SETDB1 mediated FosB expression increases the cell proliferation rate during anticancer drug therapy

  • The efficacy of anticancer drugs depends on a variety of signaling pathways, which can be positively or negatively regulated. In this study, we show that SETDB1 HMTase is down-regulated at the transcriptional level by several anticancer drugs, due to its inherent instability. Using RNA sequence analysis, we identified FosB as being regulated by SETDB1 during anticancer drug therapy. FosB expression was increased by treatment with doxorubicin, taxol and siSETDB1. Moreover, FosB was associated with an increased rate of proliferation. Combinatory transfection of siFosB and siSETDB1 was slightly increased compared to transfection of siFosB. Furthermore, FosB was regulated by multiple kinase pathways. ChIP analysis showed that SETDB1 and H3K9me3 interact with a specific region of the FosB promoter. These results suggest that SETDB1-mediated FosB expression is a common molecular phenomenon, and might be a novel pathway responsible for the increase in cell proliferation that frequently occurs during anticancer drug therapy.

Understanding the LST (Land Surface Temperature) Effects of Urban-forests in Seoul, Korea

  • Urban development and population have augmented the increase of impervious land-cover. This phenomenon has amplified the effects of climate change and increasing urban island effects due to increases in urban temperatures. Seoul, South Korea is one of the largest metropolitan cities in the world. While land uses in Seoul vary, land cover patterns have not changed much (under 2%) in the past 10 years, making the city a prime target for studying the effects of land cover types on the urban temperature. This research seeks to generalize the urban temperature of Seoul through a series of statistical tests using multi-temporal remote sensing data focusing on multiple scales and typologies of green space to determine its overall effectiveness in reducing the urban heat. The distribution of LST values was reduced as the size of urban forests increased. It means that changing temperature of large-scale green-spaces is less influenced because the broad distribution could be resulted in various external variables such as slope aspect, topographic height and density of planting areas, while small-scale urban forests are more affected from that. The large-scale green spaces contributed significantly to lowering urban temperature by showing a similar mean LST value. Both of concentration and dispersal of urban forests affected the reduction of urban temperature. Therefore, the findings of this research support that creating urban forests in an urban region could reduce urban temperature regardless of the scale.

  • Corrugation of a corrugated panel dramatically increases the bending stiffness per weight. However, corrugated panels show lower sound insulation performance than that of the flat plate having the same weight. Especially, in a particular frequency region, the sound transmission loss significantly decreases. Main reason of the problem is known as the local resonance. A number of local resonance modes occur above a certain frequency band and modal density rapidly increases. In this study, we investigate the relation of the sound transmission loss and the modal density. Finally, we propose a design methodology in terms of the modal density to improve the sound insulation performance of the corrugated panels without weight increase.

Detection and characterization of avian hepatitis E virus from broiler breeders and layers in Korea (육용종계와 산란계에서 avian hepatitis E virus의 검출 및 특성 규명)

  • The helicase genes and hypervariable regions (HVRs) of three avian hepatitis E viruses (HEVs) detected at three different farms were sequenced and characterized. Two isolates (DW-L and GI-B2) were classified as genotype 2 and one isolate (GR-B) was classified as genotype 1. A phylogenetic tree, based on the helicase gene and HVR nucleotide sequences, revealed the newly detected viruses and other avian HEVs were classified similarly. Unlike previously reported avian HEVs, the DW-L isolate detected in broiler breeders with characteristic lesions of avian HEV had no proline-rich motif in its HVR, suggesting that the proline-rich motif is non-essential for viral replication and infection.

  • This study has purpose on deducting problems of evacuation plan for vulnerable populations in disaster and suggesting improvement plan through analysis of disaster weakness in domestic rural region aiming at vulnerable populations in disaster like old people containing most of domestic rural population, sometimes being in blind spot of safety when landslide or disaster occur. As a result, we could know that rural regions have high proportion of vulnerable populations in disaster like old people, also being so weak to landslide and slope collapse. So we suggested development of manual describing prevention of disaster and evacuation for vulnerable populations in disaster like old people and disaster evacuation organization for house and minimizing solution for damage of human life through improvement of steep slope evaluation criteria.

Mobile Application based on Image Processing and a Proportion for Food Intake Measuring

  • In the paper, we propose a new reliable technique for measuring food intake based on image automatically without user intervention. First, food and bowl image before and after meal is obtained by user. The food and the bowl are divided into each region by the K-means clustering, Otsu algorithm, Morphology, etc. And the volume of food is measured by a proportional expression based on the information of the container such as it's entrance diameter, depth, and bottom diameter. Finally, our method calculates the volume of the consumed food by the difference between before and after meal. The proposed technique has higher accuracy than existing method for measuring food intake automatically. The experiment result shows that the average error rate is up to 7% for three types of containers. Computer simulation results indicate that the proposed algorithm is a convenient and accurate method of measuring the food intake.

Preparation and Characterization of Cerium Doped Titanium Dioxide Nano Powder for Photocatalyst

  • This study was aimed at synthesizing and characterizing cerium-doped titania. Cerium-doped anatase titania powders were prepared by sol-gel process, with ammonium (IV) nitrate and titanium (IV) butoxide as the raw materials. The characteristics of pure $TiO_2$ and cerium-doped $TiO_2$ were investigated by XRD, TG/DTA, FE-SEM, and UV-vis spectroscopy. The results of this study show that anatase type of $TiO_2$ was obtained in as-prepared and calcined $TiO_2$ and Ce-$TiO_2$ powder. A DTA curve was also observed as the crystallization temperature decreased with increasing cerium contents. We found that the crystallite size of the obtained anatase particles decreased from 55 nm to 25 nm and the particle size decreased with increasing cerium contents. Moreover, UV-vis spectra showed that anatase titania powders with various cerium contents effectively extend the light absorption properties to the visible region.

  • Segmentation is one of the first steps in most diagnosis systems for characterization of dental caries in an early stage. The purpose of automatic dental cavity detection system is helping dentist to make more precise diagnosis. We proposed the semi-automatic method for the segmentation of dental caries on digital x-ray images. Based on a manually and roughly selected ROI (Region of Interest), it calculated the contour for the dental cavity. A snake algorithm which is one of active contour models repetitively refined the initial contour and self-examination and correction on the segmentation result. Seven phantom tooth from incisor to molar were made for the evaluation of the developed algorithm. They contained a different form of cavities and each phantom tooth has two dental cavities. From 14 dental cavities, twelve cavities were accurately detected including small cavities. And two cavities were segmented partly. It demonstrates the practical feasibility of the dental lesion detection using Computer-aided Detection (CADe).

Anti-cancer Activity of Human Colon Cancer (HT-29) Cell Line from Different Fraction of Zanthoxylum schnifolium Fruits (산초 (Zanthoxylum schnifolium)열매 분획물의 대장암세포(HT-29)에 대한 항암 효과)

  • This study was presented to the anti-cancer activity from different fraction of Zanthoxylum schnifolium fruits. The values for human colon cancer cell(HT-29) survival rate of 0.3 mg/mL of 70% EtOH and 70% MeOH ethyl acetate fraction extracts were 7.62${\pm}$0.173%, 7.66${\pm}$0.037%, respectively. It was shown that human colon cancer cell(HT-29) survival rate was in a dose-dependent manner. The percentages of cells were increased in the sub-G0 and G0/G1 phase region, meaning that cell proliferation was decreased. The RT-PCR demonstrated that 70% EtOH and 70% MeOH ethyl acetate fraction extracts were down-regulated the expression of Bcl-2 and survivin genes in HT-29 cells. We examined that 70% EtOH and 70% MeOH ethyl acetate fraction extracts anti-cancer activities initiated through ROS generation suggesting that HT-29 cells treated with ethyl acetate fraction extracts induced ROS generation. Our results revealed that the Zanthoxylum schnifolium fruit may expect for anti-cancer activities in HT-29 cells.

Moieties of Complement iC3b Recognized by the I-domain of Integrin αXβ2

  • Complement fragment iC3b serves as a major opsonin for facilitating phagocytosis via its interaction with complement receptors CR3 and CR4, also known by their leukocyte integrin family names, αMβ2 and αXβ2, respectively. Although there is general agreement that iC3b binds to the αM and αX I-domains of the respective β2-integrins, much less is known regarding the regions of iC3b contributing to the αX I-domain binding. In this study, using recombinant αX I-domain, as well as recombinant fragments of iC3b as candidate binding partners, we have identified two distinct binding moieties of iC3b for the αX I-domain. They are the C3 convertase-generated N-terminal segment of the C3b α'-chain (α'NT) and the factor I cleavage-generated N-terminal segment in the CUBf region of α-chain. Additionally, we have found that the CUBf segment is a novel binding moiety of iC3b for the αM I-domain. The CUBf segment shows about a 2-fold higher binding activity than the α'NT for αX I-domain. We also have shown the involvement of crucial acidic residues on the iC3b side of the interface and basic residues on the I-domain side.
