Effect of Taurine Supplement on the Lipid Peroxide Formation and the Activity of Glutathione-Dependent Enzyme in the Liver and Islet of Diabetic Model Mice (당뇨 모델쥐의 간관 췌장에서 타우린이 지질과산화물 생성과 글루타티온 의존성 효소의 활성에 미치는 영향)

  • In this study we wanted to investigate the effect of taurine supplement on the lipid peroxide formation and the activities of glutathione(GSH) dependent enzyme in diabetic model mice. We induce type I diabetes mellitus with alloxan injeciton in ICR mice and type II with high calorie diet in genetically hyperglycemic KK mice. Taurine was given in drinking water at the level of 5%(w/v) for seven days. In type I diabetic model, the malondialdehyde(MDA) of liver and islet significantly increased compared to control group and they significantly decreased by taurine supplement. In type II diabetic model, the concentration of MDA was not changed by taurine supplement. The activities of GSH-peroxidase(GPX) of liver and islet increased in type I diabetic group while decreased in type II. GPX activities were not changed by taurine supplement in the liver of both types but increased in the islet of type II. Taurine supplement has no effect on the activities of GSH S-transferase(GST) in both types. From these results, we suggest that taurine supplement protect against lipid peroxide formation in diabetic model of type I.

Introduction of Saxicolous Lichens Distributed in Coastal Rocks of U-do Islet in Jeju, Korea

  • This study reports, for the first time, the ivestigation of the distribution of Korean saxicolous lichens in the coastal rocks of U-do islet, which is known as an unpolluted zone in Jeju. More than thirty lichens were obtained and investigated from the coastal rocks frequently contacted by seawater. A molecular analysis using PCR amplification of the rRNA ITS regions revealed the coastal rock lichens could be placed into 8 families and 14 genera, Ramalinaceae (Bacidia, Ramalina), Physciaceae (Buellia, Dirinaria, Phaeophyscia, Physcia, Pyxine), Lecanoraceae (Candelaria, Lecanora), Parmeliaceae (Xan­thopannelia), Graphidaceae (Graphis), Pertusariaceae (Pertusaria), Rhizocarpaceae (Rhizocarpon), and Teloschistaceae (Caloplaca), showing a diversity of lichens, with foliose (flat leaf-like), crustose (crust­like), and fruticose (miniature shrub-like) life forms might be distributed in the coastal rocks. These findings suggested the possibility that the lichens identified in the present work might be resistant to a salty environment.

Antidiabetogenic Activity of Trichosanthes kirilowii on the Alloxan-treated Mouse (괄루성 엑기스가 백서에서의 Alloxan 유발 당뇨에 미치는 영향)

  • In order to investigate the antidiabetogenic activity of T. kirilowii on the alloxan-treated mice, the effects of the oral administration of the methanol extract on the blood glucose, the survival rate and the histological changes of the Langerhans' islet were observed. The blood glucose was reached at the highest levels of $265.03\pm15.07$ and $186.41\pm0.16mg/dl at 30 min after alloxan injection, and at the lowest levels of $43.35\pm5.28$ at 4 hour and $20.40\pm3.264mg/dl at 10. hour in experimental and control group, respectively. 57.15% of the experimental group and 4.2%, of the control group were survived more than 24 hours. The life spans of the mice, which were dead within 24 hours, were $631.45\pm60.17$ for the experimental group and $414.60\pm36.24$ min for the control group. The mice from the control group, which were dead within 10 hours, showed totally necrotized $\alpha$- and $\betha$-cells. However, 3 of the 10 mice from the experimental group, which were survived until 24 hours, showed a normal histological feature of the Langerhans' islet.

A Method for the Separation of Mouse Pancreatic Islets Using Discontinuous Percoll Gradient Centrifugation

  • A discontinuous Percoll gradient was used to separate islets from the collagenase-treated mouse pancreas easily and rapidly. Since the osmolality of Percoll is very low, adjustment of its osmolality to 340 mOs/kg $H_2$O was essential for securing the optimal separation. A discontinuous gradient layering with Percoll solution of 1.09 g, 1.07g, and1.05g/m, respectively, when centrifuged at 800$\times$g for 10 min, resulted in an optimal condition for separation and yielded a banding pattern with an even distribution of islet cells. No significant difference was observed in the morphological features between the Percoll-isolated and the manually-isolated islets. In conclusion, the discontinuous Percoll gradient can be effectively used to isolate the pancreatic islets from mice with four-fold higher efficiency compared to the handpicking method.

  • 실험대상(實驗對象)으로서 유우(乳牛) 5두(頭)에게 50% 포도당(葡萄糖) $500m{\ell}$를 생리적식염수(生理的食鹽水)에 혼합(混合) 13~15회(回) 반복(反復) 경정맥(jugular vein)을 통해 주입후(注入後) 혈중(血中) insulin수준(水準)과 Langerhans islets의 조직화학(組織化學)을 관찰(觀察)한바, 혈중(血中) insulin의 수준(水準)은 포도당(葡萄糖)의 투여전(投與前)에 비(比)하여 4~11배(倍)나 증가(增加)됨이 관찰(觀察)되었다. 한편, Langerhans islet의 B cell도 포도당(葡萄糖)의 투여회수(投與回數)에 따라 특이(特異)하게 주름이 관찰(觀察)되었는데 이는 B cell에서의 insulin 합성능(合成能)을 높이는데 그 원인(原因)이 있었다고 본다.
