  • Invasion of enteric bacteria, such as Salmonella and invasive E. coli, into intestinal epithelial cells induces proinflammatory gene responses and finally epithelial cell apoptosis. In this study, we asked whether invasive bacterial infection of human intestinal epithelial cells could upregulate cyclooxygenase-2 (COX-2) gene expression and whether increased COX-2 expression could influence intestinal epithelial cell apoptosis. Expression of COX-2 mRNA and prostaglandin (PG) $E_2$ production were upregulated in HT-29 colon epithelial cells which were infected with S. dublin or invasive E. coli, as examined by quantitative RT-PCR and radioimmunoassay. Inhibition of COX-2 expression and $PGE_2$ production using NS-398, a specific COX-2 inhibitor, showed a significant increase of epithelial cell apoptosis and caspase-3 activation in HT-29 cells infected with invasive bacteria. However, the addition of valerylsalicylate, a specific COX-1 inhibitor, did not change apoptosis in S. dublin-infected HT-29 cells. These results suggest that up regulated COX-2 expression and $PGE_2$ production in response to invasive bacterial infection could contribute to host defense by inhibiting apoptosis of intestinal epithelial cells.

The Shigella Flexneri Effector OspG Interferes with Innate Immune Responses by Targeting Ubiquitin-Conjugating Enzymes

  • Bacteria of Shigella spp. are responsible for shigellosis in humans, a disease characterized by destruction of the colonic epithelium that is induced by the inflammatory response elicited by invasive bacteria. They use a type III secretion system injecting effector proteins into host cells to induce their entry into epithelial cells and triggers apoptosis in macrophages. We present evidence that the effector OspG is a protein kinase that binds various ubiquitinylated ubiquitin-conjugating enzymes (E2s) and blocks degradation of phospho-$I{\kappa}B{\alpha}$ induced upon entry of bacteria into epithelial cells. Transfection experiments confirmed that OspG interferes with the $NF-{\kappa}B$ activation patway by preventing phospho-$I{\kappa}B{\alpha}$ degradation, suggesting that OspG inactivates a component of the $SCF^{{\beta}-TrCP}$ ubiquitin ligase complex (E3) involved in phospho-$I{\kappa}B{\alpha}$ ubiquitination. Upon infection of ileal loops in rabbits, the ospG mutant induced a stronger inflammatory response compared with the wild-type strain, indicating that OspG down-regulates the host innate response induced by invasive bacteria.

Viruses, Bacteria and Helminths of Invasive Carp: Insights from an In Vitro Assay and a Survey with Native Fishes in a Large Midwestern River

  • Pathogen introductions associated with aquatic invasive species threaten ecosystems and biodiversity worldwide. Bigheaded carps (BHC), including Silver Carp Hypophthalmichthys molitrix, Bighead Carp H. nobilis, and their hybrids, are prolific, invasive pests in central US rivers. However, little is known about pathogen effects on invading BHC or how BHC affect the disease risk profile for native fishes in receiving ecosystems. We therefore conducted, from May 2013-December 2014, a systematic pathogen survey for BHC and native fishes in the Wabash River watershed, Indiana, USA. We found Pseudomonas fluorescens, P. putida, and Salmonella enterica DNA in BHC as well as native fishes, although none of these bacteria were exclusively present in BHC. DNA from other bacterial taxa was detected only in native fishes and Common Carp Cyprinus carpio. No gastrointestinal helminths were detected in BHC, although they were common in most native fishes examined. We also conducted in vitro studies on BHC tissues (skin, gill, fin, and fry) and found high sensitivity to Largemouth Bass virus, viral hemorrhagic septicemia virus, and infectious pancreatic necrosis virus. We conclude that BHC are not heavily burdened by bacteria, viruses and parasites in the invaded study ecosystems, although they do harbor native bacteria and show potential for high sensitivity to endemic viruses.

Evaluation of Biological Activities of Invasive Alien Plants for Development of Functional Biomaterials

  • This study aims to confirm the possibility of using the invasive alien plants in Jeju as a functional biomaterial. To achieve this purpose, 70% ethanol extract and solvent fractions were prepared for five invasive alien plants (Hypochaeris radicata, Rumex acetosella, Humulus japonicus Siebold & Zucc., Solanum viarum, Lactuca scariolar) and their antioxidant, antibacterial anti-inflammatory and anti-obesity effects were investigated. The DPPH radical scavenging activity of ethanol extract from invasive alien plants was shown in the order of Rumex acetosella > Hypochaeris radicata > Humulus japonicus. Antimicrobial activity of ethanol extract against food poisoning bacteria (4 species) and oral cavity-induced microorganisms (6 species) was measured. As a result, the extract of Humulus japonicus showed high antibacterial effects against food poisoning bacteria (E. coli, V. parahaemolyticus) and oral microbes (L. casei, S. epidermidis, E. faecalis). In LPS-induced RAW 264.7 cells, the anti-inflammatory effect of ethanol extract from invasive alien plants was investigated. As a result, the NO production inhibition activity was highest in the Rumex acetosella and the Humulus japonicus Siebold & Zucc. ethanol extract, and the NO production inhibition activity was concentration-dependent. In addition, the Rumex acetosella and the Humulus japonicus Siebold & Zucc. ethanol extract showed a concentration-dependent inhibitory effect on cytokine (IL-6) production. These extracts also showed inhibitory activity of COX-2, an inflammatory protein. This suggests that NO production inhibition activity by the extract of invasive alien plants is the result of inhibition of iNOS and COX-2 expression. Currently, organic solvent fractions of crude extract are manufactured and the investigation of active ingredients is continuing along with evaluation of biological activity such as anti-inflammatory. These results are expected to be a major data for the study on the separation and utilization of active ingredients with antioxidant, antibacterial and anti-inflammatory effects using foreign plant crude extract and solvent fractions, and are highly likely to be applied to the development of functional food and cosmetics materials.

  • 저자들은 객담, 기침, 발열과 호흡곤란을 보이고 단순 흉부촬영상 다발성 공동과 경변을 보이며 흉부 전산화 촬영과 폐동맥 혈관조영술상 폐 동맥류가 확인된 환자에서 폐 절제술후 만성 괴사성 침입성 폐 국균증으로 진단된 1예를 경험하였기에 이에 보고하는 바이다.

  • 편도선의 염증반응은 세균이 편도상피세포에 부착되는 것으로부터 시작되며 부착된 세균은 증식하여 집락을 형성한 후 독소를 분비하여 점막 장벽을 뚫고 조직에 손상을 일으킨다. 저자들은 급성편도선염의 병태를 이해하기 위하여 생체내에서 급성편도선염 환자군과 정상인에서 상피세포의 세균부착성에 대한 차이를 알아보고자 본 실험을 시행하였다. 급욍편도선염 환자군 20례와 정상인 대조군 20례를 대상으로 편도선부위를 면봉으로 문지른후 세포혼합물을 Acridine orange로 염색하여 형광현미경하에서 관찰하였다. 50개의 편도상피세포에서 부착된 세균수를 계산하였다. 또한 동시에 근배양을 시행하였다. 급성편도선염군은 대조군보다 상피세포에서 10% 이상 부착된 세균수가 많았으며 (p<0.05), 상피세포에 부착된 세균수는 연령과 유의한 상관관계를 보여주었다.

Shigella flexneri Inhibits Intestinal Inflammation by Modulation of Host Sphingosine-1-Phosphate in Mice

  • Infection with invasive Shigella species results in intestinal inflammation in humans but no symptoms in adult mice. To investigate why adult mice are resistant to invasive shigellae, 6~8-week-old mice were infected orally with S. flexneri 5a. Shigellae successfully colonized the small and large intestines. Mild cell death was seen but no inflammation. The infected bacteria were cleared 24 hours later. Microarray analysis of infected intestinal tissue showed that several genes that are involved with the sphingosine-1-phosphate (S1P) signaling pathway, a lipid mediator which mediates immune responses, were altered significantly. Shigella infection of a human intestinal cell line modulated host S1P-related genes to reduce S1P levels. In addition, co-administration of S1P with shigellae could induce inflammatory responses in the gut. Here we propose that Shigella species have evasion mechanisms that dampen host inflammatory responses by lowering host S1P levels in the gut of adult mice.

특이 과거력이 없는 소아에서 발생한 Streptococcus intermedius 화농 관절염 1예 (A Case of Septic Arthritis due to Streptococcus intermedius in a Healthy Child)

  • Streptococcus intermedius는 입안, 상부 호흡기, 위장관 등에 정상 상재균 중 하나로 존재하는 그람 양성 산소내성 혐기성 알균이다. 면역저하나 심장질환자에서 외상 또는 침습적 치료 후에 발생하는 기회 감염의 원인이 될 수 있다. 저자들은 최근 침습적 치료나 외상 등 특이 과거력이 없이 건강했던 7세 소아에서 S. intermedius에 의해 발생한 엉덩관절 화농 관절염을 경험하였기에 보고하고자 한다. 환자는 내원 3주전부터 왼쪽 엉덩관절 통증이 있었으며, 본원 내원 당일부터 발열이 발생하였다. 엉덩관절 자기공명영상에서 왼쪽 엉덩관절의 급성 화농 관절염으로 진단한 후 수술적 배농을 시행하였고, 혈액과 흡인 관절액에서 S. intermedius가 배양되었다. 수술 후 2주간의 ceftriaxone 정맥주사 후 3주간의 amoxicillin 경구 항생제로 치료한 후 재발없이 안정된 상태를 보이고 있다.

Detection of Enteropathogens in Human Immunodeficiency Virus and Non-Human Immunodeficiency Virus-Infected Children with Acute Diarrhea in an Indonesian Tertiary Hospital Using Multiplex Real-Time Polymerase Chain Reaction

  • Purpose: Diarrhea is one of the leading causes of mortality in children living in developing countries. The etiology of acute diarrhea in each healthcare center varies depending on place, time, and population. This study aimed to identify pathogen patterns in human immunodeficiency virus (HIV)-infected and non-HIV children suffering from acute diarrhea, using multiplex real time reverse transcriptase polymerase chain reaction (RT-PCR), in an Indonesian tertiary hospital. Methods: This cross-sectional study was conducted at Dr. Cipto Mangunkusumo National Hospital from March 2019 to April 2020. Results: The study showed that multiplex RT-PCR results were positive in 58.9% of the specimens, with more positive results in HIV-infected children than in non-HIV-infected children (70% vs. 54.7%). Altogether 72 enteropathogens were detected from all specimens. Enteropathogens in non-HIV children with acute diarrhea consisted of bacteria (70.6%) and viruses (29.4%) with a predominance of enteroaggregative Escherichia coli (25.4%), followed by Campylobacter spp. (11.8%), enteropathogenic E. coli (9.8%), Norovirus GII (7.8%), and Clostridium difficile (7.8%). Enteropathogens in HIV-infected children consisted of viruses (57.1%), bacteria (28.6%), and parasites (14.3%) comprising Norovirus GII (24%), Cryptosporidium spp. (14.3%), Campylobacter spp. (14.3%), Norovirus GI (14.3%), and Astrovirus (14.3%). Cryptosporidium spp. was the only parasite found in this study and was found only in HIV-infected children. In non-HIV children with acute diarrhea, most pathogens were invasive bacteria, while in HIV-infected children, more viral and parasite infections occurred, primarily caused by opportunistic pathogens. Conclusion: The pattern of enteropathogens can help clinicians determine further examinations and appropriate empirical antimicrobial therapy for the patient.
