Immunostimulating and Anticancer Activities of Hot-water Extracts from Acanthopanax senticosus and Glycyrrhiza uralensis

  • When 10 kinds of herbal medicines were fractionated into hexane, MeOH, cold-water, and hot-water extracts, hot-water extracts from Acanthopanax senticosus (AS), Glycyrrhiza uralensis (GU), Cichorium intybus (CI), and Polygonatum odoratum (PO) showed the potent intestinal immune system modulating activity (1.72-, 1.62-, 1.60-, and 1.53-fold of control at $100{\mu}g/mL$, respectively). Especially, hot-water extracts from AS (215% compared with the control) and GU (187%) also had macrophages stimulating activity and mitogenic activity of splenocytes (7.1- and 6.5-fold) at $100{\mu}g/mL$. In addition, the effects of hot-water extracts from herbal medicines on anticancer activities were studied in mice. Hot-water extracts from AS and GU enhanced cytotoxicity of natural killer cell against cancer cell, Yac-1 (37 and 34% cytotoxicity) at E/T ratio 100:1, and colon 26-M3.1 cancer cell lines had significantly inhibited (82.1 and 75.2%) in experimental lung metastasis. These results suggest that hot-water extracts from A. senticosus and G. uralensis can be used as biological response modifiers to stimulate immune system and inhibit tumor.

고추장 분리 균주의 혈전용해능, 면역활성능 및 세포독성 효과 조사 (Fibrinolytic, Immunostimulating, and Cytotoxic Activities of Microbial Strains Isolated from Kochujang)

  • 본 연구는 고추장에서 분리한 미생물들을 이용하여 혈전용해능, 면역세포활성능 및 세포독성과 같은 건강기능활성을 조사하고, 건강기능활성 미생물을 고추장에 응용하고자 수행되었다. 전통식 및 개량식 고추장으로부터 분리된 294균주 중 단백질분해력이 우수한 91균주를 선별하였고, 선별된 91개 균주의 혈전용해능을 측정하여 최종적으로 혈전용해능 활성이 높은 3 균주(TPP 0014, TPP 6013, TPP 6015)를 선별하였다. 이들 최종 선별균주들의 면역세포활성화능력 측정 결과 TPP 6015 균주에 의한 거식세포에 의한 NO 생성량은 3.67 ${\mu}g/mL$이었고, $TNF-{\alpha}$ 생성량은 626.3 pg/mL 그리고 $IL-{\alpha}$ 생성량은 645.9 pg/mL로 측정되어 세 균 주 중 비교적 높은 면역세포활성이 있음을 확인할 수 있었다. 또한 대장암 세포주인 SNU-C4와 정상세포주인 CHO-K1에 대한 선별된 균주의 세포독성을 MTT assay에 의해 조사한 결과 SNU-C4 세포주에 65-78%, 그리고 CHO-K1 세포주에 60-70%의 세포독성을 나타내는 것을 확인하였다. 고추장 시료는 혈전용해 활성을 나타내지 않았으나, 이들 선발 균주들을 고추장에 첨가하였을 때, 높은 혈전용해활성을 나타내는 것으로 밝혀졌다. 이들 선별 균주들을 API kit를 이용하여 동정한 결과 TPP 0014와 TPP 6015 균주는 Bacillus stearothermophilus로 동정이 되었고, TPP 6013 균주는 B. amyloliquefacience로 동정이 되었다.

  • Lentinus edodes is a well-known edible and medicinal mushroom used in Oriental cultures. Recently, L. edodes has attracted a lot of attention owing to its antifungal activity, antibacterial activity, antiviral activity, hepatoprotective effect, antitumor activities, and immunomodulatory and cytotoxic effects. In this study, the water-soluble crude polysaccharides, CPF and CPB, which were obtained from the fruiting body and culture cell-free broth of L. edodes by hot-water extraction and ethanol precipitation, were fractionated by DEAE cellulose and Sepharose CL-6B column chromatography, resulting in six polysaccharide fractions, CPFN-G-I, CPFN-G-II, CPFN-G-III, CPFA-G, CPBN-G, and CPBA-G Among these fractions, CPFN-G-I, CPBN-G, and CPBA-G were shown to stimulate the functional activation of macrophages including NO production, cytokine expression, and phagocytosis.

구름버섯의 항암성 다당류분획(Copolang)이 마우스의 면역기능에 미치는 영향에 관한 연구 (Antitumor Activity of the Polysaccharide-Fraction(Copolang) from Coriolus versicolor and its Effects on the Immune Function)

  • Polysaccharide fraction isolated from Coriolus versicolor (Copolang) was studied on the antitumor activity and immunostimulation activities with reference to PS-K. Copolang showed nearly equal antitumor activities to the PS-K and exhibited marked augmentation effects on the antibody mediated hypersensitivity reaction, delayed type hypersensitivity reaction and NK-cell activity in tumor bearing mice. But it did not show any noticeable effect on the antibody secreting cell and macrophage function in normal mice. These results indicate that the antitumor activity and immunostimulating effect of Copolang are comparable to those of PS-K.

신령버섯(Agaricus blazei)으로부터 면역증강활성 다당류의 분리 및 화학적 특성 (Chemical Features and Purification of Immunostimulating Polysaccharides from the Fruit Bodies of Agaricus blazei)

  • 인공 재배된 신령버섯(Agaricus blazei)의 자실체를 열풍 건조한 다음 다당류를 추출, 분리 정제하여 항암면역활성을 측정하고 그들의 물리화학적 특성의 일부를 조사하였다. 면역증강 다당류는 0.9% sodium chloride와 열수를 사용하여 순차적으로 추출하고 에탄을 침전, 투석 및 동결건조로 얻었으며, 이들 다당류는 이온 교환수지와 겔을 사용하여 균질한 다당류로 정제하였다. 정제된 다당류의 임파구의 항체 형성능 촉진 효과를 in vitro에서 조사한 결과, 0.9% sodium chloride 추출 분획에서 AG-2가, 열수 추출 분획에서 AG-6, -7 및 -8에서 비교적 높은 활성을 보였으며, 특히 AG-3이 양성 대조구(LPS)보다 높은 활성을 보였다. 활성이 비교적 높은 다당류의 경우 주로 glucose로 구성되어 있으며 단백질과 결합된 단백-다당체이었다. 정제된 다당류는 AG-4를 제외하고 모두 glucose를 비롯한 $2{\sim}6$종의 단당류를 갖고 있는 heteropolysaccharide이었으며, AG-1의 경우 galactose, mannose 및 fucose가 79.1:8.9:12.0의 몰비로, AG-5의 경우 glucose, mannose 및 xylose가 45.6:17.9:25.2의 몰비로 존재하였으며, 이들 두 분획은 약한 면역증강활성을 보였다.

한국인으로부터 분리한 Pediococcus pentosaceus EROM101의 면역증강 및 항암활성 (Immunostimulating and Anti-cancer Effects of Pediococcus pentosaceus EROM101 Isolated from Korea.)

  • 사람 분변으로부터 대식세포 활성 증강효능이 있는 유산균을 선별하여 이를 동정한 결과 P. pentosaceus 동정되었으며, 이 균주를 P. pentosaceus EROM101로 명명하였다. P. pentosaceus EROM101의 면역증강 및 항암활성을 알아보기 위하여 실험동물에게 3주간 생균 및 파쇄액을 경구 투여한 뒤 대식세포와 비장세포의 활성화를 측정하였으며, 동시에 소장점막상피세포에서 생산되는 IgA의 함량을 측정하였다. P. pentosaceus EROM101의 경구 투여는 대식세포 및 비장세포의 분열을 촉진하여 면역증강 기능이 있는 것으로 나타났다. 특히, 생균보다 파쇄액에서 활성이 더 뛰어난 것으로 나타나, P. pentosaceus EROM101은 장내에서 생균으로 서식하여 지속적으로 면역증강 기능을 나타내는 것이 아닌 세포 구성물에서 면역증강활성이 있는 것으로 사료되었다. 또한, 소장점막상피세포에서 IgA의 분비량에서도 대조군보다 약 4배이상의 IgA 생산량을 증가시켜 장관면역 활성화 기능이 있는 것으로 판단되었다. 또한 P. pentosaceus EROM101 파쇄액이 Sarcoma 180 복수암세포를 이식해 준 실험동물 내에서 복수암성장을 억제하였으나, Sarcoma 180 세포열에 대한 세포독성을 나타내지 않아 면역증강에 의해서 항암 활성이 나타난 것으로 추측되었다. 이상의 연구결과에서 P. pentosaceus EROM101은 면역증강 능력이 있으며, 동시에 항암 활성을 기대할 수 있을 것으로 사료된다.

Immunostimulating Effect of a Well-known Thai Folkloric Remedy in Breast Cancer Patients

  • The study aimed to evaluate immune-stimulating effects of a well-known Thai folkloric remedy when used for adjuvant therapy with conventional chemotherapeutics for treatment of breast cancer. Immunostimulating influence of the remedy (215 mg/kg body weight per day) on NK cell activity and TNF-${\alpha}$ release from the monocytes/macrophages were investigated in a total of 15 healthy women and 13 female patients with breast cancer (Group 1). The effect of breast tumor surgery on NK cell activity was further investigated in 18 female patients with breast cancer (Group 2). NK cell cytotoxic activity was determined by chromium release cytotoxic assay using K562, an erythroleukemic cell line. TNF-${\alpha}$ release from monocytes/macrophages separated from blood samples was determined through a biological assay using actinomycin D-treated L929 mouse fibroblast cells in the presence and absence of LPS. Baseline NK cell activity of the monocytes/macrophages separated from Group 2 patients expressed as %cytotoxicity was significantly lower than in the healthy subjects at E:T ratios of 100:1 and 25:1. In healthy subjects, there was no change in NK cell cytotoxic activity (%cytotoxicity or LU) following 1 and 2 weeks of treatment with the remedy compared with the baseline at various E:T ratios but the binding activity (%binding) was significantly increased after 2 weeks of treatment. The addition of one or two conventional chemotherapeutic regimens did not significantly reduce the NK cytotoxic activity but did affect release of TNF-${\alpha}$ in both unstimulated and LPS-stimulated samples. Surgery produced a significant suppressive effect on NK cell activity. The use of the remedy as an adjunct therapy may improve therapeutic efficacy and safety profiles of conventional chemotherapeutic regimens through stimulation of the immune system in cancer patients.

  • A potent immuno-stimulating activity was detected from the watersoluble and ethanol-precipitated crude extract (AG-1) of the root of Angelica gigas Nakai. The crude extract was fractionated into two fractions, an acidic AG-2 and a neutral AG-3 fraction by DEAE-cellulose adsorption. The two fractions contained polysaccharides of which M.W. were 10 Kdal and >2,000 Kdal respectively, proteins, and various inorganic components. The immunostimulating activities of two fractions were not reduced by proteinase K, acid or alkali treatment. The polysaccharides obtained from the root of A. gigas were mainly composed of arabinose, galactose, and galacturonic acid. These results indicated that immuno- stimulating components of the root of A. gigas was a kind of pectic polysaccharides or arabinogalactans.

Immunostimulating activity of maysin isolated from corn silk in murine RAW 264.7 macrophages

  • Corn silk (CS) has long been consumed as a traditional herb in Korea. Maysin is a major flavonoid of CS. The effects of maysin on macrophage activation were evaluated, using the murine macrophage RAW 264.7 cells. Maysin was isolated from CS by methanol extraction, and preparative $C_{18}$ reverse phase column chromatography. Maysin was nontoxic up to $100{\mu}g/ml$, and dose-dependently increased TNF-${\alpha}$ secretion and iNOS production by 11.2- and 4.2-fold, respectively, compared to untreated control. The activation and subsequent nuclear translocation of NF-${\kappa}B$ was substantially enhanced upon treatment with maysin ($1-100{\mu}g/ml$). Maysin also stimulated the phosphorylation of Akt and MAPKs (ERK, JNK). These results indicated that maysin activates macrophages to secrete TNF-${\alpha}$ and induce iNOS expression, via the activation of the Akt, NF-${\kappa}B$ and MAPKs signaling pathways. These results suggest for the first time that maysin can be a new immunomodulator, enhancing the early innate immunity.
