Samsoeum inhibits systemic anaphylaxis and release of histamine, cytokine in vivo and in vitro

  • Samsoeum (SSE) is used in traditional oriental medicine for various medicinal purposes. However, the exact mechanism that accounts for the anti-allergy and anti-inflammatory effects of the SSE is still not fully understood. The aim of the present study is to elucidate whether and how SSE modulates the allergic reactions in vivo, and inflammatory reaction in vitro. In this study, we showed that SSE significantly decreased compound 48/80-induced systemic anaphylaxis, ear-swelling response, histamine release from preparation of rat peritoneal mast cells and anti-dinitropheny IgE-induced passive cutaneous reaction. Also, SSE inhibited the expression of inflammatory cytokine and cyclooxygenase-2 in PMA plus A23187-stimulated human mast cells (HMC-1). In addition, we showed that anti-inflammatory mechanism of SSE is through suppression of nuclear factor-${\kappa}B$ activation and $I{\kappa}B-{\alpha}$ phosphorylation/degradation in HMC-1. These results provided new insight into the pharmacological actions of SSE as a potential molecule for therapy of inflammatory allergic diseases.

The Effects of Hangryunhaedocktang on Allergic Contact Dermatitis Based on the Morphological Changes in Epidemal Damage in Mice (황연해독탕(黃連解毒湯)이 알러지성 접촉피부염에 미치는 영향 -알러지성 접촉피부염 유발로 손상된 생쥐 상피세포의 완화를 중심으로)

  • This study was performed to investigate the effectiveness of Hangryunhaedocktang (HHT) on epidermal damages induced by allergic contact dermatitis (ACD). The sensitization were caused by one application of $25{\mu}l$ of 5% 2,4-dinitroch1orobenzene (DNCB) onto a back-lumbar skin of BALB/c mice. 2 weeks later, ACD was elicitated with $4{\mu}l$ of 2.5% DNCB and then mice were given HHT extract in doses of 3.3ml/kg/day, for 72 hours. The ACD induced epidermal damages in HHT treated ACD mice was more mitigated than non-treated ACD elicited mice. The features related with epidermal damage such as epidermal hyperplasia, infiltration of inflammatory cells, increase of nuclear shrinkages and vacuolation, and enlargement of intercellular space softened. And the distribution of soybean agglutinin (SBA) positive reaction in stratum spinosum (SS) and stratum basale (SB) were similarly maintained in a normal configuration. The numerical decrease of BrdU, TUNEL, and Fas positive cells observed were prominent in SB. Results suggest a benefit role for HHT in mitigating epidermal damages in mice with allergic contact dermatitis.

Inhibitory Effect of Galgeunhaegi-Tang on Compound48/80 Stimulated Allergic Reaction (갈근해기탕(葛根解肌湯)의 항 Allergy 및 항염증 효과)

  • The present study was conducted to investigate the anti-allergic activity of Galgeunhaegi-Tang(GHT). We investigated the anti-allergic effects of GHT in RBL-2H3 basophilic leukemia cells by compound48/80, a mast cell degranulator and compound 48/80 induced anaphylactic shock in mice. GHT significantly inhibited ${\beta}$-hexosaminidase and histamine release from compound 48/80 stimulated RBL-2H3 cells. In addition, GHT effectively inhibited anaphylactic shock in mice by 40% at a dose 100 mg/mouse versus PBS treated control after the l.p injection(8 mg/kg) of compound 48/80. The in vitro anti-inflammatory activities of GHT in LPS-stimulated RAW 264.7 cells were investigated. GHT inhibited NO production in LPS-stimulated RAW 264.7 cells and effectively dowregulated the expression of iNOS mRNA and iNOS protein expression in LPS-stimulated RAW 264.7 cells. These result provide evidences that GHT may be beneficial in the treatment of allergic inflammtory disease.

  • The present study was conducted to investigate the anti-allergic activity of Gunggwihyangso-San(GHS). Anti-inflammatory and Immunomodulatory Effects of the Botanical Product $AMP-365^{TM}$ (천연물제제 마루플랜트$-AMP-365^{TM}$의 항염증 및 면역활성에 미치는 영향)

  • The effect of aqueous extracts of medicinal plants $AMP-365^{TM}$ was tested for immune system regulating activity based on anti-inflammatory activity, anti-oxidant, macrophage proliferation and T-lymphocyte proliferation activity. $AMP-365^{TM}$ dose-dependently increased proliferation of RAW264.7 macrophage cells and its nitric oxide production as well as DPPH radical scavenging activity. On the other hand, T-lymphocyte proliferation activity was decreased on dose-dependent manner. Passive cutaneous anaphylaxis reaction was alleviated by 49% by administering 250 mg/kg of $AMP-365^{TM}$. The results suggest that $AMP-365^{TM}$ can be beneficial in the treatment of immediate allergic reactions as an adjuvant supplement material.

The Effect of Onpye-tang on the Rat Model with Allergic Rhinitis (온폐탕(溫肺湯)이 알레르기 비염 유발 흰쥐 모델에 미치는 영향)

  • Objectives : Allergic rhinitis is an inflammation or irritation of the mucous membranes that in the nose. Common symptoms include sneezing; a stuffy or runny nose; itch eyes, nose and throat; and watery eyes. We aimed to determine therapeutic effect of Onpye-tang by observing changes in blood cells and the nasal mucosal tissue. Methods : Twenty-four Sprague-Dawley rats were divided into three groups: normal, control, and sample group. Allergic rhinitis was induced in the control and sample group by intraperitoneal and intranasal sensitization with 0.l% Ovalbumin solution. Then Onpye-tang was orally administered only to the sample group for 28days, while the rats in the control group was given normal saline. We observed changes in the number of RBC and WBC; changes of neutrophils, lymphocyte, monocyte, and eosinophil proportions; total IgE level and AST & ALT change; also, changes in the nasal mucosal tissue. We used Student T-test statistically(p<0.05). Results : Onpye-tang showed significantly decreased IgE level on the serum of the rat model. Onpye-tang showed significantly decreased eosinophil level on the blood of the rat model. Onpye-tang inhibited the inflammatory reaction on the nasal mucosal tissue, according to nasal mucosal biopsy. Onpye-tang showed an inhibitory effect on the process of allergic rhinitis, according to IgE level, Eosinophil level, nasal mucosal biopsy. Onpye-tang had no hepatoxicity, according to AST and ALT on the serum. Conclusion : According to the above results, it is considered that Onpye-tang has an inhibitory effect on the process of allergic rhinitis and it can be used in relieving symptoms of allergic rhinitis.

We used ANOVA test statistically(p<0.05). Results: 1. Gudambope-tang showed an inhibitory effect on the process of allergic rhinitis. 2. Gudambope-tang inhibited the inflammatory reaction on the nasal mucosal tissue. 3. Gudambope-tang had no hepatoxicity. Conclusions: According to the above results. it is considered that Gudambope-tang has an inhibitory effect on the process of allergic rhinitis and it can be used in relieving symptoms of allergic rhinitis.

Anti-allergic Effects of Socheongyoug-tang on RBL-2H3 Mast Cell and Mice-mediated Allergy Model (RBL-2H3 비만세포와 동물병태모댈에서 소청룡탕(小靑龍湯)의 항알레르기 효과)

  • The discovery of drugs on the treatment of mast cell-mediated allergic disease is a very important subject in human heath. The Socheongyoug-tang(SCYT) has been used for centruries as a traditional medicine in Korea and is known to have an anti-inflammatory effect. However, its specific mechanism of action is still unknown. In this report, we investigated the effect of hot water extract from SCYT on RBL-2H3 mast cell-mediated allergic reaction and studied its possible mechanism of action. SCYT inhibited compound 48/80-induced systemic anaphylaxis and serum histamine release in mice. SCYT decreased the passive cutaneous anaphylaxis reaction activated by Anti-lgE antibody-HSA. SCYT dose-dependently reduced histamine release from mice peritoneal mast cells activated by Anti-lgE antibody-HSA. SCYT increased cAMP and decreased compound 48/80-induced intracellular $Ca^{2+}$ levels. Our findings provide evidence that SCYT inhibits mast-derived allergic reactions, and also demonstrate the involvement of cAMP and intracellular $Ca^{2+}$ in these effects.

Antiallergic Effects of Fermented Ixeris sonchifolia and Its Constituents in Mice

  • To evaluate the antiallergic effect of fermented Ixeris sonchifolia (IS, family Compositae), we prepared IS kimchi, isolated lactic acid bacteria (LAB) from it, fermented IS with these LAB, and investigated their antiallergic effects. IS kimchi inhibited the passive cutaneous anaphylaxis (PCA) reaction induced by an IgE-antigen complex as well as the scratching behavior induced by compound 48/80 or histamine more potently than IS. When IS was fermented with LAB isolated from IS kimchi, its antiallergic effects was also increased. Of LAB used for fermentation, Lactobacillus brevis more potently increased the antiallergic effects. Its main constituents, chlorogenic acid and luteolin, potently inhibited the PCA reaction induced by the IgE-antigen complex as well as the pruritis induced by compound 48/80 or histamine. These constituents inhibited the expression of pro inflammatory and allergic cytokines, TNF-$\alpha$. and IL-4, and transcription factor NF-${\kappa}B$ activation induced by the IgE-antigen complex in RBL-2H3 cells, as well as the degranulation of RBL-2H3 cells induced by the IgE-antigen complex. Luteolin more potently inhibited these allergic reactions than chlorogenic acid. These findings suggest that the antiallergic effect of IS can be increased by LAB fermentation, and the fermented IS might improve allergic reactions such as pruritus, anaphylaxis, and inflammation.
