Antimutagenic Effects and Isolation of Flavonoids from Humulus japonicus Extract (환삼덩굴(Humulus japonicus) 추출물의 항돌연변이 효과와 Flavonoid 성분의 분리)

  • To investigate the physiological activities of Humulus japonicus, we examined the mutagenic and antimutagenic effects of the extract and isolated the flavonoid compounds. The water and methanol extracts of Humulus japonicus did not show the mutagenicity by Ames test in the 0.5-4 mg/plate. The methanol extract showed the antimutagenic effect aganinst Salmonella typhimurium TA98 and TA100 induced by 4-nitro-o-phenylenediamine(NPD) and 4-nitroquinoline-N-oxide(NQO). The hexane fraction showed comparatively higher antimutagenic effect than other solvent fractions from the methanol extract. The 50% inhibition concentration($IC_{50}$) of hexane fraction were 2.0 mg/plate, 0.5 mg/plate in TA98 and TA100 respectively. Quercitrin and luteolin were identified in the ethyl acetate fraction from methanol extract of Humulus japonicus by TLC and HPLC.

Isolation of a Marine Bacterium Capable of Biodegrading Poly(butylene succinate)

  • We developed a poly(butylene succinate) (PBS) indicator plate and isolated a marine bacterial colony capable of biodegrading PBS based on the appearance of a clear zone. Growth of the PBS-2 isolate was observed over 4 days of culture at $37^{\circ}C$ in PBS-tryptone basal liquid medium, but not in PBS-deprived control medium. The PBS-2 isolate was named Paenibacillus sp. PBS-2 based on 16S rDNA gene sequencing. The PBS-biodegrading marine bacterium isolated in this study will contribute to the effective management of PBS waste problems in marine environments.

Isolation and Characterization of Soil Streptomyces Involved in 2,4-Dichlorophenol Oxidation

  • Over 50 morphologically distinctive soil Streptomyces were isolated from various Jocations in the Yongin area in Korea and visually screened for dye-decoloring activities on an agar plate. Two Streptomyces species (AD001 and ND002) showed strong dye-decoloring activities on the plate containing congo-red and new-fuchin dyes, respectively. Also, the liquid culture supernatants of these species showed 2,4-dicholophenol (DCP) oxidation activities only in the presence of hydrogen peroxide, a characteristic of Actinomycetes lignin-peroxidase (ALiP)-P3 isoform found in dye-degrading S. viridosporus T7A and S. badius 252. Based on their dye-decoloring capabilities and the 2,4-DCP oxidation kinetic data, it is suggested that these Streptomyces secrete not-yet-characterized extracelluar enzyme(s), whose activities are very similar to the ALiP-P3 enzyme.

  • The purpose of this study was to compare Black-pigmented Bacteroides isolated from necrotic pulp with the presence or absence of eight symptoms associated with pulpal necrosis and to identify the proportion of each Black-pigmented Bacteroides species. The canal contents of fourteen traumatically and cariously necrotized teeth were sampled with a special technique and cultured aerobically and anaerobically for growth in blood agar plate and for Black-pigmented Bacteroides on selective blood agar plate. Each Black-pigmented Bacteroides species were identified by Gram's stain, hemolysis reaction, colony color and morphology and biochemical tests. The results were as follows; 1. 60.9 percent of the bacteria isolated were anaerobic and 39.1 percent were aerobic. 2. Four Black-pigmented Bacteroides species were isolated; B. loescheii (74.1%), B. denticola (11.1%), B. intermedius (7.4%) and B. gingivalis (7.4%). 3. Black-pigmented Bacteroides was found to be significantly related to sinus tract formation and exudate.

Potent Antimutagenic and Their Anti-Lipid Peroxidative Effect of Kaikasaponin III and Tectorigenin from the Flower of Puer-aria thunbergiana

  • The MeOH extract of Pueraria thunbergiana (Leguminosae) flowers and its fractions were subjected to Ames test to test the antimutagenicity. EtOAc fraction (1 mg/plate) decreased the number of revertants of Salmonella typhymurium TA100 by 95% against aflatoxin $B_1{\;}(AFB_1)$. Phytochemical isolation of the EtOAc fraction afforded four isoflavonoids (tectorigenin, glycitein, tectoridin and glycitin) and one saponin (kaikasaponin III). Though the three isoflavonoids other than tectoridin showed significant antimutagenicity, the activity of kaikasaponin III was the most potent. Kaikasaponin III (1 mg/plate) decreased the number of revertants of S. typhymurium TA 100 by 99% against $AFB_1$ but by 75% against N-methyl-N'-nitro-N-nitrosoguanidine (MNNG). Tectorigenin (1 mg/plate) inhibited the $AFB_1$-induced mutagenicity by 90% and MNNG-induced one by 76%. Glycitein and glycitin were less active than tectorigenin and kaikasaponin III. This result suggested that kaikasponin III prevents the metabolic activation of $AFB_1$ and scavenge electrophilic intermediate capable of mutation. The two components with potent activities, tectorigenin and kaikasaonin III, significantly prevented the malondialdehyde formation caused by bromobenzene in the rat.

Evaluation of Dry Rehydratable Film Method for Enumeration of Microorganisms in Korean Traditional Foods (한국 전통식품 중 미생물 분석을 위한 건조필름법 평가)

  • Dry rehydratable film methods were compared to conventional methods for the enumeration of microorganisms in Korean traditional foods. Kimchi, doenjang, kochujang, kanjang, takju, sujeongkwa and sikhe were used as Korean traditional foods. $Petrifilm^{TM}$ aerobic count plate, $Petrifilm^{TM}$ coliform count plate, $Petrifilm^{TM}$ E. coli/coliform count plate, $Petrifilm^{TM}$ yeast and mold count plate and $Petrifilm^{TM}$ staph express count plate were compared to plate count agar, most probable number (MPN) for coliform, MPN for E. coli, potato dextrose agar and coagulase test, respectively. Regression analysis indicated that correlation coefficient values were 0.974-0.998, 0.913-0.995, 0.955-0.978, 0.968-0.986 and 0.998-0.999 for total aerobic bacteria, yeast and mold, coliform, E. coli and S. aureus, respectively. There were no significant differences between two methods, suggesting that $Petrifilm^{TM}$ plates can be used as an alternative to conventional method for the determination of microorganisms in Korean traditional foods.

식품내의 미생물 분리를 위한 dryfilm 방법의 평가연구

  • Dryfilm method by using 3M Petrifilm$^{TM}$ has been examined to replace conventional agar method for isolation of microorganisms from foods. The objectives of the present study were to evaluate suitability of dryfilm method as a microbial isolation method and to determine the effect of antimicrobial agent on dryfilm for isolation of microorganisms from foods. Five different foods, milk, ground beef, fishery surimi, Takju and wheat flour were used to isolate the natural microflora in foods and the inoculated Escheri chia coli. Standard method agar (SMA, Difco) and Petrifilm$^{TM}$ aerobic count (PAC, 3M) were used to isolate total microorganisms from foods. Violet red bile agar (VRBA), brilliant green lactose bile (BGLB) broth and Petrifilm$^{TM}$ coliform count (PCC, 3M) were used to isolate coliforms from foods. E. coli broth (EC broth) and Petrifilm$^{TM}$ E. coli count (PEC, 3M) were used to isolate E. coli from foods. Acidified potato dextrose agar (APDA) and Petrifilm$^{TM}$ yeast & mold count (PYMC, 3M) were used to isolate yeasts and molds from foods. Total aerobic plate counts isolated from five different foods by SMA and PAC (3M) were riot significantly different each other at P<0.05 level and were highly correlated each other ($\geq$0.96). Mugwort extract as an antimicrobial agent did not affect microbial enumeratiion of Dryfilm. Significantly higher number of coliform colonies were formed on VRBA than PCC (3M) from ground beef, but they were not significantly different in coliform colonies from milk samples. PCC (3M) and BGLB were not significantly different for enumeration of coliforms in milk and beef samples. Significantly higher number of E. coli were isolated by EC broth than PEC from ground beef, but these were not significontly different for enumeration of E. coli from milk. Yeast and mold counts isolated from Takju and wheat flour by APDA and PYMC (3M) were not significantly different at P<0.05 level. These data indicate that dryfilm method by using 3M Petrifilm$^{TM}$ can be successively used as an alternative to conventional agar method for enumeration of microorganisms in various foods.

Isolation of Antibiotic-producing Microorganisms Antagonistic to Soilborne Pathogenic Fungi of Bentgrass and Their Antifungal Activity (잔디 토양전염성병원진균에 대한 길항미생물의 분리 및 길항효과)

  • Recently, the importance of management and cultivation of grasses has been increased in Korea. Among these cultural practices, the appropriate control of diseases is considered more important than other cultivation techniques such as fertilization and irrigation. The damages of brown patch and large patch caused by Rhizoctonia spp. and Pythium blight caused by Pythium spp. are serious in the major cultivation area of turfgrass in Korea. Since these diseases are difficult to control by agrochemicals, the damages are very serious if these are occured. The periodic spray of agrochemicals, to protect and control these diseases could make some problems of toxicity and environmental pollution as well as rising of non-target diseases. Therefore, the biological methods to control diseases have been required to decrease problems resulted from overuse of agrochemicals, to conserve natural ecosystem, and to control effectively diseases of grasses in the long period. The number of studies about biological control using antagonistic microorganisms have been increased for last half century. However, the application of biological control method has been very limited. In this study, thirteen isolates of R. cerealis, 8 isolates of R. solani and 3 isolates of Phthyn spp. have been isolated from diseased turfgrass in golf course and grass-culture area that have patch and wilting symptoms of zoysia grass and creeping bentgrass. Isolation frequency of R. cerealis and R. solani was high in especially zoysiagrass, while Pythym spp. was isolated from bent grass at low frequency but showed high pathogenicity. Totally, 205 isolates of soil microorganisms were isolated in this study as primary antagonistic microorganism by Herr's triple agar layer plate and dual culture method using rhizosphere of grasses, soil of crop field as the source of antagonistic microorganisms. Among the 205 isolates, 23 isolates were actinomycetes and 182 isolates were bacteria. All of the actinomycetes were isolated by Herr's method. Antagonistic effect of primary isolated microorganisms was tested for in vitro mycelial growth inhibition against pathogenic fungi isolated from grasses and for inhibition of disease occurrence in 24 well tissue culture plate and pot experiment. Then, four isolated of bacteria which are BG23, BG74, BG136 and BG171 were selected as antagonistic microorganisms against soil-born pathogenic fungi of bentgrass.

Measurement of Adiabatic Wall Temperature on an Impinging Surface by Under-expanded Jet (과소팽창된 충돌제트에 의한 단열벽면 온도 측정)

  • An experimental investigation for impingement of under-expanded, axisymmetric jets on a flat plate has been conducted, and the surface pressure, the adiabatic wall temperature distributions on the plate have been measured in detail. For the explanation on the wall temperature distributions, the total temperature distributions along a free jet have also been measured with total temperature probes. In this study, the under-expansion ratio and the nozzle-to-plate distance have been considered as experimental parameters. Depending on nozzle-to-plate distances, different distributions of adiabatic wall temperature are shown by the energy separation at a jet edge and a impinged surface. Also, the recovery factor on a stagnation point decreases significantly due to the isolation of fluid particles in a central region.

Heavy-weight Impact Noise Reduction of Concrete Slab Reinforcement Using F.R.P (F.R.P 재료 보강에 의한 신개념 중량충격음 저감대책)

  • Low frequency heavy-weight impact noise is the most irritating noise in Korean high-rise reinforced concrete apartment buildings. This low frequency noise is generated by foot traffic due to the fact that Koreans do not wear shoes at home. The transmission of the noise is facilitated by a load bearing wall structural system without beams and columns which is used in these buildings. In order to control low frequency heavy-weight impact noise, floating floors using isolation materials such as glass-wool mat and poly-urethane mat are used. However, it was difficult to control low frequency heavy-weight impact sound using isolation material. In this study, reinforcement of concrete slab using beams and plate was conducted. Using the FEM analysis, the effect of concrete slab reinforcement using FRP(fiber-glass reinforced plastic) on the bang machine impact vibration acceleration level and sound were conducted at the standard floor impact sound test building. The $3{\sim}4dB$ floor impact vibration acceleration level and impact sound pressure level were reduced and the natural frequency of slabs were changed.
