no statistically significant correlation in glycogen contents among different tissues. Especially, total lipid content showed a negative correlationship between the foot muscle, adductor muscle, visceral mass and mantle. Therefore, these results indicate that the nutrient content of the foot muscle, adductor muscle, viseral mass and mantle changed in response to gonadal energy needs.

  • Reproductive cycle with the gonadal development of Ruditapes philippinarum can be classified into five successive stages by histological observations: early active stage (January to March), late active stage (February to May), ripe stage (April to August), partially spawned stage (May to October), and spent/inactive stage (August to February). Changes in total lipid contents in the adductor muscle tissues reached the maximum in the inactive and early active stages (January) and sharply decreased in the early and late active stages (February), while their contents in the visceral mass tissues reached the maximum in the ripe and partially spawned stages (April) and gradually decreased in the partially spawned stage (to October). On the whole, changes in total lipid contents showed a negative correlationship between the adductor muscle and visceral mass tissues (r = -0.699, p<0.05). Changes in glycogen contents in the adductor muscle tissues reached the maximum in the late active and ripe stages (April) and rapidly decreased in the partially spawned stage (May to October), while their contents in the visceral mass tissues reached the maximum in the early and late active stages (February) and rapidly decreased in the late active stage (March). Thereafter, their levels gradually increased in the ripe and partially spawned stages (April to July). On the whole, changes in glycogen contents appeared no correlationship between the adductor muscle and visceral mass tissues (r = 0.062, p > 0.05). These results indicate that the adductor muscle and visceral mass tissues are an important energy storage and nutrient supply organ in the Manila clams, and the nutrient contents of the adductor muscle and visceral muscle tissues change in response to gonadal energy needs.

Early Life History and Spawning Behavior of Chinese Minnow, Rhynchocypris oxycephalus Reared in the Laboratory (실험실에서 사육한 버들치의 산란습성(産卵習性) 및 초기생활사(初期生活史))

  • This study was conducted to observe the spawning behavior and early life history of chinese minnow, Rhynchocypris oxycephalus reared in the laboratory. The spawning period of Rhynchocypris oxycephalus is from May to June in nature. The eggs of Rhynchocypris oxycephalus were spawned on the sand and surface of the gravel. The fertilization eggs were demersal in shape and adhesive, released as a clump forming a thin layer and their diameter were 1.70~1.90mm (mean 1.80 mm, n=20). Hatching of eggs was started in 88 hour 45 minute after fertilization at water temperature $19{\pm}0.5^{\circ}C$ and finished in 90 hour. Newly-hatched larvae were measured 4.87~5.02 mm in total length (TL, mean 4.94 mm), mouth and anus were not opened. 6 days old larvae were 6.32~6.56 mm in TL (mean 6.44 mm). Yolk sac was almost absorbed, mouth and anus was began to open. 13 days old larvae were 6.74~6.91 mm in TL (mean 6.82 mm). Part of Dorsal fin was began to rising and myomere number was 15+23=38. 25 days after hatching, total length of larvae was 8.45~8.60 mm (mean 8.52 mm). Dorsal and anal fin rays became differentiated, and also caudal part of the notocord flexion was achieved at $45^{\circ}C$. In the time, growth rate was higher than the other stage. Aggregate numbers of all fin rays were completed at 16.39~16.57 mm in TL (60 days after hatching), at which time the larvae reached the juvenile stage, but fin-fold on ventral was remained yet. External features of adult specimens were almost completed at 80 day old juveniles (18.69~18.87 mm in TL).

Effects of Bisphenol A to interspecific hybrids between marine medaka Oryzias dancena and javanese medaka O. javanicus (바다송사리, Oryzias dancena와 자바송사리, O. javanicus 간 잡종에 대한 비스페놀 A의 효과)

  • Bisphenol A (BPA) is mainly used in the production of epoxy resins and polycarbonate plastics, which is a known endocrine disruptor and acutely toxic to aquatic organisms. In this study, estrogenic effect of BPA was investigated on hybrid between Oryzias dancena and O. javanicus (ODJ). ODJ were exposed to BPA of various concentrations (eg. 2.5 mg/L, 5.0 mg/L and 10.0 mg/L) for 56 days. The growth rate, abnormality and the ratio of female and male were observed in test group and control group. As a result, the growth was $14.7{\pm}2.0$ mm in total length (TL) in 2.5 mg/L, $13.7{\pm}2.5$ mm in 5.0 mg/L, $12.8{\pm}2.5$ mm in 10.0 mg/L in test group while it was $18.0{\pm}1.2$ mm in TL in control group which was not treated with bisphenol A. The result showed that the growth decreased as the concentration of BPA increased. The abnormality rate was 13.6% in control group, 65.4% in 2.5 mg/L, 81.3% in 5.0 mg/L and 98.1 % in 10.0 mg/L which showed increase in abnormality as an increase of BPA concentration. As a result of analyzing ratio of sex in the test group and control group, 6.0% was examined to be interspecific in controls, 76.9% in 2.5 mg/L and 100.0% in 5.0 mg/L and 10.0 mg/L. In conclusion, these results suggest that BPA has estrogenic effect on ODJ.

Evaluation of Commercial Diets and Replacement of Raw Fish with Formulated Diets in Moist Pellet for Olive Flounder (Paralichthys olivaceus) (시판 넙치사료의 평가와 모이스트펠렛사료중 냉동어류를 분말배합사료로 대치)

  • Two studies were carried out: to compare commercial flounder diets (C1-C6) with a laboratory-formulated diet (S), and to evaluate moist pellet (MP) prepared with or without raw fish for olive flounder culture. In experiment I, three replicates of 20 fish (42 g in average) each were fed S or each of C1-C6 for four weeks at $17^{\circ}C. For experiment 2, three replicates of 400 fish (78 g in average) each were fed $100\%$ laboratory-formulated grower diet (G), 50\%\;G+50\;%$ frozen mackerel containing $70\%$ moisture or $50\%\;C6$ (the same diet as that used in Exp. 1) +$50\%$ frozen mackerel for six weeks. Weight gain of fish fed S was significantly (P< 0.05) higher than that of fish fed C2 or C3 (26.7 vs 15.4 or 17.5g/fish over the 4-week period, respectively). Feed efficiency was significantly (p<; 0.05) lower in fish fed C2 than in fish fed S but was not different (P> 0.05) between the fish fed Sand the other commercial diets. No significant differences in weight gain were found among the fish fed $100\%\;G,\;50\%\;G+50\%$ frozen mackerel or $50\%$ C6+frozen mackerel. The results of our studies indicate that most of the commercial flounder diets can be improved for better growth, when we compared these diets with a laboratory-formulated diet, and the growth of flounder fed MP containing no raw fish was as good as that of those fed MP prepared with raw fish.

Evaluation of Dietary Lipid Sources for Juvenile Abalone (Haliotis disucs hannai) (참전복 치패 사료의 지질원 평가)

  • Three feeding experiments were carried out to evaluate the effects of fatty acids or lipid sources in diets on the survival, growth and body composition of junenile abalone(Haliotis discus hannai). Diets used in this study contained casein or fish meal as a protein source. Three replicate groups of abalone averaging 160 mg were fed with casein diets containing 12:0, 18:1, 18:2n-6, 18:3n-3, n-3HUFA, squid liver oil (SO), corn oil (CO), beef tallow (BT), SO+CO, and SO+BT, or fed fish meal diets containing SO, CO, BT, SO+CO, SO+BT and not supplemental oil for 20 weeks, respectively. Survival rate, weight gain and soft body weight of abalone were not significantly affected by different fatty acids in the casein diets (P>0.05). Weight gain, soft body weight and shell length of abalone fed the casein diets containing SO, SO+CO or SO+BT were significantly higher (P<0.05) than those of abalone fed the casein diets containing CO or BT. Survival rate of abalone fed the fish meal diets was not influenced by different lipid sources (P>0.05). Weight gain and soft body weight of abalone fed the fish meal diets containing beef tallow (BT or SO+BT diet) were lower than those of abalone fed the diet not added oil or diets containing SO, CO and/or SO+CO(P<0.05). These data indicated that SO or SO+CO was good dietary lipid source for juvenile abalone, and that these oil supplement in diet was not necessary when fish meal was used as a protein source.

Effects of Nursery Environmental Factors on the Growth of Pacific Oyste, Crassostrea gigas (양식어장 환경요인이 참굴 (Crassostrea gigas)의 성장에 미치는 영향)

  • This study was performed to investigate the effects of environmental parameters on the growth of Pacific oyster, Crassostrea gigas at old suspended culturing grounds of Korsung Bay and newly deveoped bottom, culturing grounds of Haenam region located in southern coast of Korea. The Kosung Bay was semiclosed and water exchange occurred through the small mouth and the current speed was very slow as the value of below 0.5kn. Haenam was opened and littoral zone. Current speed of the surroundings of this Bay about to 5.0knand transparency was low with the mean depth of 1.2m by the resuspension of the sediments. Water temperature and salinity regimes were suitable for oysters at those sites, however the mean value of those parameters and seasonal fluctuations was high at Kosung. The mean concentration of dissolved oxygen in survey area was above $5.0mg/\ell$. In Kosung, however, it was decreasing in June and showed the minimum with the concentration of $4.0mg/\ell$ in August at the bottom layer and recovered in September. Eutrophication parameters such as COD were 1.44 mg/l at this location. This mean value was higher than that of the Haenam Bay's $0.96mg/\ell$. The men value of chlorophyll-$\alpha$ at Haenam was two times higher than the other sites. Growth of shell height and meat weight of oysters at Kosung Bay were higher and the fatness was lower compared to thost of Haenam. As a result, absolute growth of oysters was high at Kosung and allometric growth, such as fatness, was good at Haenam. Low water temperature and frequent exposure of oysters to the air during ebb tide may retard the shell growth. On the other hand, relatively high food availability was profitable for the fatness at haenam site.
