  • Seven compounds, scopoletin (1), p-hydroxyphenethyl trans-ferulate (2), $1-({\alpha}-L-rhamnosyl(1\;{\rightarrow}\;6)-{\beta}-D-glucopyranosyloxy)-3,4,5-trimethoxybenzene$ (3), benzyl ${\alpha}-L-rhamnopyranosyl(1\;{\rightarrow}\;6)-{\beta}-D-glucopyranoside$ (4), suberic acid (5), sebacic acid (6) and scopolin (7) were isolated from the methanol extract of the roots of Althaea rosea Canival. Compounds $1\;{\sim}\;7$ were first isolated from this plant. The isolated compounds were tested for their cytotoxicity against human cancer cell lines using a SRB method in vitro.

  • The aim of this study was designed to determine the induction of rat growth hormone(rGH) by extracts of a popular herb, Glycyrrhizae radix(GR), roots of Glycyrrhiza glabra $\sub$Linne/, and Glycyrrhiza uralensis $\sub$Fischer/. In vitro study was carried out using primary rat pituitary cell culture for 3 days and then was treated with methanol extract corresponding to 1 mg of dried weight of herb per 1 $m\ell$ of culture solution.   • A new diterpene, agastanol[1] with dehydroagastol[2] was isolated from th root of Agastache rugosa, and their structures were elucidated by chemical and instrumental analysis. Agastanol[1], its derivatives, agastanone[3] and methylagastanol[5], and dehydroagastol[2] showed cytotoxic activites against in vitro human cancer cell lines. Agastanol[1] showed weak antifungal activity against Trichophyton rubrum.

  • To determine the appropriate concentrations of nutrients and growth regulators for shoot proliferation and root initiation, several rose hybrid tea cultivars were cultured. Cultured shoot tips and lateral buds from different cultivars proliferated multiple shoots on Murashige and Skoog (MS) medium supplemented with 0 to 4 mg/L BA and 0 to 0.05 mg/L NAA. The ability of the explants to proliferate shoots and initiate roots was affected by genotype, the nodal position of explant, the strength of MS basal medium and growth regulators used. The buds nearest the apex exhibited the slowest rate of development. Most cultivars had the highest shoot proliferation when cultured on MS medium with 2 mg/L BA and 0.01 mg/L NAA, but the degree varied by cultivars. Root development was enhanced by lowering the concentration of MS salts.

  • I. Objectives Intracanal medicament with antibacterial action is required to maximize the disinfection of the root canal system. The purpose of this study was to evaluate the efficacy of 2% chlorhexidine digluconate(CHX) and controlled release device(CRD) containing 20% CHX and chitosan coating, compared to calcium hydroxide as intracanal medicament against E. faecalis. II. Materials and Methods One hundred and twenty intact freshly extracted bovine incisors were used and were 0.5% NaOCl. Middle 1/3 portion of roots were sliced into 4mm thick section and cementum was removed using diamond burs and external diameter was approximally 6mm.(omitted)

Effects of Light, Temperature, and Sucrose on Plant Regeneration from the Flower Organ Explant in Iris ensata (꽃창포 화기조직 절편체 배양으로부터 식물체 분화에 미치는 광.온도.당의 영향)

  • A study was under taken to investigate the appropriate explant sources of flower organ and suitable cultural conditions such as light, temperature, and sucrose in plant regeneration of Iris ensata culture. Explants of perianth, ovary, pedicel, and peduncle of Iris ensata were cultured at different daylength (0, 8, 16, 24 hour), different temperatures (10, 15, 25, 3$0^{\circ}C$), and sucrose concentrations (1, 3, 6, 9%) on MS medium. Formation of adventitious roots from explants of Iris ensata was effective in the dark, while that of adventitous shoots was effective in the light. The optimum daylength for young plant regeneration was 16 hours. The optimum temperature for shoot formation of Iris ensata explants was $25^{\circ}C$ but the formation at 10 and 15$^{\circ}C$ was ineffective. Especi-ally, perianth and ovary was effective in shoot formation from flower organ expants. T-he optimum concentration of sucrose for shoots and roots formation of Iris ensata explants was 3 and 6%, respectively.

  • Rauvolfia serpentina (L.) Benth. ex Kurz is a medicinal plant and an endangered tropical rainforest plant species. Since the field cultivation that aims to fulfill the industrial needs is never accomplished, tissue culture appears to be the most feasible way to improve the quality and quantity of R. serpentina. This experiment used two kinds of explants (roots and shoots) to induce optimal root formation in different combinations of auxin and photoperiod. Each explants exhibited different responses on given treatments. Differentiated root could be produced from explants cultured in IBA 20 mg/L with and without light. The highest number of roots, root length and root weight induced from shoot explants were effective on MS medium containing IBA 20 mg/L and incubated under dark condition, while highest total weight (callus and root) from root explants cultured on MS medium supplemented 10 mg/L IBA and 10 mg/L NAA and incubated under day length (11/13 hr). The root induced from shoot explants produced the highest major alkaloid content. The highest content of ajmaline (2.17 ppm fresh weight) and reserpine (1.30 ppm fresh weight) were observed in shoot explants cultured in MS medium containing combination of IBA 10 mg/L and NAA 10 mg/L and incubated under dark condition, yohimbine (1.47 ppm fresh weight) was in the shoot explants cultured in MS medium containing NAA 20 mg/L and incubated under day length, while serpentine was absent.

AN IN VITRO STUDY OF THE THERAPEUTIC EFFECT OF ARGON LASER IRRADIATION ON HYPERSENSITIVE ROOTS (과민성치근에 대한 Argon laser조사의 치료효과에 관한 실험적 연구)

  • In order to evaluate the therapeutic effect of argon laser irradiation on hypersensitive root, 60 mandibular premolars that had been extracted due to periodontal disease were treated by thorough scaling, root planing and root conditioning with tetracycline HCI(100mg/ml, 5 min.) for induction of hypersensitive root. Within middle one third of root mesial surface(30) or distal surface(30) was randomly irradiated by 0.5W-, 0.75W- and 1.0W-power for 2 minutes argon laser(HGM Inc., USA) with or without topical application of 1.23% acidulated phosphate fluoride gel(Pascal, USA). After all teeth were maintained in 2% methylene blue solution for 48 hours and cross-sectioned through irradiated surface of dried roots, methylene blue stained area was compared between irradiated side and nonirradiated side. Following results were obtained : 1. Methylene blue stained area was significantly less in the laser irradiated group than control group(p<0.05). 2. There was no significant difference in methylene blue stained area between 0.5W-power and 0.75W-power group(p>0.1) and between 0.75W-power and 1.0W-power group(p>0.1), but there was significant difference between 0.5W-power and 1.0W-power group(p<0.05). 3. There was no significant difference in methylene blue stained area between two groups with and without topical application of 0.12% acidulated phosphate fluoride gel(p>0.1). The results suggest that argon laser irradiation on hypersensitive root may reduce the hypersensitivity by obstruction of dentinal tubules.

Evaluation of Cytotoxicity, Carbohydrate, and Lipid Inhibitory Activity of Codonopsis lanceolata using Different Solvent Fractions

  • This study was conducted to evaluate the cytotoxicity and ${\alpha}-Amylase$, ${\alpha}-Glucosidase$, pancreatic lipase inhibition in vitro by different solvent fractions from the roots of Codonopsis lanceolata. The values of $IC_{50}$ against Calu-6 cell showed a high effect in n-hexane fraction ($10.13{\mu}g/mL$) whereas DW fraction exhibited the weakest inhibition on cell viability, having an $IC_{50}$ value of over $1,000{\mu}g/mL$. The values of $IC_{50}$ against HCT-116 cell showed the highest activity in n-BuOH fraction ($102.01{\mu}g/mL$), followed by n-hexane fraction ($145.85{\mu}g/mL$), methylene chloride fraction ($332.02{\mu}g/mL$), ethyl acetate fraction ($462.93{\mu}g/mL$) and DW fracion ($>1,000{\mu}g/mL$). ${\alpha}-Amylase$ inhibitory activity in methylene chloride fraction and ethyl acetate fraction was found to have a higher inhibitory effect with 24.5% and 25.6% than the other fractions. The highest ${\alpha}-Glucosidase$ inhibitory activity was observed from the ethyl acetate fraction extract, while the extract of DW fraction showed the lowest level of inhibitory activity at given experiment concentration. The pancreatic lipase inhibitory activity of C. lanceolata was found to have a higher the effect in ethyl acetate fraction. Inhibition of lipase activity of the ethyl acetate fraction and n-hexane fraction showed a relatively high, while the extract of DW fraction showed the lowest level at given experiment concentration. These results suggested that the roots of C. lanceolata may assist in the potential biological activity on carbohydrate, lipid Inhibitory activity and anticancer activity.

Transformation of Populus alba × P. glandulosa by Agrobacterium rhizogenes (Agrobacterium rhizogenes 에 의한 현사시나무의 형질전환(形質轉換))

  • The widely cultivated hybrid poplar Populus alba ${\times}$ P. glandulosa in Korea was transformed with Agrobacterium rhizogenes agropine type strain A4. Genetic transformation was confirmed by the presence of agropine. Alteration in growth rate of hairy roots was seen following changes in the dilution rate of medium and concentration of sucrose, suggesting that improved growth might be achieved by more precise manipulation of the nutrient medium. Plant regeneration occurred from transformed hairy roots on MS medium supplemented with 0.5 mg/l BAP. Transformed plantlets grown in vitro exhibited a more developed root system characterized by fast growth behavior in comparison to normal plantlets. This work demonstrates that the root transformation would be useful in improving plantlet establishment and growth through the effective root system.
