In Vitro Rearing of parasitoids of Insect Pests in China

  • Since 1975, the studies on in vitro rearing Trichogramma spp., Anastatus japonicus Ashmead, Telenomus dendrolimusi Chu, Dibrachys cavus Walker, Habrobracon hebetor(Say), Bracon greeni Ashmead have been conducted successfully in China. The simulated “host-eggs" are made of polyethylene or polypropylene semispherical capsules, containing artificial diets, in which insectan pupal hemolymph is the main component. Manufacture of simulated “host larvae" are made of small rectangular parafilm of cotton-paper bags, containing artificial diets with insectan pupal hemolymph as the main component. Mass production of in vitro reared Trichogramma spp. and Anastatus japonicus and its utilization in the fields showed good effectiveness in controlling cotton bollworm, pine caterpillars, sugarcane borers and litchi stink bug.

Serichlor, A New Disinfectant in Indian Sericulture

  • Silkworm diseases are better prevented than cured. Disinfection and hygiene are the two important aspects in silkworm rearing to prevent the diseases. Suitable disinfectant is the primary need to disinfect the rearing house, its surroundings and appliances to eliminate the persistent pathogens from the rearing environment. In this direction, Serichlor, a new disinfectant in Indian Sericulture marketed as Serichlor-60 (contains 60,000 ppm of chlorine dioxide) and Serichlor-20 (contains 20,000 ppm of chlorine dioxide) has been evaluated for its germicidal effect against the pathogens of silkworm, viz., spores of Nosema bombycis, Bacillus thuringiensis, polyhedra of BmNPV and conidia of Beauveria bassiana both in vitro and in vivo. Results indicated that high concentration (2,500 ppm of chlorine dioxide) is required to kill all the pathogens at 100% level. The efficacy of the Serichlor was greatly enhanced by the addition of 0.5% slaked lime solution. 500 ppm of chlorine dioxide in 0.5% slaked lime solution was found effective against all the pathogens tested. This concentration of disinfectant was also found effective for disinfection of rearing house, rearing appliances and silkworm egg surface. The disinfectant is stable, non hazardous, least corrosive and most suitable for Indian Sericulture.

Biological Characteristics of Tetrastichus sp. reared on Artificial Host (인공숙주에서 증식된 Tetrastichus sp.의 생물학적 특성)

  • A gregarious pupal endoparasitoid Tetrastichsus sp. (Hymenoptera: Eulophidae) was reared in vitro from oviposition to adult emergence on an artificial host. The host pupal case was made using 0.02 mm-thick polypropylene film, and was filled with a diet consisting of powders of Antheraea pernyi pupa, chicken yolk, infant formula, royal jelly, and Neisenheimer's salt solution. Female parasitoids reared in the artificial host produced smaller sized progeny than those reared in in vivo, but the adults reproduced fertile offsprings. Furthermore in vitro second-generation (G$_2$) females showed more improved biological characteristics, compared with their parents. The fecundity (mean no. adult progeny), oviposition period (days), and longevity (days) of G$_2$ female were evaluated as 45.7, 7.8, and 13.8, respectively Female biased sex ratio was obtained with 76.9% female progeny. The results demonstrated that Tetrastichus sp. is a promising parasitoid for in vitro mass production.

Studies on the Applicability of Furazolidone to the Silkworm Rearing Industry as a Useful Remedy for Certain Silkworm Diseases (Report I) (푸라졸리돈의 누에병치료약으로서의 응용가능성에 관한 연구 (제1보))

  • The author studied the applicability of Furazolidone to the silkworm rearing industry as a useful remedy for certain silkworm diseases, at the silkworm rearing house of the college of agriculture, Seoul national university, during both the spring and the autumn silkworm rearing season of 1972. Discovering the fact that Furazolidone, when put on the mulberry leaves in a powdered form, is eaten along with the leaves by silkworms and thus the systematic administration of Furazolidone to silkworms is possible, the experimenter carried on a series of experiments (1. determining the in vitro antibacterial activity of Furazolidone to four pathogens of silkworm diseases-Bacillus thuringiensis, Aspergillus oryzae, Aspergillus flavus, and Isaria farinosa, 2. observing the prophylactic and therapeutic effect of Furazolidone against the experimental flacherie caused by inoculation of B. thuringiensis, and 3. examining the toxicity of Furazolidone to silkworm larvae). As the results of the experiments the investigator found out the fundamental fact that Furazolidone exerts a good prophylactic and therapeutic effect against flacherie which is the most common and important silkworm disease: Furazolidone, in in vitro test, inhibited completely the growth of B. thuringiensis, the pathogen of bacterial flacherie, at the concentration of 1 ${\mu}$g/mι. with the tube method and at the concentration of 5 ${\mu}$g/mι. with the plate method, and the drug showed an excellent prophylactic effect and a considerably good therapeutic effect, depending on the time of administration, on the 5th instar silkworms inoculated B. thuringiensis, at the tentative dose of 150mg. per 10 silkworms administered once a day for 2 days. For the practical administration of Furazolidone against flacherie, the dose, the time and duration of administration, and the form of preparation, will be investigated more closely.

Experience of Mothers with Babies by in Vitro Fertilization (시험관 아기를 둔 어머니의 경험)

  • A phenomenological study was conducted to investigate the experiences of mothers pregnant via in vitro fertilization (IVF). Sample of nine mothers participated. Ten theme clusters emerged when the formulated meanings were organized into categories. During the infertility period, the participants were subject to self- depreciation, envy, anxiety, and depression. It changed their priorities in life from a job-oriented life to one where having a baby was the most important thing. After trying numerous alternative therapies, IVF became their last hope in having a baby. Since the success rate for IVF is low (only 20-30%), the participants for the treatment were overwhelmed with uncertainty, and it led to further anxiety, depression and despair. Success of pregnancy gave them extreme satisfaction, but they became very cautious in their day-to-day life because of their fear of abortion and early delivery. Some were even worried about the side effect of IVF during the pregnancy. Finally, the delivery of the baby gave them relief from the obligation of having a baby. Women did not have any difficulties in rearing a "test tube baby" except in the case of twins. Most women had no ethical difficulty in having a baby by IVF. However they did not wish this information to be revealed to other people. They again turned to IVF to have a son(s) when the resultory child(s) was a daughter(s). This is because of the strong preference for sons in Korean society.

An Optimal Standardized in vitro Bioassay to Evaluate Susceptibility of Green Peach Aphid, Myzus persicae (Sulzer)(Insecta: Hemoptera: Aphididae), to Aphicides (복숭아혹진딧물, Myzus persicae (Sulzer)(Insecta: Hemoptera: Aphididae), 살진딧물 최적 in vitro 살충력 검정 방법 확립)

  • Leaf-spray in vitro bioassays appraise new aphicidal formulations for managing deleterious plant-feeding aphids. The formulation may utilize alternative and integrated strategies. However, leaf spraying even under controlled conditions may affect aphid reproduction and mortality. This study examines leaf spray applications for optimum and reproducible aphicidal results using tobacco leaves overlaid on cotton fabric or water agar surfaces. Infestation of the undersides of tobacco leaves with nymphs of green peach aphids was used in the assays. Spray distance and volume were optimized using water-sensitive paper to ascertain the best surface coverage. Overlays of the leaves on water agar caused less mortality and greater reproduction than the use of cotton fabric. The relative humidity of the insect-rearing chambers changed with the watering regime for the insect - rearing chambers with cotton fabric; 60% relative humidity was optimal. Relative humidity was not affected by the concentration of agar in the water agar chambers. Applications of the chemical aphicidal standard, Sulfoxaflor, under the optimized conditions exhibited similar times for lethality although the rate was faster with leaves on the cotton fabric than on water agar. These studies establish reproducible and sensitive techniques for assessing the lethality and effects on reproduction of potential aphicidal products.

In vivo and In vitro Digestibility of Enzyme-treated Fish Meal for Juvenile Atlantic Bluefin Tuna Thunnus thynnus (효소처리어분에 대한 치어기 대서양 참다랑어(Thunnus thynnus)의 in vivo 및 in vitro 소화율 평가)

  • This study was conducted to investigate in vivo and in vitro digestibility in juvenile Atlantic Bluefin tuna Thunnus thynnus. In vivo digestibility was compared between four experimental diets to determine the optimum dietary inclusion level of an enzyme-treated sardine fish meal (EFM) and sardine fish meal (FM). The experimental diets were as follows; EFM75 (75% EFM), EFM60 (60% EFM and 15% FM), FM75 (75% FM) and SL (frozen sand lance) as a raw fish feed. Feces of Bluefin tuna (90.3 g) were collected both by siphoning from a 700 L cage and by dissection in 69 ton concrete rearing tanks. For the siphoning method, protein digestibility was higher in the tuna fed SL diet than that of other groups. The lowest protein digestibility was observed in FM75. For the dissection method, protein digestibility was higher in tuna fed EFM75 diet than that of other groups. The lowest protein digestibility was observed in the EFM60 group. In vitro digestibility was compared in six protein sources to find an alternative source of EFM for the tuna feed. The highest in vitro digestibility was observed in EFM (92%) followed by low temperature FM (72%), meat meal (65%), feather meal (60%), sardine fish meal (57%) and poultry by-product meal (55%).

Effect of Certain Insecticides in Controlling Pesudodendrothrips mori in Mulberry

  • Mulberry plantations are prone to several kinds of insect pests including the thrips, Pesudodendrothrips mori. The thrips infestation affects the qualitative and quantitative characters of mulberry leaf, which in turn affects the silkworm cocoon crop. In the present study, four commercial insecticides viz., Rogor, Quinalphos, Confidor and Methyl parathion were tested for their efficacy in the control of P. mori thrips. These insecticides were observed to kill 68-80% within 24 hrs in vivo and 100% after 12 hrs post-treatment in vitro. Confidor (0.05%) and Quinalphos (0.2%) were most effective in controlling the thrips incidence. The insecticide-sprayed mulberry leaves did not show any adverse effect on the rearing of silkworms.

The Effects of Aqueous Fibrolytic Enzyme Products Supplementation on In vitro Fermentation and Growth Performance of Hanwoo Steers (섬유소분해효소 발효액의 첨가가 In vitro 발효 및 거세한우의 성장에 미치는 영향)

  • The objective of this study was to investigate the effects of an enriched diet supplemented with aqueous fibrolytic enzyme products (FEP) on In vitro fermentation and growth performance of Hanwoo Steers. Total mixed ration combined with fibrolytic enzyme products showed a slight increase in the pH level on In vitro fermentation. Acetate ratio in volatile fatty acids, and the concentration of total volatile fatty acids were significantly greater at 12hr and 24hr after cultivation (p<0.05). Dry matter digestibility was significantly increased in groups supplied with aqueous fibrolytic enzyme products at 24hr after cultivation (p<0.05). In a rearing trial, the average daily gain and feed efficiency improved in the treatment groups supplied with aqueous fibrolytic enzyme products. Nutrient digestibility such as DM, crude fiber, NDF, and ADF digestibility were also significantly increased with the addition of aqueous fibrolytic enzyme products (p<0.05). Therefore, the aqueous fibrolytic enzyme products supplementation on In vitro fermentation is effective in improving the condition of rumainal fermantation. Also two kinds of aqueous fibrolytic enzyme products were found to be effective in improving the average daily gain and feed efficiency in Hanwoo Steers.

Characteristics of Sawdust and Cocopeat Beddings, and Their Usefulness According to the Fan and Pen Location for Rearing Hanwoo Cattle

  • This study was designed to examine the characteristics of sawdust and cocopeat bedding materials, including physicochemical properties (Exp. I) and on-farm trial (Exp. II). In Exp. I, the proportion of particle size was in the order of sawdust>cocopeat India>cocopeat Vietnam (p<0.05), and cocopeat contained higher proportion of small particles ($250{\mu}m$+below $250{\mu}m$) than sawdust, causing a dust production problem. Bulk density was cocopeat India>cocopeat Vietnam>sawdust (p<0.05), thus cocopeat treatments showed 4.4 times higher bedding cost than sawdust. The water absorption rates were 702.0% in cocopeat India, 678.3% in cocopeat Vietnam, and 444.0% in sawdust, showing cocopeat had approximately 1.5 times higher water absorption rate than sawdust. Moisture evaporation rates after 12 h of air blowing (2.00 m/s) were higher (p<0.05) in cocopeat Vietnam (80.4%) than sawdust (71.2%) and cocopeat India (72.8%). In vitro ammonia emissions were higher (p<0.05) in sawdust ($2.71mg/m^2/h$) than cocopeat India ($1.59mg/m^2/h$) and Vietnam ($1.22mg/m^2/h$), and total ammonia emissions were higher (p<0.05) in sawdust ($37.02mg/m^2$) than cocopeat India ($22.51mg/m^2$) and Vietnam ($13.60mg/m^2$). In Exp. II, an on-farm trial was conducted with 48 Hanwoo cattle in 16 pens using the same bedding materials as in Exp. I, with fan (blowing 2.00 m/s) and no fan treatments, and feed bunk side (FB) and water supply side (WS) within a pen (4.5 m, $width{\times}9.0m$, length). Beddings were replaced with fresh bedding materials when moisture concentrations were over 65%. No interactions among treatments were detected for moisture concentration and increment rates, and ammonia concentrations, but a significant effect was observed (p<0.01) for each of the treatments. Both concentrations and increment rate of moisture were higher (p<0.01) in the beddings without fan than with fan. Moisture concentrations and increment rate within a pen were also higher (p<0.01) in FB than WS. Thus, the whole no-fan-FB and sawdust-fan-FB were replaced with fresh bedding material between 4 to 5 experimental weeks. The ammonia concentrations and pH of beddings were not significantly different among treatments. Therefore, using cocopeat bedding with a blowing fan can extend twice the bedding utilization period, and WS within a pen showed twice the bedding-life compared to FB. Despite the outstanding characteristics of cocopeat compared with sawdust, using cocopeat as an alternative for sawdust bedding is not recommended for cattle management, considering it has 4.4 times higher bedding cost and a dust production problem.
