Detection of Irradiated Fruits Using the DNA Comet Assay (DNA Comet Assay를 이용한 과일의 방사선 조사 확인)

  • The simple microgel electrophoresis of single cells, a 'comet assay', on fruit seeds enabled the rapid identification of irradiated fruits by comparing the intact non-irradiated cells and the damaged cells of irradiated fruits. Grapes and plums were irradiated with 0.1, 0.5, 0.7, 1.0 kGy and strawberries, peaches, apples, and nectarines were irradiated with only 1.0 kGy. Seeds were isolated, crushed, and the suspended cells were embedded in an agarose layer. After lysis of the cells, they were subjected to microgel electrophoresis for 2 minutes, and then stained. The DNA radiation-induced fragmentation of all the fruits stretched and migrated out of the cells forming a tail toward the anode giving the appearance of a comet, while the undamaged cells appeared as intact nuclei without tails. Grape and plum seeds irradiated at 0.5 kGy and higher showed significant increases in tail length. With increasing the irradiation doses, longer extention of the DNA from the nucleus toward the anode was observed. Strawberry, peach, apple, and nectarine seeds irradiated with 1.0 kGy also showed the longer tails than non-irradiated ones. DNA comet assay as a rapid and inexpensive screening technique could be an officially validated method for the detection of irradiated fruits.

  • The purpose of this research is to measure a gravitational attraction about urban pulbic service facilities and to evaluate the location of public service facilities. It is important in that these facilities should provide more inexpensive and convenience public service to users. To do this, the GIS's spatial analysis and gravity model were used to analyze the efficiency of the public facilities. The gravity model was conducted as the main analysis method, and another model for this analysis was the discrete model. The gravity model is originally to anticipate migration flows, traffic flows and other types of movements so that this model compares the gravitational attraction between places. The discrete model is to find the optimal location and to evaluate a location of facility regarding urban areas as the combination of node and link. In this research, these two models were adopted to compare and analyze location of Mapo-Gu Office. The results of this research indicated that the locational evaluation of urban public service facilities discovered the appropriateness of those facilities, and the public facilities was necessary to displace to other site.

Inorganic Nutrient Removal Efficiency of Aquatic Plants from Recirculating Aquaculture System (수생식물을 이용한 담수 순환여과식 양식용수내의 무기영양염 처리 효율)

  • Inorganic nutrients such as nitrogen and phosphate compounds accumulate in recirculating aquaculture systems. These nutrients must be removed from the system before they affect pH and fish health. For this purpose, aquatic plants are a simple and inexpensive method of removal. There are four commonly used aquatic plants: Eichhornia crassipes (water hyacinth), Pistia stratiotes (water lettuce), Hygrophila angustifolia, and Hydrocotyle leucocephala in freshwater recirculating aquaculture systems in Korea, but their efficiencies are not known. Therefore, removal efficiencies of inorganic nutrients from a freshwater recirculating aquaculture water with four commonly used aquatic plants were tested. Removing efficiencies of TAN, N $O_2$$^{[-10]}$ -N, and N $O_3$$^{[-10]}$ -N of the plants in 210 L aquaria for 48-hour period were tested. The removing efficiencies of TAN, N $O_3$$^{[-10]}$ -N, and P $O_4$$^{3-}$-P of the two most effective plants, water hyacinth and water lettuce, were also tested in 690 L (surface area of 1.55 $m^2$) tanks under 2 different initial stocking densities, 4 kg and 6 kg, for 22 days. Proximate analysis major nutrients and N and P contents of the all plants were analyzed for calculating net removal weight of N and P by the plants. Water lettuce was the most effective for removing TAN, N $O_2$$^{[-10]}$ -N, and N $O_3$$^{[-10]}$ -N from the water for 48-hour period tested followed by water hyacinth and Hygrophila angustifolia. Water lettuce reduced TAN, N $O_2$$^{[-10]}$ -N, and N $O_3$$^{[-10]}$ -N concentration from 2.3 mg/L, 0.197 mg/L, and 21.4 mg/L to 0.4 mg/L, 0.024 mg/L and 17.4 mg/L, respectively while water hyacinth reduced them down to 0.6 mg/L, 0.029 mg/L and 17.9 mg/L, respectively. The concentrations of TAN, N $O_2$$^{[-10]}$ -N, and N $O_3$$^{[-10]}$ -N in Hydrocotyle leucocephala group were rather increased up to 3.7 mg/L, 5.7 mg/L and 48.2 mg/L, respectively. This is because the creeping stem of Hydrocotyle leucocephala had to be cut to meet stocking weight resulting in decaying of the stem in the aquaria during experiment. The net growth in weight of water hycinth and water lettuce of 4 kg each in the 1.55 $m^2$ tanks for 22 days were 9.768 kg and 10.803 kg respectively, and those at initial weight of 6 kg each were 8.393 kg and 9.433 kg, respectively. The reason of lower net growth in the later group was restricted growth space. Nitrogen and phosphorus contents in water hyacinth were 2.89% and 0.27%, and those in water lettuce were 3.87% and 0.36%, respectively. Average quantities of removed N and P from 1.55 $m^2$ tanks by water hyacinth for 22 days were 18.9 g and 1.75 g, while those by water lettuce were 36.8 g and 3.5 g, respectively. Therefore water lettuce showed much higher efficiencies for removing both N and P from recirculating aquaculture water than water hyacinth.

Harmonic Reduction Scheme By the Advanced Auxiliary Voltage Supply (개선된 보조전원장치에 의한 고조파 저감대책)

  • Diode rectifiers are very popular in industry. However, they include large low-order harmonics in the input current and do not satisfy harmonic current content restrictions. To reduce the harmonics to the power system, several methods have been introduced. It is heavy and expensive solution to use passive filters as the solution for high power application. Another solution for the harmonic filter is utilization of active filter, but it is too expensive solution. Diode rectifiers with configurations using switching device have been introduced, but they are very complicated. The combined 12-pulse diode rectifier with the square auxiliary voltage supply has been introduced. It has the advantages that auxiliary circuit is simple and inexpensive compared to other strategies. The advanced auxiliary voltage supply in this thesis is presented as a new solution. When the square auxiliary voltage supply applied, the improvement of THD is 6~60[%] in whole load range. But when the advanced auxiliary voltage supply applied, it shows stable and excellent reduction effect of THD as 57~71[%]. Especially, for the case with 10[%] load factor, reduction effect of THD has little effect as 6[%] in the case of inserting a square auxiliary voltage supply. But when the proposed new solution applied, reduction effect has excellent effect as 71[%]. Theoretical analysis of the combined 12-pulse diode rectifier with the advanced auxiliary voltage supply is presented and control methods of the auxiliary supply is proposed. The reduction in the input current harmonics is verified by simulation using software PSIM.

Atmospheric Turbulence Simulator for Adaptive Optics Evaluation on an Optical Test Bench

  • An adaptive optics system can be simulated or analyzed to predict its closed-loop performance. However, this type of prediction based on various assumptions can occasionally produce outcomes which are far from actual experience. Thus, every adaptive optics system is desired to be tested in a closed loop on an optical test bench before its application to a telescope. In the close-loop test bench, we need an atmospheric simulator that simulates atmospheric disturbances, mostly in phase, in terms of spatial and temporal behavior. We report the development of an atmospheric turbulence simulator consisting of two point sources, a commercially available deformable mirror with a $12{\times}12$ actuator array, and two random phase plates. The simulator generates an atmospherically distorted single or binary star with varying stellar magnitudes and angular separations. We conduct a simulation of a binary star by optically combining two point sources mounted on independent precision stages. The light intensity of each source (an LED with a pin hole) is adjustable to the corresponding stellar magnitude, while its angular separation is precisely adjusted by moving the corresponding stage. First, the atmospheric phase disturbance at a single instance, i.e., a phase screen, is generated via a computer simulation based on the thin-layer Kolmogorov atmospheric model and its temporal evolution is predicted based on the frozen flow hypothesis. The deformable mirror is then continuously best-fitted to the time-sequenced phase screens based on the least square method. Similarly, we also implement another simulation by rotating two random phase plates which were manufactured to have atmospheric-disturbance-like residual aberrations. This later method is limited in its ability to simulate atmospheric disturbances, but it is easy and inexpensive to implement. With these two methods, individually or in unison, we can simulate typical atmospheric disturbances observed at the Bohyun Observatory in South Korea, which corresponds to an area from 7 to 15 cm with regard to the Fried parameter at a telescope pupil plane of 500 nm.

Radiographic Analysis of Instrumented Posterolateral Fusion Mass Using Mixture of Local Autologous Bone and b-TCP (PolyBone$^{(R)}$) in a Lumbar Spinal Fusion Surgery

  • Objective : Although iliac crest autograft is the gold standard for lumbar fusion, the morbidity of donor site leads us to find an alternatives to replace autologous bone graft. Ceramic-based synthetic bone grafts such as hydroxyapatite (HA) and b-tricalcium phosphate (b-TCP) provide scaffolds similar to those of autologous bone, are plentiful and inexpensive, and are not associated with donor morbidity. The present report describes the use of Polybone$^{(R)}$ (Kyungwon Medical, Korea), a beta-tricalcium phosphate, for lumbar posterolateral fusion and assesses clinical and radiological efficacy as a graft material. Methods : This study retrospectively analyzed data from 32 patients (11 men, 21 women) who underwent posterolateral fusion (PLF) using PolyBone$^{(R)}$ from January to August, 2008. Back and leg pain were assessed using a Numeric Rating Scale (NRS), and clinical outcome was assessed using the Oswestry Disability Index (ODI). Serial radiological X-ray follow up were done at 1, 3, 6 12 month. A computed tomography (CT) scan was done in 12 month. Radiological fusion was assessed using simple anterior-posterior (AP) X-rays and computed tomography (CT). The changes of radiodensity of fusion mass showed on the X-ray image were analyzed into 4 stages to assess PLF status. Results : The mean NRS scores for leg pain and back pain decreased over 12 months postoperatively, from 8.0 to 1.0 and from 6.7 to 1.7, respectively. The mean ODI score also decreased from 60.5 to 17.7. X-rays and CT showed that 25 cases had stage IV fusion bridges at 12 months postoperatively (83.3% success). The radiodensity of fusion mass on X-ray AP image significantly changed at 1 and 6 months. Conclusion: The present results indicate that the use of a mixture of local autologous bone and PolyBone$^{(R)}$ results in fusion rates comparable to those using autologous bone and has the advantage of reduced morbidity. In addition, the graft radiodensity ratio significantly changed at postoperative 1 and 6 months, possibly reflecting the inflammatory response and stabilization.

Characterization of Perchlorate-Removal Using Elemental Sulfur Granules and Activated Sludge (원소 황 입자와 활성슬러지를 이용한 퍼클로레이트 제거특성)

  • Perchlorate (${ClO_4}^-$) is an emerging contaminant found in surface water and soil/groundwater. Microbial removal of perchlorate is the method of choice since perchlorate-reducing bacteria (PRB) can reduce perchlorate to harmless end-products. A previous study [3] showed experimental evidence of autotrophic perchlorate removal using elemental sulfur granules and activated sludge. The granular sulfur is a relatively inexpensive electron donor, and activated sludge is easily available from a wastewater treatment plant. A batch test was performed in this study to further investigate the effect of various environmental parameters on the perchlorate degradation by sludge microorganisms when elemental sulfur was used as electron donor. Results of the batch test suggest optimum conditions for autotrophic perchlorate degradation by sludge microorganisms. The results also show that sulfur-oxidizing PRB enriched from activated sludge removed perchlorate better than activated sludge. Taken together, this study suggests that autotrophic perchlorate removal using elemental sulfur and activated sludge can be improved by employing optimized environmental conditions and enrichment culture.

Development of Composite Sensing Technology Using Internet of Things (IoT) for LID Facility Management (LID 시설 관리를 위한 사물인터넷(IoT) 활용 복합 센싱 적용기술 개발)

  • Various LIDs with natural water circulation function are applied to reduce urban environmental problems and environmental impact of development projects. However, excessive Infiltration and evaporation of LID facilities dry the LID internal soil, thus reducing plant and microbial activity and reducing environmental re duction ability. The purpose of this study was to develop a real-time measurement system with complex sensors to derive the management plan of LID facilities. The test of measurable sensors and Internet of Things (IoT) application was conducted in artificial wetlands shaped in acrylic boxes. The applied sensors were intended to be built at a low cost considering the distributed LID and were based on Arduino and Raspberry Pi, which are relatively inexpensive and commercialized. In addition, the goal was to develop complex sensor measurements to analyze the current state o f LID facilities and the effects of maintenance and abnormal weather conditions. Sensors are required to measure wind direction, wind speed, rainfall, carbon dioxide, Micro-dust, temperature and humidity, acidity, and location information in real time. Data collection devices, storage server programs, and operation programs for PC and mobile devices were developed to collect, transmit and check the results of measured data from applied sensors. The measurements obtained through each sensor are passed through the Wifi module to the management server and stored on the database server in real time. Analysis of the four-month measurement result values conducted in this study confirmed the stability and applicability of ICT technology application to LID facilities. Real-time measured values are found to be able to utilize big data to evaluate the functions of LID facilities and derive maintenance measures.

Development and Applications of Pore-filled Ion-exchange Membranes (세공충진 이온교환막의 개발 및 응용)

  • Ion-exchange membrane (IEM) has fixed charge groups and is a separation membrane which is capable of selectively transporting ions of the opposite polarity. Recently, the interest in IEMs has been increasing as the importance of the desalination and energy conversion processes using them as the key components has increased. Since the IEMs determine the efficiency of the above process, it is necessary to improve the separation performance and durability of them and also to lower the expensive membrane price, which is a hindrance to the widening application of the IEM process. Therefore, it is urgent to develop high-performance and low-cost IEMs. Among various types of IEMs, pore-filled membranes prepared by filling ionomer into a porous polymer substrate are intermediate forms of homogeneous membranes and heterogeneous membranes. The production cost would be cheap like the case of heterogeneous membranes because of the use of inexpensive supports and the reduction of the amount used of raw materials, and at the same time, they exhibit excellent electrochemical characteristics close to homogeneous membranes. In this review, major research and development trends of pore-filled IEMs, which are attracting attention as high-performance and low-cost IEMs, have been summarized and reported according to the application fields.

Studies on Potential utilization of Earthworm Cast as Vegetable Plant Growth Media - Effects of mixing ratios with earthworm cast in nursery soil on pepper(Capsicum annuum L.) seedling growth - (원예용 육모 상토로서의 지렁이 분립의 이용에 관한 연구 - 지령이 분립의 혼합비율이 고추묘의 생육에 미치는 영향 -)

  • Earthworm cast was investigated in this study in order to domestically develop inexpensive and safe plug nursery soil. 20,30,40 and 50% of earthworm cast were added to the nursery soil as vegetable plant growth media, in which peatmoss and vermiculite constituted the rest of the soil. The effects of earthworm cast on the growth of pepper (Capsicum annuum L.) seedling were obtained as follows. 1. According to the growth stages, significant differences(p=0.001) were recorded in number of leaves, leaf area and biological dry matter yield per plant(shoor or root dry matter yield per plant). In view of mixing ratios in the nursery soil, number of leaves, leaf area and stem weight per plant showed differences significantly at 1% level and leaf weight, root weight and biological dry weight per plant at 5% level, respectively. 2. The nursery soil with earthworm cast showed increased number of leaves and leaf area per plant compared to the commercial nursery soil particularly in the later stage of this study than in the earlier stage 3. Through the total period of seedling growth, leaf weight, stem weight and root weight per plant in the nursery soil with earthworm cast were genrally higher than those in the control and this trend was apparant in the treatment of more than 40% of mixing ratio with earthworm cast. 4. Although leaf weight per plant was higher than stem weight per plant till the 3rd week, from the 4th week stem weight per plant was getting higher. In the later stage of seedling growth, the stem weight was higher in the earthworm cast mixed treatment than that in the control. 5. There was no significant difference on biological dry matter yield in the earlier stage of this study, however in the later stage, it was higher in the earthworm cast mixed treatment than that in the control.
