Purification and Characterization of a New Fibrinolytic Enzyme of Bacillus licheniformis KJ-31, Isolated from Korean Traditional Jeot-gal

  • Jeot-gal is a traditional Korean fermented seafood and has long been used for seasoning. We isolated 188 strains from shrimp, anchovy, and yellow corvina Jeot-gal, and screened sixteen strains that showed strong fibrinolytic activities on a fibrin plate. Among those strains, the strain that had the largest halo zone was chosen and identified as Bacillus licheniformis by using 16S rDNA sequencing and an API CHB kit. The fibrinolytic activity of Bacillus licheniformis was characterized and designated as bpKJ-31. The active component of bpKJ-31 was identified as a 37 kDa protein, designated bacillopeptidase F, by internal peptide mapping and N-terminal sequencing. The optimum activity of bpKJ-31 was shown at pH 9 and $40^{\circ}C$, with a chromogenic substrate for plasmin. It had high degrading activity for the $B{\beta}$-chain and $A{\alpha}$-chain of fibrin(ogen), and also acted on thrombin, but not skim milk and casein. The amidolytic activity of bpKJ-31 was inhibited by 1 mM phenylmethanesulfonyl fluoride, but 1 mM EDTA did not affect the enzyme activity, indicating that bpKJ-31 is an alkaline serine protease, like a plasmin. The bpKJ-31 showed approximately 14.3% higher fibrinolytic activity than the plasmin. These features of bpKJ-31 make it attractive as a health-promoting biomaterial.

  • This study researched and compared the sedimentation environment and photosynthetic pigments at Gomso (GS) -bay, Yoja (YJ) -bay and KangJin (KJ) -bay in May, 2012. It was shown that KJ-bay consist of C (clay) and M (mud), and GS-bay consists of Zs (silty sand) and Sz (sandy silt). Averagely, IL of YJ-bay was 4.98%, KJ-bay 6.10%, and GS-bay 1.45%. As for COD concentration, there were no places that exceeded Japanese sediment contamination standard 20 mg/g-dry. As for AVS concentration, in case of KJ-bay, two places exceeded Japanese sediment contamination standard 0.2 mg/g-dry. If we look into the average C/N ratio of bays, YJ-bay showed 8.50, KJ-bay 6.60, and GS-bay 5.52, thus all of them showed the characteristic of oceanic origin. As for the plankton make-up classified by photosynthetic pigments, diatom was dominant, and both ratios of pigment and C/chlorophyll. a showed the relatively lower distribution at GS-bay than at KJ-bay and YJ-bay. It is judged that this is related to the characteristic of flow-in, and it implies that predation process and decomposition by organic matters actively occurs at YJ-bay and KJ-bay.

Incidence of Panicle Blast, Race Distribution During 1993??¡?1994 and Pathogenicity of New Races of Pyricularia grisea in Korea (벼 이삭도열병 발생정도 및 병원균 레이스의 분포 변동(1993~1994)과 새로운 레이스의 병원성)

  • Severity of panicle blast disease on the leading cultivars of rice was investigated at nine locations of Korea during summer in 1993 with frequent raining and low temperature conditions. Incidence of panicle blast was 26% on Jinmibyeo, 23% on Chuncheongbyeo and 40% on Odaebyeo. Race distribution of Pyricularia grisea was examined from 1993 to 1994. A total of 1,098 isolates of the fungus obtained from the blast nurseries and framers; fields were screened using Korean differential varieties. Twenty one races were identified in 1993 and 19 races in 1994. KI-181, KJ-103, KJ-104, KI-241 and KI-209 were identified as new races, and KJ-301 (15%), KJ-201 (13%), KJ-105 (12%) and KI-409 (12%) were major races during 1993 and 1994. Race KI-197, which has a wide spectrum of virulence on rice cultivars, was isolated in a ratio of 8.3% and evenly distributed in the whole country. Some of the major and newly identified races during 1993∼1994 were used for spray-inoculation on leading cultivars and differential varieties in a greenhouse. We found that most of the cultivars were resistant to the major races, KJ-301 and KI-409, but were very susceptible to the new races, KI-241, KI-209, KI-181 and KJ-103.

Screening of Fibrinolytic Enzyme Producing from Microorganisms and Optimum Conditions of Enzyme Production (혈전 분해효소 생산균의 탐색 및 효소생산 최적조건의 조사)

  • A strain of potential producer of fibrinolytic enzyme was isolated from Korean fermented food. The isolated bacterium was identified and named as Bacillus brevis KJ-23. The optimal condition of the medium for the production of fibrinolytic enzyme from Bacillus brevis KJ-23 was nutrient broth with 0.5% D-ribose, 0.5% malt extract and 0.3% $K_2$HPO$_4$. The optimum pH, temperature and fermentation time for the enzyme production were pH 7.0, 3$0^{\circ}C$ and 24 hr, respectively.

  • The study aims to furnish the data for the study of invigorating communities of various ages including old people living alone. Furthermore, it seems that the laddering method and kj method can be useful for the study of various attitude survey. This study attempted to extract necessary factors for space for community behavior of elderly people living alone using laddering survey method and KJ rule on a sample of elderly people who live alone in three different rental housings. The keywords that were drawn from the perception survey through laddering were divided into small, middle and large classification using KJ rule. Mostly identical keywords were extracted in the three complexes, implying that there is no difference of perception among the three complexes in this survey. According to the large classification where KJ rule was applied, there were six necessary factors for space for community activity of elderly people living alone in rental housing including spatial consideration, communication, resting space, nature-friendliness, health promotion and Secure identity, which were the same for all of the three complexes.

Distribution of Races of Pyricularia oryzae in Kyungpook Province (경북지방(慶北地方)의 벼도열병균(稻熱病菌)레이스 분포(分布)에 대하여)

  • This experiment was conducted to obtain the basic information for the effective control of rice blast. Eleven races of Pyricuiaria oryzae were identified, 5 races of KJ group and 6 races of KI group in Kyungpook province. Among them KJ301 was the most dominant one, and they were prevailed in the order of KJ401, KJ201, KJ101, KJ105, KI307 and $KI315^b$. The distribution ratio of KI group race was higher in the middle and northern region than the southern and coastal region. A positive correlation was found between the distribution ratio of KI group races and cultivated area of indica type cultivals. The total ratio of Blasticidin-S resistant isolates in P. oryzae was 31.2%. The distribution ratios of resistant isolates were 43.2% in the southern region, 29.4% middle, 25.0% nothern and 10.5% coastal region, respectively. The distribution ratio of Blasticidin-S resistant Isolates was correlated positively with amounts of the chemical sprayed, but not correlated with the race of P. oryzae.

  • In this paper we study the connection between 2-primal rings, (S,1)-rings and related conditions. And we investigate some condition which is the special case of pseudo symmetric. We also study the condition (KJ) which is given by J. Y. Kim and H. L. Jin. We introduce some condition and we prove that our condition is equivalent to the condition (KJ) when it is an (S,1)-ring.

Characterization of Bacillus luciferensis Strain KJ2C12 from Pepper Root, a Biocontrol Agent of Phytophthora Blight of Pepper

  • In this study, we characterized the bacterial strain KJ2C12 in relation with its biocontrol activity against Phytophthora capsici on pepper, and identified this strain using morphological, physiological, biochemical, fatty acid methyl ester, and 16S rRNA gene sequence analyses. Strain KJ2C12 significantly (P=0.05) reduced both final disease severity and areas under the disease progress curves of 5-week-old pepper plants inoculated with P. capsici compared to buffer-treated controls. As for the production of antibiotics, biofilms, biosurfactant, extracellular enzyme, HCN, and swarming activity, strain KJ2C12 produced an extracellular enzyme with protease activity, but no other productions or swarming activity. However, Escherichia coli produced weak biofilm only. Strain KJ2C12 could colonize pepper roots more effectively in a gnotobiotic system using sterile quartz sand compared to E. coli over 4 weeks after treatments. However, no bacterial populations were detected in 10 mM $MgSO_4$ buffer-treated controls. Strain KJ2C12 produced significantly higher microbial activity than the $MgSO_4$-treated control or E. coli over 4 weeks after treatments. Bacterial strain KJ2C12 was identified as Bacillus luciferensis based on morphological, physiological, and biochemical characteristics as well as FAME and 16S rRNA gene sequence analyses. In addition, these results suggested that B. luciferensis strain KJ2C12 could reduce Phytophthora blight of pepper by protecting infection courts through enhanced effective root colonization with protease production and an increase of soil microbial activity.

Study on the Simultaneous Production of the Bacterial Cellulose and Vinegar by Gluconacetobacter persimmonis KJ145T (Gluconacetobacter Persimmonis KJ145T를 이용한 Bacterial Cellulose및 식초의 동시 생산에 관한 연구)

  • The changes of component through simultaneous production of bacterial cellulose and vinegars by G. persimmonis KJ145$^{T}$ were examined. As a results, pH was decreased to 3.22 at 8 days of fermentation and total acidity showed 4.66 which was the highest at the 8 days of fermentation. Brix didn't show any changes during the fermentation period. Free sugars of fermentation broth were consist of fructose, glucose and sucrose. The fructose concentration of fermentation broth was maintained highly during fermentation period (until the final 10 days) without a remarkable decrease. The cell growth of G. persimmonis KJ145$^{T}$ was very rapidly increased from the 2 days of fermentation and increased most at the 4 days of fermentation. The productivity of bacterial cellulose was increased in proportion to the fermentation period. Malic acid, succinic acid and oxalic acid were detected as a organic acid of vinegar. The concentration of acetic acid was rapidly increased from the 2 days and reached highest concentration at 8 days. In conclusion, the results indicated that the 8 days was the optimal fermentation period to produce the bacterial cellulose and vinegar by G. persimmonis KJ145$^{T}$ simultaneously.

Antibacterial Activity of actobacillus sp. KJ-5 Isolated from Pig Feces (돼지분변에서 분리한 Lactobacillus sp. KJ-5의 항균특성)

  • A lactic aci bacteria producing antibacterial substance was isolated from pig feces. This strain was identified as a genus Lactobacillus, through its morphological, cultural and physiological characteristics. Lactobacillus sp. KJ-5 isolated showed the strong inhibitory effect on the growth of Salmonella paratyphi. The production of antibacterial substance was growth associated form during the batch culture of Lactobacillus sp. KJ-5 and the maximum production was obtained at the culture temperature of $37^{\circ}C$ as well as optimum temperature of cell growth. The antibacterial activity of the filtrate of culture broth was decreased by adjusting the pH 6.2 and was not affected by catalase treatment. The antibacterial substance was partially purified by methanol and acetone extraction, whtch exhibited three spots in the thin-layer chromatography and one of them showed an antibacterial activity, This substance also showed the maximum absorption of UV at 270nm and an antibacterial activity was completely inactivated by the treatment of proteolytic enzymes.
