Quality Characteristics of Garaedduk Enriched with Mealworm (Tenebrio molitor) Powder (갈색거저리 유충 분말 첨가 가래떡의 품질 특성)

  • Mealworm is the most widely used edible insect in Korean traditional food. In this study, the quality characteristics and antioxidant activity of Garaedduk containing different proportions of powdered mealworm (0%, 3%, 6%, 9%, and 12%) were evaluated in order to enhance the utilization of mealworm. The results revealed that moisture content decreased while pH and starch elution increased, with increasing amount of mealworm. Furthermore, the amount of total amino acids, essential amino acids, and non-essential amino acids increased with increasing amount of mealworm. Regarding chromaticity, the L value decreased while the a and b values increased as the amount of mealworm increased. The addition of 6% mealworm showed slight differences in the pore size and surface smoothness as compared to plain Garaedduk. Texture, hardness, chewiness, and gumminess increased, but cohesiveness decreased with increasing amount of mealworm. The preference test evaluating the appearance, flavor, taste, texture, and overall preference showed that 6% mealworm additive group had the highest preference. In addition, antioxidant activity of the newly developed Garaedduk increased with increasing amount of mealworm. In conclusion, Garaedduk enriched with 6% mealworm showed promising results for developing rice cake with improved nutrition and antioxidant activity and excellent overall preference and quality.

Expression of the VP2 protein of feline panleukopenia virus in insect cells and use thereof in a hemagglutination inhibition assay

  • Feline panleukopenia virus (FPV) causes leukopenia and severe hemorrhagic diarrhea, killing 50% of naturally infected cats. Although intact FPV can serve as an antigen in the hemagglutination inhibition (HI) test, an accidental laboratory-mediated infection is concern. A non-infectious diagnostic reagent is required for the HI test. Here, we expressed the viral protein 2 (VP2) gene of the FPV strain currently prevalent in South Korea in a baculovirus expression system; VP2 protein was identified by an indirect immunofluorescence assay, electron microscopy (EM), Western blotting (WB), and a hemagglutination assay (HA). EM showed that the recombinant VP2 protein self-assembled to form virus-like particles. WB revealed that the recombinant VP2 was 65 kDa in size. The HA activity of the recombinant VP2 protein was very high at 1:215. A total of 143 cat serum samples were tested using FPV (HI-FPV test) and the recombinant VP2 protein (HI-VP2 test) as HI antigens. The sensitivity, specificity, and accuracy of the HI-VP2 test were 99.3%, 88.9%, and 99.3%, respectively, compared to the HI-FPV test. The HI-VP2 and HI-FPV results correlated significantly (r = 0.978). Thus, recombinant VP2 can substitute for intact FPV as the serological diagnostic reagent of the HI test for FPV.

Attitude of Chuncheon Citizens on Food Waste Generation (음식물류폐기물 배출에 관한 춘천시민의 의식)

  • This study was conducted to survey the attitude of Chuncheon citizens on food waste generation. About 50% of the citizens were generating $5{\sim}10{\ell}$ of MSW (municipal solid wastes) a week. These answers were almost from below 35 years old. And the citizens answered that the ratio of FW (food waste) to MSW were 10-30%. They were generating FW enveloped in a standardized vinyl bag after dewatering. About 10% of the answers were generating FW with MSW without any dewatering. The reason for FW generation in large quantities was to cook and buy much more food materials than needed. They thought that malodor and harmful insect were the most severe problems from FW storage. The systematic management was the most needed in individual restaurants. They answered that an individual citizen should make an effort to reduce FW generation. However, in fact, they did not do their best to try to reduce FW generation. They agreed that an individual citizen should try to reform 'present food culture' and to think FW as resources.

Various Enterotoxin and Other Virulence Factor Genes Widespread Among Bacillus cereus and Bacillus thuringiensis Strains

  • Many strains of Bacillus cereus cause gastrointestinal diseases, and the closely related insect pathogen Bacillus thuringiensis has also been involved in outbreaks of diarrhea. The diarrheal diseases are attributed to enterotoxins. Sixteen reference strains of B. cereus and nine commercial and 12 reference strains of B. thuringiensis were screened by PCR for the presence of 10 enterotoxigenic genes (hblA, hblC, hblD, nheA, nheB, nheC, cytK, bceT, entFM, and entS), one emetogenic gene (ces), seven hemolytic genes (hlyA, hlyII, hlyIII, plcA, cerA, cerB, and cerO), and a pleiotropic transcriptional activator gene (plcR). These genes encode various enterotoxins and other virulence factors thought to play a role in infections of mammals. Amplicons were successfully generated from the strains of B. cereus and B. thuringiensis for each of these sequences, except the ces gene. Intriguingly, the majority of these B. cereus enterotoxin genes and other virulence factor genes appeared to be widespread among B. thuringiensis strains as well as B. cereus strains.

Analysis of Consumer Receptivity to Pet Food Containing Edible Insects in South Korea (식용곤충을 이용한 반려동물 사료에 대한 소비자 수용도 조사)

  • A survey was conducted among pet owners visiting pet hospitals, using a pet animal app, or being involved in pet breeding, who had the awareness of and intention to purchase animal feed containing edible insects, as well as functional pet food. Results showed that 89.5% of survey participants were pet owners such as those of dogs, cats etc., of whom 55.6% knew about this type of pet food. Nearly half (48.5%) of respondents had an intention to purchase edible insect-based pet food, while the rest of them (51.5%) did not. The two main reasons for deciding to buy it were the excellent nutritional value of insects, and low allergenicity. The most significant reason for refusing purchase was a strong aversion to insects as food.

Pupal Drone Extracts for Anti-wrinkle and Skin-lightening Materials (수벌번데기 추출물의 주름개선 및 미백효과 구명)

  • In this study, we created pupal stage extracts of Apis mellifera L. drones for use in cosmetic materials. The effect of the drone pupae extract (DPE) on HDF cells was assessed for analysis of anti-wrinkle activity by collagen or collagenase gene expression, and the skin-lightening effect was studied by in vitro tyrosinase inhibition and B16F10 melanoma assay; the two cells were found to be non-cellular when the concentration of DPE was 100 ㎍/ml. Albutin concentration (positive control) in the whitening test was set at a capacity of 100 ug/ml and m-melanocyte stimulating hormone (α-MSH). A melanin-producing induction material was set at a concentration of 100 nM, and the expression of collagen type I and MMP1 collagenase was measured using HDF cells. MMP1 expression was seen to reduce in a concentration-dependent manner in treatment with DPE. Inhibiting melanin generation with B16F12 cells indicated a tendency to decrease in the DPE treatment group. Both L-Tyrosine and L-DOPA as DPE were used in an in vitro tyrosinase induction test to demonstrate the effects of tyrosinase suppression on concentrations. The higher the concentration of DPE, the greater the wrinkle reduction and whitening effect. In conclusion, it was found that DPE is an effective smoothing and whitening material by increasing collagen generation and inhibiting collagenase expression and reducing melanin production.

Enzyme Production of A Protease-producing Strain, Bacillus sp SH-8 Isolated from Insect-eating Plant (식충식물로부터 Protease를 생산하는 Bacillus sp. SH-8의 분리와 효소 생산성)

  • A bacterium producing the extracellular protease was isolated from insect-eating plant and has been identified as a member of the genus Bacillus based on partial 165 rRNA sequences. In order to develop the medium composition, effects of ingredients including nitrogen sources, carbon source, metal ions and phosphate were examined for protease production of the isolate, SH-8. Soluble starch increased the protease productivity, while glucose repressed it. Yeast extract was effective nitrogen source for enzyme production, but the pretense production of Bacillus sp. SH-8 was reduced by large amount of yeast extract. The calcium was found to induce pretense activity as well as protease productivity. However, cell growth and enzyme production was completely inhibited by divalent ions such as $Zn^{2+}$, $Cu^{2+}$, $Co^{2+}$ and $Mn^{2+}$. The maximum protease productivity was reached 435 unit/ml in the optimized medium consisting of soluble starch (2%), yeast extract (0.3%), $CaCl_2$ (0.3%), $K_2HPO_4$ (0.01%) and $KH_2PO_4$ (0.01%). The pretense activity of culture filtrate was dramatically decreased after incubation for 26 h.

Effect of herbal medicine by-products on the larval growth of white-spotted flower chafer(Protaetia brevitarsis seulensis) (대체 먹이원으로 한약재 부산물이 흰점박이꽃무지 유충 생육에 미치는 영향)

  • The objective of this study was to verify the stability of nutrient composition by using herbal medicine by-products as an alternative food source and to examine the growth effect on Protaeria brevitarsis seulensis larvae. As a result of comparing the nutritional components of food source, crude protein, crude fat, and crude ash content, except crude fiber content, was high in both non-fermented and fermented medicinal herbal by-products. Especially, crude protein content was highest. Cadmium, lead, mercury, and other heavy metals were not detected and thus stability as alternative food was confirmed. The growth comparison based on the feeding sources showed no significant difference between the fermented oak sawdust fed control group and the herbal medicine by-products fed laboratory group from week 1 to week 3. The weight of a 4 week larva was 0.137 g in the control group and 0.671 g in the laboratory group and so began to reveal differences at a significant level (p<0.05). As a result of comparing weights of Protaetia brevitarsis seulensis larvae according to the level of herbal medicine by-product addition, HMB40 recorded the heaviest weight in week 7. Statistical analysis showed that there was no significant difference in each body weights of HMB40 and HMB80 at week 5 (p<0.05). These results indicate that if the shipping date of an edible insect is a third instar larva, it arrives at the time of shipment at week 5. Thus feeding HMB40 and HMB80 at the 5th week is the most effective.

Improvement of Hygienic Quality and Long-term Storage of Dried Red Pepper by Gamma Irradiation (감마선 조사에 의한 건고추의 위생화와 장기 안전저장)

  • Dried-red pepper, whole and powdered types, was subjected to a storage-study by investigation the effects of packaging methods (polycloth & polyethylene/polycloth, whole dried-red pepper; nylon/polyethylene-lam-inated film, red pepper powder), temperature (ambient, $5{\sim}10^{\circ}C$) and gamma irradiation doses (0-10 kGy). After 6 months storage in polyclith sack at ambient temperature, all whole dried-red pepper showed quality deterioration, such as weight change, insect infestation, discoloration and chemical changes, After 2 years storage in combined packaging with polyethylene/polycloth sack of 5-7.5 kGy irradiated whole dried-red pepper at ambient temperature, however, quality deterioration was not observed. Gamma-irradiated red pepper powder (7.5-10kGy) showed a good quality in hygienic, physicochemical and organoleptic evaluation after 2 years of storage at ambient temperature.

Expression of Codon Optimized β2-Adrenergic Receptor in Sf9 Insect Cells for Multianalyte Detection of β-Agonist Residues in Pork

  • ${\beta}_2$-adrenergic receptor (${\beta}_2-AR$) was expressed efficiently using Bac-to-Bac Baculovirus Expression System in Sf9 cells as a bio-recognition element for multianalyte screening of ${\beta}$-agonist residues in pork. Sf9 cells were selected as the expression system, and codon optimization of wild-type nucleic acid sequence and time-dependent screening of expression conditions were then carried out for enhancing expression level and biological activity. Under optimum conditions of multiplicity of infection (MOI) = 5 and 48 h post transfection, the protein yield was up to 1.23 mg/ml. After purification by chromatographic techniques, the purified recombinant protein was applied to develop a direct competitive enzyme-linked receptor assay (ELRA) and the efficiency and reliability of the assay was determined. The IC50 values of clenbuterol, salbutamol, and ractopamine were 28.36, 50.70, and $59.57{\mu}g/l$, and clenbuterol showed 47.61% and 55.94% cross-reactivities with ractopamine and salbutamol, respectively. The limit of detection (LOD) was $3.2{\mu}g/l$ and the relevant recoveries in pork samples were in the range of 73.0-91.2%, 69.4-84.6%, and 63.7-80.2%, respectively. The results showed that it had better performance compared with other present nonradioactive receptorbased assays, indicating that the genetically modified ${\beta}_2-AR$ would have great application potential in detection of ${\beta}$-agonist residues.
