Stimuli-responsive biomaterials that alter their function through sensing local molecular cues may enable technological advances in the fields of drug delivery, gene delivery, actuators, biosensors, and tissue engineering. In this research, pH-responsive hydrogel which is comprised of dimethylaminoethyl methacylate (DMAEMA) and 2-hydroxyethyl methacrylate (HEMA) was synthesized for the effective delivery of doxorubicin (Dox) to breast cancer cells. Cancer and tumor tissues show a lower extracellular pH than normal tissues. DMAEMA/HEMA hydrogels showed significant sensitivity by small pH changes and each formulation of hydrogels was examined by scanning electron microscopy, mechanical test, equilibrium mass swelling, controlled Dox release, and cytotoxicity. High swelling ratios and Dox release were obtained at low pH buffer condition, low cross-linker concentration, and high content of DMAEMA. Dox release was accelerated to 67.3% at pH 5.5 for 6-h incubation at $37^{\circ}C$, while it was limited to 13.8% at pH7.4 at the same time and temperature. Cell toxicity results to breast cancer cells indicate that pH-responsive DMAEMA/HEMA hydrogels may be used as an efficient matrix for anti-cancer drug delivery with various transporting manners. Also, pH-responsive DMAEMA/HEMA hydrogels may be useful in therapeutic treatment which is required a triggered release at low pH range such as gene delivery, ischemia, and diabetic ketoacidosis.
The successful synthesis of hyaluronic acid micro-threads is very promising approach for the broad application in tissue engineering such as dermal fillers. Because hyaluronic acid has the excellent biocompatibility and ability to maintain the moisture of up to several hundred times its own weight. In order to generate the hyaluronic acid micro-threads in microfluidic system, we employed two-phase flow microfluidic chip to make a rapid synthesis of the hyaluronic acid hydrogel. Hyaluronic acid was mixed with 0.02N NaOH solution and 1, 4-Butanediol diglycidyl ether (BDDE) solution and then injected into core channel. The ethanol was used for the 3-dimensional micro-thread formation in sheath channel. We manipulated the diameter of HA micro-threads using controlling of flow rates in microfluidic chip, and showed the feasibility of immobilization in HA micro-threads with florescent substances. Also, the generated HA micro-threads were evaluated and showed the suitable properties with tensile strength, bending property, and swelling profiles for dermal fillers. As a result, we suggested an innovative method for microfluidic chip-based HA micro-threads which could safely be applied as dermal filler in tissue engineering.
전기화학적 방법을 통한 요산 (Uric acid) 정량분석을 위해 수용성 고분자 (hydrogel polymer)를 배위시킨 오스뮴 고분자 화합물과 요산 산화효소 (Uricase), 가교를 위한 PEGDGE (poly(ethylene glycol) diglycidyl ether)가 혼합된 용액을 스크린 프린팅된 탄소 전극 (SPCEs) 위에 흡착하여 측정하였다. 수용성 오스뮴 고분자의 전위를 조절하기 위해 리간드인 피리딘링의 4번 위치에 다른 전기음성도의 작용기를 갖는 오스뮴 고분자 화합물을 합성하였다. 합성된 오스뮴 고분자 화합물은 PAA-PVI (Poly(acrylic acid)-poly(vinyl imidazole)-$[osmium(4,4^{\prime}-dichloro-2,2^{\prime}-bipyridine)_2Cl]^{+/2+}$), PAA-PVI-$[osmium(4,4^{\prime}-dimethyl-2,2^{\prime}-bipyridine)_2Cl]^{+/2+}$, PAA-PVI-$[osmium(4,4^{\prime}-dimethoxy-2,2^{\prime}-bipyridine)_2Cl]^{+/2+}$이다. 제작된 효소전극은 순환전압전류법 (cyclic voltammetry)을 통해 uric acid에 의한 오스뮴 고분자 화합물들의 산화 촉매 전류(oxidation catalytic current)를 측정하여 uric acid의 농도를 정량적으로 분석할 수 있었다. 오스뮴 고분자 화합물들 중 0.215 V의 산화환원 전위를 갖는 $PAA-PVI-[Os(dme-bpy)_2Cl]^{+/2+}$ (PAA-PVI-osmium$(4,4^{\prime}-dimethyl-2,2^{\prime}-bipyridine)_2Cl$]$^{+/2+}$) 화합물을 이용하여 대표적인 간섭물질인 아스코르브산 (AA)과 포도당 (glucose)의 산화 신호의 간섭효과를 피할 수 있었다. 이를 이용하여 제작된 전극은 0.33 V 전위에서 다양한 농도의 uric acid (1.0, 1.5, 2.0, and 5.0 mM)의 전류를 측정한 결과 $r^2=0.9986$의 좋은 선형성을 갖는 것을 확인하였다. 이는 복잡하지 않은 간단한 방법과 일회용의 전극을 사용하기 때문에 현장현시 검사 (point of care; POC)에 적합한 요산측정용 바이오센서로서의 가능성을 확인 할 수 있었다.
Despite the fact that porcine embryonic stem cells (ESCs) are a practical study tool, in vitro long-term maintenance of these cells is difficult in a two-dimensional (2D) microenvironment using cellular niche or extracellular matrix proteins. However, a three-dimensional (3D) microenvironment, similar to that enclosing the inner cell mass of the blastocyst, may improve in vitro maintenance of self-renewal. Accordingly, as a first step toward constructing a 3D microenvironment optimized to maintain porcine ESC self-renewal, we investigated different culture dimensions for porcine ICM-derived cells to enhance the maintenance of self-renewal. Porcine ICM-derived cells were cultured in agarose-based 3D hydrogel with self-renewal-friendly mechanics and in 2D culture plates with or without feeder cells. Subsequently, the effects of the 3D microenvironment on maintenance of self-renewal were identified by analyzing colony formation and morphology, alkaline phosphatase (AP) activity, and transcriptional and translational regulation of self-renewal-related genes. The 3D microenvironment using a 1.5% (w/v) agarose-based 3D hydrogel resulted in significantly more colonies with stereoscopic morphology, significantly improved AP activity, and increased protein expression of self-renewal-related genes compared to those in the 2D microenvironment. These results demonstrate that self-renewal of porcine ICM-derived cells can be maintained more effectively in a 3D microenvironment than in a 2D microenvironment. These results will help develop novel culture systems for ICM-derived cells derived from diverse species, which will contribute to stimulating basic and applicable studies related to ESCs.

Radiation technology is an effective way for regulating polymeric materials to physicochemical and mechanical properties. New polymeric hydrogels based on vinyl ethers have been synthesized by the $\gamma$-initiated polymerization method. In this paper, we have studied the effect of radiation on mechanical and electrochemical properties of new rubber-like polymeric composite materials based on polyvinylether of ethyleneglycol (PVEEG).
Itraconazole is one of the most potent antifungal agents available in the market today. However, the low bioavailability due to its poor-water solubility calls for an alternative formulation to the current oral type. A topical itra-conazole-containing formulation may be of use for several reasons including the opportunity to reduce adverse events and generate high local tissue levels, more rapid drug delivery, and lower systemic exposure. The purpose of the present study was to investigate the vehicles for topical skin delivery of itraconazole. The effect of formulations on the hairless mouse skin permeation and deposition of itraconazole was determined using Franz diffusion cells at $37^{\circ}C$. Benzyl alcohol in micro-emulsion significantly increased the solubility of itraconazole, thereby increasing the skin permeation rate. However, lipo-some formulation showed the lowest solubility and permeation rate of itraconazole. Although the solubility of itraconazole in hydrogel formulation was lower than that in microemulsion, skin permeation rate was significantly higher probably due to its adhesive property. Therefore, microemulsion-based hydrogel formulation is expected to synergistically increase the skin permeation rate and skin deposition of itraconazole.
대표적인 양이온성 폴리사카라이드인 키토산은 생체 친화적인 특성으로 인하여 식품 및 의약품으로의 다양한 응용성이 제시되고 있으나, 용액의 pH에 따른 급격한 용해도 변화로 인하여 실제적인 사용에 많은 제약이 따른다. 본 연구에서는 양이온성 키토산 고분자체와 음이온성 고분자 전해질인 PAA의 이온성 상호작용을 이용하여 두 고분자 전해질의 구성 비율 및 용액의 pH에 따른 polyplex 형성 과정을 관찰하였다. 특히, 두 고분자체의 조성 비율에 따라 나타나는 입자 표면의 전하량은 입자 간정전기적 반발력을 제공하여 균일한 입자 크기와 높은 콜로이드 안정성을 제공하였으며, 이와 같이 안정화된 polyplex 입자는 추가적인 가교화 반응을 통하여 하이드로젤 입자로의 형성이 가능하였다. 두 고분자 전해질의 부분적인 교차 결합으로 형성된 하이드로젤 입자 내부의 acryl amide 작용기는 특징적인 저임계 용액 온도(LCST)를 나타냄으로써 입자의 온도에 따른 수화 작용에 차이를 보이며 그에 따른 입자의 수력학적 직경 변화가 관찰되어 온도에 따른 하이드로젤 입자의 가역적인 팽윤 작용이 관찰되었다. 이와 같은 하이드로젤 입자는 생리활성적인 환경에서 제한된 용해도를 보이는 키토산의 응용성을 한 층 더 높일 수 있을 것으로 기대된다.
본 논문은 가교제의 물질전달을 통한 실시간 생체고분자의 젤화 과정으로 단분산성을 갖는 구형의 알지네이트 하이드로젤을 미세유체 채널 내에서 제조하는 방법에 관한 연구이다. 먼저 미세유체 채널 내에서 단분산성 알지네이트 액적들을 형성하고 연속상에 분산된 염화칼슘 분자들의 물질전달 과정을 통해 실시간 젤화과정이 이루어지게 하여 알지네이트 하이드로젤 입자를 제조하였다. 이때, 미세유체 채널에서 형성되는 액적의 크기는 손쉽게 케필러리 수(capillary number)와 분산상의 유속 조절을 통하여 제어할 수 있다. 본 방법은 미세유체 채널 내에서 안정적인 액적을 형성할 수 있고 칼슘 가교제로 제조된 알지네이트 하이드로젤 입자들은 균일한 크기 분포를 가지며(C.V=2.71%) 유속, 점도, 및 계면장력의 조절을 통하여 $30{\mu}m$에서 $60{\mu}m$까지의 다양한 크기의 알지네이트 하이드로젤 입자를 제조할 수 있다. 본 논문에서 제시한 간단한 미세유체 접근방법을 통해 제조되는 단분산성을 갖는 알지네이트 하이드로젤 입자는 생체물질들을 손쉽게 함입(encapsulation)할 수 있으며 이는 식품, 화장품, 잉크 및 약물 등의 전달체로 활용이 가능하고 생체적합성이 뛰어나 세포이식 분야에도 활용될 가능성이 있다.
목적: 본 연구는 3-chloropyridine-4-carboxylic acid와 3-fluoropyridine-4-carboxylic acid를 첨가제로 사용하여 소프트 콘택트렌즈 제조 후 물리적 및 광학성 특성을 분석하여 콘택트렌즈 재료로서 활용도를 알아보았다. 방법: 본 실험에서는 소프트 하이드로젤 콘택트렌즈의 주재료인 HEMA (2-hydroxyethyl methacrylate)와 AA(acylic acid), MMA(methacrylic acid) 그리고 개시제인 AIBN(azobisisobutyronitrile), 교차결합제인 EGDMA(ethylene glycol dimethacrylate)를 사용하여 공중합 하였다. 결과: 생성된 고분자의 물리적 특성을 측정한 결과, 굴절률 1.4320~1.4342, 함수율 34.54~37.15%, 접촉각 $57.82{\sim}79.57^{\circ}$, 인장강도 0.2872~0.3608로 각각 나타났고 광학적 특성 측정 결과 가시광선 영역에서는 투과율이 88.8~90.6%로 나타났으며, UV-B 76.8~82.4%, UV-A 84.6~86.6%로 각각 나타났다. 결론: 본 실험 결과 3-chloropyridine-4-carboxylic acid 및 3-fluoropyridine-4-carboxylic acid를 첨가제로 사용하였을 때 고습윤성 및 자외선 차단기능을 가진 콘택트렌즈 재료로 활용될 수 있을 것으로 판단된다.
New polymeric hydrogels based on vinyl ethers have been synthesized by the ${\gamma}$-initiated polymerization method. Their physical chemistry and physical mechanical properities have been studied. It has been shown that structure and swelling behavior of the hydrogels can be regulated by the changing of synthesis conditions nature of monomers. Novel stimuli-sensitive polymers have been synthesized by the varying of macrochains hydrophilic-hydrophobic balance. The some biomedical aspects of application of hydrogels in capacity of drain aging polymeric materials in ophthalmology surgery, implants in plastic surgery as well as drug delivery systems have been investigated.
