쇠고기 저장 중 이취발생에 대한 Indicator 탐색 (Investigation on Beef Quality Indicator of Off-Flavor Development during Storage)

  • 쇠고기의 저장 중 이취발생을 대변할 수 있는 품질인자를 탐색하고자 온도의존성의 관점에서 상호관계를 분석하였다. 특수관능기법인 R-index에 의한 이취검지시간을 탐지하였고, 이화학적, 생물학적 품질인자를 측정하였다. 먼저, 이취검지시간의 온도의존성의 관계식을 개발하여, 이에 따른 이취발생 indicator의 선발 요건을 유도하였다. 이화학적 분석은 VBN, pH, 육색, Warner-Bratzler shear force,그리고 생물학적 분석은 Pseudomonas spp., Lactic acid bacteria를 대상으로 하였다. 품질인자의 온도의존성은 Arrhenius 상수인 활성화에너지를 사용하여 분석하였다. 그 결과 VBN, pH, $a^*$값, LAB, Pseudomonas spp., WBSF, $L^*$값, $b^*$값의 순으로 이취발생과의 온도의존성이 유사하게 나타났으며, 상위권에 있는 pH와 $a^*$값에 비하여 환경적 요인에 안정한 VBN이 쇠고기 저장 중 이취발생의 최적 indicator인 것으로 탐색되었다.

Bacterial Community Variations in Hot Pepper-Sown Soil Using FAME Analysis as an Indicator of Soil Quality

  • The bacterial compositions of seven hot-pepper sown soil were compared in this study. From the 624 isolates, 95 species and 49 genera were identified by fatty acid methyl ester analysis (FAME). The FAME results of seven soil showed two distinct clusters for aerobic and Gram-negative bacteria in the high productivity and monoculture soil samples. While Arthrobacter ($17\%$), Kocuria ($11\%$), Pseudomonas ($8\%$), and Bacillus ($8\%$) were predominant among bacteria which were cultured on heterotrophic (YG) agar medium, Pseudomonas ($56\%$), Stenotrophomonas ($16\%$), and Burkholderia ($8\%$) were predominant on crystal violet agar medium. Shannon Weaver indices (H) indicated that colonies obtained from heterotrophic agar medium (3.1) were found to be more diverse than those obtained from the crystal violet media (1.9). The results suggest that FAME analysis may be a potential indicator for of soil quality.

  • The purpose of this study was to survey bacteria which was known as a fecal bacteria living in the human and animal gastro and intestinal tract. The colony density of the fecal coliform and fecal streptococci was 280 and 130 in 100ml sample amounts reapectively. Staphylococcus aureus and Pseudomonas aeruginosa which is usually exist in swimming pool and was known as indicator of the pool water contamination was detected.

Antifungal Activity of Lactic Acid Bacteria Isolated from Kimchi Against Aspergillus fumigatus

  • More than 120 isolates of lactic acid bacteria obtained from Kimchi was screened for antifungal activity against Aspergillus fumigatus. Approximately 10% of the isolates showed inhibitory activity and only 4.16% (five isolates) exhibited strong activity against the indicator fungus A. fumigatus. The five isolates showed a wide rang of antifungal activity against A. flavus, Fusarium moniliforme, Penicillium commune, and Rhizopus oryzae. They were identified by 16S rDNA sequencing as Lactobacillus cruvatus, L. lactis subsp. lactis, L. casei, L. pentosus, and L. sakei. The effect of Lactobacillus on mycelial growth and fungal biomass as well as its ability to produce toxic compounds were determined. The results indicate that the three species, Lactobacillus casei, L. lactis subsp. lactis, and L. pentosus, are active against A. fumigatus.

  • This study was performed to evaluate antibacterial performance antibiotics(RCF-95, Antibio-C) which could control biochemical corrosion of concrete used sewage facilities by sulfate oxidizing bacteria. As antibacterial methods, Broth MIC testing was used for investigating controlled growth effect of sulfate oxidizing bacteria. Also, color-changed testing by indicator was used for confirming between $H_{2}SO_{4}$ diffusion rate by bacteria and antibiotics. It confirmed that Antibio-C was superior to RCF-95 in the antibacterial performance and hence anticipated that this developed Antibio-C was enough to replace imported antibiotics from Japan.

서울시내 옹달샘물의 지표미생물 분포현황 (Distribution of Indicator bacteria in Spring Water in Seoul)

  • In order to investigate the microbiological contamination of spring water, we performed the standard plate count, coliform and psychrotrophilic bacteria in 109 spring waters in Seoul. Of 109 spring waters, geometirc mean standard plate count was 0.19 CFU/ml, and the highest in Mt. Boolam, 4..43 CFU/ml and Mt. Dobong, 3.86 CFU/ml, but not detected in Mt. Woomyun and Mt. Cheonggye. Four spring waters have shown over 100 CFU/ml in standard plate count. The geometric mean psychrotrophilic bacteria was 49.2 CFU/ml, the most prevalent spring water was Mt. Nam, 125 CFU/ml, the lowest Mt. Woomyeun. Among a total of 109, coliform was detected from 21 spring waters(19.3%) and the geometric mean of coliform was 0.005 MPN/100ml. The isolated genera of coliform were 7 isolates of E. coli(33%), 5 Klebsiella(24%), 4 Enterobacter(19%), 3 Citrobacter(14%) and 2 Serratia(10%), respectively. The 22 spring waters(20.2%) failed to meet the standard limits of drinking water based on regulation in Korea. The inappropriate rate of standard plate count in spring water was 4.5%, that of coliform was 81.1% and both of them was 13.6%. There was the significant correlation between standard plate count and psychrotrophilic bacteria in spring water(r=0.95, p<0.01).

Application of a Prototype of Microbial Time Temperature Indicator (TTI) to the Prediction of Ground Beef Qualities during Storage

  • The predictive ability for off-flavor development and quality change of ground beef was evaluated using a microbial time temperature indicator (TTI). Quality indices such as off-flavor detection (OFD) time, color, pH, volatile basic nitrogen (VBN), aerobic mesophilic bacteria (AMB) counts, and lactic acid bacteria (LAB) counts were measured during storage at 5, 10, 15, and $25^{\circ}C$, respectively. Arrhenius activation energies (Ea) were estimated for temperature dependence. The Ea values for TTI response (changes in titratable acidity (TA)), VBN, AMB counts, LAB counts, and freshness, which is defined based on OFD time for quality indices of ground beef, were 106.22 kJ/mol, 58.98 kJ/mol, 110.35 kJ/mol, 116.65 kJ/mol, and 92.73 kJ/mol, respectively. The Ea of microbial TTI was found to be closer to those of the AMB counts, LAB counts, and freshness. Therefore, AMB counts, LAB counts, and freshness could be predicted accurately by the microbial TTI response due to their Ea similarity. The microbial TTI exhibited consistent relationships between its TA change and corresponding quality indices, such as AMB counts, LAB counts, and freshness, regardless of storage temperature. Conclusively, the results established that the developed microbial TTI can be used in intelligent packaging technology for representing some selected quality indices of ground beef.

하수처리장 방류수의 계절별 미생물분포에 관한 연구 (Seasonal distribution of microorganisms in the effluent water in various sewage treatment plants)

  • 07년 1월부터 11월까지 하수처리장별로 각각의 세균에 대한 계절별 분포도를 조시하였다. 조사에 사용된 미생물은 호기성 및 통성혐기성 종속영양세균으로 대표되는 총 세균과 지표생물군인 대장균, 대장균군, 장내세균군 및 포도상규군과 효모/곰팡이로 구성하였다. 총 세균 및 대장균 대장균군, 장내세균군 등의 지표생물군의 경우엔 대체로 온도가 높은 하절기에 균체수가 증가하고 있는 것으로 나타난 반면, 식중독의 원인균인 황색포도상구균의 경우, 대부분의 하수처리장에서 오히려 11월과 1월에 균체수가 증가하는 등, 계절과 관계없이 출현하는 양상을 나타냈다. 한편, 방류수의 소독처리방법과 균체생성은 큰 상관관계가 없는 것으로 나타났으나, 작은규모의 마을하수의 경우 도시하수와 비교하여 균체수가 감소하고 있는 것으로 조사되었다.

Bacteriocin ("Vulnificin") Typing of Vibrio vulnificus

  • Vibrio vulnificus, a halophilic vibrio is an estuarine gram-negative bacteria that is associated with severe and frequently fatal wound infections and life-threatening septicemia. Bacteriocins are defined as antibacterial substance produced by various species of bacteria which are usually active against closely related organisms. Bacteriocins have found widespread application in epidemiological studies as specific markers of bacteria. It was proposed by Ha et al. (1990. J. Korean. Soc. Microbiol. 25: 586.) to give the bacteriocins produced by V. vulnificus the name "vulnificins". In the present study, a total of 72 strains of V. vulnificus isolated from patients and oysters were subjected to screen potential producers and indicators of vulnificin, applying ultraviolet induction method. Sensitivity of several strains of Serratia marcesans, Pseudomonas aeruginosa, Shigella flexneri, Salmonella typhi and Yersinia enterocolitica to vulnificins were also examined out. All the tested strains of V. vulnificus produced vulnificins active against indicator strains with various different inhibitory patterns. The spectrum of vulnificin activity and sensitive spectrum of indicator strains were considerably broad. Interestingly, almost all strains of S. marcescens, P. aeruginosa, Salmonella sp., Shigella sp. and Y. enterocolitica tested were sensitive to 1-7 vulnificin(s). Taken together, the present study demonstrated that all of the isolates of V. vulnificus produced vulnificins and that 8 good vulnificin producers and 10 good indicators were detected. These strains can be employed efficiently for establishing vulnificin typing scheme of V. vulnificus and for the detection of bacteriocinogeny and sensitivity in V. vulnificus. Biological role of vulnificin remains to be further elucidated.

심층피압지하수에서 세균군집의 분석에 의한 분변성 오염 평가 (Evaluation of Bacteriological Safety in Deep Confined Aquifer)

  • 세균군집의 분포에 따른 지하수의 세균학적인 안전성을 평가하기 위하여 음용을 목적으로 개발된 4곳의 관정에서 심층피압대수층의 지하수를 채수하여 물리화학적 환경요인, 세균의 분포와 활성, 지표세균의 분포 및 종속영양세균의 종조성을 조사하였다. 조사한 4개의 관정은 모두 빈영양생태계의 특징을 나타냈으나 관정 C-2는 무기이온의 농도, 유기탄소, 종속영양세균의 수 및 활성, 지표세균의 수가 다른 관정에 비해 매우 높았다. C-1을 제외한 모든 관정에서 분변성 오염이 존재하였으며 특히, 관정 C-2는 다른 관정들에 비해 그람양성 세균의 비율이 낮았으며, 총대장균군, 분변성대장균군, fecal streptococcus 가 각각 시료 250ml 에 최고 61, 33, 55 CFU로 존재하여 지표에서 분변성 오염물질이 지하수로 유입되고 있음이 확인되었다. 종속영양세균의 동정결과 55 CFU 로 존재하여 지표에서 분변성 오염물질이 지하수로 유입되고 있음이 확인되었다. 종속영양세균의 동정결과 지하수에 주로 존재하는 세균속은 Acinetobacter, Eikenella, Moraxella, Pseudomonas, Bacillus, Micrococcus 속이었다. 한편, 지하수에 일반적으로 존재하지 않는 Enterobacteriaceae 그룹과 Staphylococcus 속과 Enterococcus 속이 조사과정에서 검출됨에 따라 이들 세균이 지하수 분변성 오염 판정의 지표세균으로 사용할 수 있음이 확인되었다.
