Seventeen Unrecorded Species from Gayasan National Park in Korea

  • Macrofungi play important roles in forest ecology as wood decayers, symbionts, and pathogens of living trees. For the effective forest management, it is imperative to have a comprehensive overview of macrofungi diversity in specific areas. As a part of the National Institute of Biological Resources projects for discovering indigenous fungi in Korea, we collected macrofungi in Gayasan National Park from 2017 to 2018. These specimens were identified based on morphological characteristics and sequence analysis of internal transcribed spacer (ITS) or the nuclear large subunit rRNA (LSU) region. We discovered 17 macrofungi new to Korea: Butyrea japonica, Ceriporia nanlingensis, Coltricia weii, Coltriciella subglobosa, Crepidotus crocophyllus, Cylindrobasidium laeve, Fulvoderma scaurum, Laetiporus cremeiporus, Lentinellus castoreus, Leucogyrophana mollusca, Marasmius insolitus, Nidularia deformis, Phaeophlebiopsis peniophoroides, Phanerochaete angustocystidiata, Phlebiopsis pilatii, Postia coeruleivirens, and Tengioboletus fujianensis. We described their detailed morphological characteristics.

Isolation of an Indigenous Imidacloprid-Degrading Bacterium and Imidacloprid Bioremediation Under Simulated In Situ and Ex Situ Conditions

  • The Bacterial community structure and its complexity of the enrichment culture during the isolation and screening of imidacloprid-degrading strain were studied using denaturating gradient gel electrophoresis analysis. The dominant bacteria in the original tea rhizosphere soil were uncultured bacteria, Rhizobium sp., Sinorhizobium, Ochrobactrum sp., Alcaligenes, Bacillus sp., Bacterium, Klebsiella sp., and Ensifer adhaerens. The bacterial community structure was altered extensively and its complexity reduced during the enrichment process, and four culturable bacteria, Ochrobactrum sp., Rhizobium sp., Geobacillus stearothermophilus, and Alcaligenes faecalis, remained in the final enrichment. Only one indigenous strain, BCL-1, with imidacloprid-degrading potential, was isolated from the sixth enrichment culture. This isolate was a gram-negative rod-shaped bacterium and identified as the genus Ochrobactrum based on its morphological, physiological, and biochemical properties and its 16S rRNA gene sequence. The degradation test showed that approximately 67.67% of the imidacloprid (50 mg/l) was degraded within 48 h by strain BCL-1. The optimum conditions for degradation were a pH of 8 and $30^{\circ}C$. The simulation of imidacloprid bioremediation by strain BCL-1 in soil demonstrated that the best performance in situ (tea soil) resulted in the degradation of 92.44% of the imidacloprid (100 mg/g) within 20 days, which was better than those observed in the ex situ simulations that were 64.66% (cabbage soil), 41.15% (potato soil), and 54.15% (tomato soil).

Bioconversion of Soybean Curd Residues into Functional Ingredients with Probiotics

  • Soybean curd residues (SCR) obtained from hot and cold manufacturing processes were fermented by indigenous microorganisms, Lactobacillus rhamnosus LS and Bacillus firmus NA-l for 15 h at 37$^{\circ}C$. The pH, acidity, viable cell counts, and tyrosine content were evaluated in samples with variations in sugar, starter and type of SCR. The raw Doowon SCR (D-SCR, cold-processed) fermented by indigenous microorganism had a 0.9% acidity and 6.7 ${\times}$ 10$^{7}$ CFU/g viable cell counts, compared with the 0.11 % acidity and 6.7 ${\times}$ 10$^{6}$ CFU/g viable cell counts of raw fermented Pulmuwon SCR (P-SCR, hot-processed). After fermentation of raw P-SCR with 1 % glucose and 1 % L. rhamnosus LS starter, the viable cell counts, tyrosine content and acidity were 4.7 ${\times}$ 10$^{8}$ CFU/g, 16.3 mg% and 0.9%, respectively. In addition, the raw P-SCR fermented with Bacillus firmus NA-l as co-starter had a 0.45% acidity, 2.4 ${\times}$ 10$^{8}$ CFU/g lactic acid bacteria, and 3.3 ${\times}$ 10$^{6}$ CFU/g Bacillus sp. In particular, the tyrosine content was increased 5 fold. The drying of fermented SCR was completed by hot-air drying (5$0^{\circ}C$) within 12 h; the dried P-SCR and D-SCR had 1.8 ${\times}$ 10$^{7}$ CFU/g and 5.3 ${\times}$ 10$^{6}$ CFU/g viable cell counts, respectively. The concentrate of methanol extract from fermented D-SCR inhibited the initial cell growth of E. coli, Staphylococcus aureus and Pseudomonas aeruginosa in liquid culture.

친환경 제제로부터 식물병원균에 대한 길항 미생물의 선발 (Selection of Antagonistic Microorganisms against Plant Pathogens from Eco-friendly Formulations)

  • BACKGROUND: Some microorganisms extant in nature have ability to suppress various plant pathogens, and also can promote plant growth. Thus microorganisms are such great source of antimicrobial agents to develop antagonistic microorganism production and eco-friendly crop management. We isolated the microorganisms in various eco-friendly formulations. The suppressive abilities against plant pathogens have been characterized in vitro level. METHODS AND RESULTS: The indigenous microorganisms have been isolated from Cooked rice, Black sugar, Rice Bran, and Red clay using dilution plating method. Population of bacteria and fungi were above 107 in the all formulations. We isolated and pure cultured the microorganisms based on morphological characteristics. Three major plant pathogens (Fusarium oxysporum, Rhizoctonia solani, Phytophthora capsici) have been used to select antagonistic microorganisms. Total 20 bacteria and 9 fungi showed the pathogen growth suppression ability in vitro condition. The selected microorganisms were identified by ITS sequence similarity. CONCLUSION: All tested eco-friendly formulations contained high-density of the microorganisms. Among the isolated microorganisms, Bacillus spp. and Streptomyces spp. showed the most effective antifungal activity against the plant pathogens such as F. oxysporum, R. solani, and P. capsici. Among the selected fungi Trichoderma sp. demonstrated antifungal activity. Our results suggest that the currently adapted eco-friendly formulations might useful for sustain agricultural system.

배양기법을 활용한 제주도내 내산 및 호염성 미생물의 분리 및 특성 분석 (Isolation and characterization of acid-resistanct and halophilic bacteria using cultivation technique in Jeju island)

  • 본 연구에서는 제주 지역의 토양 및 해양 환경으로부터 약 70주의 미생물들을 분리하였으며, 16S ribosomal RNA 유전자 분석을 통하여 최종 21종의 미생물을 발굴하였다. 이들 미생물들은 5 강(Class) 16 속(Genus)에 속하며, 모두 국내 미기록종으로 확인되었다. 분리된 미생물의 기질 특이성 및 고분자 물질 분해능을 바탕으로 내산성과 호염성 미생물들의 생리활성 표현형은 서로 구별되는 것으로 관찰되었다. 본 연구결과는, 국내 미생물 자원활용에 기초적 정보를 제공할 것으로 기대된다.

Note of Five Unrecorded Mushrooms Including Three Rare Species on Mount Juwang in Korea

  • The Mount Juwang was designated as a national park in 1976 because of its unique bedrock geology. Although Juwang National Park has maintained its natural ecosystem well, few macrofungal surveys have been carried out. As a part of a project supported by the National Institute of Biological Resources (NIBR) for the discovery of indigenous fungal species, we surveyed the mushrooms in Juwang National Park from 2018 to 2019. The macrofungi were identified based on morphological and molecular analyses. Among these fungi, five specimens were identified as species previously unrecorded in Korea: Calocybe decolorata, Crepidotus brunnescens, Mycena pearsoniana, Psathyrella phegophila, and P. sulcatotuberculosa. Three of these species are known to be rare in the world: Crepidotus brunnescens, P. phegophila, and P. sulcatotuberculosa. In this study, we provide detailed morphological descriptions of the five unrecorded species from Mt. Juwang in Korea.

Effects of radon on soil microbial community and their growth

  • The aim of this study was to estimate the microbial metabolic activity of indigenous soil microbes under the radon exposure with different intensity and times in the secured laboratory radon chamber. For this purpose, the soil microbes were collected from radon-contaminated site located in the G county, Korea. Thereafter, their metabolic activity was determined after the radon exposure of varying radon concentrations of 185, 1,400 and 14,000 Bq/㎥. The average depth variable concentrations of soil radon in the radon-contaminated site were 707, 860 and 1,185 Bq/㎥ from 0, 15, and 30 cm in deep, respectively. Simultaneously, the soil microbial culture was mainly composed of Bacillus sp., Brevibacillus sp., Lysinibacillus sp., and Paenibacillus sp. From the radon exposure test, higher or lower radiation intensities compared to the threshold level attributed the metabolic activity of mixed microbial consortium to be reduced, whereas the moderate radiation intensity (i.e. threshold level) induced it to the pinnacle point. It was decided that radon radiation could instigate the microbial metabolic activity depending on the radon levels while they were exposed, which could consequently address that the certain extent of threshold concentration present in the ecosystem relevant to microbial diversity and population density to be more proliferated.

Penicillium from Rhizosphere Soil in Terrestrial and Coastal Environments in South Korea

  • Penicillium, the most common genus plays an important ecological role in various terrestrial and marine environments. However, only a few species have been reported from rhizosphere soil. As part of a project to excavate Korean indigenous fungi, we investigated rhizosphere soil of six plants in the forest (terrestrial habitat) and sand dunes (coastal habitat) and focused on discovering Penicillium species. A total of 64 strains were isolated and identified as 26 Penicillium species in nine sections based on morphological characteristics and the sequence analysis of β-tubulin and calmodulin. Although this is a small-scale study in a limited rhizosphere soil, eight unrecorded species and four potential new species have been identified. In addition, most Penicillium species from rhizosphere soil were unique to each plant. Penicillium halotolerans, P. scabrosum, P. samsonianum, P. jejuense, and P. janczewskii were commonly isolated from rhizosphere soil. Eight Penicillium species, P. aurantioviolaceum, P. bissettii, P. cairnsense, P. halotolerans, P. kananaskense, P. ortum, P. radiatolobatum, and P. verhagenii were recorded for the first time in Korea. Here, we provide the detailed morphological description of these unrecorded species.

MMPH-0 (Enterobacter aerogenes)에 의한 6가 크롬 오염 지하수의 생지화학적 정화 (Biogeochemical Remediation of Cr(VI)-Contaminated Groundwater using MMPH-0 (Enterobacter aerogenes))

  • 오염환경에 서식하는 토착미생물은 환경정화에 중요한 역할을 담당하며 이 연구는 6가 크롬 오염 지하수에서 분리한 미생물을 이용해 반응성, 이동성, 발암성 높은 6가 크롬을 당대사 조효소인 3가 크롬으로 환원/침전시켜 경제적, 친환경적, 생지화학적 정화의 효율성을 알아보았다. 미생물 농화배양과 조성분석, 호기와 혐기환경의 6가 크롬 환원과 내성, 전자공여체별 6가 크롬 환원, 지화학적 변화, 미생물 외형과 Cr((III) 침전물의 광물특성을 연구한 결과, 분리한 MMPH-0(Enterobacter aerogenes)는 혐기/호기환경에서 6가 크롬 내성과 환원능(유기산 주입 1주 후 70%, 주입 안한 경우 4주 후 10 ~ 20%)이 있고, Eh는 미생물의 유기산 산화로 생성된 전자에 의해 산화에서 환원환경, pH는 중성에서 약산성으로 변화되어 $Cr(OH)_3$/Cr(III)침전물이 형성되었다. SEM/TEM-EDS 결과 $2{\sim}5{\mu}m$ 막대형 미생물과 세포 밖 Cr(III) 침전물은 지화학적 환경변화와 유기산 산화에 따른 전자공여에 의한 환원의 근거가 된다. 지화학적 촉매제 토착미생물의 활성화로 산화환원에 민감한 중금속 오염 지하수 정화에 효율적 기술 응용이 기대된다.

순수 분리 미생물을 이용한 오염 토양에서의 BTEX 생분해 특성과 미생물 군집 변화 (BTEX Biodegradation in Contaminated Soil Samples Using Pure Isolates and Changes in the Mixed Microbial Community Structure)

  • 본 연구에서는 BTEX로 오염된 저질로부터 순수 분리한 BTEX를 탄소원으로 $NO_3$-N를 전자수용체로 이용하는 미생물의 오염토양에 대한 현장 적용 가능성을 평가하였다. 이와 함께 주입한 순수 분리 미생물과 토착 미생물과의 상호 관계를 관찰하기 위하여 시간 경과에 따른 오염된 토양에서의 미생물 군집 변화도 관찰하였다. 이를 위해 BTEX로 오염 가능성이 적은 지역으로부터 채취한 토양 시료 100 g에 benzene, toluene, ethylbenzene, o-xylene을 각각 일정량 주입한 후, 동정 분리한 Pseudomonas stutzeri strain 15(DQ 202712):Klebsiella sp. strain 20(DQ 202715):Citrobacter sp. strain A(DQ 202713)를 2;1:1의 비율로 주입하여 BTEX 분해 효율과 미생물 군집 변화를 관찰하였다. 실험결과, $NO_3$-N와 BTEX가 모두 존재하는 조건에서 동정 분리한 미생물에 의한 분해 효율이 가장 높게 관찰되었다. 그리고 PCR-DGGE를 통한 미생물 군집 변화 관찰 결과, 토양 내 존재하는 다양한 미생물들의 peak는 대부분 감소된 반면, 주입한 동정분리 미생물 peak는 증가되는 것을 관찰할 수 있었다. 그러나 주입한 미생물 3종 가운데 Pseudomona stutzeri만이 우점화 된 결과가 관찰되었다.
