Effect of Dietary Inclusion of Various Sources of Green Tea on Immune System and Challenging Test of Juvenile Olive Flounder Paralichthys olivaceus (사료내 녹차 첨가가 넙치 유어기의 면역성 및 세균 공격성에 미치는 영향)

  • Effect of dietary inclusion of various sources of green tea on growth, immune system and challenging test of juvenile olive flounder Paralichthys olivaceus was investigated. Five experimental diets with triplicates were prepared: control, raw leaves, dry leaves, by-product and extract. Twenty five (an initial body weight of 52.5 g) were randomly distributed into 15 of 180 L flow-through tanks. Nutrient requirements of the experimental diets satisfied growth of juvenile olive flounder. The feeding trial lasted for 7 weeks. After 7-week feeding trial, blood were sampled from three randomly chosen fish for serum analysis of Iysozyme and bactericidal activity, and ten fish were infected with Edwardsiella tarda for challenging test from each tank. Weight gain (g/fish) of fish fed the diet containing extract and control diet was significantly higher than that of fish fed the other diets. Feed efficiency ratio for fish fed the diet containing extract and control diet was significantly higher than that for fish fed the diets containing raw leaves and by-product, but not significantly different from that for fish fed the diet containing dry leaves. Serum Iysozyme activity (units/ml) of fish fed the diets containing dry leaves and extract was significantly higher than that of fish fed the diets containing raw leaves and by-product, but not significantly different from that of fish fed the control diet. Serum bactericidal activity (${\times}10^6$ bacteria/ml) of fish fed the diet containing dry leaves and extract was significantly lower than that of fish fed the diets containing raw leaves, by-product and control diet in 3 hour. However, serum bactericidal activity of fish fed the diet containing extract was significantly lower than that of fish fed the other diets in 6 hour. And serum bacterial activity was low in fish fed the diets containing dry and raw leaves, by-product, and control in 6 hour in order. Accumulative mortality (%) of fish fed the control diet was low compared to that of fish fed the diets containing raw leaves and by-product, but high compared to that of fish fed the diets containing dry leaves and extract although no significant difference was found among treatments. In considering above results, dietary inclusion of extract and dry leaves of green tea seemed to be highly effective to improve immune system and endurance against E. tarda infection of juvenile olive flounder.

Intrageneric Protoplast Fusion between Alkalophilic Bacillus sp. F204 and Bacillus sp. K 17 (호알칼리성 Bacillus sp. F204와 Bacillus sp. K 17의 원형질체 융합)

  • To develop cellulase and xylanase-producing strain by protoplast fusion, alkalophilic Bacillus sp. F204 and K17 were treated with NTG(N-methyl-N'-nitro-N-nitrosoguanidine) and isolated anti-biotics resistant strains of S20 (Km$^r$ , Cm$^r$) and G70 (Str$^r$). The frequency of protoplast formation was about 95% when cells of mid-log phase were treated with 200$\mu\textrm{g}$/ml Iysozyme at 37$^{\circ}C$ for 30-45 minutes. Under addition of 0.4-0.5M sodium succinate, 0.5% casamino acid, 1.5% polyvinylpyrrolidone, 25mM MgC1$_2$ and 50mM CaC1$_2$ to the regeneration medium, the regeneration frequency of Bacillus sp. F204 and K17 was 24.9% and 26.2%, respectively. The fusion frequency was 6.6$\times$10$^{-6}$ in the presence of 30% polyethylene glycol 6000 containing 50mM $Ca^{++}$ at 45$^{\circ}C$ for 5 minutes. Cellulase complex and xylanase activities of fusant were compared with parental strains.

Isolation of lysozyme producing bacteria capable of solubilizing microbial cells (미생물 용해가 가능한 Iysozyme 분비 균주의 분리 및 특성)

  • Lysozyme-producing microorganisms were isolated to obtain bacteria which can efficiently solubilize microbial cells. Cells of normal and chloroform-treated Escherichia coli and Micrococcus Iysodeikticus were used as model substrates to isolate lysozyme-producing microorganisms and investigate the efficiency of cell lysis. The culture supernatant of the isolate New1 (98% similarity of 16S rDNA sequence with Thermomonas haemolytica) showed different lytic characteristics for different substrates. Thermal treatment (autoclave) of substrate cells showed a significant effect on cell solubilization by culture supernatant of the New1. For autoclaved substrate cells, E. coli, M. Iysodeikticus and chloroform-treated E. coli were solubilized by 58.7%, 49.4% and 79.1%, respectively, in the culture supernatant of New1. The lytic activity of New1 was mainly caused by lysozyme produced by the isolate. It was also showed that New1 exhibited high protease activity and a little cellulase activity.

Morphology and swelling property of chitosan microapsules and microbeads prepared by W/O emulsion (W/O 에멀젼에 의한 chitosan microcapsule 및 microbead의 morphology와 팽윤성)

  • Chitosan microcapsules and microbeads were prepared by W/O emulsion method, and their morphologies were observed through SEM. The microcapsules have skin layer of 8 Um and 250 Um of mean diameter, The swelling test showed higher s welling ability in protic solvents than in aphotic solvents. After containing moth-yl violet in the microcapsules, the release patterns were investigated. The results sho wed that the addition of Iysozyme in pH 5.1 acetate buffer accelerated the re-lease rate. In case of the microbeads, the mean diameter was about 70 Um. The surface of the microbeads showed porous structures. The swelling ability of the beads revealed two times higher than the one of the microcapsules.

  • In this study, the effcts of kelp (Ascophyllum nodosum) meal on growth and immune responses of parrot fish (Oplegnathus fasciatus) were studied. Fish were fed an experimental diet supplemented with $2{\%}\;and\;5{\%}$ kelp meal in a controled diet. Several factors such as weight gain, hematological parameters and nonspecific immune responses were evaluated far 0, 2, 4 and 8 weeks after the administration of the kelp meal supplemented diet. Weight gain in the fish fed the diet supplemented with $2{\%}\;and\;5{\%}$ kelp meal was not significant among the tested groups. The NBT reaction of the phagocytes in the head kidney and the phagocytic rate/index of phagocytes in the peripheral blood were significantly higher than the control group. But, there was no significant difference both in complement bactericidal activity, mucus Iysozyme activity and hematology among each group.

Effect of Maternal Factors on the Concentrations of Minerals and Immunological Substance in Breast Milk (모유의 무기질과 면역물질함량에 영향을 미치는 요인에 관한 연구)

  • The purpose of this study was to evaluate the effect of maternal nutritional status and health behaviors on the concentrations of minerals (Zn, Fe, Ca) and the immunological substances (lactoferrin, sIgA, Iysozyme) in breast milk. Breast milk was collected from 193 healthy Korean women from obstetric clinics and postpartum care centers in Seoul. : 99 colostrum (1 - 5 days postpartum), 33 transitional milk (6 - 10 days postpartum), 61 mature milk (11 - 50 days postpartum). The concentrations of minerals and immunological substance were highest in colostrum and decreased with lactational period. Concentrations of Zn and Fe reduced significantly from colostrum to mature milk, however, Ca concentration stayed constant throughout the lactational period. Contents of lactoferrin, sIgA, and lysozyme were significantly lower in mature milk than in colostrum. Mother's nutritional status, assessed by prepregnancy BMI, had an effect only on colostrum, but not on transition and mature milk. Fe concentration of colostrum was significantly lower in underweight (prepregnancy BMI < 18.5) than in overweight mothers (prepregnancy BMI $\geq$ 23.0). Also lower tendency was observed for sIgA and lysozyme contents, even though the difference was not statistically significant. Pregnancy weight gain had no effect on the breast milk component. Since nutritional factors had some effect on colostrum, the health behaviors of mothers providing colostrum were assessed. The mother's behavior of smoking, drinking, morning sickness, parity, disease, nutrient supplement use had no significant effect on the breast milk component, however, Zn, sIgA, and lysozyme were the somewhat affected components by maternal health behavior.

  • Bradyrhizobium japonicum spheroplasts were prepared by culturing cells in the presence of glycine, follwed by treatment with lysozyme. The cells were examined by electron microscopy during the formation of spheroplast. Then Ti plasmid from Agrobacterium tumefaciens 15955 was introduced into Bradyrhizobium japonicum by glycine-lysozyme induced spheroplast transformation. After cell wall regeneration, transformants were selected by the ability of utilization of octopine. Transformation were received at a frequency of $1{\times}10^{-7}$. The transformants obtained from spheroplast transformation harbored the introduced Ti plasmid, which was identified by agarose gel electrophoresis. Furthermore, the differences in their gene products were observed between the transformant and the recipient cell by two-dimensional polyacrylamide gel electrophoresis. The transformants which still possessed the same ability nodulate soybean (Glycine max.) as that of the original host strain, acquired the ability to induce tumor on Petunia hybrida like Agrobacterium, but formed the small crown galls in size compared to those of Agrobacterium tumefaciens.

  • The cornea and conjunctiva of the human eye are exposed to external environment and thus are damageable. If the damaged part is infected with some pathogenic microorganisms. serious visual loss may be occured by inflammation. Keratitis or conjunctivitis does not always occur if the eyes are routinely exposed to pathogenic factors because lysozyme in human tears has antimicrobial activity against the microorganisms. 10 this study we have selected 5 strains causing keratitis and/or conjunctivitis. and cultured them in the optimum media. And then we have estimated the growth inhibition of the strains with the addition of various concentration of lysozyme to media to investigate the antimicrobial activity of lysozyme. The results are as follows. The growth of the strains were decreased according to the increase of lysozyme concentration. The growth of Pseudomonas. Neisseria. Klebsiella and Staphylococcus were inhibited 43%, 41%, 35% and 22% respectively by 1 mM concentration of lysozyme. The susceptibility of the gram-negative bacteria to lysozyme is 1.5~2 times higher than the Staphylococcus which is gram-positive bacteria in 1 mM concentration of lysozyme. But lysozyme inhibited the growth of Fusarium which is fungi slightly.

  • The moment analysis of lysozyme was implemented using chromatograms that were obtained from weak cation exchange column in high performance liquid chromatography system. Three elution sodium phosphate buffers containing 1.0, 0.75, 0.5M sodium chloride were used. Experiments were conducted by varying flow rate, elution sodium chloride concentration, and lysozyme solute concentration. The general rate (GR) model was employed to calculate the first moment and the second moment. By plotting $L/u_0$ vs. $({\mu}_1-t_0)/(1-{\varepsilon}_e)(1-{\varepsilon}_i)$] equilibrium constants (K) were obtained from first moment analysis. Intra-particle diffusivity was obtained from theoretical plate number data. Based on the results of moment analysis, van Deemter plots were drawn in order to investigate the contributions of $H_{ax}$, $H_f$, and $H_d$ to total Height Equivalent to a Theoretical Plate (HETP, $H_{total}$). The effect of intra-particle diffusion ($H_d$) was the most dominant factor contributing to HETP while external mass transfer ($H_f$) was negligible factor.

Stimulatory Effects of Extracellular Products of Mycobacterium spp. and Various Adjuvants on Non-specific Immune Response of Nile Tilapia, Oreochromis nilotica

  • In the present paper, the immunostimulatory effects of the extracellular products (ECP) from Mycobacterium spp. and various adjuvants on the non-specific immune responses of Nile tilapia, Oreochromis nilotica, were examined. Nile tilapia were immunized by injecting ECP of Mycobacterium spp. (strain TB40, TB267 or the type strain Mycobacterium marinum) into their swim bladders. A variety of adjuvants like as Freund s complete adjuvant (FCA), Freund's incomplete adjuvant (FIA) and Titremax were similarly injected into additional groups of tilapia. The number of nitroblue tetrazolium (NBT)-positive cells observed in the swim bladder of the immunized fish was signigicantly increased by the fourth day post-immunization. By day 8, the numbers of NBT-positive cells were fewer in fish immunized with ECP from mycobacteria strains TB40 or TB267 than those immunized with ECP from M. marinum or fish injected with FCA or FIA. The level of Iysozyme activity detected in the serum of fish 40 alter immunization with ECP from various Mycobacterium spp. was also significantly higher than that found in the serum of the control fish. Head kidney macrophages showed enhanced reduction of NBT when cultured in vitro with 1 $\mu$ g/ml of ECP. Concentrations greater than this (10 or 100 $\mu$g/ml) were found to suppress the reduction of NBT by the macrophages. ECP from Mycobacterium spp. and the various adjuvants used in the study all appear to be good activators of the non-specific immune responses of Nile tilapia.
