Physiological Characteristics and Anti-Diabetic Effect of Pediococcus pentosaceus KI62

  • The purpose of this study is to examine the physiological characteristics and anti-diabetic effects of Pediococcus pentosaceus KI62. The α-amylase and α-glucosidase inhibitory activity of P. pentosaceus KI62 was 94.86±3.30% and 98.59±0.52%, respectively. In MRS broth containing 3% maltodextrin inoculated by P. pentosaceus KI62, the amounts of short chain fatty acids (SCFA) were propionic acid 18.05±1.85 mg/kg, acetic acid 1.12±0.07 g/100 mL, and butyric acid 2.19±0.061 g/kg, and those of medium chain fatty acids (MCFA) were C8 0.262±0.031 mg/kg, C10 0.279±0.021 mg/kg, and C12 0.203±0.009 mg/kg. Compared to sixteen antibiotics, P. pentosaceus KI62 had the highest sensitivity to penicillin-G and rifampicin, as well as the highest resistance to vancomycin and ampicillin. The strain also showed higher leucine arylamidase and valine arylamidase activities than other enzyme activities, but it did not produce β-glucuronidase which is carcinogenic enzymes. The survival rate of P. pentosaceus KI62 in 0.3% bile was 91.67%. Moreover, the strain showed a 98.63% survival rate in pH 2.0. P. pentosaceus KI62 exhibits resistance to Escherichia coli, Salmonella Typhimurium, Listeria monocytogenes, and Staphylococcus aureus at rates of 29.41%, 38.10%, 51.72%, and 50.47%, respectively. P. pentosaceus (23.31%) showed a similar adhesion ability to L. rhamnosus GG, the positive control (24.49%). These results show that P. pentosaceus KI62 has possibility as a probiotic with anti-diabetic effects.

Inhibitory Effect of Citrus Unshiu Peel and Cimicifuga Dahurica Extracts on Melanogenesis accroding to the Various Extraction Methods (추출조건에 따른 진피(陳皮) 및 승마(升麻) 추출물의 미백 활성효과)

  • Objectives: Excessive melanin production leads to skin pigmentation, which causes various cosmetic and health problems. Citrus unshiu Markovich peel (CS) and Cimicifuga dahurica (CD) have long been widely used as a oriental medicinal plant because of their pharmacological properties including anti-ulcer, anti-oxidant, and anti-inflammatory properties. Methods: In this study, we investigated the inhibitory effects of CS, CD extracts or CS:CD=1:2 mixture on melanogenesis according to the various extraction methods. CS and CD extracted were prepared by ethanol extraction (EE), ultrasonification extraction (USE), Supercritical extraction (SCE), reflux extraction (RE), respectively. Results: DPPH radical scavenging and tyrosinase inhibitory activity of CD extracts or CS:CD=1:2 mixture were increased in dose-dependent manners. In addition, we evaluated the effect of CS, CD extracts or CS:CD=1:2 mixture on tyrosinase activity and melanogenesis in α-melanocyte stimulating hormone-induced B16-F10 melanoma cells. CS, CD extracts or CS:CD=1:2 mixture significantly inhibited tyrosinase activity and melanogenesis at 10-200 ㎍/mL. Conclusions: Therefore, our study suggests that CS and CD extracts have potential as a safe treatment for excessive pigmentation or as a natural ingredient in cosmetics.

Basic research for exploring anti-obesity activity of several medicinal plants from Mongolia (몽골산 약용식물 수종의 항비만 활성 탐색을 위한 기초 연구)

  • Objective : Obesity in modern society has a significant impact on pathological, psychological and social problems. Therefore, many studies on obesity treatment using herbal extracts with fewer side effects have been conducted. This study was designed to investigate the effect of inhibiting fat accumulation in vitro in order to Mongolian medicinal plants find anti-obesity candidate among. Methods : We measured pancreatic lipase inhibitory activity in Mongolian medicinal plants extract. Cytotoxicity of these extracts was monitored in 3T3-L1 cells by MTT assay. In addition, the anti-obesity effects was confirmed at concentrations of 0.2, 0.1 and 0.05 mg/㎖ through Oil red O staining. Results : Among Mongolian medicinal plants, Rheum undulatum roots in September (RURS), Paeonia anomala L. (PAL), and Fragaria orientalis (FO) showed the highest pancreatic lipase inhibitory activity. As a result of the MTT assay, more than 80% was judged to be non-toxic, and the concentration was determined, and as a result of evaluating the lipid accumulation inhibitory effect, 6 types were selected as candidates. Conclusion : Based on these results, the top 7 species expected to be used as anti-obesity functional materials were selected. However, additional efficacy verification and mechanism of action need to be established in the future. So, it is expected that the medicinal plants verified through this will be used as functional materials for the prevention and treatment of obesity.

Design, Synthesis, and Functional Evaluation of 1, 5-Disubstituted Tetrazoles as Monoamine Neurotransmitter Reuptake Inhibitors

  • Tetrazoles were designed and synthesized as potential inhibitors of triple monoamine neurotransmitters (dopamine, norepinephrine, serotonin) reuptake based on the functional and docking simulation of compound 6 which were performed in a previous study. The compound structure consisted of a tetrazole-linker (n)-piperidine/piperazine-spacer (m)-phenyl ring, with tetrazole attached to two phenyl rings (R1 and R2). Altering the carbon number in the linker (n) from 3 to 4 and in the spacer (m) from 0 to 1 increased the potency of serotonin reuptake inhibition. Depending on the nature of piperidine/piperazine, the substituents at R1 and R2 exerted various effects in determining their inhibitory effects on monoamine reuptake. Docking study showed that the selectivity of tetrazole for different transporters was determined based on multiple interactions with various residues on transporters, including hydrophobic residues on transmembrane domains 1, 3, 6, and 8. Co-expression of dopamine transporter, which lowers dopamine concentration in the biophase by uptaking dopamine into the cells, inhibited the dopamine-induced endoctytosis of dopamine D2 receptor. When tested for compound 40 and 56, compound 40 which has more potent inhibitory activity on dopamine reuptake more strongly disinhibited the inhibitory activity of dopamine transporter on the endocytosis of dopamine D2 receptor. Overall, we identified candidate inhibitors of triple monoamine neurotransmitter reuptake and provided a theoretical background for identifying such neurotransmitter modifiers for developing novel therapeutic agents of various neuropsychiatric disorders.

Addictive Effects of Ecklonia cava Methanol and its Solvent-soluble Extract Against Antibiotic-resistant Cutibacterium acnes Causing Acne Vulgaris (항생제 내성 Cutibacterium acnes에 대한 감태(Ecklonia cava) 추출물의 항균 시너지 효과)

  • Acne vulgaris is often associated with acne-related bacteria such as Cutibacterium acnes. In this study, we investigated the antibacterial effects of the methanol extract of the algae Ecklonia cava and its solvent-soluble extract against C. acnes. Among five solvent fractions, the ethyl acetate (EtOAc) fraction exhibited the strongest antibacterial activity against C. acnes. Furthermore, the EtOAc-soluble extract exhibited the highest total phenolic contents among the five solvent fractions tested. The EtOAc subfraction 07 (Fr. 07) extract showed the highest antibacterial effect against C. acnes and isolated C. acnes along with minimum inhibitory concentration (MIC) ranging from 32 to 64 ㎍/mL. Additionally, MICs of antibiotics against antibiotic-resistant C. acnes strains were substantially reduced when antibiotics were combined with Fr. 07, suggesting that Fr. 07 restore the antibacterial activity of the antibiotics. The fractional inhibitory concentration indices clearly revealed an additive synergistic effect of Fr. 07 with antibiotics. The results of the present study suggest a potential role for E. cava in the control of infections related to acne vulgaris.

Inhibitory Activity of Garlic Fermented by Pediococcus pentosaceus KACC 91419 against Antibiotic-resistant Pathogens

  • The aim of this study was to screen lactic acid bacteria for the fermentation of garlic and to assess the increase in inhibitory activity of garlic fermented against antibiotic-resistant pathogens for use as an animal feed supplement. We screened 45 strains of lactobacillus for the fermentation of garlic. Of these strains, 23 showed similar growth rates with or without allicin. Cultures of the 23 strains were mixed with an equivalent amount of garlic juice and incubated overnight at $37^{\circ}C$. The three strains with the lowest pH values were Lactobacillus paracasei KCTC 3169, L5 strain, and L. reuteri SW. Garlic juice fermented by the L5 strain more strongly inhibited antibiotic-resistant pathogenic bacteria than L. paracasei KCTC 3169, L. reuteri SW, or garlic juice itself. By examining carbohydrate utilization, morphologic properties and 16S rRNA gene sequences, we identified the L5 strain as Pediococcus pentosaceus and deposited it in the name of P. pentosaceus KACC 91419 into the Korea Agricultural Culture Collection. To identify the antimicrobial compound from the garlic filtrate fermented by P. pentosaceus KACC 91419, we fractionated P. pentosaceus KACC 91419 culture on a C18 column and checked the antimicrobial activity of fractions A6 to A10. Only fraction A9 showed inhibitory activity on Staphylococcus aureus. Comparing the mass spectra of the fractions with and without antimicrobial activity, we observed a single dominant product ion (m/z 157.99) from the fraction showing antimicrobial activity. Its molecular mass (157.99) was 2 atomic mass units less than that of allicin (162.02). This suggests that allicin might be converted to its derivative, which has antimicrobial activity, during fermentation by P. pentosaceus KACC 91419.

Change of Biological Activity of Melon (Cucumis melo L.) according to Frozen Storage Period (냉동저장기간에 따른 멜론(Cucumis melo L.)의 생리활성 변화)

  • The change of biological activities of melon were investigated during frozen storage. The total phenolic concentrations in melon juice and water extract were 296.25 and $433.25\;{\mu}g/mL$, respectively. The total flavonoid contents in melon juice and water extract were 20.83 and $53.58\;{\mu}g/mL$, respectively. Antioxidant activities of melon juice and water extract were determined. The DPPH of water extract of melon (85.84%) was higher than the melon juice (60.58%). ABTS of melon juice and water extract were 94.50 and 99.30%, respectively. SOD-like activity and xanthine oxidase inhibitory activity of melon of water extracts were higher than those of melon juice. $\alpha$-Glucosidase inhibitory activity of melon juice and water extract were 22.42 and 23.43%, respectively. The changes in the antioxidant activity of melon was insignificant until 6 months of frozen storage. Therefore, it was expected that frozen storage of melon was useful preservation expedient for consistent supply of raw materials.

Functional Chemical Components and Their Biological Activities of Houttuynia cordata and Lespedeza cuneata (어성초와 야관문의 기능성 성분 분석과 항산화, 항고혈압, 및 항당뇨 활성)

  • For this study, we prepared organic solvent fractions from methanol extracts of Houttuynia cordata and Lespedeza cuneate, and analyzed their chemical components and various biological functions such as anti-oxidation, angiotensin-converting enzyme (ACE) inhibition, and α-glucosidase inhibitory activities. We found that DPPH radical scavenging activity was highest in the ethyl acetate fractions of Houttuynia cordata (90.8%) and Lespedeza cuneata (91.2%), whereas ABTS radical scavenging activity was highest in the ethyl acetate fractions of Houttuynia cordata (86.1%) and the chloroform fractions of Lespedeza cuneata (95.6%). FRAP activity was highest in the ethyl acetate fraction of Houttuynia cordata (360.1 mg TE/g) and Lespedeza cuneata (239.2 mg TE/g). ACE inhibitory activity was highest in the chloroform fraction of Houttuynia cordata (13.2%) and Lespedeza cuneata (35.2%). And, α-glucosidase inhibitory activity was highest in the ethyl acetate fraction of Houttuynia cordata (56.3%), and the water residue of Lespedeza cuneata (93.6%). Finally, we investigated the DPPH radical scavenging activity of 20 types of pure compounds identified in Houttuynia cordata and Lespedeza cuneate. The results show that quercetin demonstrates the highest DPPH radical scavenging activity. Overall, these results help us to understand the functional chemical components of Houttuynia cordata and Lespedeza cuneate and the biological effects of these components.

Beauty activity of extracts from Saurus chinensis by ultra-fine ground (초미세 분쇄한 삼백초(Saurus chinensis) 추출물의 미용 활성)

  • In this study, beauty food activities were determined for extracts of Saurus chinensis as functional beauty food. Tyrosinase inhibitory effect was increased as more 4 folds by ultra-fine grind technology than normal grind technology in ethanol extracts from Saurus chinensis. A ultra-fine ground technology was used to increase the extraction yield for bioactive compounds from medicinal plants. Tyrosinase inhibitory activity, which is related to skin-whitening, was confirmed to be 100% for ethanol extracts at 1,000 ppm of phenolics. Anti-wrinkle activity was determined showing 30% of inhibition effect at above 1,000 ppm phenolic concentration. The elastase inhibitory activity of water and ethanol extracts from ultra-fine ground Saurus chinensis showed higher inhibition activity than compared to extracts from normal ground Saurus chinensis. Astringent activity in ethanol extracts was determined showing over 70% at 250 ppm of phenolics. Based on these results, Saurus chinensis extracts can be used as a functional beauty food and cosmetic with whitening, anti-wrinkle and pore contraction effects.

In vitro Antioxidant Activity and α-Glucosidase and Pancreatic Lipase Inhibitory Activities of Several Korean Sanchae (우리나라 일부 산채의 in vitro 산화 방지 활성, 알파-글루코시데이스와 췌장 라이페이스 저해 활성)

  • This study evaluated in vitro antioxidant activity, antioxidant content, and ${\alpha}$-glucosidase and pancreatic lipase inhibitory activities of ethanol extracts of samnamul (shoot of Aruncus dioicus), miyeokchwi (Solidago virgaurea), daraesoon (shoot of Actinidia arguta Planchon), and bangpungnamul (leaves of Ledebouriella seseloides), as muknamul, and fresh chamnamul (Pimpinella brachycarpa). Tocopherol content (4.8-78.3 mg/100 g) of sanchae was lower than polyphenols (4.4-12.2 g/100 g). Daraesoon with high tocopherol contents showed high antioxidant activity and ${\alpha}$-glucosidase and pancreatic lipase inhibitory activities. Samnamul had the highest levels of polyphenols and flavonoids, the highest antioxidant activity, and ${\alpha}$-glucosidase inhibition. Antioxidant activity was correlated with flavonoid content ($r^2=0.8895$), but was not correlated with the levels of other antioxidants tested, suggesting that polyphenol content in samnamul, miyeokchwi, daraesoon, bangpung, and chamnamul might not be critical determinant of antioxidant activity. Our results strongly suggest that samnamul and daraesoon could be useful in the treatment of diabetes and obesity.
