Plant Growth-Promoting Capabilities of Diazotrophs from Wild Gramineous Crops (야생 벼과식물 유래 질소고정세균의 식물생장촉진 관련 특성)

  • Since there could be more and rather various diazotrophs in rhizosphere of wild crops than those in rhizosphere of cultivars, some wild gramineous crops grown in Korea were collected for isolating nitrogen-fixing bacteria. Six diazotrophs were purified from their roots using nitrogen-free media. The isolated bacteria were partially identified as 4 genera by 16S rDNA sequence analysis: Stenotrophomonas sp., Bosea sp., Klebsiella sp., and Azorhizobium sp. By PCR amplification and sequence analysis, DNA fragments extracted from all isolates turned out to have an individual nifH homologous gene. Five isolates (KNUC163, KNUC165, KNUC169, KNUC170, and KNUC171) showed auxin activity and four isolates (KNUC163, KNUC166, KNUC170, and KNUC171) produced siderophores. Especially,3 strains of S. maltophilia showed both auxin and siderophore activities. In conclusion, the isolated nitrogen-fixing bacteria might have capabilities for plant growth promotion.

Fermentative Bio-Hydrogen Production of Food Waste in the Presence of Different Concentrations of Salt (Na+) and Nitrogen

  • Fermentation of food waste in the presence of different concentrations of salt ($Na^+$) and ammonia was conducted to investigate the interrelation of $Na^+$ and ammonia content in bio-hydrogen production. Analysis of the experimental results showed that peak hydrogen production differed according to the ammonia and $Na^+$ concentration. The peak hydrogen production levels achieved were (97.60, 91.94, and 49.31) ml/g COD at (291.41, 768.75, and 1,037.89) mg-N/L of ammonia and (600, 1,000, and 4,000) $mg-Na^+/L$ of salt concentration, respectively. At peak hydrogen production, the ammonia concentration increased along with increasing salt concentration in the medium. This means that for peak hydrogen production, the C/N ratio decreased with increasing salt content in the medium. The butyrate/acetate (B/A) ratio was higher in proportion to the bio-hydrogen production (r-square: 0.71, p-value: 0.0006). Different concentrations of $Na^+$ and ammonia in the medium also produced diverse microbial communities. Klebsiella sp., Enterobacter sp., and Clostridium sp. were predominant with high bio-hydrogen production, while Lactococcus sp. was found with low bio-hydrogen production.

Evaluation and Isolation of Phytin Phosphohydrolyzing Bacterial Population in the Rumen

  • A series of experiments was conducted to evaluate phytin phosphohydrolysis actlVlty in the rumen and to isolate phytase positive rumen bacteria. Endogenous phytase activity of wheat bran was estimated and compared with that of bacterial phytin phosphohydrolysis. Substantial phytase activity was detected in wheat bran during in vitro rumen incubation. Bacterial phytase activity was suggested not to be high. Only two facultative anaerobes, Klebsiella sp. and Corynebacterium sp. were isolated as phytase producing organisms. These belonged to a minor microbial group in the rumen population. Protozoal fraction showed an initial velocity of phytin phosphohydrolysis 7 times higher than the bacterial fraction.

Isolation and Cultural Characteristics of Styrene Dimer [Endocrine Disrupter] Biodegrading Microorganism (Styrene dimer [환경호르몬 물질] 분해균주의 분리 및 배양특성)

  • We examined the culture conditions and degrading characteristics of styrene dimer (endocrine disrupter) using microorganism. The isolated microbe were consisted of 3 kinds of strain. The strains were identified to Pseudomonas sp. and Klebsiella pneumoniae by API 20E kit, but one was not identified. Single strain was not grown on the C-medium containing styrene dimer. However the complex strain YH3 could grow and we confirmed it by the broth color and O.D$_{660nm}$ (optical density 660 nm). The optimal culture conditions of complex strain YH3 were 35$^{\circ}C$, 1,000 ppm (v/v) of styrene dimer and pH 7.0, respectively. In tolerance test against the organic solvents, the complex strain YH3 could grow above log P=3.1, and could degrade ethyl benzene and 2,4-D, one kind of herbicide. As a result of TLC (Thin Layer Chromatography) analysis, we confirmed that the metabolite of styrene dimer was created by YH3 after 5th day, but not at control samples.

The microbial diversity analysis of the Korea traditional post-fermented tea (Chungtaejeon) (한국 전통 미생물발효차(청태전)의 미생물 군집분석)

  • Tea is the most popular beverage in the world. In fact, there are mainly three different kinds of tea (Green tea, black tea, and post-fermented tea). Post-fermented tea is produced by the microbial fermentation process using sun-dried green tea leaves (Camellia sinensis) as the raw material. Chungtaejeon was a traditional tea introduced in the age of the ancient three states and is the only "Ddeok-cha or Don-cha" culture in the world that survived on the southwestern shore of Republic of Korea. In this study, the structures of the bacterial community involved in the production of oriental traditional post-fermented tea (Chungtaejeon) were investigated using 16S rRNA gene analysis. The 16S rRNA gene analysis of dominant microbial bacteria in post-fermented tea confirmed the presence of Pantoea sp., and Klebsiella oxytoca. Phylogenetic analysis suggested that the taxonomic affiliation of the dominant species in the post-fermented tea was ${\gamma}$-proteobacteria. As a result of the microbial community size analysis, it was confirmed that the size of the microbial communities of Chungtaejeon was the largest compared to other teas
