Molecular phylogeny of Indonesian Lymantria Tussock Moths (Lepidoptera: Erebidae) based on CO I gene sequences

  • Many species of Lymantria are important forestry pests, including L. dispar which is well known distributed from Asia to North America as an invasive species. Like of most other genera of moths, the systematic of this genus is still in dispute, especially on the monophyly and the relationship within this genus due to the fact that genus is very large and varied. This genus was morphologically defined only by a single aphomorphy. To clarify the monophyly of the genus Lymantria, to reveal the phylogenetic relationship among the Indonesian species, and to establish the genetic characters of Indonesian Lymantria, we analyzed 9 species of Indonesian Lymantria involving 33 other species distributed around the world based on nucleotide sequence variation across a 516-bp region in the CO I gene. The results showed that the base composition of this region was a high A+T biased (C: 0.3333). The results also showed that the monophyly of Lymantria was not supported by bootstrap tests at any tree building methods. Indonesian species was distributed into four different groups but the relationship among them was still in dispute. It indicates that relationships among the basal nodes (groups) proposed here were least valid due to the fact that the number of species may not be enough to represent the real number of species in the nature. Moreover CO I gene sequences alone were not able to resolve their relationships at the basal nodes. More investigations were needed by including more species and other genes that the more conserved.

Molecular phylogeny of Indonesian Zeuzera(Lepidoptera:Cossidae) wood borer moths based on CO I gene sequence

  • Zeuzera is one of the most important wood borer pests in South East Asia. Like of most other genera of moths, the systematic of this genus is still in dispute, especially on the monophyly and the relationship within this genus due to the fact that genus is very varied. This genus was defined based on external characters only such as cross vein Sc-Rs present, humeral plate approximately triangular in shape and anal plate moderately long to moderately short. Therefore, the monophyly of this genus need to be evaluated based on more comprehensive data. To clarify the monophyly of the genus Zeuzera, to reveal the phylogenetic relationships among the Indonesian species, and to establish the genetic characters of Indonesian Zeuzera, we analyzed seven species of Indonesian Zeuzera including three other species distributed around the world based on nucleotide sequence variation across a 580-bp region in the CO I gene. The results showed that the monophyly of Zeuzera was supported by bootstrap tests at the MP and ML tree building methods (>95%). Genus Zeuzera was divided into two groups (A and B) with Z. borneana was excluded from the two groups and occupied at the basal node. Indonesian species was distributed into two different clades. CO I gene alone was able to fully resolve the relationships among species within clade B. However, further investigations were needed by including more species and other genes that the more conserved to test the validity of the phylogenetic hypothesis proposed here.

Molecular detection of bat coronaviruses in three bat species in Indonesia

  • Bats are an important reservoir of several zoonotic diseases. However, the circulation of bat coronaviruses (BatCoV) in live animal markets in Indonesia has not been reported. Genetic characterization of BatCoV was performed by sequencing partial RdRp genes. Real-time polymerase chain reaction based on nucleocapsid protein (N) gene and Enzyme-linked immunosorbent assay against the N protein were conducted to detect the presence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) viral RNA and antibody, respectively. We identified the presence of BatCoV on Cynopterus brachyotis, Macroglossus minimus, and Rousettus amplexicaudatus. The results showed that the BatCoV included in this study are from an unclassified coronavirus group. Notably, SARS-CoV-2 viral RNA and antibodies were not detected in the sampled bats.

  • Sixteen species of macrochelid mites were collected on the surface of scarab dung beetles in Mount Gede-Pangrango National Park, West Java, Indonesia. Of these, one species, Macrocheles gedeensis sp. nov. is described here as new to science. The other fifteen species were Holostaspella katakurai Hartini and Takaku, 2003, H. oblonga Hartini and Takaku, 2010, H. villosa Hartini and Takaku, 2010, Neopodocinum halimunense Hartini and Takaku, 2003, N. subjaspersi Hartini and Takaku, 2003, Neopodocinum sp., Macrocheles dispar (Berlese, 1910), M. entetiensis Hartini and Takaku, 2005, M. hallidayi Walter and Krantz, 1986, M. jabarensis Hartini and Takaku, 2003, M. kraepelini (Berlese, 1905), M. pumilus Hartini, Dwibadra and Takaku, 2009, M. turgoensis Hartini, Dwibadra and Takaku, 2009, M. sukabumiensis Hartini and Takaku, 2003, and M. sukaramiensis Takaku, 2001.

Comparison of ecophysiological and leaf anatomical traits of native and invasive plant species

  • Background: To address the lack of evidence supporting invasion by three invasive plant species (Imperata cylindrica, Lantana camara, and Chromolaena odorata) in tropical ecosystems, we compared the ecophysiological and leaf anatomical traits of these three invasive alien species with those of species native to Sempu Island, Indonesia. Data on four plant traits were obtained from the TRY Plant Trait Database, and leaf anatomical traits were measured using transverse leaf sections. Results: Two ecophysiological traits including specific leaf area (SLA) and seed dry weight showed significant association with plant invasion in the Sempu Island Nature Reserve. Invasive species showed higher SLA and lower seed dry weight than non-invasive species. Moreover, invasive species showed superior leaf anatomical traits including sclerenchymatous tissue thickness, vascular bundle area, chlorophyll content, and bundle sheath area. Principal component analysis (PCA) showed that leaf anatomical traits strongly influenced with cumulative variances (100% in grass and 88.92% in shrubs), where I. cylindrica and C. odorata outperformed non-invasive species in these traits. Conclusions: These data suggest that the traits studied are important for plant invasiveness since ecophysiological traits influence of light capture, plant growth, and reproduction while leaf anatomical traits affect herbivory, photosynthetic assimilate transport, and photosynthetic activity.

인도네시아산 주요 조림 및 유망 수종의 해부학적 특성 (Anatomical Properties of Major Planted and Promising Species Growing in Indonesia)

  • 본 연구에서는 인도네시아의 목재자원 중 새로운 유망 목재자원을 개발하고 최근 주요 조림 수종들의 효율적 이용을 위한 기초자료를 수집하기 위해 인도네시아산 10수종의 해부학적 특성을 조사하였다. 모든 수종은 산공재로써 고립관공, 방사복합관공이 관찰되었는데, Mangium에서는 위의 특성과 더불어 접선복합관공이 관찰되었다. 접선방향의 관공 직경은 Jeunjing과 Nangka를 제외한 전 수종에서 방사방향보다 큰 값을 보였다. 특히 Nangka는 공시수종 중 가장 큰 접선방향의 관공 직경을 보였다. 단위면적($mm^2$)당 관공의 수는 Mangium, Gandaria와 Kupa가 다른 수종보다 많았으며, 특히 Kupa가 가장 많게 나타났다. 유세포의 접선방향 폭은 Gmelina, Mangium, Kupa와 Mangga가 목섬유의 접선방향 폭보다 크게 나타났으며, 그 중 Mangium이 가장 큰 폭을 보였다. 이 밖의 6수종은 반대로 목섬유의 접선방향 폭이 유세포의 접선방향 폭보다 크게 나타났으나 Durian과 Nangka는 그 차이가 타 수종에 비해 미미했다. Gmelina, Jeunjing과 Rambutan은 평복세포로 이루어진 동성방사조직이, Gandaria는 평복세포와 단열의 직립세포로 구성된 이성방사조직이, Mangium, Durian, Jengkol, Kupa, Mangga와 Nangka는 평복세포와 약 1~2열의 직립세포로 구성된 형태의 이성방사조직이 관찰되었다. Durian, Gandaria, Jengkol, Jeunjing, Mangga, Rambutan은 결정을, Jeunjing과 Mangga는 실리카를 함유하는 특징을 나타냈다.

Molecular Analysis of Intraspecific Variations of the Indonesian Cochliobolus heterostrophus

  • The primary objective of the current research was to detect genetic variations within the Indonesian isolates of Cochliobolus heterostrophus collected from ecologically different places of the country at molecular level using PCR-RFLP analyses. The primer pair of NS3 and NS6 produced amplification fragment in all of the isolates tested. A single fragment of estimated 907 bp was observed in the PCR product pattern. RFLP analysis of the PCR product employing three restriction enzymes, HaeIII, HhaI, and RsaI, respectively, did not reveal intraspecific variations within the fungus. Similarly, nucleotide sequences of portion of small subunit of the ribosomal DNA gene of two of the isolates collected showed no appreciable differences, indicating the absence of genetic diversities among the isolates tested. A phylogenetic tree was constructed and the Indonesian C. heterostrophus, represented by SM-1 isolate, was found to be phylogenetically located near C. sativus, a closely related species.

Preliminary Study of the Ecological Impact of Forest Fires in G. Massigit, G. Gede-Pangrango National Park, West Java

  • Gunung Gede Pangrango National Park is one of the Long-term Ecological Research Site in Indonesia. In the late 1997, the fires have burnt and destroyed nearly 300 ha forest in this park. and G. Masigit was the largest burnt area (250 ha) of nine locations of hot spot recognized. Undergrowth vegetation got the most severe impacts. Almost undergrowth vegetation in various location were totally burnt. However, within three months following burning new seedlings such as Omalanthus populneus, Macaranga, Trema orientalis and Eupatorium appeared in the forest floor- The number of mycoflora recorded in burnt forest was interestingly increased in post forest fires site. Forest fires in G. Masigit had also affected the wild life population and diversity. For example, the number of bird species and the number of soil insects in burnt forest was significantly reduced. The forest fires had also great impact on soil. such as on soil organic contents, bulk density, colour, consistency, permeability and the activity of soil microorganisms.

The Expression of Codon Optimised Hepatitis B Core Antigen (HBcAg) of Subgenotype B3 Open Reading Frame in Lactococcus lactis

  • Hepatitis B treatments using immune therapy are gaining interest because of the improvements in dendritic cell performance for antigen presentation, which induces an appropriate immune response and raises patient survival rates. This research aims to produce a significant amount of the HBcAg antigen, which can induce an immune response and have a curative effect on HBV infection. In this study, the HBV subgenotype B3 of the HBcAg gene was used, which is dominant in Indonesia. Further, Lactococcus lactis bacteria was used as the host because of its safety and tightly regulated protein expression. The codon usage for the HBcAg gene was optimized to improve protein expression in L. lactis, which is important because a codon is not random between species. The HBcAg gene is attached to a pNZ8148 plasmid and transformed into the L. lactis NZ3900 expression host. The results confirm that a positive protein band (21 kDa) in two fractions of purified HBcAg was recognized by both western blotting and dot blot hybridization, even if the HBcAg optimized codon has higher GC contents than that suggested for L. lactis expression. Overall, this research strengthens the broad use of L. lactis bacteria for any protein expression, including higher protein expression of codon optimized HBcAg gene compared to non-optimized genes. Furthermore, the improvement in the codon optimization of the HBcAg gene significantly increases the total protein expression by 10-20%, and the expression level of the codon optimized HBcAg increases 1.5 to 3.2-times that of the native HBcAg.

인도네시아산 주요 조림 및 유망 수종의 물리적 및 역학적 특성(I) (Physical and Mechanical Properties of Major Plantation and Promising Tree Species Grown in Indonesia (I))

  • 인도네시아산 주요 조림수종(Albizia, Gmelina, Mangium)과 유망수종(Durian, Gandaria, Jengkol, Kupa, Mangga, Nangka, Rambutan)의 밀도, 수축률, 종압축강도와 경도 등 물리적 및 역학적 특성을 조사하였다. 그 결과, Mangium, Gandaria와 Rambutan은 높은 밀도를, Albizia, Durian과 Jengkol은 낮은 밀도를 보였다. 공시재료 중 Mangium, Gandaria와 Mangga의 수축률은 다른 수종에 비해 작았으며, Albizia, Jengkol과 Rambutan의 수축률은 다른 수종에 비해 높았다. 이방도(T/R)는 Albizia, Kupa와 Mangga가 낮았고, Gmelina, Durian과 Jengkol이 다른 수종에 비해 높았다. Mangium과 Nangka는 종압축강도와 경도가 높았으며, Durian, Jengkol과 Mangga는 낮은 종압축강도를, Albizia와 Gmelina는 낮은 경도를 보였다. 10수종 중 Albizia, Durian, Jenkol, Mangga의 4수종을 제외한 6수종의 물리적 및 역학적 성질은 비교적 우수한 것으로 판단되었다. 본 실험의 결과는 인도네시아산 조림목의 효율적인 이용 및 장래 조림을 위한 유용한 기초자료로 활용될 수 있을 것이라 생각된다.
