Antioxidant and Anticancer Activities of Artemisia annua L. and Determination of Functional Compounds (개똥쑥의 항산화 및 항암활성과 기능성 물질의 탐색)

  • The antioxidant and anticancer activities of leaves and stems from Gaeddongssuk (Artemisia annua L.) were investigated. The leaves and stems were extracted with water and 80% ethanol, respectively. The antioxidant and growth inhibition activities toward cancer cells by the 4 kinds of extracts were tested. In addition, phenolic compounds from A. annua were identified through quantitative analysis using HPLC. Antioxidant activities significantly increased in a does-dependent manner, and those of water and ethanol extracts of the leaves were stronger than those of the stems. Growth inhibition activities of the leaf extracts on HeLa and AGS cells were higher than those of the stem extracts. In particular, the ethanol extract of the leaves had growth inhibition activities of 61.07% and 57.24% against HeLa and AGS cells, at the concentration of $500\;{\mu}g$/mL, respectively, which were the highest among all the extracts. Phenolic acid and catechin contents of the A. annua extracts as determined by HPLC were higher in the leaves than in the stems. Flavonols were identified only in the leaves. The data suggest that the antioxidant and anticancer activities of A. annua extracts were due to phenolic compounds as well as unknown biological compounds in A. annua.

Extractives from the Bark of Platycarya strobilacea (굴피나무(Platycarya strobilancea) 수피의 Flavonol glycosides)

  • The dried bark of Platycarya strobilacea were ground, extracted with 95% EtOH, concentrated, and one of EtOH extracts was fractionated with a series of n-hexane, dichloromethane and another was fractionated with a series of petroleumether, $Et_2O$, ethyl acetate on a separatory funnel. A portion of dichloromethane soluble was chromatographed on a Sephadex LH-20 column ($72.0{\times}5.0cm$) using EtOH-$CHCl_3$ (7:3, v/v) as eluent and A portion of $Et_2O$ soluble was chromatographed on a silica gel column ($42.0{\times}3.5cm$) using $CHCl_3$-MeOH (9:3, v/v) as eluent. The isolated compounds were identified by TLC, $^1H$-, $^{13}C$-NMR, HMBC and EI-MS. Two flavonoids and three flavonoid glycosides were isolated from the bark of P strobilacea. The structures were determined to quercetin (compound 1), myricetin (compound 2) as flavonol compounds and afzelin (compound 3), quercitrin (compound 4), myricitrin (compound 5) as flavonol glycosides, respectively, on the basis of spectrosopic data.

  • Flavonoids, non-nutrient secondary metabolites of plants, are widely distributed in commonly consumed agro-food resources. Flavonoids include aglycones, and their glycosides are reported to have potential health-promoting compounds. The aim of this study was to investigate flavonoid glycosides in the fruit and leaves of Zizyphus jujuba var. inermis (Bunge) Rehder (jujube). A total of six flavonoids (five flavonols and one chalcone) were identified in jujube fruit and leaves by using ultra-performance liquid chromatography-diode array detector-quadrupole time of flight mass spectrometry along with chemical library and an internal standard. In positive ion mode, six flavonoids were linked to the C- and O-glycosides which were conjugated with sugar moieties based on kaempferol, quercetin, and phloretin aglycones. Total flavonoid contents of leaves (8,356.5 mg/100 g dry weight (DW)) was approximately 900-fold higher than that of fruit (fresh fruit, 13.6 mg/100 g dry DW; sun-dried fruits, 9.2 mg/100 g dry DW). Quercetin 3-O-rutinoside (rutin) and quercetin 3-O-robinobioside were the predominant flavonols in fruit and leaves of jujube. In particular, rutin had the highest content (6,735.2 mg/100 g DW) in leaves, and rutin is a widely reported bioactive compound. Phloretin 3',5'-di-C-glucoside (chalcone type) was detected only in leaves. The leaves of jujube contain a high content of flavonoids and the results of this study indicate that jujube leaves may be a source of bioactive flavonoids.

Protective Effects of Cellular Membrane and Component Analysis of Polygonum aviculare Extracts (마디풀 추출물의 세포 보호 효과 및 주성분 분석)

  • In this study, the antioxidative effects and component analysis of Polygonum aviculare (P. aviculare) extracts were investigated. The ethyl acetate and the aglycone fraction from P. aviculare extracts were more active than (+)-${\alpha}$-tocopherol and $\small{L}$-ascorbic acid, which are known as strong antioxidants for their antioxidative activity by the DPPH method and chemiluminescence assay. The cellular protective effects of fractions of P. aviculare on the rose-bengal sensitized photohemolysis of human erythrocytes, increased in a concentration dependent manner ($1-10{\mu}l$). In particular, the ethyl acetate fraction at a concentration of $10{\mu}l$ showed the most prominent protective effect among all the extracts (${\tau}_{50}$, 314.70 min). TLC and HPLC chromatogram of the ethyl acetate fraction of P. aviculare extracts revealed 3 main bands (PA8, PA5, PA6) and peaks (peak 1, peak 2, peak 3), which were identified as myricetin-3-O-rhamnoside (myricitrin, PA8, peak 1), quercetin-3-${\alpha}$-rhamnoside (quercitrin, PA6, peak 3) by LC/ESI-MS/MS and $^1H$-NMR respectively. These results indicate that fractions from P. aviculare could be applicable to new functional cosmetics as antioxidants.

Comparison of Chemical Properties and Phenolic Compound for Ethanol Extract of Blueberry, Bokbunja and Mulberry and their Pomaces (블루베리·복분자와 오디 그리고 이들 부산물 주정 추출물의 이화학적 특성 및 페놀화합물 함량 비교)

  • In this study, the chemical properties and phenolic compound of blueberry, bokbunja and mulberry and their pomace were determined to develop them as functional food materials. Water content of individual whole berry was ranged from 84.25-86.20%, and water content was significantly high in whole berries rather than their pomace (p<0.01). Additionally, each berry and its pomace's pH was 3.32-5.18. Among them, whole mulberry showed the highest pH which is 5.18 (p<0.01). Total polyphenol and flavonoid contents were the greatest in blueberry pomace and they were 24.81 mg/g and 2.13 mg/g, respectively (p<0.01). However, mulberry pomace generated the greatest anthocyanin content compared to others (p<0.01). In phenolic compound profiles, cyanin chloride was detected in mulberry and bokbunja. Epigallocatechin, gallocatechin and isorhamnetin were found only in blueberry. Catechin (hydrate) and epicatechin were greater in pomaces than whole berries except blueberry (p<0.01), otherwise, significantly great rutin (trihydrate) and quercetin contents were found in whole berries as compared to their pomace except blueberry (p<0.01). Gallic acid was significantly greatest in mulberry (p<0.01) and quercetin 3-D-galactoside was significantly greatest in blueberry (p<0.01). Apigenin and luteolin were traced in mulberry, and mulberry pomace showed greater apigenin and luteolin contents than whole mulberry (p<0.01). Naringenin was greater in pomaces than whole berries (p<0.01). As a result, it was found that all berry extracts used in this study were able to be applied as functional food materials and their pomace contained high phenolic compound enough to be a good source of phytochemical for nutraceutical use.

  • Prior studies of physiologically active substances in grapes have focused only on the fruit, pericarp, and branches, as well as the pulp and seeds. The present study assessed the changes in quercetin-3-O-glucuronide (Q3OG) and flavonol glycoside content using grape leaves from Vitis labruscana B. and V. coignetiae at different growth stages and provided basic data for quality control. Content analysis showed that both varieties differed in Q3OG and flavonol glycoside content by growth stage, and the components were found to be high in the order of fruit, maturation, and coloration. Also, Vitis labruscana B. has a better vascular relaxation effect than Vitis coignetiae. These results suggest that in the use of grape leaves as a functional raw material, Q3OG and flavonol glycosides can be used as indicator components. In addition, if raw materials for each growth stage are mixed in a particular ratio, it will be a way to manage the specific efficacy and content of indicator components.

Neuroprotective effects of antioxidant constituents isolated from Opuntia ficus-indica var. saboten Makino

  • Opuntia ficus-indicavar. saboten Makino (Cactaceae) is a tropical or subtropical plant that has been widely used as folk medicine for the treatment of diabetes, asthma, burn, edema and gastritis. The purposes of the present study were to identify antioxidant constituents from fruits and stems of the plant cultivated in Cheju island, Korea, and examine their in vitro neuroprotective activities. Using a chromatographic fractionation method, ten chemical constituents were isolated from ethyl acetate extracts. By means of chemical and spectroscopic methods, those were identified as eight flavonoids such as kaempherol (a), quercetin (b), kaempferol 3-methyl ether (c), quercetin 3-methyl ether (d), narcissin (e), dihydrokaernpferol (f), dihydroquercetin (g) and erioclictyol (h), and two terpenoids such as 3-oxo-${\alpha}$-ionol-${\beta}$-d-glucopyranoside (i) and roseoside (j). Among the isolated compounds, comrounds c~e and h~j were those reported for the first titre from the plant. Compounds b, d and g showed DPPH free radical scavenging activities with IC$\sub$50/ values of 28, 19 and 31, ${\mu}$M respectively. Compounds d and g also inhibited iron-dependent lipid peroxidation with IC$\sub$50/ values of 2.4 and 3.5 ${\mu}$M. In a primary rat cortical neuronal cell culture system, compounds b, d and g inhibited xanthine/xanthine oxidase-induced (IC$\sub$50/ values of 18.2, 2.1 and 54.6 ${\mu}$M) and H$_2$O$_2$-induced (IC$\sub$50/ values of 13.6, 1.9 and 25.7 ${\mu}$M) cytotoxicities. In addition, compounds d and g inhibited NMDA-induced excitotoxicity by 21 and 33%, and only compound d inhibited growth factor withdrawal-induced apoptosis by 31% at a tested concentration of 3 ${\mu}$M. The results suggest that the antioxidant constituents with in vitroneuroprotective activities may serve as lead chemicals for the development of neuroprotective agent.

  • The aim of this study was to evaluate the change of phenolic compounds after bordeaux mixture treatment on Cheorwon onion. Onion is a perennial plant belonging to the lily family. It is native to Persia of Southwest Asia. It is cultivated much in the temperate regions of the world. Onion is a good name for the 'Okchong' to drop blood cholesterol and cardiovascular blood flow to increase the prevention of adult diseases. Cheorwon area is inland, but it has high continental climate due to its high altitude. After the treatment with bordeaux mixture, the weight, width and plant height of the onion were affected to some extent. In the phenolic compounds, caffeic acid, ferulic acid, rutin, and kaempferol except benzoic acid, p-coumaric acid and quercetin were similar to or less than the control. Therefore, it was concluded that the treatment with bordeaux mixture had some effect on onion growth and phenolic compound contents.

Onion is a good name for the 'Okchong' to drop blood cholesterol and cardiovascular blood flow to increase the prevention of adult diseases. Cheorwon area is inland, but it has high continental climate due to its high altitude. Therefore it is said that the onion cultivated in this region has higher sugar content and higher taste than onion grown in the southern region. Phosphorus components are particularly important ingredients for promoting muscle development. However, if the phosphoric acid content of the soil part is maintained to a large extent until the harvest, the competition of the nutrients tends to cause decay of the root part. Therefore, it is important to improve the quality and shelf life of onion by inducing nutrient balance by applying foliar fertilization method on the reducing phosphorus at harvest time. In this study, acidity was controlled by diluting phosphorous acid(H3PO3) and potassium hydroxide(KOH), followed by leaf surface treatment with phosphite on onion. In this study, the concentration of phosphite was diluted to 500, 1,000, 1,500ppm and sprayed three times over the onion leaves in May 2018 using an atomizer and harvested at the end of June, and the phenolic compounds were analyzed by HPLC. As a result, the content of quercetin, one of the important substances in onion, was phosphite 500ppm(179.70㎍/g), 1,000(150.27), 1,500(105.95). The contents of caffeic acid, p-coumaric acid, ferulic acid, rutin, kaempferol, and sugar content were higher in the treatments than in the control. Therefore, the phosphite does not have a great influence on the growth, but it may play a role as a method of achieving balance with nitrogen in the rainy season by supplying the role of the material catalyst and the water soluble phosphoric acid and the potassium in the influence of the material change.

Correlation between Antioxidant Capacities and Color Values in Korean Red Grape Juices (국내 적포도주스의 항산화능과 색도의 상관성)

  • Grape juice is consumed worldwide and studied due to the high antioxidant activities and contents. The color of grape juice is due to the presence of phenolic compounds such as anthocyanins. Therefore, color values can be an indicator of antioxidant capacities of grape juice. However, the correlation between color values and antioxidant activities in grape juice has not been well studied. In this study, the physicochemical characteristics and correlation between color values and antioxidant capacities of Korean red grape juices (five commercial juices from market and two juices prepared in the laboratory) were investigated to estimate antioxidant capacities. Antioxidant capacities were determined by 1,1-diphenlyl-2-picrylhydrazyl radical scavenging assay, ferric ion reducing antioxidant power assay, and oxygen radical absorbance capacity assay. Total phenolic contents, total flavonoid contents, and total anthocyanin contents, including five kinds of polyphenolic compounds, were examined by high performance liquid chromatography (HPLC). The results for physicochemical properties showed similar values, except titratable acidity. The color hue values of the prepared juices were higher than those of commercial juices, which was in contrast to the lower color intensity values (P<0.05). The Hunter L, a, and b values showed almost no difference between commercial and prepared juices. The antioxidant activities and total phenolic contents of commercial juices were higher than those of prepared ones. Gallic acid, catechin, and quercetin were confirmed by HPLC in all samples. The total phenolic and total flavonoid contents showed positive correlation with antioxidant activities. In addition, antioxidant activities and contents correlated with color values. Thus, estimation of antioxidant capacity could be feasible through the spectrophotometric measurement of color values.
