Promoted Growth of Maize by the Phosphate Solubilizing Bacteria Isolated from North-east China

  • A strain of phosphate solubilizing bacterium was isolated from rhizosphere and identified as Burkholderia sp. by 16S-rRNA gene sequence analyses. The bacterium was found to release gluconic acid and the solubilization of hydroxyapatite in the liquid medium by a significant drop in pH to 3.7 from an initial pH 7.0. The soluble-P concentration continuously increased during the incubation periods and the total amount of soluble P released in culture filtrate was detected at 990 mg $L^{-1}$ after 10 days of inoculation. Most promoted maize growth was found in the standard NPK (240-120-120 kg $ha^{-1}$) soil inoculation with Burkholderia sp. (Twenty milliliters/plant, 106 CFU) and also in the absence of Burkholderia sp. inoculation, the soil amended with only 2/3 levels of P gave significant higher plant yield compared to 1/3 levels of P or without P supplementation.

Studies on Biodegradation of Synthetic Detergents (합성세제의 성분해에 관한 조사연구)

  • ABS, Alkyl benzene sulfonate, persists for long periods in stream because of its resistance to biologic degradation. Its bio-degradation is very varied in the environments. This investigation was therefore undertaken in order to know the biodegradation of synthetic detergents which are comprising soft and hard forms from February 1976 to September 1976. The biodegradations by spontaneuos settling and aeration were determined. The reduction rates of ABS, DBS, and LAS were 9.8%, 13.7% and 10.4% by the settlings for 5 days at $25^{\circ}$ and 63.3%, 27.2%, and 43.9% by aeration respectively. It was not contributed effectively to biodegradate the synthetic detergents by the simulaneous incubation with micro-organisms isolated from sewage such as Enterobacter sp., E. coli, Bacillus sp., Flavobacterium sp., Pseudomonas sp., Staphylococcus sp., and etc. Tricking filter could also eliminate some amount of detergent. By the application of this investigation, it could be designed that detergents would be regulated in abuse in context with water pollution and be treated at a proper process in the sewage treatment plant to be installed.

Mineralization-inducing potentials of calcium silicate-based pulp capping materials in human dental pulp cells

  • Background: This study was performed to provide a long-term bacterial seal through the formation of reparative dentin bridge, calcium silicate-based pulp capping materials have been used at sites of pulpal exposure. The aim of this study was to evaluate the mineralization-inducing potentials of calcium silicate-based pulp capping materials (ProRoot MTA [PR], Biodentine [BD], and TheraCal LC [TC]) in human dental pulp cells (HDPCs). Methods: Specimens of test materials were placed in deionized water for various incubation times to measure the pH variation and the concentration of calcium released. The morphology of HDPCs cultured on the specimens was examined using a confocal laser scanning microscope (CLSM). Alizarin red S staining and alkaline phosphatase assays were used to evaluate mineralization-inducing potentials of the capping materials. Results: BD showed the highest calcium release in all test periods, followed by PR and TC. (p<0.05). All experimental groups showed high alkalinity after 1 day, except at 14 days. BD showed the highest cell viability compared with PR and TC after 1 and 3 days, while TC showed the lowest value (p<0.05). The CLSM analysis showed that cells were well adhered and expressed actin filaments for all pulp capping materials. Mineralization by PR and BD groups was higher than that by TC group based on alizarin red S staining. BD showed significantly higher alkaline phosphatase activity than PR and TC, while TC showed the lowest value (p<0.05). Conclusion: Within the limitations of the in vitro study, BD had higher mineralization-inducing potential than PR and TC.

  • This study investigates the population model of the spread of HIV/AIDS which the infection is generated by an infectious individual in a population of susceptible. A mathematical model is presented for the transmission dynamics of HIV infection within the communities of homosexual males. The pattern on the epidemic character of HIV, the causative agent of AIDS, was analysed by the mathematical model of AIDS system which is derived according to the ecological relationship between five epidemilogic states of individuals. The computer simulation was performed using real data and the following conclusions are drawn on the basis of the simulations. 1. The model structure and the algorithm described n the thesis is good. 2. In proportion to increase Ro, the population of AIDS patient increases and the time of its widespread reaches earlier. 3. The AIDS patients will be maximum between 7 and 21 years after an attack of AIDS and widespread between 10 and 20 years. 4. Considering the properties of the incubation periods, the maximum number of infected person is increased, and the attack rate is decreased.

Effects of glycerol and seminal plasma in characteristics of preserved canine spermatozoa (개 정액의 보존시 glycerol 첨가 및 정장제거가 정자의 성상에 미치는 영향)

  • Multiple ejaculates were collected from four male mongrel dogs. The second fraction and the small volume of third fraction from the ejaculates were divided and treated as follows : control; addition of the egg-yolk Tris extender to the semen at $37^{\circ}C$. group I; Removal of seminal plasma, group II; addition of the glycerolated extender at $4^{\circ}C$, group III Removal of seminal plasma and addition of glycerolated extender at $4^{\circ}C$. The semen cooled to $4^{\circ}C$ was equlibrated for 2hrs and preserved in refrigerator at $4^{\circ}C$. The preserved semen was evaluated for kinetics, morphology, motility and thermoresistance daily for 3 days. 1. The kinectics after preserved days 2 and 3 of group I was significantly higher than that of control(p<0.05). 2. There were no significant difference in abnormal morphology of each group between the periods of storage. 3. The motility after preserved day 1 and days 3 of group I was significantly higher than that of others(p<0.05), and the molity after preserved days 2 of group I and III was signficantly higher than that of others(p<0.05). 4. When the molity of preserved semen was measured during incubation at $37^{\circ}C$, the motility of four groups was declined at similar rates. There was no effect of removal of seminal plasma and glycerol addition on thermoresistance.

Effect of Co-Culture with Mammalian Spermatozoa on In Vitro Maturation of Porcine Cumulus-Enclosed Germinal Vesicle Oocytes

  • In vitro maturation of denuded porcine immature oocytes can be enhanced by co-incubation with spermatozoa even before fertilization. This study was to determine whether the addition of spermatozoa into the culture medium could influence the nuclear maturation of porcine cumulus-enclosed germinal vesicle (GV) oocytes. Cumulus-oocyte complexes (COCs) were collected from follicles of 3- to 5-mm diameter. Porcine COCs were cultured in tissue culture medium containing spermatozoa. After 48 h culture, oocytes were examined for evidence of GV breakdown, metaphase I, anaphase-telophase I, and metaphase II. The proportion of oocytes reaching at metaphase II was significantly (P < 0.05) increased in the oocytes cultured in media containing spermatozoa compared to those in media without spermatozoa (52.3% vs 12.5%). No difference in the percentage of metaphase II was observed among the different periods of spermatozoa exposure and among the spermatozoa from different species. The proportion of oocytes reaching metaphase II was significantly different between high and low concentrations of spermatozoa. The present study suggests that manunalian spermatozoa contain a substance(s) that improves nuclear in vitro maturation of porcine cumulus-enclosed GV oocytes. Enhancing effect of spermatozoa for in vitro maturation of oocytes is a highly dose-dependent.

Daily Amperometric Monitoring of Immunoglobulin E in a Mouse Whole Blood: Model of Ovalbumin Induced Asthma

  • There is an increasing interest in monitoring of specific biomarker for determining progression of a disease or efficacy of a treatment. Conventional method for quantification of specific biomarkers as enzyme linked immunosorbent assay (ELISA) has high material costs, long incubation periods, requires large volume of samples and involves special instruments, which necessitates clinical samples to be sent to a lab. This paper reports on the development of an electrochemical biosensor to measure total immunoglobulin E (IgE), a marker of asthma disease that varies with age, gender, and disease in concentrations from 0.3-1000 ng/mL with consuming 20 µL volume of whole blood sample. The sensor provides rapid, accurate, easy, point-of-care measurement of IgE, also, sequential monitoring of total IgE with ovalbumin (OVA) induced mice is another application of sensor. Taken together, these results provide an alternative way for detection of biomarkers in whole blood with low volumes and long-term ex-vivo assessments for understanding the progression of a disease.

  • The purpose of this study was to elucidate the chemical changes and formation of sulfur minerals following reduction and subsequent oxidation of the acid sulfate soils derived from the fluvio-marine plains in Gimhae area. Changes in pH, Eh and water soluble $SO_4$, Fe, Al, K, na and Ca were determined in the soil under the reduced and oxidized conditions. These chemical properties were related to the formation of the pyrite and jarosite, the major sulfur minerals in the acid sulfate soils. On incubation, suspension pH tended to increase with decreaseing Eh in the reduction periods. Jarosite formation was favored by maintaining continuous low pH(below 4.0) and high Eh(above 400mV) during the oxidation periods, however, the conditions were not favorable for the soils with $Ca(OH)_2$. Water soluble K increased by reduction but decreased by oxidation, while the jarosite of the soil with $Ca(OH)_2$ was dissolved even under the oxidation conditon, resulting in rapid increase of water soluble K. The water soluble Ca decreased rapidly, indicating that gypsum was formed with $Ca(OH)_2$ during the oxidation periods. The formation of jarosite was favored by the oxidation condition, and hindered by the reduction condition. But the formation of pyrite was favored by reduction and hindered by oxidation. When the troll was treated with $Ca(OH)_2$, Jarosite was dissolved in both oxidized and reduced conditions.

Growth and substance transfer effect of Pleurotus cornucopiae by low concentration treatments of selenium(I) (저농도 셀레늄 처리가 노랑느타리버섯(Pleurotus cornucopiae) 생육과 물질전이에 미치는 영향(I))

  • The research for incubation period, mycelial density, day required for primordial formation after inoculation(below DPI), number of valid stipes, individual weight and accumulation amounts of organic selenium for P. cornucopiae by treating 100, 200, 300, 400, 500(${\mu}g/50g$) of $Na_2SeO_3$ is following. Incubation periods of P. cornucopiae are 20~23 days per each low concentration treatment with $Na_2SeO_3$. Compared to the control which took 22 days of incubation period, it is reduced 1 or 2 days. Mycelial density of P. cornucopiae treated with $Na_2SeO_3$ between 100 and $500{\mu}g/50g$ is very compact. DPI of P. cornucopiae treated with $Na_2SeO_3$ between 100 and $400{\mu}g/50g$ was reduced 1 or 2days, but $500{\mu}g/50g$ was increased 1 day. Number of valid stipes of P. cornucopiae treated with $Na_2SeO_3$ between 100 and $400{\mu}g/50g$ is between 19 and 20. It was increased 1 or 2, as compared to 18 of control, but $500{\mu}g/50g$ was reduced to 1. Individual weight of P. cornucopiae treated with $Na_2SeO_3$ between 100 and $400{\mu}g/50g$ was between 129 and 138g/850cc. It was increased 4.9~12.2% as compared to 123g/850cc of the control but $500{\mu}g/50g$ was 122g/50g. Accumulation amount of organic selenium for P. cornucopiae treated with $Na_2SeO_3$ between 100 and $500{\mu}g/50g$ was $2.73{\sim}8.19{\mu}g/g/dry$. It was increased 55~164 times as the concentration increased when compared to $0.05{\mu}g/g/dry$ of the control. In conclusion, incubation period, mycelial density, DPI, number of valid stipes, individual weight and accumulation amounts of organic selenium for P. cornucopiae by treating 100, ${\sim}400{\mu}g$ of $Na_2SeO_3$ was increased, but $500{\mu}g/50g$ was reduced. So more than $500{\mu}g/50g$ concentration treatments are required research.

Incubation periods of P. cornucopiae are 25~30 days per each treatment with $Na_2SeO_3$. Compared to the control which took 22 days of incubation period, it is increased 3 or 8 days for the treatment of $600{\sim}1000{\mu}g/50g$. Mycelial density of P. cornucopiae treated with $Na_2SeO_3$ between 600 and $1000{\mu}g/50g$ is very similar with control. DPI of P. cornucopiae treated with $Na_2SeO_3$ between 600 and $1000{\mu}g/50g$ was increased 3 or 8 days. Number of valid stipes of P. cornucopiae treated with $Na_2SeO_3$ between 600 and $1000{\mu}g/50g$ was between 10 and 16. It was decreased 2 or 8 as compared to 18 of control. Individual weight of P. cornucopiae treated with $Na_2SeO_3$ between 600 and $1000{\mu}g/50g$ was between 94 and 116g/850cc. It was decreased 5.7~23.5% as compared to 123g/850cc of the control. Accumulation amount of organic selenium for P. cornucopiae treated with $Na_2SeO_3$ between 600 and $1000{\mu}g/50g$ was $9.1{\sim}10.8{\mu}g/g/dry$. It was increased 182~216 times as the concentration increased when compared to $0.05{\mu}g/g/dry$ of the control. In conclusion, incubation period, mycelial density, DPI, number of valid stipes, individual weight and accumulation amounts of organic selenium for P. cornucopiae by treating $600{\sim}1000{\mu}g/g$ of $Na_2SeO_3$ was decreased. So that the optimal treatment was less $400{\mu}g/g$ than $600{\sim}1000{\mu}g/g$.
