The Effect of Cyclosporin A on Osteoblast in vitro (Cyclosporin A가 in vitro에서 조골세포에 미치는 영향)

  • Cyclosporin A(CsA) is an immunosuppressive agent widely used for preventing graft rejecting response in organ transplantation. The basic properties of CsA to osteoblast has not been well known yet. A better understanding of the mechanisms of CsA function on bone could provide valuable information regarding basic properties of bone remodeling, pharmacotherapeutic intervention in metabolic bone disease, and the consequences of immunosuppression in bone physiology. The purpose of this study was to investigate the effect of CsA on osteoblast by evaluating parameters of proliferation, collagen synthetic activity, alkaline phosphatase activity, and ALP mRNA expression in mouse calvarial cell. 1. CsA ($3{\mu}g/m{\ell}$) treated mouse calvarial cell showed statistically significant increase in cell proliferation.(P<0.05) 2. CsA($1,\; 3{\mu}g/m{\ell}$) treated MC3T3 cell line showed statistically significant increase in cell proliferation. 3. The amount of collagen of CsA($3{\mu}g/m{\ell}$) treated mouse calvarial cell was decreased statistically significantly. 4. Alkaline phosphatase activity was increased statistically significantly in CsA treated group($1{\mu}g/m{\ell}$). 5. mRNA expression of ALP was increased in CsA treated group These results suggest that CsA could affect bone remodeling by modulating proliferation & differentiation of osteoblast.

  • The purpose of this study is to make the internet website on the order-made fashion design so as to do meet the need of netizen in the 21st century. It made the consumer do the design which he or she wants by the combination of the compositions that do select the pattern of the skirt design to fit his or her size and body shape. For the set-up of website, the elements of composition must be considered, and also the planning and design must be done fur the order-made fashion design. With the internet website, it's possible to do reach the below points; first, it can make the consumer do the design which he or she wants by himself or herself; second, it can provide him or her the information of measuring his or her body shape to buy the design pattern which he or she wants. Third, it can be contributed to make the brand more active by improving his or her satisfaction with the design and pattern which he or she wants. Also it can bring up the effects as fellows; first, lashing power of the design can be increased by the order-made fashion design to attract his or her interest, and also it can make him or her more satisfied. Second, the competitiveness can be strengthened by the differentiation of the fashion site between the existing design and the order-made fashion design. Third, the production of design can be higher and also its expense can be reduced even for the small and medium-sized clothes maker or merchants. Finally, it is able to enlarge the applicable field into the design of small articles, home fashion, and textile which they want to sell the differentiated and order-made products.

An Exploratory Study on the Factors of Difference in Trust Levels for Internet-Only Banks by Age Groups (인터넷전문은행의 신뢰 결정요인의 연령대별 차이에 관한 탐색 연구)

  • This paper aims to empirically analyze the factors of difference in trust level for Internet-only bank by age groups. The purpose of this study is to provide clues and a research data in developing differentiation strategies that can increase consumer's trust by age group. In this study, above all, the financial consumer group was divided into the 2030s and 4050s based on age. The study explored the trust determinants of the two groups and compared the differences between the two groups using a multi-group analysis method. As results, in all age groups, financial consumer's trust was seen as a determinant of convenience, security and benefit of financial transaction costs. The difference of the determinant of trust between 2030s and 4050s occurred in the factor of benefit. Moreover, it reconfirmed that consumer's trust is later strongly extended to relationship commitment. These findings suggest that trust is of paramount importance to the Internet-only bank, whose market competition will intensify in the future.

  • The primary purpose of this study is to compare and analyze the performance of local spatial autocorrelation techniques in identifying spatial distribution patterns of green spaces. To achieve the objective, this researcher uses satellite image analysis and spatial autocorrelation techniques. The result of the study shows that the LISA cluster map with the spatial outlier cluster is superior to other analytical methods in identifying the spatial distribution pattern of urban green space. This study can contribute to the related fields in that it uses several different research methods than the existing ones. Despite this differentiation and usefulness, this study has limitations in using low-resolution satellite imagery and NDVI among vegetation indices in identifying spatial distribution patterns of green areas. These limitations may be overcome in future studies by using UAV images or by simultaneously using several vegetation indices.

Design of a RANK-Mimetic Peptide Inhibitor of Osteoclastogenesis with Enhanced RANKL-Binding Affinity

  • The receptor activator of nuclear factor ${\kappa}B$ (RANK) and its ligand RANKL are key regulators of osteoclastogenesis and well-recognized targets in developing treatments for bone disorders associated with excessive bone resorption, such as osteoporosis. Our previous work on the structure of the RANK-RANKL complex revealed that Loop3 of RANK, specifically the non-canonical disulfide bond at the tip, performs a crucial role in specific recognition of RANKL. It also demonstrated that peptide mimics of Loop3 were capable of interfering with the function of RANKL in osteoclastogenesis. Here, we reported the structure-based design of a smaller peptide with enhanced inhibitory efficiency. The kinetic analysis and osteoclast differentiation assay showed that in addition to the sharp turn induced by the disulfide bond, two consecutive arginine residues were also important for binding to RANKL and inhibiting osteoclastogenesis. Docking and molecular dynamics simulations proposed the binding mode of the peptide to the RANKL trimer, showing that the arginine residues provide electrostatic interactions with RANKL and contribute to stabilizing the complex. These findings provided useful information for the rational design of therapeutics for bone diseases associated with RANK/RANKL function.

Genetic and Expression Analysis of the SIRT1 Gene in Gastric Cancers

  • Purpose: Silent mating-type information regulation 2 homologue 1 (SIRT1) is a nicotinamide adenine dinucleotide-dependent deacetylase. SIRT1 plays an important role in the regulation of cell death/survival and stress response in mammals. The aim of this study was to investigate whether the SIRT1 gene is involved in the development or progression of gastric cancers. Materials and Methods: SIRT1 and p53 genes in 86 gastric cancers were examined for genetic alterations by PCR-single strand conformation polymorphism sequencing, as well as SIRT1 protein expression in 170 gastric cancers by immunohistochemistry. Results: In the genetic analysis, we found SIRT1 and p53 mutations in two and 12 cases, respectively. Two missense mutations, c.599 C>T (T200I) and c.1258 G>A (E420K), were detected in the SIRT1 gene coding region. The SIRT1 and p53 mutation were found in mutually exclusive gastric cancers. The immunohistochemistry revealed that SIRT1 overexpression was found in 95 (55.9%) of 170 gastric cancers. Altered SIRT1 expression was not statistically associated with clinicopathological parameters, including tumor differentiation, location, lymph node metastasis, or p53 expression. Two cases with an SIRT1 mutation showed increased SIRT1 expression. Conclusions: These results suggest that genetic alterations and overexpression of the SIRT1 gene may contribute to gastric cancer development.

Using DGPS as An Acceleration Sensor for Airborne Gravimetry

  • In airborne gravimetry, there are two data streams. One is the specific force measured by an air/sea gravimeter or accelerometers, the other is kinematic acceleration measured by DGPS. And the difference of them provides the gravity disturbance information. To satisfy the requirement of most applications, an accuracy of 1mGal $(1mCal=10^{-5}m/s^{2})$ with a spatial resolution of 1km is the aim of current airborne gravimetry. There are two different methods to derive the kinematic acceleration. The generally used method is to differentiate the position twice, and the position can be calculated by commercial DGPS software. The main defect of this method is that integer ambiguities need to be fixed to get the precise position solution, but it's not a trivial thing for long base line. And to fix integer ambiguities, the noisier iono-free measurement is used. When differentiation is applied, noise is amplified and will influence the accuracy of acceleration. The other method is to get carrier phase acceleration by differentiate the carrier phase first, and then using the acceleration of GPS satellite to derive the vehicle acceleration. The main advantages include that fixing integer ambiguities is not needed anymore, position can be relaxed to about 10 meters, and smoother acceleration can be got since iono-free measurement is not needed. In some literatures, it's considered that the dynamic performance of the second method is inferior to that of the first. Through analysis, it is found that the performance degradation in dynamic environment results from the simplification of the GPS carrier phase observable model. And an iterative algorithm is presented to compensate the model error. Using a dynamic GPS data from an aeromagnetic survey, the importance of this compensation is showed at last.

  • Amphibians play a pivotal role in the ecosystem as a mediator between aquatic and terrestrial environment. Currently they are directly exposed to a variety of chemicals in the aquatic environment throughout their life cycle. Azole fungicides have been widely used in medical applications and agricultural activities. The direct exposure of azole fungicides causes an alarming situation for various ecosystem. Recently, teratogenesis and endocrine disruption by azole fungicides have been reported in amphibians. In an effort to provide the current information for amphibian toxicity of azole fungicides and to make the guidelines for safe usage of azole-based materials, the effects of azole fungicides including imidazole, triazole, thiazole, oxazole, and pyrazoleon on early development, differentiation and reproduction of amphibians were reviewed.

Effects of Added Chongmyung-tang on Behavior and Molecular Factors in the Alzheimer's Disease Model (ACM의 알츠하이머 생쥐 모델의 행동과 생체인자에 미치는 영향)

  • This experiment was designed to investigate the effect of Added Chongmyung-tang (ACM) on Alzheimer's disease mouse model. Effects of ACM on learning behavior were investigated using the Morris water maze method. Expression levels of molecular factors related to Alzheimer's disease such as glial fibrillary acidic protein (GFAP), cluster of differentiation antigen 68 (CD68), and tau protein in the hippocampus of APP-SWE Tg2576 mice were analyzed by immunofluorescence staining method. ACM reduced escape latency in the Morris water maze test. ACM decreased the expression level of GFAP and tau protein in the hippocampus. These results suggest that ACM may be involved in regulating molecules that are known to play an important role in the pathogenesis of Alzheimer's disease.

Micropropagation through Callus Culture in Chinese Foxglove (Rehmannia glutinosa) (지황의 캘러스 배양에 의한 기내 대량증식)

  • Chinese foxglove (Rehmannia glutinosa) is receiving much attention as one of the principal medicinal crops and the crude drug damand expands rapidly.This study was conducted to obtain the basic breeding information of Chinese foxglove. Effects of supplemental plant growth regulators were investigated on leaf tissue for proliferation. 100% callus formation, 31% plantlet regeneration and 6% root differentiation were obtained by adding 0.5 mg/L NAA and 2.0 mg/L BA. 2,4-D and Zeatin treatment also resulted in 95% increase in callus formation, but shoot was not formed. During the subculture, callus propagation rate recorded 15.4% with 0.2 mg/L NAA and 1.0 mg/L BA and plant regeneration improved on MS medium supplemented with 0.2 mg/L NAA and 0.5 mg/L kinetin. The number of shoot formed ranged from 1.7 on WPM medium to 3.4 on MS medium with 0.1 mg/L NAA and 0.5 mg/L BA. Supplementation of 1.0 g/L activated charcoal improved the In vitro plant growth.
