Spatial and Temporal Variability of Water Quality in Geum-River Watershed and Their Influences by Landuse Pattern (금강 수계의 시.공간적 수질특성과 토지이용도의 영향)

  • The objective of this study was to analyze long term temporal trends of water chemistry and spatial heterogeneity for 83 sampling sites of Geum-River watershed using water quality dataset during 2003~2007 (obtained from the Ministry of Environment, Korea). The water quality, based on multi-parameters of temperature, dissolved oxygen (DO), biochemical oxygen demand (BOD), chemical oxygen demand (COD), suspended solids (SS), total nitrogen (TN), total phosphorus (TP), and electric conductivity (EC), largely varied depending on the landuse patterns, years and seasons. The watershed was classified into three different landuse types: forest stream (Fo), agricultural stream (Ag), and urban stream (Ur). Largest seasonal variabilities in most parameters occurred during the two months of July to August and these were closely associated with large spate of summer monsoon rain. Conductivity, used as a key indicator for an ionic dilution during rainy season, and nutrients of TN and TP had inverse functions of precipitation. BOD, COD decrease during the rainy season. Minimum values in the conductivity, TN, and TP were observed during the summer monsoon, indicating an ionic and nutrient dilution of river water by the rainwater. In contrast, major inputs of suspended solids (SS) occurred during the period of summer monsoon. The landuse patterns analyses, based on the variables of BOD, COD, TN, TP and SS, showed that the values were greater in the agricultural stream (Ag) than in the forest stream (Fo) and urban stream (Ur) and that water quality was worst in the urban stream (Ur). The overall dataset suggest that efficient water quality management, especially in Gap-Stream and Miho-Stream, which showed worst water quality is required along with some of urban stream (Ur), based on the analysis of landuse patterns.

Anti-adipogenic Activity of Cortex ulmi pumilae Extract in 3T3-L1 Preadipocytes (유근피 추출물의 3T3-L1지방전구세포의 분화 억제 효능에 관한 연구)

  • Cortex ulmi pumilae, the cortex of Ulmus davidiana var. japonica, has been used in traditional folk medicine for its anti-inflammatory effect. Although its various bioactivities such as anti-inflammatory, anti-microbial, and anti-cancer, have been reported, the anti-adipogenic activity of cortex ulmi pumilae remains unclarified. In the present study, we investigated the effect of cortex ulmi pumilae extract on adipocyte differentiation in 3T3-L1 preadipocytes. Treatment with cortex ulmi pumilae extract significantly reduced the formation of lipid droplets and triglyceride content in a dose-dependent manner; this is associated with an inhibition of the adipogenic transcription factors, CCAAT/enhancer binding protein ${\alpha}$ ($C/EBP{\alpha}$), $C/EBP{\beta}$, and peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$). In addition, cortex ulmi pumilae extract treatment during the early stage of adipogenesis showed more efficient anti-adipogenic activity than treatment during other stages of adipogenesis. Cortex ulmi pumilae extract also inhibited cell proliferation and induced G1 arrest of 3T3-L1 cells in the early stage of adipogenesis. This was associated with upregulated expression of Cdk inhibitor p21 and downregulated expression of cyclin E and phospho-Rb, indicating that cortex ulmi pumilae extract blocks mitotic clonal expansion by cell cycle regulation. Taken together, these results suggest that cortex ulmi pumilae extract possesses anti-adipogenic activity through the inhibition of adipocyte differentiation by blocking mitotic clonal expansion.

Identification of Equine Heat Shock Proteins Gene and Their mRNA Expression Analysis after Exercise (말의 열충격 단백질(heat shock proteins)의 특성 구명과 운동 후 유전자의 발현 분석)

  • The purpose of this study was to characterize equine heat-shock protein (Hsp) genes and analyze their expression pattern in various horse tissues and blood leukocytes after exercise. In a previous study, RNA sequencing of blood and skeletal muscles of thoroughbreds before and after exercise was performed using differently expressed gene (DEG) analysis. Three Hsp genes (HspH1, Hsp90${\alpha}$ and Hsp70) were selected by DEG analysis and were found to be differentially expressed in either blood or muscle. To validate and extend previous observations on these genes, we performed RT-PCR analyses of horse tissue as well as real-time qPCR analyses of blood leukocytes after exercise. mRNA expression of these Hsp genes was found to be ubiquitous in the analyzed tissues (including thyroid, colon, skeletal muscle, cecum, kidney, spinal cord, heart, and lung). In addition, Hsp mRNA expression of these genes in extracted whole blood increased after 120 minutes of exercise compared to the baseline condition. These results are in agreement with the results of human and other experimental animals, suggesting that regulatory mechanisms that are responsible for upregulation of Hsp gene transcription may be conserved among species. Further investigations to correlate Hsp gene expression patterns with athletic performance or recovery processes after exercise are warranted.

Antioxidant Activities of Solvent Extracts from Rosa multiflora (찔레꽃 추출물의 용매별 항산화 활성)

  • This study was attempted to investigate antioxidant activities of water and ethanol extract from Rosa multiflora (RM) by in vitro assays measuring 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, 2,2'-azino-di-2-ethyl-benzthiazoline sulphonate (ABTS) radical scavenging activity, reducing power activity, nitric oxide (NO) radical scavenging activity, total phenol and total flavonoid content. The water and ethanol extracts from RM scavenged the DPPH radical and ABTS radical in a dose-dependent manner at the concentration range from 10 to $500{\mu}g/ml$. The DPPH radical scavenging activity of water extract was higher than that of ethanol extract. $IC_{50}$ of DPPH radical scavenging activity of water and ethanol extract were $79.73{\mu}g/ml$ and $145.85{\mu}g/ml$. The reducing power activity of water extract was higher than that of ethanol extract. The nitric oxide (NO) radical scavenging activity of the RM extract was similar to their DPPH radical scavenging activity. $IC_{50}$ of ABTs radical scavenging activity of water and ethanol extract was $79.82{\mu}g/ml$ and $159.03{\mu}g/ml$. The reducing power activity of water extract (0.775) was higher than that of ethanol extract (0.568). Total phenolic content of water extract (140.74 mg/g) was higher than that of the ethanol extracts (37.83 mg/g). Total flavonoid content of water extract (45.31 mg/g) was higher than that of the ethanol extracts (42.68 mg/g). This results suggest that water extract of RM may be useful as potential antioxidant sources.

Characterization of Symptom and Determination of Tissue Critical Concentration for Diagnostic Criteria in 'Maehyang' Strawberry (Fragaria $\times$ ananassa Duch.) as Influenced by Calcium Concentrations in the Fertigation Solution ('매향' 딸기의 칼슘 영양진단을 위한 결핍증상 및 식물체 내 한계농도)

  • Objective of this research was to investigate the effect of calcium concentrations in the fertilizer solution on growth and development of Ca deficiency in 'Maehyang' strawberry (Fragaria $\times$ ananassa Duch.). The margins of the youngest leaves were scorched and they developed a cupped shape as they were expanded in Ca deficient plants. The vein area of the youngest leaves became brown when the deficiency became severe. The quadratic responses were observed in dry weight production to elevated Ca concentrations in fertilizer solutions with the highest growth in 4.5 mM treatment. The regression equation was y=2.4026+1.0209x-$0.098x^2$ $(R^2=0.3546^{***})$. However, tissue Ca contents increased lineally as the Ca concentrations in fertilizer solutions were elevated (y=1.2108+0.1333x, $R^2=0.9189^{***}$). In changes of the fresh weight and Ca concentrations in petiole sap, fresh weight production showed quadratic responses to elevated Ca concentrations in fertilizer solution, but Ca concentration increased lineally. The equations in changes of fresh weight and Ca concentrations were y=9.273+4.882x-$0.4245x^2$ $(R^2=0.4935^{***})$ and y=52.311+3.2917x $(R^2=0.6918^{***})$, respectively. When the concentration of calcium at which plant growth was retarded by 10% is regarded as critical concentration level, the calcium contents based on dry weight of above ground plant tissue and in petiole sap should be in the range between 1.6 to 2.25% and 63 to $79mg{\cdot}kg^{-1}$, respectively.

Detection of Dopamine and Serotonin by Competitive Enzyme-Linked Immunosorbent Assay (경쟁적 ELISA를 이용한 도파민과 세로토닌의 검출)

  • Dopamine (DA) and serotonin (5-Hydroxytryptamine, 5-HT) are neurotransmitters and hormones that exist in small amounts but have important role in the body. Serum and 24-hour urine are used as specimens, and are usually examined by HPLC-MS. In this study, we tried to detect DA and 5-HT by competitive ELISA using antigen-antibody (Ab) reaction. After immobilizing $5{\mu}g/mL$ BSA conjugate on a 96-well surface, hormone and primary Ab, which are respectively diluted to different concentrations, were treated. Then, HRP-conjugated secondary Ab and TMB were added to measure absorbance. The regression equation and $R^2$ value were calculated based on absorbance, and sensitivity of Ab to hormone as well as the correlation between hormone concentration and absorbance were determined. In DA ELISA, $R^2$, the correlation between the concentration of hormone and absorbance, was the highest by 0.91 when anti-dopamine Ab was diluted 6,000 times and 7,000 times. In 5-HT ELISA, $R^2$ was bigger than 0.90 in every concentration except 3,000 times and 6,000 times. Both DA and 5-HT were not effectively detected at low concentrations (less than $1.0{\times}10^{-7}M$); and because reference value of serum DA is lower than this, HPLC-MS was required to detect serum DA. However, competitive ELISA may be effective in detecting 24-hour urine DA, serum, and 24-hour 5-HT. Further studies are needed to detect hormones more accurately at lower concentrations.

Nutritional and Microbiol Quality Assessment of Chungmukimbab (충무김밥의 영양적 및 미생물적 품질 평가)

  • This research was designed to evaluate the nutritional an microbiol quality assessment of Chungmukimbab purchased from market in Tongyeoung area. Contents of calories, calcium, iron, thiamin and riboflavin in ordinary kimbab and Chungmukimbab were lower than the recommended levels of Korean adult men. So, We suggested that a fruit, beverage and ffod stuff were supplemented to maintain nutritional balance. Total aerobic bacteria and coliform group of just prepared ordinary kimbab and Chungmukimbab samples from market were not significantly different, showing approximately $5.50{\pm}0.38 log_{10} CFU/g,\;2.10{\pm}0.47log_{10}MPN/100g$ in ordinary kimbab, $5.61{\pm}0.42log_{10}CFU/g,\;1.75{\pm}0.34 log_{10} MPN/100g$ in Chungmukimbab, respectively. Total aerobic bacteria of law ingredients of chungmukimbab sample were 3 to $4 log_{10}CFU/g$ in kimbab, seasoning squid and radish roots kimchi, 4 to $5 log_{10}CFU/g$ in boiled fish paste. The coliform groups were 1 to $2 log_{10}$ MPN/100 g in kimbab, seasoning squid and radish roots kimchi, 2 to $3 log_{10}$ MPN/100g in boiled fish paste. Detection rate of E. coli and Staphylococcus aureus counts were 10.0, 12.5% in Chungmu-kimbab, 15.0, 10.0% in seasoning squid, 0, 10.0% in radish roots kimchi respectively, not detected in boiled fish paste samples. During storage at $15^{\circ}C$ for 24 hours, total aerobic counts and coliform groups in ordinary kimbab and Chungmukimbab were increased by the 1.94, $0.97log_{10}CFU/g$, 0.60, 0.72 log10 MPN/100g respectively. Monitoring on Microbiological Contamination of Packed Ice Creams from Manufacturing Factories in Korea (국내 제조공장에서 생산된 아이스크림류의 미생물학적 오염실태 조사)

  • In this study, the bacteriological survey was examined on ice creams at manufacturing factories in Korea during the summer season of 2011. The nineteen selected among 166 samples by preliminary test were collected from 11 different manufacturing factories in four major manufacturers in May 2011. Samples from ice milk, ice creams, sherbets, and non milk fat ice creams were tested for the total aerobic bacteria, coliform bacteria, and five food borne pathogens, respectively. The results showed that the coliforms including E. coli O157:H7, Salmonella spp., Staphylococcus aureus, Clostridium perfringens, and Listeria monocytogenes were not detected on all the ice creams. The total aerobic bacteria of the packed samples examined ranged between $2.5{\times}10^3$ and $5.5{\times}10^5cfu/g$. One ice cream, two sherbets, and four ice milk samples exceeded the acceptable limits of total aerobic bacteria according to the Korean standards for ice cream ($1.0{\times}10^5cfu/g$) and others ($5.0{\times}10^4cfu/g$). The levels of these microorganisms from ice creams were higher in three original equipment manufacturers than seven self-manufacturers. Three of ten ice creams (30.0%), three of six ice milks (50.0%), and one of two sherbets (50%) exceeded the acceptable limits of total aerobic bacteria, respectively. The personnel hygiene procedures with chocolate and vanilla chip addition from the manufacturing process were the main sources of the microbial contamination of stick-bar type ice creams when being produced in a factory. Improvement of the hazard analysis critical control points (HACCP) system should be introduced into the ice cream factory to improve the microbial quality of the ice cream products in Korea.

Formaldehyde Monitoring of Hygiene Products in Domestic Market (국내 유통 위생용품 중 포름알데히드 잔류량 모니터링)

  • By the standards and specifications for hygiene products, three test methods for formaldehyde are specified for each item type of hygiene product. After derivatization using acetylacetone and 2,4-dinitrophenylhydrazine (2,4-DNPH), formaldehyde is analyzed by spectrophotometer and high-performance liquid chromatography (HPLC). Validation of the three test methods was performed on tissue, diaper lining and waterproof layer, and panty liner products. The results of linearity (R2), limit of detection (LOD), limit of quantification (LOQ), recovery rate (%) and reproducibility (%), showed that all three methods are suitable for analyzing formaldehyde in hygiene products. After derivatization with 2,4-DNPH and cetylacetone, formaldehyde was analyzed at 0, 3, 6, 9, 24 and 48 hours by HPLC. Formaldehyde derivatized with 2,4-DNPH showed no statistically significant change in formaldehyde peak area over time (P>0.05). But, acetylacetone-derivatizated formaldehyde showed a negative correlation coefficient (r) over time (P<0.01). We investigated the residual amounts of formaldehyde in 205 hygiene products distributed in Busan. Among 74 disposable diaper products tested, 73 had low concentrations of formaldehyde (0.13-29.87 mg/kg). Moreover, formaldehyde was not detected in any of 78 tissue, 27 disposable paper towel, 12 disposable dishcloth, 7 paper cup, one brand of paper straw and 6 disposable napkin products.

Drug Resistance and R Plasmids of Lactobacilli Isolated from Fermented Milk (유산균음료(乳酸菌飮料)로부터 분리(分離)한 유산간균(乳酸桿菌)의 R-Plasmids의 중개(仲介)에 의(依)한 대장균(大腸菌)에로의 항생제내성(抗生劑耐性) 전달(傳達))

  • Eleven strains of lactobacilli were isolated from fermented milk of 9 companies. They were classified 3 strains as L. bulgaricus, 2 strains as L. plantarum, 2 strains as L. cellobiosus, 1 strain as L. lactis, 1 strain as L. acidophilus, 1 strain as L. casei subsp. casei and 1 strain as L. casei subsp. tolerans. And these strains were examined for drug resistance, transferability and transfer frequency of R plasmid. Most of isolates were sensitive to tetracycline(TC), penicillin(PC), and erythromycin(EM), but some strains were resistant to streptomycin(SM), chloramphenicol(CP), ampicillin(AP), kanamycin(KM), and nalidixic acid(NA). All of isolates were resistant to two or more drugs and 6 different drug resistant patterns were found. The most frequently encountered patterns were NA AP CP SM KM(5 strains) followed by NA AP CP KM(2 strains), NA AP CP SM(1 strain), NA AP CP(1 strain), NA CP(1 strain) and NA AP(1 strain). Tranfer experiment of drug resistance showed that of all 11 resistant strains, 9 strains transferred R plasmid determining AP(6 strains) or AP SM(3 strains) to a recipient, E. coli ML 1410 strain with $2.8{\times}10^{-5}-1.5{\times}10^{-1}%$ of transfer frequency. These results indicate that lactobacilli conjugally transfer their resistance to E. coli.
