This study was performed to evaluate the activities of glutathione S-transferase (GST) and lactate dehydrogenase (LDH) in Maddin-Darby canine kidney (MDCK) cells infected with virus and/or treated with amantadine. On cell morphological findings, monolayer fractions in MDCK cells infected with virus were exfolated more than 80% in 1 TCID$_{50}$ group and that in 10 TCID$_{50}$ were completely exfolated after 3 days during infectious process. In proportion to the dose of amantadine, activities of GST and LDH of MDCK cells were significantly decreased and those of LDH in medium fraction were more significantly increased compared with control. According to in both dose and time of virus innoculation, activities of GST and LDH in MDCK cells were significantly decreased in 1 and 10 TCID$_{50}$ infected cells after 3 days. LDH activities in infectious medium were remarkably rised at 10 fold. In case of the cell line inoculated with type A 100 TCID$_{50}$ and additionally treated with amantadine, the decreasing rate to the control in activities of GST and LDH was lower than that in those in case of that infected with virus only. These results suggested that virus infection and amantadine treatment may effect the activity of the detoxicating enzyme in the target cells.
The optimal condition of in vitro regeneration and transformation were investigated for newly bred potato varieties in Korea. Leaf and internodal stem tissues of 'Chubak', 'Namsuh', 'Jasim', 'Jopung' and 'Jowon' were used to investigate the influence of growth regulator on regeneration efficiency The effect of phenolic compound acetosyringone on gene transformation efficiency was also investigated. Leaf tissue of 'Jowon' and internodal stem tissues of 'Jopung' were showed high regeneration efficiency on M5 medium supplemented with 0.1 mg/L GA₃, 2.0 mg/L Zeatin and 0.01 mg/L NAA. The other three potato cultivars were showed low regeneration efficiency less than 25%. The effect of acetosyringone on Agrobacterium-mediated gene transformation with leaf and internodal stem tissues were noticeable. By adding the 75 μM of acetosyringone during the Agrobacterium innoculation, the transformation efficiency was increased up to 1.5∼4.0 fold compare to non-treated control. In case of 'Jowon' the transformation efficiency was 87.9% in leaf tissue and 'Jopung' was 68.4% in internodal stem tissues.
The effect of arbuscular mycorrhizae(AM) on the growth of trifoliate orange seeding were investigated in volcanic ash soil. Trifoliate orange is used as a root stock of citrus in Cheju island. Seedings innoculated with AM fungi were grown for 16 weeks in pots of various levels of fertilizer. Growth characteristics and mineral nutrient contents were measured and spores of AM fungi colonized were identified. Seventy % of the replicsted pots of seedings were colonized by AM in the treatment of high level fertilizer and additional phosphate (40g of 21-17-17 complex fertilizer and 50g of fused phosphate added to 50L of soil). In treatments of low levels fertilizer or without fused phosphate addition, the pots colonized were less than 20 %. Colonization of trifoliate seedings with AM fungi greatly increased the growth of seedings. Shoot length and weight of shoot and root positively regressed on AM colonization ratio. AM colonization caused higher concentrations of P, Cu and Mg in plant, and the relations were significant at 5 % level. Contents of N and Zn in plants also tended to increase, while that of Ca to decrease, with increasing colonization ratio. Four species of AM fungi - Glomus deserticola, G. rubiforme, G. vesiculiferum and Acaulospora sp, - were found in the soil where roots of trifoliate orange as an innoculation materials were collected. All of the 4 species were found in the inoculated pot soils after the seedling growth, indicating that these species can be colonized in trifoliate orange roots.
A series of experiments have been made on the mass culture of Phaeodactylum tricornutum, Platymonas sp. and Chlorella sp. in the laboratory. The shortest lag phase was found In the culture of Ph. tricornutum followed by Platymonas sp. and Chlorella sp. As compared to the aeration culture, the stagnant culture, in general, showed long duration of the lag phase, short period of the exponential phase and extremely small daily increment. The relative growth constants of Ph. tricornutum, Chlorella sp. and Platymonas sp. were $0.302{\pm}0.028$, $0.226{\pm}0.013$, and $0.151{\pm}0.008$, respectively The maximum daily increment of the three species and then daily ages are as follows: Ph. tricornutum Maximum daily increment : 47.5, Daily age : 10, Platymonas sp. Maximum daily increment : 5.6, Daily age : 14, Chlorella sp. Maximum daily increment : 21.1, Daily age : 14 Comparing the packed cell volume with a certain number of cells, the largest value was found in the population of Ph. tricornutum followed by Platymenas sp. and Chlorella sp. A straight line relationship exists between the two values, and the magnitude of the relationship coincides well with the size of the cells. The culture of Ph. tricornutum was proved satisfactory for feeding the larvae of bivalves at about 12 days after innoculation and both of Platymenas sp. and Chlorella sp. were about 16 days respectively.
Calli and suspension cultures were obtained following inoculation of the explant from leaves of Ginkgo biloba L on the supplemented MS basal medium. The obtained calli and suspension cultured cells were able to produce detectable amounts of ginkgolides which are known as natural specific PAF antagonists. The production of ginkgolides in the calli and suspension cultured celles were identified using GC/MS, GC and HPLC with authentic ocmpounds. Since the production of ginkgolides A and B the calli and suspension cultured cells had been confirmed, effects of types and concentration of plant growth regulators, media and illumination on the induction and growth of the callus were studied. The concentrations of growth regulators for optimal callus were studied. The concentrations of growth regulators for optimal callus induction were studied. The concentrations of growth regulators for optimal callus induction were 1.0 to 2.0 mg/L for NAA and o.1 mg/L for kinetin. The growth of the Callus seemed to be more simnultaed with the combination of NAA and kinetin than NAA and BA with illumination at all concentration ranges of 1.0 to 4.0 mg/l for NAA and o.1 to 1.0 mg/L for kinetin or BA studied. Amogn 8 different media used, the induction rate of callus on Anderson, Eriksson, and Shenk and Hildebrant at 4 weeks after the innoculation was almost the same as that of MS. However, callus was rarely induced on Heller or White medium. Suspension cultures were easily initiated with 3 g of callus (fresh weight) derived from ginkgo leaves on supplemented MS medium. A typical growth curve of suspension cultured cells could be obtained by measuring the fresh weight of the suspension cultured cells at every 3 days. To improve the growth of suspension cultured cells of ginkgo, effects of concentrations of NAA, sucrose, phosphate ions and molar ratio of $NH_{4}^+\;to\;NO_{3}^-$ ions in the culture medium were studied. The maximum growth of the cells was achieved when the culture medium contained 1.0 mg/L of NAA, 30 g/L sucrose, 1.75 mM phosphate ions and 1:5 molar ratio of $NH_{4}\;to\;NO_{3}^-$ ions.
Economic injury levels and economic thresholds were estimated for the american serpentine leafminer (Liriomyza trifolii) on greenhouse eggplant. Liriomyza trifolii density was increased until the late June and decreased after the July in innoculation on may 17. Growth of an aerial plants and fruits were not different in treatment respectively. But total number of fruits and yields were decreased on higher inoculation density. Whereas the rate of yield loss was increased. The rates of damaged leaf by L. trifolii were increased on higher inoculation density and the peak was 65%. The number of commodity fruits and the rates of commodity fruit were become lower than non-treatment (72.2%). The rates of damaged leaf area were 5.3, 11.7, 19.7, 25.7% on inoculation densities and the rates of yield loss were 0.6, 4.8, 9.8, 14.7%, respectively. There existed close correlation between rate of yield loss and inoculation density (Y = 0.76779X + 0.298354, $R^2\;=\;0.9599$). Considerated of the results, the economic injury levels of L. trifolii on eggplant greenhouse was 6.1 adults per 4 plant and the economic thresholds was 4.9 adults per 4 plant.
Soybean seed injury was analyzed in the experiments in which the bean bug, Riptortus pedestris(Hemiptera: Alydidae), was released into screen-caged pots containing full seed stage(R6) of soybean. When the different stages of insects, from the 3rd instar nymphs to adults, were released into pots during 8 days in soybean R6 stage, soybean seeds with injury marks (B-type seeds) increased. The weight reduction ratio in B-type seeds was highest in the injury by the 5th instar nymphs, while the daily-produced number ratio of B-type seeds was small in the treatment. In the injury by the different number of adult released into pots, 4, 8, 16 adults caused a significant increase of B-type seeds, while 16 adults caused the significant increase of the deformed (C-type) seeds. In the injury by the different release period of adults, the total number of pods was not significantly different among treatments, while the total seed number harvested was significantly small in the release for 48 days. The release for 8 and 16 days caused a significant increase of B-type seeds, while the release for 48 days caused the increase of C-type seeds. The results indicated that injury of soybean R6 stage by the bean bug produced soybean seeds with distinct injury marks at relatively low density and during short term attack period, while it produced deformed seeds at high density and during long attack period.

The occurrences of the major diseases and the densities of air-born microbes were surveyed in the cultivation facilities for oyster mushroom (Pleurotus ostreatus), king oyster mushroom (Pleurotus eryngii), and enoki mushroom (Flammulina velutipes) in different areas of Korea. Green mold disease was most often developed in oyster mushroom bed cultivation with the disease incidence rate of approximate 10% while the disease incidences from bottle and plastic envelop cultivation were less than 1~2%. In the bed cultivation, the major air-born microbes in the growth room were Aspergillus, Penicillium, Trichoderma, and Curvularia with the total fungal population density of 567~1,297 CFU/$m^3$ . However, only Trichoderma and Penicillium were detected in the growth rooms and innoculation rooms of bottle and plastic envelop cultivation with the densities of 350~700 CFU/$m^3$ and 160~260 CFU/$m^3$, respectively. The bacterial diseases become evident in the growth rooms of bottle and plastic envelop cultivation with the approximate incidence rate of 10%. The identified bacterial species were Brevibacillus levelkil, Rhizobium radiobacter, Brevundimonas vesicularis, Pseudomonas mosselii, Microbacterium testaceum. Sphingomonas panmi, Sphingomonas yabuuchiae, Paracocus dinitrificans, Curtobacterium flaccumfaciens pv. flaccumfaciens and some unidentified bacteria with the densities of 40~6,359 CFU/$m^3$ in the growth rooms and 9 CFU/$m^3$ in the inoculation room. This study indicated that the green mold disease by fungal strains was the major mushroom disease in the bed cultivation and suggested that the contamination of bacteria and fungi together in the growth media could result in severe production loss. The plastic envelope and bottle cultivation were evidenced to be less susceptible to such contaminations.
This study was conducted to investigate the hypoglycemic effects and the production conditions of the Korean organic native rice cultured by Ganoderma lucidum. The broth culture of Ganoderma lucidum, the innoculation and culture of Ganoderma lucidum to the Korean organic native rice, and the oral administration of the Korean organic native rice cultured by Ganoderma lucidum to the streptozotocin-induced diabetic rats, were carried out. Then, the blood glucose level, the serum total cholesterol and triglyceride, and the activities of alanine aminotransferase and aspartate aminotransferase, were analyzed. The pH 4.5 at $30^{\circ}C$ was the optimal condition of the broth culture of Ganoderma lucidum. When the physical shape and the smell of the produced rice were considered, the optimal conditions to produce the Korean organic native rice cultured by Ganoderma lucidum, were the seeding of Ganoderma lucidum to Korean organic native rice in the rate of 7.5% weight, and the culture period of 9days at $28^{\circ}C$. In streptozotocin-induced diabetic rats, the blood glucose level of the dieted group by the Korean organic native rice cultured by Ganoderma lucidum for 2 weeks, were significantly decreased when compared with the control. And, the levels of the total cholesterol and triglyceride in serum were ranked less than those of the control. Also, in streptozotocin-induced diabetic rats, the activities of alanine aminotransferase and aspartate aminotransferase in the serum of the dieted group by the Korean organic native rice cultured by Ganoderma lucidum for 2 weeks, were lower than those of the control.

Antimicrobial effects of the specific IgY separated from eggs which were laid by hens vaccinated with Streptococcus mutans were investigated. The comparison tests of vaccination, addition levels of crude specific IgY, and innoculation concentration were applied by microscopic observation and turbidity test. Ten% addition of crude specific IgY obtained from vaccinated hens showed agglomerative clusters of S. mutans cells in supernatants and sediments, while crude IgY produced by non-vaccinated hens showed no cluster. IgY addition above 5% showed agglutinating clusters of most S. mutans cells and there was definite difference between IgY addition below 2.5% and above 5%. Concentration tests of crude IgY revealed that antimicrobial effects were differentiated by addition level and addition over 10% produced satisfactory results with turbidity test. The cluster size was dependent upon concentration of crude IgY addition. $10^5\;cfu/mL$ inoculation showed agglutinated cells and extent of agglomeration was proportional to cell numbers. Study of inoculation levels showed that 10% addition of crude IgY decreased turbidity effectively regardless of number of S. mutans cells. Plaque formation decreased to 75% with 15% addition of specific IgY concentration. These results implied that IgY separated from eggs laid by S. mutans-vaccinated hens might prevent dental caries caused by S. mutans.
