Effects of Astragalus Membranaceus, Innamomum Cassia, Phellodendron Amurensis(BHH10) on MC3T3-E1 Cells Proliferation, Differntiation and Bone Mineralized Formation (MC3T3-E1 세포주에서 황기.계지.황백 처방(BHH10)의 골형성 촉진 효능 연구)

  • Objectives : BHH10 is traditional medicine herb used for enhancing body resistance against various diseases. The aim of this study was to identify BHH10 extract induces osteogenic activity in human osteoblast-like MC3T3-E1 cells. Methods : MC3T3-E1, pre-osteoblast cell line, were treated with BHH10 of various concentrations($0.1{\mu}g/mL$, $1{\mu}g/mL$, $10{\mu}g/mL$). And then, the effect of BHH10 on osteoblast differentiation was examined by alkaline phosphatase(ALP) activity, von Kossa staining and RT-PCR for osteoblast differentiation markers such as osteocalcin(OCN), osteopontin(OPN). Results : BHH10 had dose-dependent effect on the viability of osteoblastic cells, and dose-dependently increased alkaline phosphatase(ALP) activity. BHH10 markedly increased mRNA expression for OCN, OPN in MC3T3-E1 cells. Also, BHH10 significantly induced mineralization in the culture of MC3T3-E1 cells. Conclusions : In conclusion, these results propose that BHH10 can play an important role in osteoblastic bone formation, osteogenesis, and may possibly lead to the development of bone-forming drugs.

  • A method based on high-performance liquid chromatography (HPLC) with ultraviolet detection has been developed to quantify coenzyme Q10 (CoQ10) in raw materials and dietary supplements. Single-laboratory validation has been performed on the method to determine linearity, selectivity, accuracy, limits of quantification (LOQ) and repeatability for CoQ10. An excellent linearity (r=0.999) was observed for CoQ10 in the concentration range $15.625{\sim}500\;{\mu}g/ml$ in dietary supplement. Observed recovery of CoQ10 was found to be between 98.33 and 99.38%. LOQ was found to be $250\;{\mu}g/ml$ Repeatability precision for CoQ10 was between 0.15 and 0.21% relative standard deviation (RSD). Further, limited studies showed that some adulterants and degraded material could be satisfactorily separated from CoQ10 and identified.

The Effect of Platform Screen Doors on PM10 Levels in a Subway Station and a Trial to Reduce PM10 in Tunnels

  • $PM_{10}$ concentrations were measured at four monitoring sites at the Daechaung station of the Seoul subway. The four locations included two tunnels, a platform, and a waiting room. The outside site of the subway was also monitored for comparison purposes. In addition, the effect of the platform screen doors (PSDs) recently installed to isolate the $PM_{10}$ in a platform from a tunnel were evaluated, and a comparison between $PM_{10}$ levels during rush and non-rush hours was performed. It was observed that $PM_{10}$ levels in the tunnels were generally higher than those in the other locations. This might be associated with the generation of $PM_{10}$ within the tunnel due to the train braking and wear of the subway lines with the motion of the trains, which promotes the mixing and suspension of particulate matter. During this tunnel study, it was observed that the particle size of $PM_{10}$ ranged from 1.8 to 5.6 ${\mu}m$. It was revealed that the $PM_{10}$ levels in the tunnels were significantly increased by the PSDs, while those in the platform and waiting room decreased. As a result, in order to estimate the effect of ventilation system on $PM_{10}$ levels in the tunnels, fans with inverters were operated. It was found that the concentration of $PM_{10}$ was below 150 ${\mu}g/m^3$ when the air flow rate into a tunnel was approximately 210,000-216,000 CMH.

Fatigue and Retention Characteristics of PLZT(10/y/z) Thin films with Various Zr/Ti Concentrations Ratio (PLZT(10/y/z) 박막에서 Zr/Ti 농도에 따른 피로와 리텐션 특성)

  • The effects of Zr/Ti concentration ratio in PLZT (10/y/z) thin films prepared by sol-gel method are investigated for the NVFRAM application. As Ti amount of Zr/Ti concentration ratio increases, the dielectric constants at 10 kHz decrease from 550 to 400, while the loss tangents increase from 0.028 to 0.053 and the leakage current densities at 170 kV/cm decrease from $1.64\times10^{-6}$ to $1.26\times10^{-7}\;A/cm^2$. In the results of hysteresis loops measured at $\pm170kV/cm$, the remanent polarization and the coercive field increase from 6.62 to $12.86{\mu}C/cm^2$ and from 32.15 to 56.45 kV/cm, respectively, according to the change from 40/60 to 0/100 in Zr/Ti concentration ratio. Fatigue and retention properties also improve much as the Zr/Ti concentration ratio change from 40/60 to 0/100. After applying $10^9$ square pulses with $\pm5V$, the remanent polarization of the PLZT (10/40/60) thin film decreases $50\%$ from the initial state while that of the PLZT (10/0/100) thin film decreases $28\%$. In the results of retention measurements of 10s s, the remanent polarization of the PLZT (10/0/100) thin film dec.eases only $10\%$ from the initial state, while that of the PLZT (10/40/60) thin film decreases $40\%$.

Spontaneous Oscillatory Rhythm in Retinal Activities of Two Retinal Degeneration (rd1 and rd10) Mice

  • Previously, we reported that besides retinal ganglion cell (RGC) spike, there is ~10 Hz oscillatory rhythmic activity in local field potential (LFP) in retinal degeneration model, rd1 mice. The more recently identified rd10 mice have a later onset and slower rate of photoreceptor degeneration than the rd1 mice, providing more therapeutic potential. In this study, before adapting rd10 mice as a new animal model for our electrical stimulation study, we investigated electrical characteristics of rd10 mice. From the raw waveform of recording using $8{\times}8$ microelectrode array (MEA) from in vitro-whole mount retina, RGC spikes and LFP were isolated by using different filter setting. Fourier transform was performed for detection of frequency of bursting RGC spikes and oscillatory field potential (OFP). In rd1 mice, ~10 Hz rhythmic burst of spontaneous RGC spikes is always phase-locked with the OFP and this phase-locking property is preserved regardless of postnatal ages. However, in rd10 mice, there is a strong phase-locking tendency between the spectral peak of bursting RGC spikes (~5 Hz) and the first peak of OFP (~5 Hz) across different age groups. But this phase-locking property is not robust as in rd1 retina, but maintains for a few seconds. Since rd1 and rd10 retina show phase-locking property at different frequency (~10 Hz vs. ~5 Hz), we expect different response patterns to electrical stimulus between rd1 and rd10 retina. Therefore, to extract optimal stimulation parameters in rd10 retina, first we might define selection criteria for responding rd10 ganglion cells to electrical stimulus.

Changes in Microbial and Chemical Composition and Sensory Characteristics of Fermented Soybean Paste, Chungkukjang, by High Dose Gamma Irradiation $(10{\sim}120\;kGy)$ (고선량$(10{\sim}120\;kGy)$ 감마선 조사 청국장의 미생물 변화와 화학적 성분과 관능적 특성 변화)

  • Changes in the number of microorganisms, chemical composition, and sensory quality of chungkukjang by gamma-irradiation up to 120 kGy were investigated. The total viable cell counts in chungkukjang irradiated with 10 kGy decreased from $10^9\;CFU/g$ of the control to $10^5\;CFU/g$. Proximate chemical composition and pH of chungkukjang were not much affected by irradiation. Acidity of chungkukjang decreased by irradiation with over 20 kGy. Fatty acid contents of chungkukjang irradiated under 20 kGy were the same as those of the nonirradiated ones. Polyunsaturated fatty acids content, decreased at over 40 kGy. The color and odor of chungkukjang were not significantly affected by irradiation up to 20 kGy. Unpalatable odor (p<0.01), off-flavor (p<0.001), and fish-odor (p<0.001) increased, and brown intensity (p<0.05) and acceptability (p<0.001) decreased at over 20 kGy. Results indicate 20 kGy is sufficient to decrease the microbial count of chungkukjang and prevent decrease in the content of polyunsaturated fatty acids.

Effects of Molecular Weights of Sodium Hyaluronate on the Collagen Synthesis, Anti-inflammation and Transdermal Absorption (히알루론산나트륨의 분자량 크기에 따른 Collagen 합성, 항염증 및 피부 흡수에 미치는 영향)

  • In this study, we examined the effects of various molecular weights (1, 10, 50, 100, 660, and 1500 kDa) of sodium hyaluronate (HA), which were prepared by enzyme hydrolysis, on the collagen synthesis, anti-inflammation and skin absorption. These HA did not significantly affect the viability of human dermal fibroblast Hs68 cells. Among them, 1500 kDa, 50 kDa HA most significantly increased collagen production by 59%, and 50% in the Hs 68 cells, respectively. Whereas 1500 and 660 kDa HA hardly pass through mouse transdermis membrane, lower molecular weights (1, 10, or 50 kDa) of HA showed time-dependent increase in skin permeation. HA of 50 kDa showed highest anti-inflammatory effects by reducing nitric oxide and tumor necrosis factor-${alpha}$ production in the RAW 264.7 cells, comparing to other HA (1, 10, and 100 kDa HA). Recently, there is no report about anti-wrinkle and anti-inflammatory effects and skin permeation of different molecular weights HA (1, 10, 50, 100, 660 and 1500 kDa), which were produced by enzyme hydrolysis. These results suggested that 50 kDa HA can be potent candidates for the development of effective anti-aging and anti-wrinkle cosmetic agents. The results of this study demonstrated that among those HA with different molecular weights, 50 kDa HA showed highest anti-inflammatory activity, significant capability to induce collagen synthesis and high level of skin permeation.

Reliability of Scapular Winging Measurement Using a Scapulometer (익상견갑의 평가를 위한 "Scapulometer"의 신뢰도 연구)

  • This study was undertaken to develop the scapulometer, a measuring tool for scapular winging, and to determine the reliability of scapulometer. Thirty subjects with scapular winging greater than 2 cm participated in this study. Two examiners measured scapular winging using the scapulometer. Standardized measurement position of each subject was as follows: neutral position of shoulder and wrist, $90^{\circ}$ flexion of elbow, and cuff weight of 5% and 10% of body weight (BW) to neutral wrist. The interrater and intrarater reliability were calculated with an intraclass correlation coefficient (ICC) with 95% confidence interval (CI) and standard error of measurement (SEM) at 5% BW and 10% BW. The level of scapular winging was tested between 5% BW and 10% BW with a paired t-test. The interrater reliability was shown to be high at 5% BW (ICC(2,k)=.955, 95% CI=.900~.979, SEM=.07) and at 10% BW (ICC(2,k)=.968, 95% CI=.930~.985, SEM=.06). The intrarater reliability was shown to be high at 5% BW (ICC(2,k)=.921, 95% CI=.842~.961, SEM=.09) and at 10% BW (ICC(2,k)=.906, 95% CI=.813~.954, SEM=.10). The level of scapular winging was significantly different between 5% BW and 10% BW. Conclusively, it is demonstrated that the scapulometer is an objective and qualitative measurement tool for scapular winging.

Influence of Temperature and pH on the Stability of Dimethoxy Biphenyl Monocarboxylate${\cdot}$HCl Solutions

  • The accelerated stability of dimethoxy biphenyl monocarboxylate.HCl (DDB-S) was investigated in 6 mg/mL water solution in the pH ranging 2-10 and the temperature of $45-85^{\circ}C$. The observed rate of degradation followed first-order kinetics. The energy of activation for DDB-S degradation was calculated to be 14.1 and 16.5 $Kcal/mole$ at pH 5 and in distilled watery respectively. The degradation rate constant ($K_{25^{\circ}C}$) obtained by trending line analysis of Arrhenius plots for DDB-S was $5.3{\times}10^{-6}h^{-1}$. The times to degrade 10% ($t_{10}$) and 50% $t_{500}$) at $K_{25^{\circ}C}$ were 829 and 5,416 days, respectively. DDB-S exhibited the fastest degradation at pH 10 and the slowest rate at pH 5. In addition, at $K_{65^{\circ}C}$, degradation rate constants of DDB-S were 0.066, 0.059, 5.460, 32.171, and $1.4{\times}10^{-6}h^{-1}$ at pH 2, 5, 8, 10 and in distilled water, respectively. These observations indicated that the rate-pH profile of DDB-S showed general acid-base catalysis reaction in the range of pH 2-10.

  • A Spinning Rotor Gauge was calibrated between $4.04\times10^{-3}$ Pa and $1.11\times10^{-2}$ Pa at the high vacuum standard by static expansion method. The results were analysed according to the document of 'Guide to the Expression of Uncertainty in Measurement' of ISO. The expanded uncertainty was $3.0035\times10^{-3}$ Pa at $7.5448\times10^{-3}$ Pa. $95\%$ confidence level, and coverage factor of k = 1.
