해표이진탕가녹용(解表二陳湯加鹿茸)이 Xylene으로 유발된 마우스의 급성 염증에 미치는 영향 (Anti-Inflammatory Effect of Haepyoejin-tang plus Antler water extract in Xylene-Application Mouse Ear Acute Inflammation Model)

  • In the present study, the anti-inflammatory effect of Haepyoeiin-tang plus Antler water extract water extract was tested in Xylene-Application mouse ear acute inflammation model. The test articles were once dosed before Xylene-Application, and the changes on Dody weight and weights and histopathological observation of induced ear were conducted with ear histomorphometry, The increases of absolute and relative ear weight detected in vehicle control compared to that of sham, were significantly and dose-dependently inhibited by Haepyoejin-tang plus Antler in the present study, A classic acute inflammatory histological changes such as subcutaneous edema, hypertrophy and infiltration of inflammatory cells, was detected in vehicle control. However, these histological changes were significantly and dose-dependently inhibited by Haepyoeiin-tang plus Antler. In addition, the increases of ear thickness half and thickness full detected in the vehicle control, were also dose-dependently decreased in the all Haepyoeiin-tang plus Antler-dosing groups. Base on these results, it is concluded that Haepyoeiin-tang plus Antler water extracts have a clear anti-inflammatory effect on the acute inflammation. However, somewhat lower anti-inflammatory effects were detected in Haepyoeiin-tang plus Antler water extracts compare to that of Dethamethason and Dicrofenac. About 500 mg/kg of Haepyoeiin-tang plus Antler water extracts have similar effect compared to that of Dicrofenac 15 mg/kg.

만분방(慢盆方)이 염증 관련 cytokines의 유전자 발현과 생성량에 미치는 영향 (The Experimental Study on Anti-inflammatory Effect of Manbunbang)

  • Purpose: This study was performed to evaluate the anti-inflammatory effect of Manbunbang extract (MBB). Methods: In order to understand the mechanism of anti-inflammatory effect of MBB, expression of cytokines and its levels in RAW 264.7 cell lines, as well as changes of cytokine gene expressions in serum, spleen, and liver tissues in acute inflammation induced mouse model were investigated. Results: 1. MBB significantly suppressed the expression levels of IL-1${\beta}$, TNF-${\alpha}$ and COX-2 mRNAs at 100 and 50 ${\mu}$g/m${\ell}$ concentrations, and IL-6 and NOS-II genes at 100, 50 and 10 ${\mu}$g/m${\ell}$ concentrations in RAW 264.7 cell lines, compared to those of the control. 2. MBB significantly reduced the production level of IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ at 100 and 50 ${\mu}$g/m${\ell}$ concentrations in RAW 264.7 cell lines compared the those of the control. 3. MBB significantly reduced the production of IL-1${\beta}$, IL-6 and TNF-${\alpha}$ levels in sera of acute inflammation induced mice. 4. MBB significanlty suppressed the expression level of IL-1${\beta}$, TNF-${\alpha}$ mRNA in spleen tissues as well as IL-6 mRNA in liver tissues in acute inflammation induced mice. Conclusion: From the results above, anti-inflammatory effect of MBB through its immune regulation could be experimentally explained. Wide treatment of inflammatory diseases such as pelvic inflammation using MBB are recommended.

RAW 264.7 Cell에서 리포폴리사카라이드로 유도된 염증성 매개인자들의 생산에 있어서 Corticosterone 전처리 효과 (Effect of Corticosterone Pretreatment on the Production of LPS-Induced Inflammatory Mediators in RAW 264.7 Cells)

  • Glucocorticoids are known to have anti-inflammatory effect. To investigate whether corticosterone pretreatment enhances or not lipopolysaccharide (LPS)-induced production of inflammatory mediators, RAW 264.7 cells were pretreated with various concentrations of corticosterone for 24 h and then cultured without corticosterone in the presence or absence of LPS. Our results demonstrated that LPS remarkably increased production of TNF-${\alpha}$, IL-6, IL-$1{\beta}$, vascular endothelial growth factor (VEGF), and NO (nitric oxide). Corticosterone pretreatment significantly attenuated LPS-induced production of TNF-${\alpha}$, IL-$1{\beta}$, and VEGF, while significantly enhanced IL-6 and NO. These findings suggest that corticosterone pretreatment may contribute to LPS-induced inflammatory responses in macrophages via pro- and anti-inflammatory imbalance of inflammatory mediators.

The inhibition of inflammatory molecule expression on 3T3-L1 adipocytes by berberine is not mediated by leptin signaling

  • In our previous study, we have shown that berberine has both anti-adipogenic and anti-inflammatory effects on 3T3-L1 adipocytes, and the anti-adipogenic effect is due to the down-regulation of adipogenic enzymes and transcription factors. Here we focused more on anti-inflammatory effect of berberine using real time RT-PCR and found it changes expressions of adipokines. We hypothesized that anti-adipogenicity of berberine mediates anti-inflammtory effect and explored leptin as a candidate mediator of this signaling. We studied this hypothesis by western blot analysis, but our results showed that berberine has no effect on the phosphorylations of STAT-3 and ERK which have important roles on leptin signaling. These results led us to conclude that the anti-inflammatory effect of berberine is not mediated by the inhibition of leptin signal transduction. Moreover, we have found that berberine down-regulates NF-${\kappa}B$ signaling, one of the inflammation-related signaling pathway, through western blot analysis. Taken together, the anti-inflammatory effect of berberine is not mediated by leptin, and berberine induces anti-inflammatory effect independent of leptin signaling.

連翹敗毒散加味方이 炎症狀態의 面胞에 미치는 影響 (Effect of the addition temperament drugs of Yeongyopaedock-san(連翹敗靑散加味方)on acne in the state of inflammation)

  • In the age of puberty or 20-30th young people who are sensitive to outward appearance, acne is serious problem at beauty and has social and psychological influence on that people. So this experiment is carried out for test whether the addition temperament drugs of Yungyopaedock-san(YP) have an anti-inflammatory effect and have suppression effect on immunocyte in the state of inflammation which induced by acne. The results was as follows. 1. YP has suppress inflammatory reaction induced by carageenan. 2. YP has suppress increasing activation of abdominal cavity macrophage in the carageen and zymosan induced inflammation. 3. YP has suppress increasing activation of spleen cell in the carageenan and zymosan induced inflammation. Based on the above result, YP was improved its suppression effect to the inflammatory reaction through the suppression of spleen cell and macrophage activation. So we concluded that YP is prospected as a anti-inflammatory agent to cure inflammation induced by ance.

Anti-inflammatory Activity of Propolis

  • Propolid (bee-glue), known as a folk medicine, is a lipo;hilic material found in honeybee hives. In the present study on the anti-inflammatory effect of Korean propolis, it was extracted with ethanol, and used as a test material. The $LD_{50}$ value with the oral administration of ethanolic extract of Korean propolis (EEKP) was higher than 2g/kg in mice. The oral administration of the propolis extract (100mg/kg) significantly inhibited the development of hind paw edema induced by carrageenin in rats. the oral pretreatment of the propolis extract markedly inhibited the increase in vascular permeability and the number of writhing induced by acetic acetic acid in mice. Propolis extract, 50 and 100 mg/kg p.o. per day for 7 days, produced a significant inhibitory effect on granuloma and exudate formation in rats. This inhibitory effect was enhanced with the concomitant use of prednisolone (2.5 mg/kg). These results suggest that Korean propolis apparently has a strong anti-inflammatory activity.

  • Experimental studies were done to research the clinical effects of Sopoongyangjetang on the Anti-allergic effect, Analgesic action and Anti-inflammatory action. The results obtained as follows ; 1. In effect of Sopoongyangjetang on vascular permeability responses to intradermal serotonin, it had tendency to decrease, but was none significant effect. 2. In effect of Sopoongyangjetang on vascular permeability responses to intradermal histamine, it revealed significant effect. 3. In the homologous PCA provoked by the IgE-like antibody against white egg albumin, Sopoonyangjetang showed significant effect. 4. In the delayed type hypersensitivity responses to picryl chloride, Sopoongyangjetang was proved significant effect. 5. In the delayed type hypersensitivity responses to SRBC, Sopoongyangjetang revealed significant effect. 6. In analgesic action by acetic acid method, Sopoongyangjetang was recognized significantly. 7. In Anti-inflammatory action by carrageenine method, Sopoongyangjetang showed significant effect.

PLGA/헤스페리딘 함량별 필름에서 염증 완화 효과: In vitro, In vivo 결과 (Effect of Inflammatory Responses to PLGA Films Incorporated Hesperidin: In vitro and In vivo Results)

  • 헤스페리딘은 혈관 순환계 보호 효과, 항암 효과, 항산화 효과, 항염증 효과 등의 작용이 있다고 알려져 있다. 본 연구에서는 헤스페리딘을 함유한 poly(lactic-co-glycolic acid)(PLGA) 필름을 조직공학적 세포담체로 이용하고 생체 내/외의 실험을 통하여 헤스페리딘과 염증 반응의 상관관계를 알아보고자 하였다. 세포담체에서의 세포의 증식과 부착을 확인하고자 PLGA/헤스페리딘 필름에 세포(RAW 264.7, NIH/3T3)를 파종하여 1, 3일 후에 MTT와 SEM을 통하여 분석하였다. 또한 항산화 효과와 염증 완화 효과를 알아보고자 ELISA, RT-PCR, FACS 등을 평가한 결과 PLGA 필름보다는 헤스페리딘이 첨가된 PLGA 필름에서 염증 반응이 감소하는 것을 확인하였다. In vivo 실험에서도 헤스페리딘이 PLGA에 의해 유도된 염증세포와 섬유피막 축적을 감소시켜 헤스페리딘의 항염증 효과를 볼 수 있었다. 이 실험 결과 PLGA 필름보다는 PLGA/헤스페리딘 필름에서 염증 반응이 감소함을 확인하였다.

Anti-inflammatory Effect of Branches Extracts from Quercus mongolica in LPS-induced RAW264.7 Cells

  • Quercus mongolica (QM), which belongs to fagaceae, is one of the oak native to Korea. We evaluated the anti-inflammatory effect of branches extracted with 70% ethanol of QM (QM-B) and elucidated the potential signaling pathway in LPS-induced RAW264.7 cells. The QM-B showed anti-inflammatory activity through inhibition of NO production. The QM-B dose-dependently suppressed NO production by inhibiting iNOS, COX-2 and IL-6 expression in LPS-induced RAW264.7 cells. The QM-B inhibited the degradation and phosphorylation of IκB-α and NF-κB activation. The QM-B suppressed the phosphorylation of p38 and ERK1/2. Also, the QM-B increased HO-1 expression. These results suggested that QM-B may utilize anti-inflammatory activity by suppressing NF-κB and MAPK signaling pathway and inducing HO-1 expression indicated that the QM-B can be used as a natural anti-inflammatory drugs.

Anti-inflammatory Effect of Gyulpidaehwangbakcho-tang (Jupidahuangpoxiao-tang) in the Collagen-induced Arthritis Mouse Model

  • Objectives: To investigate anti-inflammatory and anti-arthritic effects of Gyulpidaehwangbakcho-tang (GDBT) extract in a murine model of rheumatoid arthritis. Methods: The mice received $100{\mu}g$ of bovine type II collagen in Freund's complete adjuvant by intradermal injection at the base of the tail on day 0 and a booster injection on day 21. The mice were orally administered with GDBT (200 or 50mg/kg dissolved in distilled water) daily from day 1 to day 21 after arthritis incidence, and monitored for disease incidence and the severity of arthritis up to day 21. In order to evaluate the effect of GDBT on disease progression, we examined pro-inflammatory cytokines including IL-$1{\beta}$, IL-6, TNF-${\alpha}$, COX-2 and NOS-II. Results: GDBT produced a significant and dose dependent inhibition of arthritis and inflammation during the entire duration of the study. This action was characterized by the decreased production of IL-$1{\beta}$, IL-6, TNF-${\alpha}$, COX-2, and NOS-II in vivo. Conclusion: We believe that the anti-arthritic activity of GDBT is due to its modulatory effect on the expression of pro-inflammatory cytokine in the synovium. Our results contribute towards validation of the traditional use of GDBT in the treatment of RA and other inflammatory joint disorders.
