The Protective and Inhibitory Effect of Antioxidants Found in Broussonetia kazinoki Siebold against Oxidative DNA Damage

  • Oxidative DNA damage negatively affects humans and the research is currently ongoing to find ways to reduce oxidative stress. Oxidative stress has been identified as a key factor in triggering various diseases. Thus, its alleviation is important for human health. Broussonetia kazinoki (B. kazinoki) has been used in traditional Korean medicine as a dermatological therapy to treat burns, pruritus, and acne. B. kazinoki is generally segregated into peeled root (PR), root bark (RB), peeled stem (PS), and stem bark (SB). To assess these components for their antioxidant activity and protection against DNA damage, their ethyl acetate fractions were examined by 1,1-diphenyl-2-picryl hydrazyl (DPPH) and 2,2'-Azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging assay. As a result of confirming the expression of factors involved in attenuating DNA damage, the protective effect of SB on oxidative stress suppressed the expression of p-p53 and γ-H2AX. Additionally, the levels of p53 and H2AX mRNA were significantly downregulated. In conclusion, these results indicated that the SB component of B. kazinoki had the potential to be used as an effective natural antioxidant compared to the other parts of the plant.

The characteristics of treatment of smallpox and its significance recorded in the Duchanggyeongheombang (『두창경험방(痘瘡經驗方)』에 나타난 두창 치료의 특징과 그 의의)

  • Objectives : The paper seeks to delve into a study of smallpox, specifically as one of the most representative contagious diseases during the Chosun era and how it was treated during that time. The lack of traditional medical texts that deal with smallpox has led this paper to dive into Duchanggyeongheombang. Methods : Everything related to edition and copy, and the author was organized based on pre-existing studies. The copy that is assumed as the original copy was considered and studied. The book was compared to and reviewed with Donguibogam, and the differences between the book and Korean translation were given attention as well. Results : Park Jin-hee is assumed to have authored Duchanggyeongheombang, and it is like that was had access to texts published by the state, one of which includes Donguibogam. He utilized the periods with smallpox outbreaks as his frame of study and put together a series of treatment for different symptoms. To this, he attached various medical charts, and added Korean translation for higher usability. In his book, he placed emphasis on spleen and stomach, but when needed, he did not shy away from using cold and coolness herbs. Conclusions : Duchanggyeongheombang was influenced partially by Donguibogam, but the book was given new life through the author Park Jin-hee's edition. Medical charts were added to increase its usability, and Korean translation was provided for wider audience. Its scope of prescription is very wide, and it provided the reasonings behind its medical judgements based on actual clinical studies instead of being tied down to traditions or taboos.

여성들의 찜질방 이용 현상에 관한 일상 생활 기술적 연구

  • The ZZimzilbang is a room where many women eagerly go due to the special meaning women give to it related to their health. It is a different type of sauna room which maintains low degree of temperature, consisting of an Ondol room(Korean under-floor heating system, hypocaust) built with mineral stone radiating ultrared rays. Even though many women mention that they utilize it for health, there is no precise evidence of the effects on their health. This ethnographic study sought to define the phenomenon from the perspective of the women who experienced the ZZimzilbang. A convenience sample of 27 women was interviewed during a 15 month period from December 1995 to July 1997 in 12 ZZimzilbangs located in Seoul, Korea. The mean ages of the women 57.3 years ; seventeen women were housekeeper and only eight women had job : twenty women were married and three women among them were widows. The main reasons women patronize the place are : for mitigation and healing of physical signs and symptoms : composure ; safe lodging and boarding : control of outward appearance : control of health ; meeting and fellowship with friends : and custom. The outcomes of the utilization of the place were : mitigation of physical signs and symptoms : psychological tranquility : cosmetic and diet ; good use of spare time : and utilization of services provided there. Most women who visited ZZimzilbang for relief of physical signs and symptoms strongly mentioned a correlation to inadequate Sanhujori, the traditional postpartal and postabortal care for woman. Some of specific kinds of services provided in that place were alternative therapy such as acupuncture, negative cupping, finger-pressure, mugwort steam and various kinds of massage including massage of blood vessels that are influenced by Oriental medicine ; health education of breathing such as abdominal breathing or Danjeon(단전호흡) : and selling of many things including health foods, drugs for osteoporosis, and eutrophics. This study suggests that professional caregivers should further study this phenomenon for the development of adequate care of women with a resulting important in their quality of life.

Antigastritic and Anti Helicobacter pylori of Trifolirhizin from Sophora Radix (고삼 Trifolirhizin의 항위염 및 항 H. pylori 균 효과)

  • Sophorae Radix, the dried roots of Sophora flavescens Aiton (Leguminosae), has been used in oriental traditional medicine for treatment of skin and mucosal ulcers, sores, gastrointestinal hemorrhage, diarrhea, inflammation and arrhythmia. Present study was carried out for the gastroprotective effect of trifolirhizin from Sophora flavescens. This reports were evaluated antibacterial activity against Helicobacter pylori and HCl ethanol-induced gastric lesion in rats and showed the significant effectiveness. In pylorus ligated rats, the treatments of trifolirhizin showed decrease in the volume of gastric secretion and acid output. Also we evaluated the antibacterial activity against H. pylori treated with methanol extract and trifolirhizin from Sophora flavescens had a equivalent antibacterial activity with ampicillin against H. pylori at the dose of $100{\mu}g/mL$. It may be regarded that the antigastritic effects and antibacterial activity of trifolirhizin from Sophora flavescens are originated from reduction of total acid output identified by gastric secretion reduce, free radical scavenging effects and the antibacterial activity against H. pylori.

Lindera obtusiloba Extends Lifespan of Caenorhabditis elegans

  • Lindera obtusiloba has been widely used as a traditional medicine for the treatment of lots of diseases, including abdominal pain, bruise, and hepatocirrhosis. Here in this study, we elucidated the lifespan-extending effect of methanolic extract of Lindera obtusiloba (MLO) using Caenorhabditis elegans model system. We found that MLO has potent lifespan extension activities under normal culture condition. Then, we determined the protective effects of MLO on the stress conditions such as osmotic, thermal and oxidative stress. To reveal possible mechanism of MLO-mediated lifespan, we further investigated the effect of MLO on the antioxidant enzyme activities and intracellular ROS levels. Our results demonstrated that superoxide dismutase and catalase activities were significantly up-regulated by MLO treatment, resulted in reduced intracellular ROS levels. In this work, we also tested whether MLO-mediated longevity activity was associated with aging-related factors such as food intake and growth. Our data revealed that both of pharyngeal pumping rate and body length were significantly shifted by MLO treatment, indicating these factors were involved in MLO's lifespan-extension effects. Although MLO induces reduction in food intake, the body movement of MLO-fed aged worms was not decreased, compared to untreated control worms, indicating MLO might extend lifespan without affecting healthspan.

  • With the advanced learning and the diversity of topics, researchers on each area keenly feel the need of precise and a quick discovery of required information at any time. This study presents a way of constructing the automatic mapping system that can compare and analyze duplicated data and that describes the result by building an effective reference extraction method and another way of correcting the wrong form of used Chinese characters with Traditional Korean Medicine dictionary. With this innovation, data duplication on references and Chinese characters errors can be fixed. Under the situation that a number of references of newly published papers that can continuously be extracted.

Hepatoprotective Effect of Fermented Artemisia princeps PAMPANINI by Lactic Acid Bacteria (발효강화쑥의 간장해 보호효과)

  • Artemisia princeps PAMPANINI has been used in traditional medicine of the treatment of inflammatory, liver dysfunction and order disorder in the far east countries including Korea. The present study was carried out to investigate the hepatoprotective effects of ethanol extract of Artemisia princeps (AP) and its fermented agents (AP-F) by lactic acid bacteria derived from human intestinal bacteria on liver injured rat induced by $CCl_4$ and d-galactosamine. Hepatoprotective activity was monitored by estimating serum ALT, AST, ALP, LDH, superoxide dismutase (SOD), glutathione redeuctase (GR) and glutathione peroxidase (Gpx) activities in the liver injured by hepatotoxin. Pretreating rats with AP or AP-F at the same dosage regimen significantly suppressed the acute elevation of serum transaminase, ALP, LDH and GR activities, and significantly increased the lowering of blood SOD and GR activites induced by hepatoxin. Based on these findings, it is presumed that AP and APF may have the hepatoprotective effect on $CCl_4$ and d-galactosamine-induced hepatotoxicity rat.

Smilax guianensis Vitman Extract Prevents LPS-Induced Inflammation by Inhibiting the NF-κB Pathway in RAW 264.7 Cells

  • Nutraceutical treatments can reduce inflammation and prevent the development of inflammatory diseases. In this study, the anti-inflammatory effects of Smilax guianensis Vitman extract (SGE) were examined. SGE suppressed lipopolysaccharide (LPS)-mediated nitrite production in RAW 264.7 cells. SGE also prevented the LPS-induced expression of inducible nitric oxide synthase (iNOS) but not cyclooxygenase (COX)-2. Western blot analysis showed that SGE attenuated LPS-induced phosphorylation of IκB kinase (IKK), inhibitor of kappa B (IκB), and p65. Additionally, SGE inhibited LPS-induced IκB degradation in RAW 264.7 cells. Western blot analysis of the cytosolic and nuclear fractions, as well as immunofluorescence assay results, revealed that SGE suppressed LPS-induced p65 nuclear translocation in RAW 264.7 cells. Moreover, SGE reduced LPS-induced interleukin (IL)-1β, IL-6, and tumor necrosis factor-α (TNF-α) mRNA expression and IL-1β and IL-6 protein expression in RAW 264.7 cells. Collectively, these results indicate that SGE suppresses the NF-κB signaling pathway and thereby inhibits the production of NO, IL-1β, and IL-6.

Anti-inflammatory Activity of Extracts from Ultra-Fine Ground Saururus chinensis Leaves in Lipopolysaccharide-Stimulated Raw 264.7 Cells

  • Bioactive components of ultra-fine ground Saururus, the extraction yield increases when the leaves are ultra-fine ground. Comparison of normal-ground and ultra-fine ground Saururus chinensis leaves showed that the solid content and antiinflammatory activity of ultra-fine ground extracts was higher than that of normal-ground extracts. Lipopolysaccharide (LPS)-stimulated Raw 264.7 cells were treated with different concentrations of Saururus chinensis extract and the amount of nitric oxide (NO) was determined; LPS-treated cells produced 2 times more NO than cells that were not treated with LPS. Moreover, the NO production in cells treated with Saururus chinensis extract was inhibited in a concentration-dependent manner. Because the stimulant-induced NO production is regulated by the inducible nitric oxide synthase (iNOS), we measured the iNOS protein level to elucidate the mechanism by which the NO production was inhibited. We found that the amount of iNOS decreased dose-dependently. It was reduced by 53% at a Saururus chinensis extract concentration of $100{\mu}g/mL$. The protein expression of cyclooxygenase-2 (COX-2) in LPS-treated Raw 264.7 cells was inhibited by 31% at $100{\mu}g/mL$ of Saururus chinensis extract. Gel shift of the nuclear factor kappa B-DNA complex occurred in LPS-treated cells and the intensity of the band decreased gradually in a concentration-dependent manner. Ultra-fine ground Saururus chinensis extract had a concentration-dependent inhibitory effect on the production of prostaglandin $E_2$, tumor necrosis factor ${\alpha}$, interleukin $1{\beta}$ (IL-$1{\beta}$), IL-6, and IL-8 in LPS-treated Raw 264.7 cells, i.e., at $50{\mu}g/mL$ of Saururus chinensis extract, their levels were decreased by 53, 67, 52, 37, and 21% respectively.

Quality Characteristics of Inner Beauty Food (Mosidae Tofu) by the Addition of Adenophora remotiflora Powder (모시대 분말을 첨가한 미용식품(모시대 두부)의 품질 특성)

  • In this study, physio-chemical, mechanical, and sensory characteristics of Tofu containing 0, 0.5, 1.0, and 2.0% Adenophora remotiflora powder were examined. In addition, we examined the potential of utilizing Adenophora remotiflora powder as a functional food material by estimating total phenol contents, electron-donating abilities, and hydroxyl radical scavenging activities of the hot water and ethanol extracts of Adenophora remotiflora powder in the range from 0~2.0%. The total phenol content of the ethanol extracts of Adenophora remotiflora powder was $487.93{\mu}g/mL$ while the that of the water extract of Adenophora remotiflora powder was $403.70{\mu}/mL$. The electron-donating abilities of the ethanol and water extracts of Adenophora remotiflora powder were 75.37 and 86.10%, respectively. The hydroxyl radical scavenging activities of the ethanol and water extracts of Adenophora remotiflora powder were 65.50 and 66.22%, respectively. We also evaluated the quality characteristics of Tofu containing Adenophora remotiflora powder. In the case of color values, as the level of Adenophora remotiflora powder increased, the values of L (lightness) and a (redness) decreased, whereas that of b (yellowness) increased. In the case of mechanical properties, as the level of Adenophora remotiflora powder increased, hardness, gumminess, and chewiness values increased (p<0.05), whereas springiness and cohesiveness values decreased (p<0.05). In the case of sensory evaluation, MPT1.0 scored significantly higher in color, flavor, tenderness, texture, and overall quality. To sum up, Tofu containing 1% Adenophora remotiflora powder showed the highest overall preference.
