Eupafolin, a constituent of the aerial parts of Phyla nodiflora, has neuroprotective property. Because reducing the synaptic release of glutamate is crucial to achieving pharmacotherapeutic effects of neuroprotectants, we investigated the effect of eupafolin on glutamate release in rat cerebrocortical synaptosomes and explored the possible mechanism. We discovered that eupafolin depressed 4-aminopyridine (4-AP)-induced glutamate release, and this phenomenon was prevented in the absence of extracellular calcium. Eupafolin inhibition of glutamate release from synaptic vesicles was confirmed through measurement of the release of the fluorescent dye FM 1-43. Eupafolin decreased 4-AP-induced [Ca2+]i elevation and had no effect on synaptosomal membrane potential. The inhibition of P/Q-type Ca2+ channels reduced the decrease in glutamate release that was caused by eupafolin, and docking data revealed that eupafolin interacted with P/Q-type Ca2+ channels. Additionally, the inhibition of calcium/calmodulin-dependent protein kinase II (CaMKII) prevented the effect of eupafolin on evoked glutamate release. Eupafolin also reduced the 4-AP-induced activation of CaMK II and the subsequent phosphorylation of synapsin I, which is the main presynaptic target of CaMKII. Therefore, eupafolin suppresses P/Q-type Ca2+ channels and thereby inhibits CaMKII/synapsin I pathways and the release of glutamate from rat cerebrocortical synaptosomes.
Purpose: Barely sprout is a well-known oriental herbal medicine with a wide range of health benefits. Recent studies have provided scientific evidence of its therapeutic effects with expanded application. This study investigated anti-melanogenic effect of barley sprout water extract (BSE) in murine melanocyte B16F10. Methods: Various concentrations (0, 50, 125, and $250{\mu}g/mL$) of BSE and arbutin (150 ppm) were applied to B16F10 stimulated with or without alpha-melanocyte stimulating hormone (100 nM) for 72 hours. The whitening potency of BSE was determined altered cellular melanin contents. Activity and expression of tyrosinase and microphthalmia-associated transcription factor (MITF) were also assayed. Results: Experimental results revealed that treatment with BSE reduced cellular melanin production by approximately 40% compared to the control. Molecular findings supported that suppressed activity and expression of tyrosinase and MITF proteins by BSE were associated with declined cellular melanogenesis. Furthermore, anti-melanogenic effect of BSE ($250{\mu}g/mL$) was similar to that of arbutin, a commonly used whitening agent. Lastly, polyphenols including p-coumaric, ferulic, and vanillic acids were identified in BSE using HPLC analyses. They might be potential active ingredients showing such melanogenesis-reducing effect. Conclusion: BSE was evident to possess favorable anti-melanogenic potency in an in vitro model. As a natural food sourced material, BSE could be an effective depigmentation agent with potential application in pharmaceutical and cosmetic industries.
Obesity is considered as a primary health problem over the past century in line with life environmental changes. It is mainly associated with increased risk of numerous chronic diseases which may significantly reduce health-related quality of life. Therefore, efforts to reduce weight should be performed. This study suggested a novel approach to reduce body fat by applying external stimulation which is micro-current stimulation (MCS). In this clinical study, we evaluated the potential effects of MCS for reduction of abdominal fat and weight loss. Prior to the clinical test, computational simulation was conducted to find the proper MCS conditions that allow externally applied stimulation to reach the internal fat section from the external skin. Particularly, the clinical study evaluated the unilateral effects of MCS for body fat loss and lipolysis without any additional limitations such as physical exercise and dietary therapy. The results showed that whole body fat, waist circumferences, and abdominal fat are gradually decreased after intervention in proportion to the time. From the results, we can estimate that MCS can be effective on the body fat loss by activation of lipolysis in human adipose.
본 연구는 단풍잎돼지풀 70% 에탄올 추출물의 항염증 활성을 검증하기 위해 수행되었다. 단풍잎돼지풀 70% 에탄올 추출물의 전자공여능 측정과 ABTS+ 라디칼 소거능을 측정한 결과, 1,000 ㎍/ml 농도에서 각각 84.1%와 92.5%의 효과를 나타냈고, 수렴활성 측정을 한 결과 1,000 ㎍/ml 농도에서 94.7%의 효과를 보였다. 단풍잎돼지풀 70% 에탄올 추출물의 항염증 효과를 측정하기 위해 lipopolysaccharide (LPS)로 유도된 RAW 264.7세포를 사용하여 효과를 측정하였다. 세포에서 단풍잎돼지풀 추출물의 세포독성을 측정하기 위해 MTT assay를 수행하였다. 그 결과, 500 ㎍/ml 농도에서 90% 이상의 생존율을 보였다. Nitric oxide 생산을 억제하는 효과를 측정한 결과, 단풍잎돼지풀 추출물에서 농도가 증가할수록 NO 생성이 감소되는 효과를 확인하였다. 단풍잎돼지풀 추출물의 단백질 발현효과를 western blot을 통해 25, 50, 100 ㎍/ml 농도에서 측정하였고, 양성 대조군으로 β-actin을 사용하였다. 그 결과, inducible nitric oxide synthase (iNOS), cyclooxygenase (COX)-2 단백질 발현효과는 100 ㎍/ml 농도에서 8.6%, 25.1%의 감소됨을 확인하였다. ERK1/2, p38, JNK와 Iκ-Bα의 단백질발현 효과는 인산화를 통해 확인하였고, 농도의존적으로 감소하였음을 확인하였다. mRNA 발현 억제 효과를 RT-PCR을 통해 25, 50, 100 ㎍/ml의 농도에서 측정하였고, 양성 대조군으로 GAPDH를 사용하였다. 그 결과, LPS로 유도된 대식세포에서 iNOS, COX-2, interleukin (IL)-1β, IL-6, TNF-α의 mRNA 발현 억제 효과는 농도가 증가할수록 발현이 감소됨을 확인하였다. 결론적으로 단풍잎돼지풀 추출물은 염증을 억제할 수 있는 가능성이 있는 항염증 소재의 가능성을 증명하였다.
복합처방인 승마갈근탕을 이용하여 화장품 및 식품에 응용하고자 항산화 및 항균효과를 검토하였다. 승마갈근탕의 전자공여능은 모든 추출물 100 ppm에서 50% 이상의 효과를 나타내었고, 특히 초임계 추출물의 경우 500 ppm에서 88%로 가장 높은 효과를 나타내었다. SOD 유사활성은 승마갈근탕 1,000 ppm에서 50% 이하의 다소 낮은 경향을 보였으나, 그 중 에탄올과 초임계 추출물의 경우 각각 46, 45의 효능을 나타내었으며, xanthine oxidase 저해활성의 경우 초임계 추출물 1,000 ppm에서 76%로 대조군인 vitamin C의 39%보다 높은 효과를 나타내었다. 승마갈근탕의 지방산패 억제능을 측정하기 위해 산화촉진제인 $Fe^{2+}$, $Cu^{2+}$ 이온을 첨가하여 측정한 결과 초임계 추출물의 $Cu^{2+}$ 첨가 시 500 ppm에서 68%의 효과를 나타내었고, $Fe^{2+}$를 첨가 시 100 ppm에서 60%의 지방산패 억제능을 나타내었다. 승마갈근탕의 clear zone 형성을 관찰한 결과, 추출물 모두 Escherichia coli를 제외한 모든 균주에 대해서 항균 효과가 나타내었다. Staphylococcus aureus와 Propionibacterium acnes의 경우 4 mg/disc에서 초임계 추출물에서는 1.45, 2.10 cm로 가장 높은 항균 효과를 나타내었고, Staphylococcus epidermidis의 경우, 에탄올 추출물이 1.82 cm의 항균효과를 나타내었다.
백련을 기능성 화장품 소재로 활용하기 위하여 항산화능, 미백 및 주름개선 효과를 검증한 후 W/O/W multiple emulsion에 첨가하여 기능성 화장품 소재로서의 가능성을 검증하였다. 백련 부위별 추출물의 총 폴리페놀 및 플라보노이드 함량을 측정한 결과 백련 잎 추출물이 213.5 mg/g 및 76.1 mg/g로 가장 높은 함량을 나타내었다. 백련 부위별 추출물의 전자공여능 측정 결과 백련 꽃과 잎 추출물 500 ppm에서 85% 이상의 효과를 나타내었으며, SOD 유사활성능 측정 결과 백련 잎 추출물 1,000 ppm에서 60%로 가장 높은 유사활성능을 나타내었다. 백련 부위별 추출물의 xanthine oxidase 저해활성 측정 결과 전반적으로 낮은 활성을 나타내었으나 백련 잎 추출물이 백련 꽃과 뿌리줄기 추출물에 비해 저해능이 높았다. 미백효과 검증으로 tyrosinase 저해활성을 측정한 결과 백련 꽃 추출물 1,000 ppm에서 36%의 저해능을 나타내었다. 주름억제 효과 측정 결과 백련 잎 추출물 1,000 ppm에서 30%의 elastase 저해활성을 나타내었다. 백련 부위별 추출물을 W/O/W multiple emulsion에 첨가하여 활성을 검증한 결과 전자공여능 측정 결과 백련 꽃과 잎 추출물 100 ppm에서 60% 이상의 활성을 나타내었다. 백련 부위별 추출물을 첨가한 W/O/W multiple emulsion의 SOD 유사활성능 측정 결과 백련 잎 추출물 1,000 ppm에서 30%의 활성을 나타내었으며, xanthine oxidase 저해활성 측정 결과 백련 부위별 추출물 모두 비교적 낮은 저해능을 나타내었다. 백련 부위별 추출물을 첨가한 W/O/W multiple emulsion의 tyrosinase 저해활성 측정 결과 백련 꽃 추출물 1,000 ppm에서 34%의 활성을 나타내어 추출물과 유사한 효능을 나타내었으며, elastase 저해활성 측정 결과 백련 잎 추출물 1,000 ppm에서 55%의 활성을 나타내었다.
본 연구에서는 1-chloro-2,4-dinitrobenzene (DNCB)에 의해 유도된 아토피성 피부염 마우스 모델에서 고삼, 감초 및 백선피 추출물과 그들 복합 추출물(GGB)의 항아토피 효능을 평가하였다. 아토피성 피부염의 병리학적 형태인 홍반, 귀부종, 부종, 각질과 상처는 복합 추출물 중 고삼, 감초 및 백선피 추출물이 3:1:1로 혼합된 GGB-1 처리군에서 다른 시료 처리군들에 비하여 유의적인 억제 효능을 나타내었다. 또한 면역 기관인 비장의 무게도 GGB-1 처리군에서 감소하였고, 간의 무게는 정상군과 시료 처리군의 값이 유사한 결과를 나타내었다. 아울러 GGB-1 처리군에서 비만 세포의 침윤이 감소되었으며, 혈청 중 IgE의 농도 역시 GGB-1 처리군에서 가장 낮은 값을 보였다. 비록 단일 한약재와 그들의 혼합물 처리군에서는 면역 사이토카인(IFN-${\gamma}$, IL- 1, 4, 5, 6, 13, $1{\beta}$, and TNF-${\alpha}$) 의 값이 유의적으로 감소되었으나, 여러 가지 결과들을 종합하여 볼 때 고삼, 감초 및 백선피 추출물이 3:1:1로 혼합된 복합 추출물인 GGB-1이 아토피성 피부염의 개선에 효과적인 약물로 판단된다.
The purpose of this study is to evaluate the antioxidant activity and hair growth effects of a natural herbal ethanol extract, Samhwang-Sasimtang(SS). In case of antioxidant ability of SS, the content of phenolic compounds was 28.44mg/g. The extract showed strong electron donating ability and free radical scavenging activity in a concentration-dependent manner. SOD-like activity also rose through increasing the concentrations of SS. In order to estimate the hair growth effects, the extract was applied to the back of seven-week-old C57BL/6 male mice ($150{\mu}{\ell}$ a day, five days a week, for four weeks) in four groups (C, control, saline; PC, positive control, 3% minoxidil; E1, experimental 1, 1% SS; E2, experimental 2, 2% SS). Ten mice were assigned to each group and five mice in each group were sacrificed at weeks 2 and 4, respectively. There was no a significant difference in body weight change among experimental groups. In macroscopic observation of hair growth at week 4, the scores of hair growth on the backs of mice were 80, 60, 40 and 20% in the PC, E2, E1 and C groups. In terms of histological observation, the ratio and thickness of hair follicles, the enzyme activities of ALP and ${\gamma}$-GT, immunohistological examination of IGF-1 and VEGF which are the hair growth factors, in each group were significantly high in this order: PC, E2, E1 and C group, at week 4. Meanwhile, hair growth inhibition factors, TGF-${\beta}1$ and Caspase-3, were reduced in PC, E1 and E2 groups compared with C group. These results indicate that SS extract may be effective in promoting hair growth, and suggest that it can be used practically as a superior natural agent for hair growth promotion.
목 적: 본 연구는 길경(Platycodon grandiflorum root)를 함유한 복합 한약재 추출물(ExMH-PGR)의 골다공증 예방 및 치료효과를 알아보기 위해 MC3T3-E1 조골세포주와 RAW 264.7 파골세포주를 이용하여 in vitro 수준에서 검증하였다. 방 법: 배양액에 다양한 농도의 ExMH-PGR를 첨가하여 MC3T3-E1 세포와 RAW 264.7 세포의 세포증식을 비교하였다. MC3T3-E1 세포에서 Alkaline phosphatase (ALP) 활성, 콜라겐 합성, 오스테올칼신 생성, 무기질 축적을 분석하였다. RAW 264.7 세포에서 Tartrate-resistant acid phosphatase (TRAP) 활성과 actin ring 형성 정도를 분석하였다. 결 과: ExMH-PGR는 $25{\mu}g/mL$ 농도까지 유의한 수준으로 ALP 활성, 콜라겐 합성, 그리고 오스테올칼신 형성을 증가시켰다. $50{\sim}200{\mu}g/mL$의 ExMH-PGR은 TRAP 활성과 actin ring 형성을 유의하게 억제했다. 결 론: ExMH-PGR은 조골세포의 활성을 촉진하고 파골세포의 활성을 억제하는 효과가 있어 골다공증 치료에 유용할 가능성이 있다.
목적 : 한의학에서 골절이나 신장질환에 사용되어 왔던 골쇄보열수추출물 약침액이 항염증 및 항산화활성에 대한 효능이 있는지를 연구하고자 한다. 방법 : 골쇄보열수추출물 약침액이 세포독성에 미치는 영향을 관찰하기 위하여 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay를 실시하였다. 골쇄보열수추출물 약침액이 항염증효능에 미치는 영향을 관찰하기 위하여 lipopolysaccharide(LPS)로 유도된 RAW264.7 대식세포에서의 산화질소(NO) 및 프로스타글란딘($PGE_2$) 생성 억제력을 관찰하였다. 또한 골쇄보열수추출물 약침액이 항산화활성에 미치는 영향을 관찰하기 위하여 1,1-diphenyl-2-pciry hydrazyl(DPPH) radical 소거능을 관찰하였다. 결과 : 골쇄보열수추출물 약침액은 $50{\sim}400{\mu}g/ml$ 농도에서 세포독성이 나타나지 않았다. 골쇄보열수추출물 약침액은 200 및 $400{\mu}g/ml$ 농도에서 LPS로 유도된 NO 및 $PGE_2$ 생성을 통계학적으로 유의하게 감소시켰다. 골쇄보추출물은 $50{\sim}400{\mu}g/ml$ 농도에서 DPPH 소거능을 관찰한 결과 농도의존적으로 활성화되는 것을 관찰하였다. 결론 : 골쇄보열수추출물 약침액은 항염증 및 항산화 효과가 있을 것으로 사료된다.
